Studies on the Production of Elderberry Wine (Elderberry Wine 제조(製造)에 관(關)하여)

  • This experiment was carried out to evaluate the possibility of wine-making with Elderberry cultivated in Korea. The results obtained are summarized as follows: 1. The Elderberry fruits were composed of 82.8% moisture, 0.82% crude fat, 3.73% crude protein and 0.83% crude ash. 2. The Elderberry juices were composed of 9.2% sugar, 0.62% total acidity (as tartaric acid), pH 4. 46 and the percentage juice extraction ratio from fruits was 61.4%. 3. In fermentation experiments carried out with two strains, Saccharomyces cerevisiae ellipsoideus showed better characteristics for wine-making than Saccharomyces cerevisiae montrachet. 4. In fermentation experiments carried out with Saccharomyces cerevisiae ellipsoideus under different storage conditions, frozen fruits showed better characteristics for wine-making than dried fruits. 5. In fermentation experiments carried out with Saccharomyces cerevisiae ellipsoideus under different pressing times, red wine and pink wine showed little difference. 6. The results of organic acid contents determined by gas chromatography showed that citric acid was the main acid for frozen fruit, and lactic acid for both dried fruit and wines. 7. Sensory evaluation showed that the wine pressed at the 3rd day had the best palatability but the wine fermented with the dried fruits had the worst among the tested.

Wine Quality Properties with Reference to the Temperature of Grape-Must Prior to Fermentation (발효전 포도 으깸이의 열처리 온도에 따른 와인 품질특성)

  • We investigated the effect of grape-must temperature ($50^{\circ}C$, $60^{\circ}C$, and $70^{\circ}C$) on wine quality. We used Muscat Bailey A (MBA) grapes; these are often used in wine making in Korea. The numbers of viable bacteria, yeasts and molds in grape-must decreased with an increase in temperature. Polyphenol oxidase activities in grape-must were reduced at high temperatures. Redness; and, total anthocyanin, polyphenol, and tannin contents of MBA grape wine were higher after heating at $70^{\circ}C$ than after use of lower temperatures. Antioxidant activity was also enhanced after exposure to high temperatures, which may be attributable to the presence of increased levels of polyphenolic compounds in the grape-must. Thus, the heat treatment to prepare grape-must before wine fermentation greatly affects wine quality.

The Chemical Characteristics of Korean Red Wine with Different Grape Varieties (포도품종을 달리한 적포도주의 이화학적 성분변화 (I))

  • Five kinds of red wine from three different grape varieties, Gerbong (G), Campbell (C), Moru (M), Gerbong+Moru (70:30, GM) and Gerbong+Campbell (70:30, GC), were prepared. Their chemical and microbiological changes were monitored during alcohol fermentation and aging. The changes of pH, brix and total acidity were $3.2{\sim}3.6,\;17.9{\sim}6.0$ and $2.4{\sim}4.6\;g/L$, respectively. The viable cell numbers of initial, stationary and death phases were $6.0{\times}10^6,\;1.0{\times}10^8$ and $7.0{\times}10^5\;cfu/mL$ during alcohol fermentation. The sugar fermentability, glucose and fructose contents were greatly decreased less than 0.2 g/L, and the final contents of ethanol and sulfur dioxide were $11.4{\sim}12.3%$ and $40{\sim}62\;mg/L$, respectively. The conversion ratios of malic acid to lactic acid were 23% (G), 67% (M), 28% (C), 33% (GM) and 39% (GC). The chemical characteristics of five red wine were significantly different in pH, total acidity, sulfur dioxide and lactic acid contents (p<0.05).

Study on the Color Characteristics of Korean Red (한국산 적포도주의 색도 변화에 관한 연구 (II))

