  • 본 연구에서는 국산배의 기능학적 우수성 증명을 위해 국산배로부터 생리활성 화합물을 밝히고자 하였다. 이에 배 과피 MeOH 추출물을 용매분획하여 얻은 EtOAc-산성획분을 대상으로 Sephadex LH-20 column chromatography와 ODS-HPLC를 이용하여 순차적으로 정제를 행하여 2종의 화합물을 단리하였다. 단리된 화합물 1과 2는 MS 및 NMR 분석을 통하여 각각 quercetin 3-O-${\beta}$-Dglucopyranoside(화합물 1)와 3,5,6,7,8,3',4'-heptahydroxyflavan [(-)-dulcisflavan, 화합물 2)]으로 동정되었다. 단리된 화합물 1은 동양배로부터, 그리고 화합물 2는 배로부터 처음 동정된 화합물이다. 본 연구결과가 국산배의 기능학적 우수성 증명을 위한 기초자료로 활용되길 기대한다.

조생종 추석용 단감 '초시' 육성 ('Chosi', Early Maturing Sweet Persimmon (Diospyros kaki Thunb.) for 'Chuseok')

  • 조생종 추석용 품종을 육성하기 위해서 2000년 '조홍시'에 '서촌조생'을 교배하여 얻은 실생묘를 전라남도 영암군 소재 배연구소 교배육성 포장에 식재하였고 그 중에서 과실 특성이 우수한 '00-10-135' 계통을 1차 선발하여 검토한 결과, 연차간 변이가 적고 특성이 우수하여 최종 선발하여 '초시'로 명명하였다. 과실의 숙기는 9월 19일경으로 조생종이며, 평균 과중은 161 g이고, 당도는 15.3 °Bx이었으며, 과즙이 많고, 과육이 아삭하여 식미가 우수하다. 과형은 편원형이고 과피색은 오렌지색으로 외관이 수려하고, 과정부 열과, 과피미세균열 등 생리장해가 발생하지 않아 재배관리가 쉽다(등록번호: 제7075호).

  • Finite element method(FEM) was used to obtain an approximate solution, since the mathematical formulations for the problem are complex and cannot be solved analytically. In this study, the fruit as well as the aluminum support on vibrator are discretized into small elements, and the approximate functions are used to describe the displacements in each element in terms of nodal values, and because of the complexity of the problem of viscoelastic materials such as the fruit and vegetables, it was necessary to validate the modeling approach before pear simulations were performed, and the finite element modeling approach was first validated by comparing the results obtained from simulation and experiment for the pear in the frequency range 3 to 150 Hz and acceleration level of 0.25 G-rms. Based on the relatively good agreement between simulated and measured frequencies for the pear, finite element models of tomato and oriental melon were created to study the vibration characteristics of the fruit and vegetables. The resonance frequencies of the pear, tomato and oriental melon using FEM were 62.50, 39.45 and 62.73 Hz and the peak accelerations of them using FEM were 2.21, 1.38 and 1.98 G-rms.

Scab of Balsam Pear (Momordica charantia) Caused by Cladosporium cucumerinum in Korea

  • During winter season of 2004 to 2006, a scab disease on balsam pear (Momordica charantia) caused by Cladosporium cucumerinum was observed in Daesan-myon, Changwon-city, Gyeongnam province, Koyea. The disease symptom started with small dark brown speck on the fruits and leaves then the lesions expanded and spreaded irregularly. The aggregated mycelial mass and conidia of the fungus formed sooty scab. The colony of purely isolated fungus grew in greenish black to velvety on potato dextrose agar (PDA). Conidia were ellipsoidal, fusiform or subspherical, mostly one-celled but occasionally septated and $3{\sim}32{\times}2{\sim}6{\mu}m$ in size. The conidiophores were erected and had long branch, chains pale olivaceous brown in color and $6{\times}280{\mu}m$ in size. Ramoconidia were $10{\sim}34{\times}3{\sim}8{\mu}m$ in size. The fungus was identified as Cladosporium cucumerinum based on the morphological characteristics. The pathogenicity of the fungus was confirmed according to Koch's postulate. The optimum temperature of the isolate was about $20^{\circ}C$. This is the first report on scab of balsam pear caused by C. cucumerinum in Korea.

