Role of Intergenic and 3'-Proximal Noncoding Regions in Coat Protein Expression and Replication of Barley yellow dwarf virus PAV

  • Barley yellow dwarf virus PAV (BYDV-PAV) has a 5.7-kb positive-sense single-stranded RNA genome that contains six open reading frames (ORFs). BYDV-PAV produces three subgenomic RNAs (sgRNAs). The largest of which encodes the coat, 17-kDa, and readthrough proteins from two initiation codons. To investigate the role of intergenic and 3'-proximal noncoding regions (NCRs) in coat protein (CP) expression and BYDV-PAV replication, a full-length infectious cDNA of the RNA genome of an Illinois isolate of BYDV-PAV was constructed downstream of the Cauliflower mosaic virus-35S promoter. Linear DNA molecules of these cDNAs were infectious, expressed the 22-kDa CP, and produced both genomic RNA sgRNAs in ratios similar to those observed in protoplasts inoculated with viral RNA. The portion of 5'NCR of sgRNA1 between ORFs 2 and 3 was not required for, but enhanced translation of CP from ORF3. Mutants containing deletions in the NCR downstream of ORF5 failed to replicate in oat protoplasts. These results indicate that an intact 3$^1$NCR is required for BYDV-PAV replication.

Effect of Wave Load on the Member Force of Steel Structure of Floating Buildings

  • For floating buildings may fl oat on the water for a long time, they are constantly affected by various environmental loads such as wind and wave loads. In this study to find the wave effect on the floating building, five models are designed using steel moment resisting frame. It is assumed that the lower part of the floating building is a reinforced concrete pontoon, while the upper part is a three-story steel frame. To analyze floating buildings affected by wind and wave loads, hydro-dynamic and substructure analysis are performed. As input loads, this study set limits that the mean wind velocity is 35 m/s and the significant wave height is 0.5 m for the residential building. From the hydrodynamic analysis, the time-history acceleration of building is obtained and transformed into a base ground input for a substructure analysis of the superstructure of the building. Finally the mean of the maximum from 30 dynamic analysis of the floating buildings are used to be compared with the results of the same model on the ground. It was shown that the dynamic results with wind and wave loads are not always lesser than the static results which are calculated with static equivalent wind load for a building that is located on the ground.

Effect of Abiotic Factors on Fumosorinone Production from Cordyceps fumosorosea via Solid-State Fermentation

  • Cordyceps fumosorosea is an important species in the genus of Cordyceps, containing a variety of bioactive compounds, including fumosorinone (FU). This study was a ground-breaking assessment of FU levels in liquid and solid cultures. The present study focused on the impacts of solid-state fermentation (SSF) using solid substrates (wheat, oat, and rice), as well as the effects of fermentation parameters (pH, temperature, and incubation period), on the generation of FU. All the fermentation parameters had significant effects on the synthesis of FU. In a study of 25 ℃, 5.5 pH, and 21 days of incubation period combinations calculated -to give maximal FU production, it was found that the optimal values were 25 ℃, 5.5 pH, and 21 days, respectively. In a solid substrate medium culture, FU could be produced from SSF. At 30 days, a medium composed of rice yielded the most FU (798.50 mg/L), followed by a medium composed of wheat and oats (640.50 and 450.50 mg/L), respectively. An efficient method for increasing FU production on a large scale could be found in this approach. The results of this study might have multiple applications in different industrial fermentation processes.

Influence of Companion Planting on Microbial Compositions and Their Symbiotic Network in Pepper Continuous Cropping Soil