  • Five kinds of red wine from three different grape varieties, such as Gerbong (G), Campbell (C), Moru (M), Gerbong+Moru (70:30, GM), and Gerbong+Campbell(70:30, GC) were prepared in the year of 2000. The total phenolic content and color changes were monitored using hunter colorimeter and spectrophotometry methods during wine processing. The total phenolic content of Gerbong (G), Campbell (C), Moru (M), Gerbong+Moru(70:30, GM), and Gerbong+Campbell(70:30, GC) were 712.6 mg/L, 3472.9 mg/L, 2209.4 mg/L, 2019.4 mg/L, and 1184.5 mg/L, respectively. Between total phenolic content and hunter colorimeter value (L, a, and b) of red wine showed no significant difference, but that of spectrophotometry color value (hue and intensity) were significantly different (p<0.001). The total phenolic content, hue, and intensity values represented significantly different between grape varieties (p<0.01, p<0.001). the hue showed a negative correlation relationship $(r^2=0.8660,\;p<0.0001)$, and that of intensity was a positive $(r^2=0.8304,\;p<0.0001)$ between total phenolic contents. The total phenolic content of red wine could be estimated simply using these equations, $Y_{Total\;phenolic\;content\;(mg/L)}=3319.3-X_{Hue}/2208.36,\;and\;Y_{Total\;phenolic\;content\;(mg/L)}=1230.97-X_{intensity}/98.93$

Evaluation of Lysozyme to Control Vinification Process and Histamine Production in Rioja Wines

  • Lysozyme and the reduction of metabisulfite addition to red wine were evaluated during a winemaking process and after malolactic fermentation (MLF). Treatment, with lysozyme, of the must from Tempranillo grapes and at the end of alcoholic fermentation (AF) caused the 100% implantation of the inoculated bacterial strain and shortened the duration of MLF by 7 days. At the end of the MLF, wines treated with lysozyme showed lower volatile acidity, color intensity, and biogenic amine content. The differences in color intensity disappeared during wine stabilization. The lysozyme addition after MLF led to lower histamine concentrations in wines. These phenomena occurred irrespective of the lactic acid bacteria (LAB) proliferation control and of the Oenococcus oeni dominant strain identified at this period. The results of this study show the significance of preventive use of lysozyme in vinification of red wine to maintain low histamine levels and ensure a successful implantation of inoculated O. oeni starters.

Anti-Thrombosis Activity of Sinapic Acid Isolated from the Lees of Bokbunja Wine

  • From the lees of bokbunja wine (LBW) made from Rubus coreanus Miquel, we have identified six compounds (1: trans-4-hydroxycinnamic acid; 2: trans-4-hydroxy-3-methoxycinnamic acid; 3: 3,4-dihydroxycinnamic acid; 4: 4-hydroxy-3-methoxybenzoic acid; 5: 3,5-dimethoxy-4-hydroxybenzoic acid; and 6: 3,5-dimethoxy-4-hydroxycinnamic acid (sinapic acid)) through silica gel chromatography and UHPLC-MS. The compounds 1-6 showed strong anticoagulation and platelet aggregation inhibitory activities without hemolytic effect against human red blood cells. To date, this is the first report of the in vitro anti-thrombosis activity of sinapic acid. Our results suggest that different cinnamic and benzoic acid derivatives are closely linked to the anti-thrombosis activity of LBW, and sinapic acid could be developed as a promising anti-thrombosis agent.

A Study on the Sensory Characteristics of Korean Red Wine (한국산 적포도주의 관능적 특성에 관한 연구 (III))

  • The sensory characteristics of red wine Gerbong (G), Campbell (C), Moru (M), Gerbong + Moru (70 : 30, GM), Gerbong + Campbell (70 : 30, GC) and French wine (F, Carbernet Sauvignon, 1998) were evaluated. The preferences of color, flavor, taste and total evaluation were determined by a ranking test, and the organoleptic characteristics were evaluated by a quantitative descriptive analysis (QDA) method. The mean color scores of C, GM, F, GC, M and G were 4.74, 3.94, 4.67, 3.70, 2.65 and 1.47, respectively (p<0.001). The order for the mean score for flavor was GM (4.12) = M (3.94) = C (3.76) = F (3.76) ${\geq}$ GC (3.12)>G (2.29) (p<0.01), and the order for taste was F (4.75) ${\geq}$ C (4.25) ${\geq}$ GM (3.37) = GC (3.50) ${\geq}$ G (2.75) = M (2.37) (p<0.001). The total evaluation of mean scores showed G, M, C, GM, GC and F were 237, 2.44, 4.06, 3.87, 3.64 and 4.81, respectively (p<0.001). Influences of sensory characteristics on the total evaluation, in percentages, were 69.3% for taste, 3.7% for color, and 1.5% for flavor. The influences of taste, color, and flavor in red wine were 17% for sweet, acid, bitter and salty taste, 28.9% for purple and red color, and 14.4% for grape flavor. The attributes of the purple and red colors showed a positive correlation with grape flavor, oak flavor, grape taste, and floral tastes, but a negative correlation with $SO_2$, flavor. The attribute of sweet taste showed a positive correlation with grape flavorand floral flavor, but a negative correlation with bitter and astringency tastes, according to Pearsons correlation analysis (p<0.01).

