The Nutritional Components of Buckwheat Flours and Physicochemical Properties of Freeze-dried Buckwheat Noodles (메밀의 영양성분과 냉동건조 막국수의 이화학적 성질)

  • To investigate nutritional and biochemical values of buckwheat, amino acids and minerals in buckwheat harvested from Kangweon-do were analysed. Mixed flour between buckwheat and wheat were made to be used for buckwheat noodle and were also analysed minerals and major nutrients in composite flours. When three different levels of flour mix were compared, major nutrients were about same among those mixes but minerals were higher by adding more buckwheat flour, especially by adding more imported buckwheat flour. From this mixed flour, 9 minerals and 16 amino acids including 9 essential amino acids were analysed. Contents of selenium, sodium and magnessium were very high in buckwheat flour, Therefore, nutritional value of domestic buckwheat was highly evaluated. BAP method was used to determine the change of gelatinization in the noodles prepared by extruder at 8$0^{\circ}C$, and in the noodles during freeze drying and refrigeration. 70% of gelatinization was done during noolding process and retrogradation was severe during refrigeration.

Current Research Status on the Development of Genetically Modified Plants in Korea (유전자변형식물의 국내 연구 현황)

  • In an attempt to evaluate the current research status of genetically modified (GM) plants, the scientific research publications in Korea as well as in international SCI journals were screened. About 190 research articles related to the development of GM plants were searched from 10 different domestic journals in the last 12 years (Jan. 1990 to Sept. 2002), The researches in 65 articles were carried out with tobacco plant, 20 with rice, 19 with potatoes, and less then 9 articles from each other plant species, respectively, In total, 38 different plant species were being subjected for the development of GM plants. In particular, there was only one article for each major staple grains such as wheat, barley, soybean, and maize. In more than 47% of total published articles, scientists mainly focused on the basic research such as developing transformation system (46 articles), gene expression study in transgenic plants (34), and vector constructions (10). In addition, 28 articles which main authors are Korean scientists were searched from 11 different international SCI journals. Again, major plants for GM research were tobacco (10) and rice (7). More than 50% of published articles were focused on the basic research, gene expression study with transgenic plants (16). The publications on the research of disease-resistant plants were 7 articles, 3 for the development of stress-resistant and 2 for the herbicide-resistant plants, respectively. It is believed that the last 10 year's investment through government organizations has just strengthen the capacity for the next big stride on agricultural biotechnology in Korea.

Xylanase Production from Bacillus safensis Isolate by Xylan or Xylan Hydrolyzed Products (Xylan과 Xylan 가수분해물에 의한 Bacillus safensis 분리균의 Xylanase 생산)

  • A bacterial strain capable of hydrolyzing xylan was isolated from fermented soybean paste obtained from a domestic Buddhist temple, using enrichment culture with rice straw as a carbon source. The isolate, named YB-1301, was identified as Bacillus safensis on the basis of its DNA gyrase subunit B gene (gyrB) sequence. The xylanase productivity of strain YB-1301 was drastically increased when it was grown in the presence of wheat bran or various xylans. In particular, the maximum xylanase productivity reached above 340 U/ml in the culture filtrate from LB broth supplemented with only birchwood xylan at shake-flask level. The xylanase production was significantly induced by xylans at the stationary growth phase in LB medium containing xylan, whereas only a small amount of xylanase was constitutively produced from cells grown in LB medium with no addition of xylan. Furthermore, xylanase biosynthesis was induced more rapidly by the enzymatically hydrolyzed products of xylan than by the non-hydrolyzed xylan. In addition, the xylanase in the culture filtrate of B. safensis YB-1301 was found to have optimal activity at 55℃ and pH 6.5–7.0.