  • Continuous cropping obstacles have become a serious factor restricting sustainable development in modern agriculture, while companion planting is one of the most common and effective methods for solving this problem. Here, we monitored the effects of companion planting on soil fertility and the microbial community distribution pattern in pepper monoculture and companion plantings. Soil microbial communities were analyzed using high-throughput sequencing technology. Companion plants included garlic (T1), oat (T2), cabbage (T3), celery (T4), and white clover (T5). The results showed that compared with the monoculture system, companion planting significantly increased the activities of soil urease (except for T5) and sucrase, but decreased catalase activity. In addition, T2 significantly improved microbial diversity (Shannon index) while T1 resulted in a decrease of bacterial OTUs and an increase of fungal OTUs. Companion planting also significantly changed soil microbial community structures and compositions. Correlation analysis showed that soil enzyme activities were closely correlated with bacterial and fungal community structures. Moreover, the companion system weakened the complexity of microbial networks. These findings indicated that companion plants can provide nutrition to microbes and weaken the competition among them, which offers a theoretical basis and data for further research into methods for reducing continuous cropping obstacles in agriculture.

Comparison of Breeding and Cultural Contribution to Yield Gains of Korean Rice

  • Analysis of breeding gains in grain yield has been intensively conducted in wheat, barley, oat, maize, and soybean. Such information is limited in rice. The objective of this study was to compare the breeding gains and cultural gains contributed to yield gains of Korean rice varieties since early 1900s. Two sets of yield data were used for analysis; the historical yield data of 1908 for old japonica cultivars, and present yield data in the years from 1996 to 1998 for the six cultivars, consisting of previous two old cultivars and four contemporary cultivars. The old cultivars were two native cultivars, Jodongi and Damageum, while contemporary cultivars were two premium quality japonica cultivars, Hwaseongbyeo and Dongjinbyeo, and two Tongil-type cultivars, high yielding cultivars developed from indica/japonica hybridization, Milyang23 and Dasanbyeo. The yield differences of old cultivars between the experiments in 1908 and the experiments from 1996 to 1998 were estimated as cultural gains (1.84 tons $\textrm{ha}^{-1}$) due to the improvement of cultivation technology. Yield differences between the old cultivars and contemporary cultivars were considered total yield gains during the periods. These were 2.51 tons $\textrm{ha}^{-1}$ for japonica cultivars and 3.81 tons $\textrm{ha}^{-1}$ for Tongil-type cultivars. From these data, the genetic gain of 0.67 tons $\textrm{ha}^{-1}$ and 1.97 tons $\textrm{ha}^{-1}$ were estimated for japonica cultivars and Tongil-type cultivars respectively. The ratio between cultural gain and genetic gain appeared to be 2.7:1 for japonica cultivars and 1:1 for Tongil-type cultivars. This analysis clearly showed the higher genetic contribution in Tongil-type cultivars than in japonica cultivars, suggesting a guideline to be used when planning new yield improvement programs. Additional implication has emerged when a better yield response to modem cultivation technology was found in one of the old cultivars, suggesting the combined improvement between breeding and cultural improvement is necessary for attaining the maximum yield capacity of a crop.

Prospect of new variety breeding of Italian ryegrass in South Korea

  • The objective of this study was to know prospect and present state of new variety breeding of Forage and Grassland in South Korea. The industry of forage in Korea is going up to expend through utilization of good quality forage, Italian ryegrass, forage corn and oat rather than rice straw. Especially, since 2007, Italian ryegrass(IRG) had been very important major winter forage crop in South Korea and developed 13 varieties including very early maturity variety (three varieties), early maturity variety (three varieties), medium maturity variety (one variety) and late maturity variety (six varieties). But the disadvantage of Italian ryegrass was weak winter hardness and drought but has good advantage of forage nutritive value, high-yielding and high sugar content, and like livestock as like hanwoo, dairy cattle, goat so on. The Ko-variety (Korea developed variety) of Italian ryegrass has high cold-tolerant and adaptability more than any other country developed variety, and expend to cultivation area from southern area (below Daejeon) to middle-northern area (upper Han river). Although the cultivation area of Italian ryegrass of South Korea was 21,700 ha in 2007, right now, that of Italian ryegrass is about 123,600ha due to expend cultivation area and know famer to good forage crop and have a various maturity varieties (very early, early, medium, late) according to local situation (before-crop harvesting stage or double cropping system). The seed market of Italian ryegrass in South Korea becoming extended to around 100 million Korean won and seed export get nearer to foreign country. We are going to develop of new variety for stress tolerant and high yield and quality forage variety, good adaptability to the Korean environmental conditions including reclaimed area, make self-sufficiency system for forage seed (Italian ryegrass), export our seed to foreign countries.

