A Study on the Contents of Tryptophan and Available Lysine in Korean Foods (수종한국상용식품중(數種韓國常用食品中)의 Tryptophan 및 Available lysine 함량(含量)에 대(對)하여)

  • In order to observe the contents of some essential amino acids in Korean foods, total and free tryptophan, and available lysine in fifty kinds of Korean foods were analyzed by the sfectrophotometry. The results obtained are summarized as follows : 1) The tryptophan contents per 100g of soybean, wet green laver and skim milk were over 250 mg, and the contents Per g nitrogen in ginger, dried persimmon and chestnut were over 150 mg. 2) The free tryptophan contents per 100 g of soybean, small red bean, dried persimmon, ginseng stem were over 100 mg, ana the contents per g nitrogen in dried persimmon was over 150 mg. 3) The available lysine contents Per 100g of soybean, wet green laver and skim milk, fish(auchovy), dried yeast, casein and silkworm pupa were over 1000 mg, and the contents per g nitrogen in potato, perilla(wild sesame), red pepper, sausage and skim milk were over 300mg. 4) The contents of tryptophan and available lysine in soybean, green laver and skim milk were higher than in other samples. 5) In general, the contents of tryptophan and available lysine were abundant in seaweeds.

Potential Role of Polyphenolics and Polyphenol Oxidase on the Induction of Browning in Ginseng Roots (인삼 적변유기에 대한 폴리페놀과 Polyphenol Oxidase의 잠재적 역할)

  • This study was conducted to investigate the potential of polyphenols and polyphenol oxidase activity on the induction of rusty symptom development in ginseng root. When rusty inducing bacteria were inoculated on fresh ginseng root, the hue value of the inoculated root increased from 101.2 (white yellow) at 1 day after innoculation to 60.9 (brownish red) at 30 days after innoculation. Lysobacter gummosus, Pseudomonas veronii and Agrobacterium tumefaciens enhanced the accumulation of total phenolics. Along with the increase of total phenolics, total activity of polyphenol oxidase concomitantly increased but the specific activity of the enzyme was not.

Bioconversion of Ginsenosides by Bifidobacterium CBT BG7, BR3 and BL3 (비피도박테리움 CBT BG7, BR3, BL3의 진세노사이드 전환능)

  • In this study, we identified that the fermentation of Korean indigenous probiotics and red ginseng produced ginsenoside compound K (CK) from major ginsenosides. Based on whole genome sequencing of 19 probiotics species, β-glucosidase, α-arabinofuranosidase, β-xylosidase, and α-rhamnosidase related to bioconversion of ginsenosides are identified in the genome of 19 species, 3 species, 6 species, and 8 species, respectively. Among the 19 probiotics species, Bifidobacterium longum CBT BG7 converted from ginsenoside Rb1 to CK, and both B. breve CBT BR3 and B. lactis CBT BL3 converted ginsenoside Rb1 to Rd. The final concentration and yield of ginsenoside F2 and CK were higher in the fermentation with the nondisrupted cells than with disrupted cells. The combination of both CBT BG7 and BL3, and CBT BG7 and BR3 showed higher amounts of F2 than CBT BG7 only. CBT BG7 with adding α-amylase increased the amounts of F2. In this study, we identified that the fermentation of both Korean indigenous probiotic bacteria CBT BG7, BR3 and BL3, and red gingseng is able to produce CK, a bioactive compound that promotes health benefits.

Preparation and Quality Characteristics of the Fermentation product of Ginseng by Lactic Acid Bacteria (FGL) (유산균을 이용한 발효인삼 제조 및 품질 특성)

  • Ginseng as a raw material for production of probiotic ginseng product by lactic acid bacteria (LAB) was evaluated in this study. Either white ginseng (WG) or red ginseng (RG) (1% or 5%, w/v) were directly inoculated with a 24 hold seed culture of twenty seven substrains of four different LAB ($1.0{\times}10^6CFU/ml$); Lactobacillus spp., Streptococcus/Enterococcus spp., Leuconostoc/Lactococcus spp. and Bifidobacterium spp., and incubated at $37^{\circ}C$ for 24 or 48 h. Among 27 kinds of LAB, seven substrains of Lactobacillus (MG208, MG311, MG315, MG501, MG501C, MG505, MG590) and one Bifidobacterium (MG723) were selected based on their dose dependent stimulation of the growth of LAB in the presence of ginseng and changes in pH, acidity and viable cell counts during fermentation were examined. Lactobacillus MG208 specifically was found to show the best growth on 5% RG and reached nearly $14.0{\times}10^8CFU/ml$ after 48 h of fermentation and produced the titratable acidity as $0.84{\pm}0.02%$, whereas the pH was significantly lowered from $6.80{\pm}0.01\;to\;3.42{\pm}0.02$. These results indicated that ginseng can be an appropriate material to prepare the fermentation product by several strains of LAB. Therefore we should further check whether probiotic ginseng product may have synergistic health benefits of both probiotics and ginseng to serve for vegetarians and lactose-allergic consumers.