Analysis of Correlation Between Wonhyo Tunnel(section of KTX line) Works and Swamp (경부고속철도 천성산구간 원효터널공사와 늪지와의 상관성 분석)

  • The Wonhyo Tunnel on KTX railroad line is a section of latest concerns from domestic environmental NGOs, which focus on potential destruction of ecosystem or the like due to ever-depleted swamp water at about 300m upward from the tunnel under construction. As a result of study, out of all swamps in the vicinity of the tunnel, it was found that Mujechi 1st and 2nd swamps have been getting smaller in their area little by little since 50 years ago primarily under the influence of eroded streams around lower swamp and even ever-increasing annual mean temperature. As the result of monitoring about swamp before work, it was found that swamp water depends absolutely on amount of rainfall. Besides, the results of monitoring during work also didn't show any leakage generated in the tunnel during and after excavation works with regard to a wheat field swamp in the most vicinity of the tunnel (80m away). On the other hand, it was found that the range affected by ground water sink in tunnel section without grouting process amounted to about 100m around the tunnel, which indicates that such ground water sink has no significant impact upon most of swamps near the tunnel. As the result of testing by two well tracer test around swamps, it was noted that swamp water didn't run out from the bottom of swamp even with adjacent ground water level sunk in factitious ways. And the results of physical survey showed that swamp kept saturated even in dry season when ground water level becomes lower than the bottom of swamp. Therefore, even supposing that ground water level becomes sunk due to tunnel works, it is estimated that the water level of swamps would be still kept owing to impervious layer(peat beds).

Improvement in Seasonal Prediction of Precipitation and Drought over the United States Based on Regional Climate Model Using Empirical Quantile Mapping (경험적 분위사상법을 이용한 지역기후모형 기반 미국 강수 및 가뭄의 계절 예측 성능 개선)

  • The United States has been known as the world's major producer of crops such as wheat, corn, and soybeans. Therefore, using meteorological long-term forecast data to project reliable crop yields in the United States is important for planning domestic food policies. The current study is part of an effort to improve the seasonal predictability of regional-scale precipitation across the United States for estimating crop production in the country. For the purpose, a dynamic downscaling method using Weather Research and Forecasting (WRF) model is utilized. The WRF simulation covers the crop-growing period (March to October) during 2000-2020. The initial and lateral boundary conditions of WRF are derived from the Pusan National University Coupled General Circulation Model (PNU CGCM), a participant model of Asia-Pacific Economic Cooperation Climate Center (APCC) Long-Term Multi-Model Ensemble Prediction System. For bias correction of downscaled daily precipitation, empirical quantile mapping (EQM) is applied. The downscaled data set without and with correction are called WRF_UC and WRF_C, respectively. In terms of mean precipitation, the EQM effectively reduces the wet biases over most of the United States and improves the spatial correlation coefficient with observation. The daily precipitation of WRF_C shows the better performance in terms of frequency and extreme precipitation intensity compared to WRF_UC. In addition, WRF_C shows a more reasonable performance in predicting drought frequency according to intensity than WRF_UC.

Physical Properties of Pan Bread Made with Various Amounts of Squeezed Danggui Leaf (Angelica acutiloba Kitagawa) Juice (당귀잎 착즙액을 이용하여 제조한 식빵의 물리적 특성)

  • Danggui leaf (Angelica acutiloba Kitagawa) has numerous dietary fiber and healthy functional properties such as anti-inflammatory activity. However, it is usually discarded after its roots are harvested. For wide application of danggui leaf, squeezed danggui leaf juice was used for making pan bread. Treatments included imported wheat flour (IMWF) and domestic wheat flour (DOWF) as a control, 1% soluble solid contents of squeezed danggui leaf juice instead of wheat flour (1% SDLJ), 2% SDLJ, 3% SDLJ, and 4% SDLJ. In the fermentation expansion, dough volume was not significantly different between DOWF and 1% SDLJ, whereas it decreased according to increased amount of squeezed danggui leaf juice. Regarding physical properties, springiness and cohesiveness decreased according to increased amount of squeezed danggui leaf juice and longer storage period. Gumminess, brittleness, and hardness increased according to increased amount of squeezed danggui leaf juice, although these were not statistically different among IMWF, DOWF, and 1% SDLJ after making pan bread. Principal component analysis (PCA) was performed to assess the correlation between storage period and physical properties. In the PCA, variance proportion of principal component (PC) 1 was 0.87 while that of PC 2 was 0.10. Further, 1% SDLJ showed similar results as DOWF and INWF after making pan bread and after 3 days of storage. In conclusion, use of 1% SDLJ is desirable for making squeezed danggui leaf juice.