주재료에 따른 조선시대 떡류의 문헌적 고찰 (A Literature Review on the Types of Joseon Dynasty Tteok (Korean Rice Cake) according to its Main Ingredient)

  • This article examines the types of tteok (Korean rice cake) recorded in 21 old literatures of the Joseon dynasty (1392-1909) according to its main ingredients. Tteok varieties of the Joseon dynasty were categorized into jjin-tteok, chin-tteok, jijin-tteok, salmeun-tteok and guun-tteok and their changes in cooking method were discussed from the early to late eras of the Joseon dynasty. These can be summarized as follows. In the early, middle and late eras of the Joseon dynasty, there were 1, 15 and 84 kinds of tteok using non-glutinous rice as the main ingredient, and 6, 24 and 120 kinds using glutinous rice, respectively. Tteok using wheat flour was not found in the early Joseon dynasty, whereas 6 and 32 kinds were found in the middle and late eras, respectively. There were 1, 4 and 5 kinds of tteoks using buckwheat, and 5, 11 and 19 kinds using other ingredients such as yam, barley, elephant's ear, oat, and arrowroot flour, in the early, middle and late eras, respectively. The frequency of the main ingredient increased in the order of glutinous rice>non-glutinous rice>wheat flour>other ingredient>buckwheat flour during the Joseon dynasty and the ratio of tteoks using non-glutinous and glutinous rice flours was 1:1.5. The number and types of tteok were noted to increase abruptly throughout the Joseon dynasty. This may be associated with the commercial industrial development that prevailed in the late Joseon dynasty. Further study will be conducted on their recipes and ingredients recorded in these old literatures to develop a standardized recipe for the globalization of tteok.

수입건초의 품질 및 기호성에 관한 연구 (Studies on the Quality and Palatability of Imported Hay and Straw)

  • 본 연구는 2008년 1월부터 12월까지 충남대 학교 농업생명과학대학과 가축유전자원시험장에서 수행하였다. 공시가축은 유산양(Sannen, Capra aegagrus) 12두(female, $30{\pm}1.8kg$), 재래산양(Korean native goats, Capra hircus) 12두(female, $24{\pm}2.4kg$), 꽃사슴(sika deer, Servus nippon) 5두(female, $92{\pm}5.2kg$)를 공시하였다. 공시재료는 2006년에 미국에서 수입된 annual ryegrass straw, perennial ryegrass straw, tall fescue straw 및 Kentucky bluegrass straw 등 4종과, 2007년에 미국에서 수입된 alfalfa hay, bermudagrass hay, timothy hay, klein grass hay, oat hay 및 orchardgrass hay 등 6종 등 총 10종과 대조구로 국내산 혼합 목건초(mixed hay) 등 총 11종을 공시하였으며 얻어진 결과는 다음과 같다. 수입건초는 종류에 따라 화학적 성분과 건물소화율(in vitro digestibility, IVDMD)는 현저한 차이를 보였으며 (p<0.05), alfalfa hay와 orchard-grass hay를 제외하고는 대부분의 수입건초는 대조구에 비하여 조단백질(crude protein, CP)함량과 IVDMD가 낮은 반면에, NDF와 ADF 같은 섬유소물질의 함량은 상당히 높은 결과를 나타내었다. RFV(relative feed value)는 수입건초의 종류에 따라 차이를 보였으며, 대부분의 수입건초의 RFV는 $4{\sim}5$등급에 해당되었다. 유산양, 재래산양 및 꽃사슴에 의한 채식량과 기호성은 수입건초의 종류에 따라 현저한 차이를 나타내었으며, 공통적으로 orchardgrass hay와 bermudagrass hay가 높았던 반면에, Kentucky bluegrass straw, tall fescue straw, annual ryegrass straw 및 perennial ryegrass straw 등은 채식량과 기호성 순위가 상당히 낮은 것으로 밝혀졌다. 이상의 결과로 보아, 공시된 수입건초는 종류에 따라 품질과 기호성이 현저한 차이를 보이고 있어 수입건초의 품질평가에 대한 기준설정이 중요하다고 하겠다.