Hypocholesterolemic Effect of Panaxydol in High Cholesterol Diet Fed Rats and Mice (콜레스테롤 식이한 흰쥐와 생쥐에서 Panaxydol의 고콜레스테롤혈증 억제 효과)

  • The effect of petroleum ether soluble fraction (PESF) and panaxydol extracted from red ginseng on serum cholesterol levels was examined in rats and mice fed on a high cholesterol diet. When 2% cholesterol diet containing PESF (800mg/kg diet) or panaxydol (100mg/kg diet) was fed to mice for two weeks, total cholesterol and LDL-cholesterol in the serum were significantly decreased in PESF and panaxydol treated group. Panaxydol (5, 10mg/kg b.w./day) was administered for 3 consecutive days by intraperitoneal injection in rats fed on 1% cholesterol diet. Serum total cholesterol, triglyceride and LDL-cholesterol levels of panaxydol administered group (5 mg/kg b.w./day) were decreased by 55%, 33% and 67% compared with those of the control group, respectively. However, a significant increase of serum HDL-cholesterol was observed on panaxydol administered group. Furthermore, hepatic cholestrol levels was significantly decreased in panaxydol administered group. These results suggest that panaxydol could protect hypercholesterolemia.

Role of microRNA-520h in 20(R)-ginsenoside-Rg3-mediated angiosuppression

  • Background: Ginsenoside-Rg3, the pharmacologically active component of red ginseng, has been found to inhibit tumor growth, invasion, metastasis, and angiogenesis in various cancer models. Previously, we found that 20(R)-ginsenoside-Rg3 (Rg3) could inhibit angiogenesis. Since microRNAs (miRNAs) have been shown to affect many biological processes, they might play an important role in ginsenoside-mediated angiomodulation. Methods: In this study, we examined the underlying mechanisms of Rg3-induced angiosuppression through modulating the miRNA expression. In the miRNA-expression profiling analysis, six miRNAs and three miRNAs were found to be up- or down-regulated in vascular-endothelial-growth-factor-induced human-umbilical-vein endothelial cells (HUVECs) after Rg3 treatment, respectively. Results: A computational prediction suggested that mature hsa-miR-520h (miR-520h) targets ephrin receptor (Eph) B2 and EphB4, and hence, affecting angiogenesis. The up-regulation of miR-520h after Rg3 treatment was validated by quantitative real-time polymerase chain reaction, while the protein expressions of EphB2 and EphB4 were found to decrease, respectively. The mimics and inhibitors of miR- 520h were transfected into HUVECs and injected into zebra-fish embryos. The results showed that overexpression of miR-520h could significantly suppress the EphB2 and EphB4 protein expression, proliferation, and tubulogenesis of HUVECs, and the subintestinal-vessel formation of the zebra fish. Conclusion: These results might provide further information on the mechanism of Rg3-induced angiosuppression and the involvement of miRNAs in angiogenesis.

Plant Growth-promoting Activity of Acremonium strictum MJN1 Isolated from Roots of Panax ginseng

  • The plant growth-promoting activity of Acremonium strictum MJN1 isolated from roots of Panox ginseng was explored. The myceliaI extract of A. strictum MJN1 enhanced the rice seedling growth by 14.5 and 9.0% in the dried weight of shoots and roots, and the growth of red pepper by 54 and 85% in the top length and the dried weight in pot experiments, respectively. The plant growth-promoting substances in the myceliaI extract of Acremonium strictum MJN1 were identified as D-adenosine and glycerol. Both commercial D-adenosine and glycerol also promoted significantly the rice seedling growth but, unlike the mycelial extract of A. strictum MJN1, hardly affected the yields of plants grown in pots or field. Therefore, it is possible that other plant growth-promoting substances are produced by A. strictum MJN1. However, this study shows that A. strictum MJN1 has a great potential as a biofertilizer.

Ginsenoside Rg3 protects against iE-DAP-induced endothelial-to-mesenchymal transition by regulating the miR-139-5p-NF-κB axis