  • In order to investigate the possibility of wine-making with the grape variety cultivating in Korea, the basic analysises were carried out; analysis of the grape maturity during ripening period, the composition of grape juice, and the chemical components of wine from different varieties. Also the yeasts existing naturally on the grape were isolated and identified. For the selection of strain, the characteristics of 6 strains were studied. The results obtained were summarized as follows. 1. The reducing sugar content increased considerably during two weeks after the ordinary grape-gathering period. 2. The sugar content was highest in Muscat bailey A, which could be fermented naturally. The other varieties, Campbell Early, Steuben, and Alden needed chaptalisation for wine-making. 3. The permanaganate number and the methanol content of wine from all varieties were lower than french wine. 4. The sensory evaluation of wine showed that the pink wine was appreciated better than the red wine of same variety and the foxy taste of wine from hybrid grape influenced little to Korean. 5. The selected 6 strains were identified as Saccharomyces chevaliers, Saccharomyces capensis and Saccharomyces globosus. The strain No. 3 and No. 4 showed the most excellent characters for wine-making.

Manufacture of the Red Ginseng Vinegar Fermented with Red Ginseng Concentrate and Rice Wine, and its Quality Evaluation (홍삼 농축액과 쌀막걸리의 동시 발효를 통한 홍삼 식초의 제조 및 품질평가)

  • The objectives of this study were to manufacture the red ginseng vinegar based on rice wine and red ginseng concentrate (RGC) using $Acetobacter$ $aceti$ and to evaluate its quality with remaining crude saponin contents and sensory score. The maximum prosapogenin (ginsenoside-Rh1, Rh2, Rg2, and Rg3) content in RGC regarding ginseng was obtained from such processes as steaming, drying, and extraction. When RGC was added into a rice wine in the range of 0-1% before acetic fermentation, pH decreased slowly during 20 days depending on RGC contents, but total acidity was not dependent on RGC contents. Compared to the crude saponin content (71.75 mg/g) of ginseng vinegar added RGC after acetic fermentation, the fermentation with RGC produced a lower crude saponin content (16.95 mg/g) in red ginseng vinegar. Sensory scores such as odor, taste, and overall preference, however, vinegar fermented with RGC were higher than those of vinegar added RGC after acetic fermentation.

Effect of Dietary Phenols on Body Tissue Oxidative State and Cancer Prevention (식이내 페놀류들이 생체조직의 산화상태와 항암작용에 미치는 영향)

  • In this study, we tried to figure out how phenol effects on cancer prevention, and for this purpose we focused on phenol effects on TBARS and the relationship between TBARS(thiobarbituric acid-reactive substances) and cancer. A protocol using a nutritionally adequate amino acid-based diet and a transgenic mouse model of neurofibromatosis was used to evaluate the effect of dietary phenols on body tissue oxidation and tumor onset. The mice carry the human T-lymphotropic virus type-1 transactivator(texl) gene and spontaneously develop externally visible tumors. Twenty-five male transgenic mice were systematically assigned into five groups, control group, 2 mmol, 4 mmol, 8 mmol catechin/kg diet groups and wine solid group. Mice in control group were without catechin, Mice in wine solid group received red wine 750 mL/kg diet, Mice were examined daily, and the age at which a first tumor appeared was recorded. Transgenic mice consuming catechin and wine solid were older when a first tumor appeared. No tumor was found in one mouse of 4 mmol catechin/kg diet and one mouse of 8 mmol catechin diet group. Levels of TBARS in brain and spleen of 8 mmol catechin group and wine solid group were significantly decreased as compared to the same tissue in control group. TBARS levels in tissues were significantly correlated with tumor onset. Results from this study suggest that dietary phenol effects on cancer prevention through tissue antioxidation in spite of different kinds of phenols.