Effect of moisture content on some physical properties of domestic wheat (함수율에 따른 우리밀의 물리적 특성)

  • The physical properties of wheat kernels were determined as a function of moisture content to optimize the design of equipment for post-harvest management. Several properties, including bulk density, dynamic repose angle, one thousand-kernel weight, specific gravity, length, width, thickness, and area of two wheat cultivars (Baekjjung: B and Jogyeong: J ), were studied in the moisture range from approximately 9% to 30% wet basis. As moisture increased, length increased linearly from 6.42 to 7.20 mm (B) and 8.71 to 9.15 mm (J), width increased from 2.90 to 3.49 mm (B) and 4.12 to 4.43 mm (J), thickness from 2.94 to 3.20 mm (B) and 3.29 to 3.63 mm (J), and area from 14.13 to $19.44mm^2$ (B) and 27.75 to $31.25mm^2$ (J). Additionally, the dynamic repose angle and one thousand-kernel weight increased linearly from $46.3^{\circ}$ to $54.0^{\circ}$ (B) and $46.3^{\circ}$ to $54.5^{\circ}$ (J) and from 32.26 to 41.51 g (B) and 45.30 to 63.07 g (J), respectively, as the moisture content increased. Based on the experimental measurements, only the bulk density and specific gravity decreased from 754.0 to $664.1kgm^{-3}$ (B) and 776.1 to $660.0kgm^{-3}$ (J) and from 1.2950 to 1.2265 (B) and 1.3379 to 1.2671 (J), respectively, as moisture content increased.

Antifungal activities of coumarins isolated from Angelica gigas and Angelica dahurica against Plant pathogenic fungi (당귀와 백지로부터 분리한 Coumarin계 물질들의 식물병원균에 대한 항균활성)

  • In order to search potent antifungal substances from domestic plants, 40 plants cultivated in Korea were collected. After extracting with methanol (MeOH) and concentrating to dryness, the MeOH extracts were screened for in vivo antifungal activity against six plant diseases at a concentration of $2000{\mu}g/mL$. Fourteen extracts showed disease-controlling activity more than 90% against at least one of the 6 plant diseases tested; eight, seven, and three extracts controlled more than 90% the development of rice blast, tomato late blight, and wheat leaf rust, respectively. However, none of the extracts exhibited in vivo antifungal activity more than 90% against rice sheath blight, tomato gray mold, and barley powdery mildew. From the MeOH extracts of Angelica gigas and A. dahurica showing potent controlling activity against rice blast, 1 and 2 antifungal substances, respectively, were isolated by solvent partitioning and column chromatography. The three compounds were identified to be coumarins, namely, decursin, imperatorin, and isoimperatorin, by mass spectrometry and NMR spectroscopy. They were examined for in vitro and in vivo antifungal activities together with umbelliferone (7-bydroxycournarin) and scopoletin (6-methoxy-7-hydroxycoumarin) containing a free hydroxyl group at position 7 to investigate the structure-activity relationship. In vitro, most of 50% growth inhibitory concentrations ($IC_{50}$) were over $200{\mu}g/mL$, indicating that they have relatively weak antifungal activity. The antifungal activity of decursin and scopoletin, containing cyclic alkoxy groups instead of free hydroxyl group at position 7, was stronger than umbelliferone and scopoletin. Especially, decursin and imperatorin showed potent antifungal activities against Pythium ultimum and Magnaporthe grisea, respectively, with $IC_{50}$ values less than $25{\mu}g/mL$. In vivo, decursin and imperatorin showed potent antifungal activity against rice blast, whereas other coumarins hardly controlled the development of 6 plant diseases tested. These results suggest that the antifungal activity of 7-hydroxycoumarin derivative is substantially increased when the hydroxyl group at position 7 is protected by a stable cyclic alkoxy grouping.