  • Background: Emerging evidence suggests that endothelial-to-mesenchymal transition (EndMT) in endothelial dysfunction due to persistent inflammation is a key component and emerging concept in the pathogenesis of vascular diseases. Ginsenoside Rg3 (Rg3), an active compound from red ginseng, has been known to be important for vascular homeostasis. However, the effect of Rg3 on inflammation-induced EndMT has never been reported. Here, we hypothesize that Rg3 might reverse the inflammation-induced EndMT and serve as a novel therapeutic strategy for vascular diseases. Methods: EndMT was examined under an inflammatory condition mediated by the NOD1 agonist, γ-d-glutamyl-meso-diaminopimelic acid (iE-DAP), treatment in human umbilical vein endothelial cells. The expression of EndMT markers was determined by Western blot analysis, real-time polymerase chain reaction, and immunocytochemistry. The underlying mechanisms of Rg3-mediated EndMT regulation were investigated by modulating the microRNA expression. Results: The NOD1 agonist, iE-DAP, led to a fibroblast-like morphology change with a decrease in the expression of endothelial markers and an increase in the expression of the mesenchymal marker, namely EndMT. On the other hand, Rg3 markedly attenuated the iE-DAP-induced EndMT and preserved the endothelial phenotype. Mechanically, miR-139 was downregulated in cells with iE-DAP-induced EndMT and partly reversed in response to Rg3 via the regulation of NF-κB signaling, suggesting that the Rg3-miR-139-5p-NF-κB axis is a key mediator in iE-DAP-induced EndMT. Conclusion: These results suggest, for the first time, that Rg3 can be used to inhibit inflammation-induced EndMT and may be a novel therapeutic option against EndMT-associated vascular diseases.

Ginsenoside Rg1 suppresses early stage of adipocyte development via activation of C/EBP homologous protein-10 in 3T3-L1 and attenuates fat accumulation in high fat diet-induced obese zebrafish

  • Background: Ginsenoside Rg1 is a class of steroid glycoside and triterpene saponin in Panax ginseng. Many studies suggest that Rg1 suppresses adipocyte differentiation in 3T3-L1. However, the detail molecular mechanism of Rg1 on adipogenesis in 3T3-L1 is still not fully understood. Methods: 3T3-L1 preadipocyte was used to evaluate the effect of Rg1 on adipocyte development in the differentiation in a stage-dependent manner in vitro. Oil Red O staining and Nile red staining were conducted to measure intracellular lipid accumulation and superoxide production, respectively. We analyzed the protein expression using Western blot in vitro. The zebrafish model was used to investigate whether Rg1 suppresses the early stage of fat accumulation in vivo. Results: Rg1 decreased lipid accumulation in early-stage differentiation of 3T3-L1 compared with intermediate and later stages of adipocyte differentiation. Rg1 dramatically increased CAAT/enhancer binding protein (C/EBP) homologous protein-10 (CHOP10) and subsequently reduced the $C/EBP{\beta}$ transcriptional activity that prohibited the initiation of adipogenic marker expression as well as triglyceride synthase. Rg1 decreased the expression of extracellular signal-regulated kinase 1/2 and glycogen synthase kinase $3{\beta}$, which are also essential for stimulating the expression of $CEBP{\beta}$. Rg1 also reduced reactive oxygen species production because of the downregulated protein level of nicotinamide adenine dinucleotide phosphate hydrogen (NADPH) oxidase 4 (NOX4). While Rg1 increased the endogenous antioxidant enzymes, it also dramatically decreased the accumulation of lipid and triglyceride in high fat diet-induced obese zebrafish. Conclusion: We demonstrated that Rg1 suppresses early-stage differentiation via the activation of CHOP10 and attenuates fat accumulation in vivo. These results indicate that Rg1 might have the potential to reduce body fat accumulation in the early stage of obesity.

Effect of Cultivation Using Plastic-Film House on Yield and Quality of Ginseng in Paddy Field (논토양에서 비닐하우스를 이용한 재배방법이 인삼의 수량 및 품질에 미치는 영향)

  • The average and maximum temperature were $29.5^{\circ}C$ and $33.2^{\circ}C$ at 2:00 p.m. respectively, in the plastic-film house covered with shade net, and both of temperature were lower $0.6^{\circ}C$ and $1.3^{\circ}C$ than those of conventional shade. Light transmittance was 14% in the plastic-film house, while 9.9% in conventional shade during growing season from May to October. Withering time of aboveground part was on October 3rd in conventional shade with 60% of withering leaf, while it was on November 10th with 3.7% of withering leaf in the plastic-film house, about 40 days longer survival. The main disease incidence were 15% of anthracnose, 17% of leaf spot, 5% of phytophthora blight and 3% of gray mold in the conventional shade, while 0 ~ 0.1% disease incidence and 95% of emergence rate in the plastic-film house. The growth in the aboveground and underground part of ginseng was totally better, particularly characteristics affecting yield such as root length, main root length and diameter in the plastic-film house. The fresh weight was increased by 128% compared to the conventional shade and harvested roots per $3.3m^2$ were 36 roots in the conventional shade and 58 roots in the plastic-film house and futhermore yield per $3.3m^2$ was increased by 216% compared to the conventional shade. As covering materials, the rice straw in the plastic-film house was excellent. The ginsenoside contents affecting the quality of ginseng were higher in the plastic-film house indicating 0.333% of Rg1, 0.672% of Rb1, 0.730% of Rc and rate of red rusty root was less than 4.0 ~ 6.1%. Above the results, the quality of ginseng grown in the plastic-film house covered with shade net was improved than that of the conventional shade.