마이크로파를 이용한 잔류 살충제 Imidacloprid 분석용 시료전처리 기술개발 (Development of the Sample Pretreatment Technique using Microwave for Analysis of Insecticide Imidacloprid Residues)

  • This study was conducted to elucidate extraction efficiency by microwave technique in comparison with sonication technique for extraction of insecticide residue in pear. In the analysis of the extraction efficiency of microwave for a pear spiked with imidacloprid, the extraction efficiency by microwave power of 300 W with extraction temperature of $80^{\circ}C$, heating time of 1 to 3 minute was shown to be similar with the extraction time 20 minutes by sonication. The optimal condition. in consideration of economical condition and treatment time, for microwave extraction of imidacloprid in the pear were 300 watts of power supply, $100^{\circ}C$ of extraction temperature, 1 minute of heating time and 10 mL of acetone volume. A new microwave vessel was developed to rapidly process the sample of the insecticide imidacloprid residues in the pear. This vessel was designed to include a reaction chamber and a filtration equipment, and a gathering chamber. The system could curtail a pretreatment time to 21 minutes than sonication and 7.9 minutes than the previous microwave vessel.

Mycelial Yield of Pleurotus ostreatus Using Thinned Apple, Pear, and Peach on Submerged Culture

  • The effect of thinned fruits, apple, pear and peach, on the mycelial growth of mushrooms was investigated. The growth of mycelia with the addition of thinned fruit was clearly better than that in the control for all the tested mushrooms. The growth rate of Pleurotus ostreatus was faster than any other mushroom. The optimal concentrations of thinned apple, pear, and peach in a solid culture were 1.0%, 1.0%, and 3%, respectively, while in a liquid culture the optimal concentrations were 5,0%, 3.0%, and 5.0%, respectively. When Pleurotus ostreatus was incubated in a 20-L pilot scale fermenter with 10 L of a liquid medium containing 3% thinned fruit at 25$^{\circ}C$ and 6 vvm for 10 days, the mass-production of mycelia was 74.2 g/10 L (apple), 96.2 g/10 L (pear), and 86.3 g/10 L (peach). The mycelial yield of Pleurotus ostreatus in a medium containing thinned fruit was 2 ∼ 3 times higher than that in the control.

Blue Mold of Pear Caused by Penicillium aurantiogriseum in Korea

  • The isolation of a causal pathogen from pear fruits showing lesions of blue mold was carried out, which were preserved in the packinghouse of Anseong for the export to the United States. When the lesion of pears(Pyrus pyrifolia Niitaka) was checked in packinghouse, their fruit epidermis over decayed parts varied from light tan to dark brown. Also, the decayed flesh was soft and watery, and separated easily from the healthy tissue. Based on the cultural and morphological characteristics, the fungus(IL-12 isolate) was identified as Penicillium aurantiogriseum. This is the first report associated with blue mold of pear caused by P. aurantiogriseum in Korea.

Gas Chromatography Residue Analysis of Bifenthrin in Pears Treated with 2% Wettable Powder

  • This study was conducted to monitor the level of bifenthrin residues in pear sprayed with 2% bifenthrin wettable powder (WP) at the recommended rate at four different schedules prior to harvest. The target analyte was extracted with acetone, partitioned into dichloromethane, and then purified by florisil chromatographic column. The residue determination was performed on a DB-5 capillary column using GC with electron capture detector (ECD). Linearity of this method was quite good ($r^2$ = 0.9951) in the concentration ranged from 0.2 mg/kg to 10 mg/kg. Recovery test was carried out at two concentration levels, 0.2 mg/kg and 1.0 mg/kg, in three replicates, and their rates were from 82.9% to 107.2%. No quantitative bifenthrin was detected in pear of all kinds of treatments including the treatment sprayed 4 times until 7 days before harvest. This sensitive and selective method can be used to monitor the trace residual amounts of bifenthrin in pear in a quite low concentration level.

