The dinoflagellate genus Pelagodinium is genetically classified in distinct sub-clades and subgroups. However, it is difficult to determine whether this genetic diversity represents intra- or interspecific divergence within the genus since only the morphology of the type strain of the genus Pelagodinium, Pelagodinium bei, is available. An isolate associated with the genus Pelagodinium from Shiwha Bay, Korea, was recently cultured. This isolate was clustered with 3 to 4 strains from the Atlantic Ocean, Mediterranean Sea, and Indian Ocean. This cluster was distinct from the subgroup more closely associated with P. bei. The morphology of the isolate was analyzed using optical and scanning electron microscopy and was almost identical to that of P. bei except that this isolate had two series of amphiesmal vesicles (AVs) in the cingulum, unlike P. bei that has one series. When the pigment compositions of the isolate and P. bei were analyzed using high-performance liquid chromatography, these two strains had peridinin as a major accessory pigment and their pigment compositions were almost identical. In addition, the swimming behaviors of these two strains were very similar. The reexamination of the type culture of P. bei revealed two series in the cingulum as for the isolate. The new findings on the number of series of AVs in the cingulum, the pigment composition, and the swimming behaviors suggest that P. bei and the isolate are conspecific despite their genetic divergence. This study provides a basis to further understand the molecular classification within Pelagodinium combining genetic, morphological, pigment, and behavioral data.
Fusarium을 간단한 방법으로 DNA를 추출한 후, 유전적 다형성을 검출하는 방법인 RAPD(Randomly Amplified Polymorphic DNA)를 이용하여 Fusarium균의 유전적 다형성과 계통분류학적 분석을 실시하였다. 20개의 분리균을 배양하여 얻어진 균사체로부터 genomic DNA를 추출하여 PCR을 실시 15개의 primer에서 $0.2{\sim}3\;Kb$ 크기의 band를 총 180개 얻었다. 180개의 band 중 다형화 현상을 보이는 band는 126개로 이를 이용하여 bionominal matrix code(0,1)을 작성한 후 UPGMA법을 이용하여 각 균주간의 관계를 분석하였다. Fusarium oxysporum 균주중 355번(F. oxysporum)과 358번(F. oxysporum)은 0.9603의 높은 상동성을 보인 반면, F. roseum (87번)과 F. oxyspoyum (358번), 69번 (F. o. f. sp. lycopersici)과 68번(F. o. f. sp. melongena)은 0.3809의 낮은 상동성을 보였다. 361번 (F. oxysporum)과 218번 (F. o. f. sp. ruphani)은 0.8730의 유사도를 354번(F. oxysporum), 228번(Fusarium sp.)은 0.7936의 유사도, 87번(F. roseum)과 57번(F. o. f. sp. raphani)은 0.5873 유사도를 가진다.
Two groups of phytoplasma were identified in chrysanthemum(Dendranthema grandiflorum) cv. Chunkwang showing distinct symptoms. Isolate Ph-ch1 showed symptoms of dwarf, witches'-broom, rosette and root death. The other isolate, Ph-ch2, revealed symptoms of dwarf, yellowing, leaf cupping, vein clearing and root death. The presence of phytoplasma structures in chrysanthemum leaf tissue was confirmed by transmission electron microscopy. The 16S rRNA gene was amplified from isolates Ph-ch1 and Ph-ch2 by PCR and cloned, and the nucleotide sequences were determined. In RFLP analysis, isolate Ph-ch2 showed profiles identical to Ph-ch1, except with restriction enzymes HhaI and MseI. The sequence data showed that isolate Ph-ch1 was most closely related to the aster yellows (AY) phytoplasma, and isolate Ph-ch2 was more closely related to stolbur phytoplasma than to AY phytoplasma. This is the first reported observation of stolbur phytoplasma in chrysanthemum species.
