  • Effects of 4-nonylphenol (4-NP) on mixed function oxygenase (MFO) in the kidneies of rockfish (Sebastes schlegeli) were investigated. The cytochrome P45O (CYP) contents and NADPH cytochrome P450 reductase, NADH cytochrome b5 reductase and 7-ethoxyredorufin-O-deethylase (EROD) activities of microsome, glutathione S-transferase (GST) activity of cytosol in rockfish exposed to 4-NP for 7 days using an intraperitoneal injection (25 mg/kg) were quantitatively determined. The GST activity of rockfish exposed to 4-NP were higher, up to 5.2 times higher, than those in the control fish. The activities of NADPH-and NADH-dependent reductases were inhibited. On the other hand, CYP contents and EROD activity of the 4-NP exposed fish demonstrated neither an increasing or decreasing trend.

  • This study was designed to verify the shelf-life of a freeze dried-block type of convenience food for rockfish Sebastes schlegeli seaweed soup product stored at different storing temperatures (25, 35, and 45℃) for 5 months. The polyunsaturated:saturated fatty acid ratio of the product stored at 25℃ was higher than that of products stored at 35℃ and 45℃ for 5 months. The colorimetric assessment indicated a noticeable decrease in the brightness of product color after 5months of storage at 35℃ and 45℃. Increased storage temperature and time negatively affected the product color. The products stored at 35℃ and/or 45℃ for more than 3 months tended to be more yellowish-red in color than those stored at 25℃ for shorter periods. No disease-causing microorganisms, including Escherichia coli and Staphylococcus aureus, posing health hazards to the human, were detected on food safety evaluation, regardless of storage conditions. Based on food visual shelf life simulator the shelf life of the rockfish seaweed soup was estimated approximately 22 months, considering the data from yellowness the safety factor of 0.7.

A Review of the Optimum Feeding Rates and Feeding Frequency in Korean Rockfish Sebastes schlegeli Reared at Seven Different Water Temperatures

  • Seven feeding trials were conducted to determine the effects of feeding rate and frequency in the Korean rockfish Sebastes schlegeli at seven different water temperatures. Two feeding-rate experiments for 5-g Korean rockfish at $17^{\circ}C$ and $20^{\circ}C$; three feeding rate experiments for 16-g Korean rockfish at $16^{\circ}C$ and $24^{\circ}C$; and finally, two feeding frequency experiments for 93- and 100-g at $15^{\circ}C$ and $20^{\circ}C$ were conducted. Twenty fish averaging $5.5{\pm}0.2$ g ($mean{\pm}SD$) were randomly distributed into 18 indoor tanks containing 40-L seawater from a semi-recirculation system. Fish were fed a commercial diet for 4 weeks at water temperatures of $17^{\circ}C$ and $20^{\circ}C$. Two feeding trials were conducted to determine the optimum feeding frequency in growing Korean rockfish reared at temperatures of $15^{\circ}C$ and $19^{\circ}C$. Broken line regression analysis of weight gain (WG) indicated that the optimum feeding rates of 5-g growing Korean rockfish were 4.48% (BW/day; BW, body weight) at $17^{\circ}C$ and 4.83% (BW/day) at $20^{\circ}C$. Broken line regression analysis of WG showed optimum feeding rates of 16-g juvenile Korean rockfish of 3.41% (BW/day) at $16^{\circ}C$, 3.75% (BW/day) at $20^{\circ}C$, and 3.34% (BW/day) at $24^{\circ}C$. The biological performance, results, along with morphological indices and serological characteristics, suggest that a feeding frequency of 1 meal/day was optimal to improve WG in growing Korean rockfish grown from 93 to 133 g at a water temperature of $15^{\circ}C$, and 100 to 132 g at $19^{\circ}C$.

