Screening of Korean Herbal Medicines with Inhibitory Activity on Advanced Glycation End Products Formation (VII) (한국약용식물의 최종당화산물 생성저해활성 검색(VII))

  • In this study, 49 Korean herbal medicines have been investigated with an in vitro evaluation system using glycation end products (AGEs) formation inhibitory activity. Of these, 18 herbal medicines ($IC_{50}$ < $50{\mu}g/ml$) were found to have significant AGEs formation inhibitory activity. Of these, five herbal medicines ($IC_{50}$ < $50{\mu}g/ml$) were found to have significant AGEs formation inhibitory activity. Particularly, Mallotus japonicus (twigs and leaves), Rhus javanica (twigs and leaves), Boehmeria nivea (whole plants), Quercus acuta (stems), and Eurya japonica (stems) showed more potent inhibitory activity (approximately 9-37 fold) than the positive control aminoguanidine ($IC_{50}=76.47{\mu}g/ml$).

Molecular analysis on the pharmacological actions of oriental medicines in stomach cancer cells (수종 한약제의 위암세포에 대한 항암작용 효능 검색 및 약리작용에 관한 분자생물학적 연구)

  • Traditional oriental medicines have been used for treatment of various kinds of human cancers for long times and some of them proven to be effective clinically. However, the pharmacological actions and mechanisms related to cancer treatment are generally unknown. In an effort to clarify the action mechanisms of several oriental medicines used for cancer treatments, we planned this experimental procedures. We selected Cordyceps sinensis (冬蟲下草), Punellae Herba (夏枯草), Rehmanniae Radix (熟地黃), Paeoniae Radix (白芍藥), Oldenlandiae Herba (白花蛇舌草), Partulaceae Herba (馬齒? ), Scdopendra subspinipes mutilans (蜈蚣), Mylabris Phalerara (班蟄), Phellinus igniarius(桑黃), Ganodermae Lignum(靈芝) for evaluation, which have been used for patients of gastric cancers. The twenty grams of medicines were boiled in 100ml of water for 1 hour and filtered with $0.2\;{\mu}m$ pore-sized filter unit to remove insoluble particles. Initially we evaluated the effects oriental medicines on growth inhibition in stomach cancer cells. The gastric cancer cell line, AGS, was cultured in RPMI 1640 supplemented with l0% heat-inactivated fetal bovine serum and treated with $10{\mu}l$ oriental medicines per 1ml of medium up to 48 hours. The specimens were subjected to MTT assay for evaluation of growth inhibition. We found mat Mylabris phalerata (班蟄) markedly suppressed the growth of cancer cells as shown in results. Next, we checked the effects of oriental medicines on cancer on cell cycles and apptosis. Mylabrls phalerata (班蟄) inhibited cell cycle progression of cancer cells a compared with control cells and cells treated with other medicines. In addition, Mylabri phalerata (班蟄) induced apoptosis in 30-40% of stomch cancer cells within 24 hours. Other oriental medicines used for this experiments did not show apoptosis-inducing effects on cancer cells. Finally, we determined the expression levels of genes associated with cell cycle and apoptosis. The expressions of Bcl-2 and bcl-XL were downregulated by the treatment of Mylabris phalerata (班蟄). However, the expression levels of genes related to cell cycles were not altered significantly. In conclusion, we found that Mylabris phalerata (班蟄) has in vivo gowth-inhibiting and apptosis-inducing effects on stomach cancer cells. However, we think that at least animal experiments are necessay for evduations.

  • All medicines are poisonous, and there are none free from poison. Thus, all things may be regarded as poisonous. And poison and medicine is the same one of which actions differ from each other. Medicine may be good medicine or poison according to doses, whether chemical medicine or herbal one. Herbal medicine falls into the category of natural materials. From ancient times humankind has Been using animals and plants as food, but on the other hand, chemical medicine is unfamiliar relatively to natural materials. Somecine is unfamilis, absorbed into the human boan, are transformed and help deemicfication. Other medicines protect the gastric mucous membrane and are demulcent, and inhibithemica materials from being absorbed. In some cases, ine is unfamilis function as deemicfiers in combination withhemica materials, and remove the toxicity and side effect caused by drug properties. Herbal medicine causes less side effects, as compared to chemical one, and thus can work effectively. As the case stands, some deem that herbal medicine does not cause problems despite high dose and long-term use because it is nontoxic or low-toxic. However, herbal medicine may be also poisonous though it is used at a very low dose. Even a deadly poison may produce therapeutic results satisfactorily without side effects, on condition that it is used properly.