연구목적: $Isolate^{(R)}$ gradient와 swim-up 방법이 정자의 형상 및 정밀정자형태 (strict morphology)에 미치는 영창을 비교분석하고, 이러한 정자처리방법이 정자의 동결-융해과정에 미치는 영향을 비교하고자 하였다. 연구재료 및 방법: 20명의 정상 정자를 대상으로 하였으며 각각의 정자는 두 가지 정자처리방법으로 나누어 정자의 형상과 정밀정자형태를 컴퓨터를 이용한 정자자동분석기를 통하여 측정하였고, 동결보호제의는 TYB 용액을 사용하였으며, 동결 및 융해는 cryo Magic사의 기계를 사용하였다. 통계는 SPSS PC+(version7.0)를 이용하였으며 통계학적인 유의성은 p<0.05로 하였다. 결과: 정자의 농도는 $Isolate^{(R)}$ gradient 처리군이 swim-up 처리군보다 유의성 있게 높았으나 ($51.2{\pm}40.1,\;156.6{\pm}64.3$), 운동성 VCL, VSL, VAP, Linearity, 및 ALH는 swim-up 처리군에서 유의성 있게 높았다. 정밀 정자형태는 swim-up 처리군과 $Isolate^{(R)}$ gradient 처리군에서 차이가 없었다 ($53.7{\pm}6.8$ vs $50.3{\pm}9.1%$). 동결-융해과정 중 두 가지 정자 처리군에서 정자의 형상들은 swim-up 처리군에서 전반적으로 높은 양상을 보였으나, 정밀정자형태는 $Isolate^{(R)}$ gradient 처리군이 swim-up 처리군 보다 감소율이 컸지만 두 군간에 유의한 차이는 없었다 ($12.8{\pm}8.5$ vs $8.6{\pm}6.6$). 결론: 정상 정자에서 swim-up 방법이 $Isolate^{(R)}$ gradient 방법보다 정자 회수율은 우수하였으나, 동결-융해과정 중 정밀정자형태에는 차이가 없어 두 방법을 상호보완적으로 사용할 수 있을 것으로 사료된다.

This study reports the findings of a distinct Potato virus Y (PVY) isolate found in Northeast China. One hundred and ten samples (leaves and tubers) were collected from potato plants showing mosaic symptoms around the city of Harbin in Heilongjiang province of China. The collected tubers were planted and let to grow in a greenhouse. New potato plants generated from these tubers showed similar symptoms, except for one plant. Subsequent serological analyses revealed PVY as the causing agent of the disease. A novel PVY isolate (referred to as HLJ-C-44 in this study) was isolated from this sample showing unique mild mosaic and crisped leaf margin symptoms. The complete genome of this isolate was analyzed and determined. The results showed that HLJ-C-44 is a typical PVY isolate. Phylogenetic analysis indicated that this isolate belongs to the N-Wi strain group of PVY recombinants ($PVY^{N-Wi}$) and also shared the highest overall sequence identity (nucleotide and amino acid) with other members of this strain group. However, recombination analysis of isolate HLJ-C-44 revealed a recombination pattern that differed from that of other $PVY^{N-Wi}$ isolates. Moreover, biological assays in four different potato cultivars and in Nicotiana tabacum also revealed a different phenotypic response than that of a typical $PVY^{N-Wi}$ isolate. This data, combined, suggest that HLJ-C-44 is a novel PVY recombinant with distinct biological properties.
The effect of dietary protein on growth and lipid levels of plasma and liver was studied in weanling male rats fed diets differing protein sources and amino acid balance. Rats were devided into 9 experimental diets which were grouped into 3 categories ; 1) Simple protein category includes gluten-, soy protein isolate-, and casein-containing diet groups, 2) Supplemented category includes casein supplemented with methionine, soy protein isolate supplemented with methionine, and gluten supplemented with lysine and methionine, 3) Mixed protein category includes diet groups containing gluten (2/3), casein (1/3), soy protein isolate (2/3) and casein (1/3), and casein (1/3), soy protein isolate (1/3) and gluten (1/3). The experimental diets composed of 15% protein, 65.8% carbohydrate, 10% fat and 1% cholesterol. The body wt. gain and P.E.R. were greater in rats of supplemented and mixed protein groups than simple protein groups. No statistical differences were found in plasma cholesterol among gluten, soy protein isolate and casein groups. Consumption of diets supplemented with limiting amino acid to gluten or soy protein isolate reduced the plasma cholesterol level by 23.2% and 34.2% respectively. However there was no difference between casein and the supplemented casein groups. The mixed protein groups shows relatively high plasma cholesterol concentration and low liver cholesterol levels. On the other hand gluten group showed low plasma cholesterol and high liver cholesterol levels, which means body cholesterol pool may not have been changed by the dietary protein. Feeding soy protein meal and the supplemented soy protein isolate resulted in lower plasma cholesterol, plasma triglycerides, liver cholesterol and liver triglycerides levels. This hypolipidemic effect is considered to see unique to soy protein isolate. Rats in gluten and the supplemented gluten groups showed lower plasma protein levels and a tendency of fatty liver.
