Genetic Relationships of Four Korean Oysters Based on RAPD and Nuclear rDNA ITS Sequence Analyses

  • Random amplified polymorphic DNA (RAPD) marker and sequence analyses of the internal transcribed spacer (ITS) region of ribosomal DNA were used to assess phylogenetic relationships of four Korean oyster species. The average number of species-specific markers identified from five universal rice primers (URPs) by RAPD-PCR was 1.8 for Crassostrea gigas, 3.2 for C. nippona, 3.6 for C. ariakensis, and 4.6 for Ostrea denselamellosa. The length of the ITS (ITS1-5.8S-ITS2) region ranged from 1,001 to 1,206 bp (ITS1, 426-518 bp; 5.8S, 157 bp; and ITS2, 418-536 bp), while the GC content ranged from 55.5-61.1% (ITS1, 56.8-61.8%; 5.8S, 56-57.3%; and ITS2, 54.1-62.2%). A phylogenetic analysis of the oysters based on our RAPD, ITS1, and ITS2 sequence data revealed a close relationship between C. gigas and C. nippona and a distant relationship between the genera Crassostrea and Ostrea. Our results indicated that RAPD and ITS sequence analysis was a useful tool for the elucidation of phylogenetic relationships and for the selection of species-specific markers in Korean oysters.

Community structures of the benthic macrofaunal assemblages in Kyonggi Bay and Han Estuary, Korea

  • The species composition and distribution patterns of the subtidal benthic faunal assemblage in Sokmo and Yomha Channels and Kyonggi Bay were studied in spring and fall of 1989 and correlated to some environmental variables: temperature, salinity and substrate type. For both seasons, a total of 7,779 organisms were counted belonging to 244 species. Of these, 3,647 organisms in 185 species were counted for spring (March 1989) and 4,132 in 189 species for fall (September 1989). The oyster, Cressostrea gigas and the tellinidean bivalve, Moerella rutilla were the two most abundant species in the spring, constituting 17.22%and 6.47% of the total abundance respectively. While in the fall, the barnacle, Balanus reticulatus and macrura, Ogyrides orientalis dominated comprising 13.72% and 6.87 % of the total number of individuals. Community structure analysis revealed good correlations with the variations of salinity in Yomha and Sokmo Channels and sedimentary facies in Kyonggi Bay, Korea.

Tributyltin Compound in Sediments and Tissues of Oysters and Rock Shell in Gwangyang Bay, Korea

  • Tributyltin (TBT) and its degradation products, dibutyltin (DBT) and monobutyltin (MBT) were quantitatively determined in surface sediments and two molluscan species, Pacific oyster (Crassostrea gigas) and rock shell (Thais ctavigera), from Gwangyang Bay, Korea. Butyltin compounds were detectable in almost all sediment and biota samples. Tributyltin concentrations in surface sediment ranged<2∼33 ng g$\^$-1/, which is at a lower end of TBT concentrations in industrialized bays in Korea. However, TBT levels in sediments were related to boating activities around the bay. In biota samples, TBT concentrations were in the range of 178∼2,458 ng g$\^$-1/ toy oyster and 47∼236 ng g$\^$-1/ for rock shell. Relatively high TBT concentrations in biota were found near wharves for fisherboats and harbor areas. About 90∼100% of the female T. clavigera displayed imposex, and relative penis length index of the imposexed-female was in the range of 20.9∼107.9%. Furthermore, TBT body residue had a significant positive relationship with degree of imposex in T. clavigera. Overall, TBT concentrations in Gwanyang Bay were much lower than other major bays in Korea.

  • 균사생장(菌絲生長)이 부진(不進)한 사철느타리버섯 (P. florida), 느타리버섯 (P. ostreatus) 및 이들 두 종(種)의 교배균주(交配菌株)로부터 30nm 및 23nm 의 구형(求刑) virus입자(粒子)를 단일(單一) 혹은 혼합형태(混合形態)로 분리(分離)하였다. 사철 느타리버섯으로부터 분리(分離)한 virus의 최적(最適) spectrum은 259.5nm 였다.

Development and Characterization of New Microsatellite Markers for the Oyster Mushroom (Pleurotus ostreatus)

  • We developed and characterized 36 polymorphic microsatellite markers for the oyster mushroom (Pleurotus ostreatus). In total, 169 alleles were identified with an average of 4.7 alleles per locus. Values for observed ($H_o$) and expected ($H_E$) heterozygosities ranged from 0.027 to 0.946 and from 0.027 to 0.810, respectively. Nineteen loci deviated from Hardy-Weinberg equilibrium. Significant (P<0.05) excess heterozygosity was observed at nine loci. Linkage disequilibrium (LD) was significant (P<0.05) between pairs of locus alleles. Cluster analysis revealed that five species of genus Pleurotus made a distinct group, and the individual cultivars were grouped into major five groups from G-1 to G-5. The diverse cultivars of P. ostreatus were discriminated and the other four species revealed a different section in the UPGMA tree. These microsatellite markers proved to be very useful tools for genetic studies, including assessment of the diversity and population structure of P. ostreatus.