The Literature Review on Procedure of Historical Changes on Herb Medicines in Chapter Sinmun Jeonkwang of 『Donguibogam』 (『동의보감(東醫寶鑑)』 신문(神門) 전광(癲狂) 처방의 전사(轉寫)에 대한 연구)

  • Objectives : The purpose of this study is to study as reference for practical application in clinics, examine the procedure of historical changes and compare components and their doses of herb medicines, which are recorded in the chapter, Sinmun Jeonkwang of "Donguibogam". Methods : I examined 19 Herb Medicines (Ed note: no need to capitalize the 'h' and 'm') in Chapter Sinmun Jeonkwang of "Donguibogam". Fourteen books, which were noted in Chapter Sinmun Jeonkwang of "Donguibogam" and 16 books in Jeonkwang part of "The Eastern Medical Textbook of neuropsychiatry" and 25 books were mentioned in same part of "Uibujeonrok", were selected as reference. Results and Conclusions : Fourteen documents were referred to the 19 Herb Medicines in Chapter Sinmun Jeonkwang of "Donguibogam". Seventeen Herb Medicines were recorded in the source book but 2 Herb Medicines were not recorded. Fourteen Herb Medicines among 19 were recorded in the source book, which were directly quoted from the firstly appeared books, and 3 Herb Medicines were re-quoted from the other books that succeeded the source books. I suspect that the components and dosages of the Herb Medicines were revised, according to the author's clinical experience and circumstances.

  • To obtain a specific pharmacological effect, herbal medicines are i)recessed based on the principle of traditional korean medicines. However, the problem in processing is the variety of methods, technics, and drug quality. In order to establish the base of standardization of processing methods, some herbal medicines were processed and studied the change of a color tone before and after processing.

Study on the Use of 'Saengsukron' in 'Tangaekpyeon of Donguibogam' (『동의보감(東醫寶鑑)·탕액편(湯液編)』에서의 생숙론(生熟論) 활용에 관한 고찰)

  • Objectives : Systematize concept of 'Saengsukron(生熟論)', providing theoretical basis for clinical and research purpose. Methods : Herbal medicines listed in 'Tangaekpyeon of Dongeuibogam' were studied in the following order ; (i) Search for herbal medicines that has applied heat. (ii) Choose herbal medicine with reference of change before and after heating. And choose herbal medicine with reference on purpose of heating. (iii) Classify herbal medicines with similar patterns. (iv) Catagorize herbal medicine by its effects, side effects, nature of herbal medicines, tastes of herbal medicines, meridian tropism of the herbal medicines, and others(color of herbal medicines, formation of herbal medicines, storage of herbal medicines) (v) Systematize concept of 'Saengsukron'. Results : We were able to obtain systematized concept of 'Saengsukron(生熟論)'. They are classified by 'Saengsasukbo(生瀉熟補)', 'Saenghwalsukji(生活熟止)', 'Saengmusukyu(生無熟有)', 'Sukjeukhyojeung(熟則效增)', 'Sukjeukhyogam(熟則效減)', 'Sukjeukyudok(熟則有毒)', 'Saengdoksukgam(生毒熟減)', 'Saengjunsukwan(生峻熟緩)', 'Saengchangsukso(生脹熟消)', 'Sukjeukwiseong(熟則爲升)', 'Sukjeukwigang(熟則爲降)', 'Sukjeukwion(熟則爲溫)', 'Sukjeukwiryang(熟則爲凉)', 'Saengjosukgam(生燥熟減)', 'Sukjeukbyeonmi(熟則變味)', 'Sukjeukbyeonchwi(熟則變臭)', 'Sukjeukbui(熟則部異)', 'Sukjeukjongsa(熟則從邪)', 'Sukjeukbyeonsaek(熟則變色)', 'Sukjeukbyeonhyeng(熟則變形)' and 'Sukjeukbojang(熟則保長)'. Conclusions : In this study, heat processing of herbal medicines confirmed that certain regular changes occur. Based on this, concept of 'Saengsukron' could be systematized. Research on 'Saengsukron' will help practitioners and researchers.

