Regeneration of Glyphosate-Resistant Plant from Tobacco (Nicotiana tabacum) Cell Culture (담배(Nicotiana tabacum) 세포배양에 의한 Glyphosate-저항성 개체의 재분화)

  • From the single cell cultures of haploid tobacco (Nicotiana tabacum L. cv. NC 2326) glyphosate-resistant plants were regenerated. After treated with N-methyl-N'-nitro-N-nitrosoguanidine (MNNG) and then inoculated onto the LS medium supplemented with 1 mM glyphosate, the single cells survived formed colonies and calluses in 65 and 95 days after culture, respectively, and then whole plants were regenerated in 0.1 mM glyphosate-containing medium from the selected calluses. There was no difference in fresh weight and shikimate content between the selected and normal haploid calluses. When sprayed with 0.1 mM glyphosate, the shikimate contents in the regenerated and normal plants were 0.659 and 20.816 mol/g fr. wt., while that in other normal plants which were not sprayed was 0.921. In addition, the calluses induced from the regenerated plants grew without showing any retardation when treated with glyphosate. These results indicate that the secelcted calluses and regenerated plants are resistant to glyphosate.

Identification of Aquatic Plants in the Muncheon Water Reservoir Using Drone-based Information (드론원격정보를 활용한 저수지 수생식물 분포 파악: 경북 문천저수지에서의 적용 예)

  • Aquatic plants serve the crucial function of helping to balance water reservoir ecosystem, as they filter and remove major minerals required for algal growth such as nitrogen, ammonia, and nitrates. Aquatic plants provide food, shade, and protection for the aquatic biome in and around the reservoir. Thus, it is important to accurately determine the existence and areal extent of the aquatic plants. In the present study drone-based facilities were used for this purpose. In the Muncheon water reservoir, Gyeongbuk, the Normalized Difference Vegetation Index (NDVI) and Surface Algal Bloom Index (SABI) were used to determine the existence status of the aquatic plants. The data so obtained exhibited reasonable accuracy; drone-based facilities can be used in future to identify the areal extent of aquatic plants.

Stabilization of photosynthetic machinery against low-temperature photoinhibition by fatty acid unsaturation of membrane lipids in plants

  • CHilling tolereance of plants are closely correlated with the degree of fatty acid unsaturation of membrane lipids. We studied the effects of low-temperature photoinhibition on the photochemical efficiency of photosystem II in terms of fatty acid unsaturation of thylakoid membranes lipids isolated from chilling -sensitive plants and chilling -resistant ones. To directly test the chilling tolerance of photosynthetic machinery in relation to membrane lipids, we further compared wild type tobacco plants with that of transgenic tobacco plants, in which the sensitivity to chilling had been enhanced by genetic modification of fatty acid unsaturation of chloroplast membrane lipids. The transgenic tobacco plants were found to contain reduced levels of unsaturated membrane fatty acids after being transformed with cDNA for glycerol-3-phophate acyltransferase from squash. The functional integrity of photosystem II during and recovery of photosynthesis from low-temperature photoinhibition will be discussed in connection with the degree of fatty acid unsaturation of chlorophast membranes lipids.

Regulation of methionine biosynthesis in plants; transgenic study

  • The committing step in Met and S-adenosyl-L-methionine (SAM) synthesis is catalyzed by cystathionine ${\gamma}$ -synthase (CGS). Transgenic Arabidopsis thaliana overexpressing CGS under control of 35S promoter show increased soluble Met and its metabolite S-methylmethionine, but only at specific stages of development. CGS-overexpressing seedlings are resistant to ethionine. Similar results were obtained with transgenic potato plants overexpressing Arabidopsis CGS. Several of the transgenic lines show silencing of CGS resulting in deformed p]ants with a reduced capacity for reproductive growth similar as transgenic plants by antisense RNA (CGS[-]). Exogenous feeding of Met to the CGS[-] and CGS[+] silenced plants partially restores their growth. Similar morphological deformities are observed in plants cosuppressed for SAM synthetase, even though such plants accumulate 250 fold more soluble Met than wild type and they overexpress CGS. The results suggest that the abnormalities associated with CGS and SAM synthetase silencing are due in part to a reduced ability to produce SAM, and that SAM may be a regulator of CGS expression.

COMPUTATIONAL INTELLIGENCE IN NUCLEAR ENGINEERING

  • Approaches to several recent issues in the operation of nuclear power plants using computational intelligence are discussed. These issues include 1) noise analysis techniques, 2) on-line monitoring and sensor validation, 3) regularization of ill-posed surveillance and diagnostic measurements, 4) transient identification, 5) artificial intelligence-based core monitoring and diagnostic system, 6) continuous efficiency improvement of nuclear power plants, and 7) autonomous anticipatory control and intelligent-agents. Several changes to the focus of Computational Intelligence in Nuclear Engineering have occurred in the past few years. With earlier activities focusing on the development of condition monitoring and diagnostic techniques for current nuclear power plants, recent activities have focused on the implementation of those methods and the development of methods for next generation plants and space reactors. These advanced techniques are expected to become increasingly important as current generation nuclear power plants have their licenses extended to 60 years and next generation reactors are being designed to operate for extended fuel cycles (up to 25 years), with less operator oversight, and especially for nuclear plants operating in severe environments such as space or ice-bound locations.