Microbial Diversity in Korean Traditional Fermenting Starter, Nuruk, Collected in 2013 and 2014

  • A total of sixty-six samples of Nuruk, a fermention starter used to make the Korean traditional rice wine, Makgeolli, were collected from central and southern regions of Korea in 2013 and 2014. We classified two groups of the Nuruk samples, "commercial" and "home-made", according to the manufacturing procedure and purpose of use. Commercial Nuruks were made in a controlled environment where the temperature and humidity are fixed and the final product is supplied to Makgeolli manufacturers. Home-made Nuruks were made under uncontrolled conditions in the naturally opened environment and were intended for use in the production of small amounts of home-brewed Makgeolli. We obtained more than five hundred isolates including filamentous fungi and yeasts from the Nuruk samples followed by identification of fungal species. Also we stored glycerol stocks of each single isolate at $-70^{\circ}C$. We identified the species of each isolate based on the sequences of ITS regions amplified with two different universal primer pairs. We also performed morphological characterization of the filamentous fungi and yeast species through observations under the microscope. We investigated the major fungal species of commercial and home-made Nuruks by counting the colony forming units (CFU) and analyzing the occurrence tendency of fungal species. While commercial Nuruks contained mostly high CFU of yeasts, home-made Nuruks showed relatively high occurrence of filamentous fungi. One of the representative Nuruk manufacturers used both domestic wheat bran and imported ones, mainly from US, as raw material. Depending on the source of ingredient, the fungal diversity was somewhat different. Another commercial Nuruk sample was collected twice, once in 2013 and again in 2014, and showed different diversity of fungal species in each year. Nuruks obtained from the southern regions of Korea and Jeju island showed high frequency of yeast such as Saccharomycopsis fibuligera and Pichia species as well as unique filamentous fungus, Monascus species. S. fibuligera was easily found in many Nuruk samples with high CFU. The major filamentous fungi were Aspergillus, Lichtheimia, Mucor and Penicillium species. In order to further our understanding of the isolates and their potential industrial applications, we assayed three enzymes, alpha amylase, glucoamylase and acid protease from 140 isolates out of about five hundred isolates and selected about 10 excellent strains with high enzyme activities. With these fungal isolates, we will perform omics analyses including genomics, transcriptomics, metabolic pathway analyses, and metabolomics followed by whole genome sequencing of unique isolates associated with the basic research of Nuruk and that also has applications in the Makgeolli making process.

Cultural Characteristics of Cauliflower Mushroom, Sparassis crispa (꽃송이버섯(Sparassis crispa)의 배양적 특성)

  • This study was carried out to obtain the basic data for the mycelial growth conditions of cauliflower mushroom, Sparassis crispa. Twenty-one isolates were collected from domestic and abroad. The optimal temperature and pH for the mycelial growth of these isolates were $23^{\circ}C$ and pH 6.0, respectively. The mycelial growth was the best in the HBA medium, but very poor in the Lilly medium. However no mycelial growth in the CzapekDox medium. The utilization of carbon source was the best with fructose, and that of nitrogen source was the best with glutamine when compared to other tested sources. The selected isolate ASI150010 produced the highest mycelial weight in liquid culture containing soybean mill ($15\;g/{\ell}$) and potato ($200\;g/{\ell}$) extract. And uncleaned rice, wheat and barley were found to be good substrates for the mycelial growth S. crispa.