배나무(Pyrus spp.) 유전체 연구 현황 (Researches of pear tree (Pyrus spp.) genomics)

  • 배나무는 원산지와 분화방향에 따라 유럽, 미국, 호주 등에서 주로 재배되는 서양배와 중국, 일본, 한국 등 동남 아시아 지역을 중심으로 분포 및 재배되고 있는 동양배로 구분된다. 17개의 기본염색체를 가진 배나무는 대부분 이배성(2n=2x=34)이며, 단일 S 유전자좌에 의해 조절되는 자가불화합성과 과수 작물의 주요 특징인 유년성으로 인해 유전 연구 및 정밀한 품종 육성에 큰 제한을 받고 있다. 배나무속 식물의 유전연구는 분자생물학 관련 기술의 발달로 다양한 형태의 분자 표지의 개발이 이루어짐과 동시에 유연관계분석, 유전자지도작성, QTL 분석과 같은 다양한 유전연구에 활발히 이용되었다. 또한 배나무의 유전자지도는 병 저항성이나 다양한 유용형질과 연관된 QTL 확인을 위한 연구로 이어지고 있다. 대량 병렬 반응 및 다중처리를 토대로 획기적인 염기서열 분석 비용의 감소를 이뤄낸 NGS 기술은 대용량, 고효율, 저비용으로 식물 유전체 해독을 가능하게 하여, 중국배 'Danshansuli'와 유럽배 'Bartlett'에서 유전체 분석이 완료되었다. 최근 국내에서는 황금배, 청실리 및 미니배의 resequencing 및 GBS를 통한 SNP 탐색 등의 연구를 통해 화기, 숙기 당도 등 농업적으로 유용형질에 대한 게놈전체 연관분석을 수행하고 있다.

동양 배 '진황'에 있어 1-MCP 처리에 따른 다양한 온도 환경에서의 유통력 비교 (Comparison of Fruit Maketability at Various Temperature Environment in Asian Pear 'Jinhwang' Treated with 1-Methylcyclopropene)

  • 본 연구에서는 동양배 '진황'의 1-MCP처리 후 유통온도에 따른 과실의 품질 변화를 관찰하였다. 과실의 감모율의 경우 저장온도가 낮을수록 적었으며 1-MCP처리구가 무처리구 보다 유의하게 적은 감모를 보였다. 수확 후 1-MCP처리는 과육경도에서 유통온도(18, 25 및 $30^{\circ}C$)와 관계없이 20N이상으로 무처리구보다 높게 유지되었다. 과피색의 변화는 모든 처리구에서 저장기간이 길어질수록 어두워졌으며, $a^*$값은 18 및 $25^{\circ}C$에서는 처리구간 차이를 보이지 않았지만, $30^{\circ}C$에서는 1-MCP처리구가 유의하게 적은 변화를 보였다. 과실의 생리장해에서 과심갈변, 분질과 발생은 저장온도가 낮을수록 발생이 적었으며, 1-MCP 처리구가 무처리구 보다 낮은 발생을 보였다. '진황'과실에서 1-MCP처리후 유통온도에 따른 과실의 품위유지 기간은 18 및 $25^{\circ}C$에서는 28일 내외이며, $30^{\circ}C$에서는 21일 내외로 1-MCP 처리로 유통기간을 약 7일 이상의 품위유지에 효과를 보였다. 이러한 결과는 특히 고온의 시장조건에서는 조생종 '진황' 과실에 1-MCP처리가 품위유지에 효과적인 것으로 사료된다.