This study was carried out to investigate the functional properties such as nitrogen solubility, emulsifying property , foaming capapcity , water and oil absorption of Camellia (Camellia japonica .) seed protein isolate in condition of distilled water and 0.5M NaCl solution at pH 2.0∼10.0. Nitrogen solubility of Camellia protein isolate in distilled water showed the minimum value at pH 4.0 and increased at pH lower or higher than the isoelectric point(pH 4.0). It was 90.0 %at pH 10.0 Nitrogen solubility of 0.5M NaCl solution showed a similar pattern with that of distrille dwater but was higher than that of distilled water except pH 2.0 and pH 10.0. Emulsifying activity of Camellia seed protein islate showed the minimum value at pH 4.0, but was higher at ether value of pH. Emulsifying stability of protein isolate was stable by heat treatment for 30min, at 80℃ and increased in 0.5M NaCl solution more than that of distille dwater. Foaming capacity of Camellia seed protein isolate in distill3ed water showed the minimum value near the isoelectric point, While it changed little at other values of pH. Foaming stability slowly decreased as, but didn't make a significant difference as time was delayed . Oil absorption was 1.4ml per a sample of 1g and water absorption was 0.9ml per a sample of 1g. The former was higher than the latter . The content of total amino acid of Camellia protein isolate was 43.67% and the major total amino acid of Camellia protein isolate was 43.67% and the major total amino acid was in the order of glutamic acid , arginine, aspartic acid, and leucine.
This study reports a cyanide resistant and/or tolerant fungus, isolated from the rhizosphere of Zea mays contaminated with cyanide-based pesticides. The isolate was characterised using molecular biology. The effect of free cyanide and heavy metals on the growth of isolate in a synthetic gold mine wastewater was examined. The molecular analyses identified the isolate as Fusarium oxysporum EKT01/02 (KU985430/KU985431). The isolate had a free cyanide degradation efficiency of 77.6%. The results indicated greater growth impairment in culture containing Arsenic (optical density 1.28 and 1.458) and cyanide (optical density 1.315 and 1.385). Higher growth was observed in all cultures supplemented with extracellular polymeric substance. This study showed that the isolate possesses wide substrate utilisation mechanism that could be deployed in environmental engineering applications.
The first human case with trichinellosis was reported in 1964 in Tibet, China. However, up to the present, the etiological agent of trichinellosis has been unclear. The aim of this study was to identify a Tibet Trichinella isolate at a species level by PCR-based methods. Multiplex PCR revealed amplicon of the expected size (173 bp) for Trichinella spiralis in assays containing larval DNA from Tibet Trichinella isolate from a naturally infected pig. The Tibet Trichinella isolate was also identified by PCR amplification of the 5S ribosomal DNA intergenic spacer region (5S ISR) and mitochondrial largesubunit ribosomal RNA (mt-lsrDNA) gene sequences. The results showed that 2 DNA fragments (749 bp and 445 bp) of the Tibet Trichinella isolate were identical to that of the reference isolates of T. spiralis. The Tibet Trichinella isolate might be classifiable to T. spiralis. This is the first report on T. spiralis in southwestern China.
For wastewater treatment and utilization of the biomass, a photosynthetic bacterium was isolated based on its cell growth rate, cell mass, and assimilating ability of organic acids. The isolate was a Gram-negative rod-shaped bacterium that contained a single polar flagellum and formed a lamellar intracytoplasmic membrane (ICM) system, including bacteriochlorophyll $\alpha$. The major isoprenoid quinone component was identified as ubiquinone Q-10, and the fatty acid composition was characterized as to contain relatively large amount of C-16:0 (18.74%) and C-18:1 (59.23%). Based on its morphology, phototrophic properties, quinone component, and fatty acid composition, the isolate appeared to be closely related to the Rhodopseudomonas subgroup of purple nonsulfur bacteria. A phylogenetic analysis of the isolate using its 16S rRNA gene sequence data also supported the phenotypic findings, and classified the isolate closely related to Rhodopseudomonas palustris. Accordingly, the nomenclature of the isolate was proposed as Rhodopseudomonas palustris KUGB306. A bench-scale photosynthetic bacteria (PSB) reactor using the isolate was designed and operated for the treatment of soybean curd wastewater.