식이섬유와 콜레스테롤 대사 (Dietary Fiber and Cholesterol Metabolism)

  • 우리나라 주요 사망원인중의 하나가 되고 있는 혈관질환의 주된 위험인자로 고콜레스테롤 혈증이 지적되고 있다. 콜레스테롤 저하효과가 있는 식이 인자에는 단백질의 종류, 지방의 종류와 양, 식이섬유, 우유, 칼슘, flavonoid 등이 있다. 특히 수용성 식이 섬유의 콜레스테롤 저하효과는 현저하며 동물실험과 인체실험을 통하여 확인되었다. 혈장 콜레스테롤 저하효과가 현저한 식이섬유에는 psyllium husk, pectin, oat fiber, guar gum, sodium alginate 등이 있으며, 농도에 비례하여 그 효과가 커지는 것은 아닌듯하다. 수용성 식이섬유의 콜레스테롤 저하효과에 대한 정확한 기작은 알려져 있지 않으나 몇 가지 가설이 제시되고 있다. 첫째, 식이섬유가 장에서 점성용액을 형성하여 지질 흡수를 저해함으로써 혈장과 간 콜레스테롤 농도를 낮춘다는 것이며, 둘째, 체내 콜레스테롤이 체외로 배설되는 유일한 경로인 담즙산의 소장흡수를 방해하고 변으로의 배설을 증가시켜 체내 콜레스테롤 pool 크기를 감소시킨다는 것이다. 세째, 대장미생물에 의해 생성된 식이섬유의 발효부산물인 short chain fatty acids가 콜레스테롤의 합성을 저해한다는 것이다. 이외에도 여러 가설들이 제시되고 있으며, 이러한 가설들이 서로 상호 배제적인 것은 아니며, 단독으로 혹은 복합적으로 작용하는 것으로 보인다.

림프절의 전이성 소세포암종의 세침흡인 세포학적 소견 - 악성 림프종과의 감별을 중심으로 5예 분석 - (Fine Needle Aspiration Cytology of Metastatic Small Cell Carcinoma of Lymph Nodes - Comparison to Non-Hodgkin's Lymphoma on 5 Cases -)

  • Small cell carcinoma of the lung is characterized by cells with finely stippled chromatin and scanty cytoplasm as well as a particularly aggressive clinical course and favorable response to the chemotherapy. Recently percutaneous fine needle aspiration (FNA) biopsy has become both widely established and highly respected for the diagnosis of lung cancer. However metastatic small cell carcinoma of lymph node should be cytologically differentiated from the small round cell tumor of particular sites, especially malignant lymphoma, because small ceil carcinoma of classic oat cell type nay simulate small cell non-Hodgkin's lymphoma. We report five cases of metastatic small cell carcinoma of in-termediate cell type diagnosed by FNA of the enlarged lymph nodes of the neck and axilla. The cytologic smears contained diffuse small neoplastic cells larger than lymphocytes with dense, pyknotic nuclei and extremely scanty cytoplasm. Apparently viable large tumor cells have vesicular nuclei with granular, sometimes very coarse chromatin. The characteristic cytologic features of small cell carcinoma as compared to malignant lymphoma were as follows.: 1) small cells with dense pyknotic nuclei are evenly distributed in the background of apparently viable larger tumor cells, admixed with mature lymphocytes and phagocytic macrophages. 2) small loose aggregates of cells with nuclear melding are indicative of small cell carcinoma rather than non-Hodgkin's lymphoma. 3) the cytoplasmic and nuclear fragments of tumor necrosis are more dominant in the smears of small cell carcinoma. 4) nuclear membrane and nucleoli are generally indistinct in small cell carcinoma due to condensation of chromatin.