A tendency of herbal medicines on anti-viral effect (항바이러스 효과에 응용되는 한약의 연구 동향)

  • In order to find antiviral effect against Human immunodeficiency virus(HIV), Herpes simplex virus type I(HSV-1) and II(HSV-2) from herb medicines, publicated 29 paters on anti-viral effect of herbal medicines and a convenient virus-induced cytopathic effect (CEP) inhibition assay was introduced. The major virus on experiment are HIV, Hepatitis B virus and HSV-1,2. Those of other studies showed inhibition of infected virus DNA replication and screening test of herbal medicines. More than 15 extractions were prepared by pure water boiling from herbal medicines, and their toxicity of infected cell and anti-viral activities were evaluated. Among them, the major part of herbal medicines showed cell stability compared with the contrast. Cytotoxic concentration (CC) of the $H_2O$ extracts of Padoo against HIV was <4.0, Hyungbangpaedoksan against HIV was 9.3, Whangyonhaedoktang against HIV-1 and HSV-2 was 15.3. These are high level cytotoxic concentration compared with the contrast. But antiviral effect was unable to figure out for selective $index(SI)=CC_{50}/EC_{50}$. The other herbal medicines were unable to showed potent anti-HIV and anti-HSV activity. The antiviral activation using herbs in this thesis have unlimited objects, to select research object will help to show the direction of antiviral drug development that have less side effect and more excellent efficiency.

A Review of Experimental Study for Herbal Medicines of Anti-allergic Effects (항알레르기 효능을 가진 한약물의 실험 연구 결과에 대한 고찰)

  • Objective : The purpose of this study is to survey the experimental studies for Herbal Medicines of Anti-allergic Effects by reviewing oriental medicine journals. Methods : We collected 48 research papers on Oriental herbal medicines (prescriptions, herbs and aqua-acupunctures) of Anti-allergic effects in the internet site OASIS using the keyword 'Antiallergy'. Then we analyzed them according to published journals, years, and research methods, etc. Results : 48 papers in relation to Oriental herbal medicines of anti-allergic effects have been published in many kinds of journals and shows a tendency to increase every year. There are 23 cases of using herb simplexes, 21 cases of herb-combined prescription, and 4 cases of aqua-acupuncture. Conclusions : This result will provide useful information of field of Oriental herbal medicine for the development of anti-allergic related herb simplexes and herb combined prescriptions. In the future, we have to perform more studies such as clinical trial and research papers on combined medication of the oriental and the western medicines for anti-allergic effects.

Antimicrobial Effects of Ethanol Extract of Yongdamgosam-hwan against Streptococcus mutans (용담고삼환(龍膽苦參丸) 에탄올 추출물의 Streptococcus mutans에 대한 항균활성에 관한 연구)

  • Objectives : Yongdamgosam-hwan(YGH) has been used as a traditional medicine from old times for antiinflammatory effects. Streptococcus mutans(S. mutans) is known as a prime bacteria responsible for causing caries by forming a biofilm referred to as dental plaque on the tooth surface. But antimicrobial activity of YGH with dental disease is not sufficiently understood. This study was designed to investigate the effects of YGH ethanol extract on antimicrobial effect against Streptococcus mutans.Methods : The antimicrobial effect of YGH ethanol extract was assessed by the paper disk diffusion method and optical density method to determine minimum inhibition concentration(MIC), also observed by fractional inhibitory concentration index(FICI) and time-kill assay to figure out the synergic effect on the combination of YGH ethanol extract with antibiotics.Results : The YGH ethanol extract 500 μg was 7.5-8.5 mm diameter of clear zone of inhibition against Streptococcus mutans in a concentration-dependent manner and MIC was 250 μg/mL. The administration of the ethanol extract in combination with gentamicin and streptomycin induced a reduction of ≥4-8-fold in all tested bacteria. Furthermore, time-kill study was found that a combination of YGH ethanol extract with oxacillin and streptomycin produced a more rapid decrease in the concentration of bacteria CFU/mL than the YGH ethanol extract or antibiotics alone.Conclusions : As a result, the YGH ethanol extract has good antimicrobial effects. And the results suggest that YGH could be employed as a natural antibacterial agent in dental care products.