Molecular Parameters for Assessing Marine Biotoxicity: Gene Expressions of Rockfish (Sebastes schlegeli) Exposed to Polycyclic Aromatic Hydrocarbons

  • Environmental and anthropogenic changes affect the health and stability of marine ecosystem. In this study we aimed to identify molecular biomarkers for ecotoxicological pollutants risk assessment in the rockfish (Sebastes schlegeli). We designed primers based on conserved sequences by multiple alignments of target genes from related species, and cloned the partial cDNAs of cytochrome P450 (CYP1A1), glutathione S-transferase (GST), metallothionein (MT), superoxide dismutase (SOD), ubiquitin (UB), vitellogenin (VTG) and $\beta$-actin by reverse transcription polymerase chain reaction (RT-PCR) from S. schlegeli. Northern blot results indicated that these six genes expressions were significantly induced by benzo[a]pyrene (BaP, 1 ${\mu}M$) and that the level of each of their transcripts increased in BaP-exposed rockfish in a time-dependent manner. This study suggests that transcriptional changes in these six genes may be used for monitoring environmental exposure to polycyclic aromatic hydrocarbons (PAHs).

Fluctuation of Temperature Induces Pathogenicity of Streptococcus iniae and Changes of Immunology Related Genes of Korean Rockfish, Sebastes schlegeli

  • This study was designed to examine the immune response in Korean rockfish during water temperature fluctuation and to elucidate the factors contributing to streptococcal pathogenesis in cultured Korean rockfish, S. schlegeli. We investigated cumulative mortality against Streptococcus iniae (FP5228 strain) infection in the exposed Korean rockfish (39.7±5.8 g) to environmentally relevant temperature (Control, 23℃; High temperature, 28℃ and 23℃ and 28℃ with 12 hours interval exchange, 23↔28℃) for 48 hours. Also, the expression of the mRNA related to the immune response genes (heat shock protein 70, interleukin1β, lysozyme g-type and thioredoxin-like 1) were measured in spleen and head kidney by real-time PCR analysis in the exposed fish to thermal stress. In this study, the combined stress with bacterial challenge in fishes exposed to thermal stress lowered the survival rate than that of control (23℃). The cumulative mortality in the group of control, 28℃ and 23↔28℃ was 24%, 24% and 40% (P<0.05), respectively. Also, thermal stress modulated the mRNA level of immune related genes; heat shock protein 70, interleukin-1β, lysozyme g-type and thioredoxin-like 1 in Korean rockfish. The present study indicates that a high and sudden water temperature change affect immune responses and reduce the disease resistance in Korean rockfish.

  • This study examined the effects of the dietary inclusion of Porphyra and sea tangle Laminaria japonica on the growth, feed utilization, body composition, and plasma chemistry of juvenile Korean rockfish Sebastes schlegeli. Eight hundred and forty juvenile fish averaging 5.0 g were allocated 40 fish per tank to 21 180-L flow-through tanks. Seven experimental diets were prepared: control (Con) without additive, 0.5 and 1% Porphyra extract (PE), 3% Porphyra powder (PP), 0.5 and 1% sea tangle extract (STE) and 3% sea tangle powder (STP), referred to as PE-0.5, PE-1, PP-3, STE-0.5, STE-1, and STP-3, respectively. Each additive was included in the experimental diet at the expense of the same amount of wheat flour. Each experimental diet was fed to triplicate groups of fish. The experimental diets had no effect on the survival, weight gain or specific growth rate of the fish, feed consumption, feed efficiency ratio, protein efficiency ratio, protein retention, hepatosomatic index, condition factor, moisture or crude protein content of the entire body excluding the liver or moisture, crude protein or crude lipid content of the liver. None of the plasma parameters were affected by the experimental diets. Based on these results, the dietary inclusion of Porphyra and sea tangle did not affect the growth, feed utilization, body composition or plasma chemistry of juvenile Korean rockfish.