Application of the rpoS Gene for Species-Specific Detection of Vibrio vulnificus by Real-Time PCR

  • Vibrio vulnificus is a causative agent of serious diseases in humans, resulting from the contact of wound with seawater or consumption of raw seafood. Several studies aimed at detecting V. vulnificus have targeted vvh as a representative virulence toxin gene belonging to the bacterium. In this study, we targeted the rpoS gene, a general stress regulator, to detect V. vulnificus. PCR specificity was identified by amplification of 8 V. vulnificus templates and by the loss of a PCR product with 36 non-V. vulnificus strains. The PCR assay had the 273-bp fragment and the sensitivity of 10 pg DNA from V. vulnificus. SYBR Green I-based real-time PCR assay targeting the rpoS gene showed a melting temperature of approximately $84^{\circ}C$ for the V. vulnificus strains. The minimum level of detection by real-time PCR was 2 pg of purified genomic DNA, or $10^3$ V. vulnificus cells from pure cultured broth and $10^3$ cells in 1 g of oyster tissue homogenates. These data indicate that real-time PCR is a sensitive, species-specific, and rapid method for detecting this bacterium, using the rpoS gene in pure cultures and in infected oyster tissues.

Genomic Differentiation Among Oyster Mushroom Cultivars Released in Korea by URP-PCR Fingerprinting

  • URP primers of 20 mer derived from repetitive sequence of rice were used to assess genetic variation of oyster mushroom consisting of 10 cultivars of Pleurotus ostreatus, two cultivars of P. florida and two cultivars of P. sajor-caju which were registered in Korea. URP2F and URP38F primers produced cultivar-specific PCR polymorphic bands in the Pleurotus species. UPGMA cluster analysis using the URP-PCR data showed that 14 Pleurotus cultivars are genetically clustered into large three groups. The URP-PCR data provided important information for more efficient breeding strategies of Pleurotus cultivars.

Haemocytes responses of the pearl oyster, Pincdata fucata, at different temperatures

  • The effects of temperatures on pearl oyster, Pincdata fucata were studied by evaluating some functional immune responses of the haemocytes. Water temperature is one of the most important factors in bivalve immune defense. Haemocytes comprise a primary line by inflammation, encapsulation and phagocytosis. These phagocytic abilities of haemocytes were observed in different temperatures. The number of the circulating haemocytes by migratory assay, phagocytic activities by MTT assay and reactive oxygen species production of haemocytes by CL assay were measured at different temperatures. Results showed that pearl oyster maintained at 20℃ and 25℃ displayed significantly higher values for all the measured immune parameters in comparison to maintained at 10, 15, and 30℃.

느타리버섯 유해균 Hypocrea sp.의 형태 및 URP-PCR 분석 (Morphological Characteristics and URP-PCR Analysis of Hypocrea sp., a Weed Mould of Oyster Mushroom Cultivation)

  • 느타리버섯 균상에 발생하는 Hypocrea속 균의 분류학적 위치를 확립하기 위하여 유성세대 및 무성세대의 형태적 특징을 조사하였고, 국내 분리 균주의 RAPD, RFLP, URP-PCR 분석 등 분자생물학적 특징을 조사하였다. 1. 충남과 전남 일대의 8개 시, 군의 느타리 재배사에서 19 균주를 분리하였으며 이들 균주간의 형태적, 발생생태학적 변이는 없었고, 기존 보고된 Hypocrea 종들과는 다른 형태적 특징을 나타내었다. 2. Hypocrea 자좌의 크기는 $6.0{\sim}13.0{\times}3.0{\sim}11.0mm$이었고, 위유조직으로 구성되어 있으며 대부분 군생하고 처음엔 백색이지만 성숙되면 노란빛을 띤 갈색을 나타내었다. 자낭각은 구형으로 크기가 평균 $239.9{\times}204.1{\mu}m$이었으며, 자낭에는 16개의 자낭포자(part ascospores)를 형성하였고, 자낭과 장낭포자의 크기는 각각 $103.3{\times}4.9{\mu}m$$6.5{\times}4.7{\mu}m$이었다. 3. 무성세대의 phialide는 보통 Trichoderma virens와 유사한 형태로 모여서 형성되나 때때로 penicillate의 형태도 관찰되었다. T. virens형 phialide의 크기는 $10.8{\times}3.5{\mu}m$이었고 penicilliale형은 $14.7{\times}3.4{\mu}m$이었다. 분생포자는 계란형에서 타원형으로 표면은 매끈하고 크기는 $4.5{\times}2.9{\mu}m$이었다. 4. Hypocrea 균주들의 배양적 특징 조사한 결과, PDA 배지에서 균사생장 최적 온도는 $25^{\circ}C$이었고 자좌는 $15{\sim}25^{\circ}C$ 범위에서 발생하였으며 자좌 발생 최적 온도는 $20^{\circ}C$였다. 5. RAPD 및 RFLP 분석을 실시한 결과 Hypocrea속 균은 공시균주는 모두 단일 유전 개체군으로 느타리 재배사에서 발생하는 Trichoderma속 균 및 기존에 보고된 Hypocrea spp.와는 다른 분류군으로 나타났다.