  • Three feeding trials were conducted to investigate the optimal feeding rates of subadult Korean rockfish Sebastes schlegeli at low, optimal and high water temperatures. In Exp-I, triplicate groups of fish (248 g) were fed a commercial diet (CD) with five different feeding rates of 0.1%, 0.2%, 0.4%, 0.6% and 0.8% body weight per day for 10 weeks at the low temperature ($11.6-15.2^{\circ}C$). In Exp-II, fish (129 g) were fed the same CD with five different feeding rates of 0.4%, 0.8%, 1.2%, 1.6% and 2.0% body weight per day for 10 weeks at the optimal temperature ($16.0-18.8^{\circ}C$). In Exp-III, fish (85.7 g) were fed the same CD with five different feeding rates of 0.4%, 0.8%, 1.2%, 1.6% and 2.0% body weight per day for 12 weeks at the high temperature ($21.0-28.5^{\circ}C$). The optimal feeding rates in the low, optimal, and high water temperatures were estimated to be 0.56%, 0.93% and 1.17% body weight per day, respectively, for subadult Korean rockfish.

  • The effects of noise stress response on hematological parameters (hemoglobin, hematocrit and MCHC) and plasma parameters (cortisol, glucose and albumin) in Korean rockfish (Sebastes schlegeli), a very important commercial marine fish in Korea, were investigated. These parameters were analyzed on fish exposed to an explosion of noise. There were no significant differences or trends in hematological parameters (hematocrit; control $29.7{\pm}4.8%$, experiment 32.0 35.5%; hemoglobin; control $6.5{\pm}0.7g/dL$, experiment 6.2 7.8 g/dL; MCHC; control $19.6{\pm}0.6g/dL$, experiment 19.9~22.2 g/dL). However, plasma cortisol and glucose exhibited significant differences from start to finish and displayed the following patterns (cortisol; control $180.7{\pm}35.4ng/mL$, experiment 247.0 444.5 ng/mL; glucose; control $32.5{\pm}6.3mg/dL$, experiment 50.5 109.0 mg/dL). In addition, the glucocorticoid receptor (GR) mRNA expression and basal levels of various tissues (eye, gills, liver, intestine, skin and gonads) were investigated for the first time in this marine fish. When the Korean rockfish was exposed to explosive noise stress, the GR mRNA was expressed more in the gonads than in other tissues tested and was elevated significantly from two and four times in the liver and gills, respectively, after noise exposure.

  • This study investigated the survival rates and physiological responses of Korean rockfish, Sebastes schlegeli, in live fish containers ($8^{\circ}C$, 33 psu) for 18 days. The survival rate was 99% and 97% in the control and experimental groups, respectively. Hematocrit and hemoglobin did not differ significantly between the control and experimental groups. Glucose and cortisol rose immediately on the first day of containment, but both gradually normalized. Aspartate aminotransferase and alanine aminotransferase did not differ significantly between the two groups after recovery. $NH_3$ continued to rise after the first day, but decreased to a level not significantly different from that of the control group during the recovery period. Plasma ions and osmolality did not change abnormally. The hemocyte population was not significantly different from that of the control. The ratios of apoptotic and necrotic cells indicated no specific variation in hemocyte viability. The histological changes in the skin and gills were not significantly different from those seen in the control. The experimental data obtained in this study suggest that live fish containers may be used to transport Korean rockfish without significantly affecting their physiology or survival.

조피볼락 (Sebastes schiegeii)에서 다환성방향족탄화수소 fluoranthene의 축적과 배설 (Accumulation and Depuration of Fluoranthene, a Polycyclic Aromatic Hydrocarbon, in Rockfish Sebastes schlegeli)

  • Rockfish Sebastes schlegeli was exposed to fluoranthene, a ubiquitous polycyclic aromatic hydrocarbon, at 1 and 10 $\mu$g/L for 4 weeks followed by depuration period of 8 weeks. Although the fluoranthene in the p]asma reached only 1.8$\~$1.9 times seawater concentration, it was 6.5 $\~$ 15.7 times higher in the liver, spleen and bile indicating efficient accumulation in the lipid -containing body tissues. When the exposed fish were then maintained in clean water, rapid fluoranthene decline occurred in the initial 2 weeks followed by a rather slow phase. This result suggests that fluoranthene accumulates efficiently provided the existence in the culture medium, but the contaminant disappears rapidly once the chemical source is removed. The fluoranthne residue in fish tissues my be a good indifator for relent PAHs exposure.
