Novel Cultivation of six-year-old Korean Ginseng (Panax ginseng) in pot: From Non-Agrochemical Management to Increased Ginsenoside

  • Background: Ginseng (Panax ginseng Meyer) is a perennial plant belonging to the Araliaceae family that is known to have various beneficial effects including improving memory loss and spatial cognitive ability, and anti-cancer and anti-diabetes activity. Its functional benefits also include improving liver function, regulating blood pressure, stress, and providing antioxidant activity. Usually, various agrochemicals are used in cultivating ginseng preventing from many diseases. Methods: FCGP (field cultivated ginseng in pot) was implemented by imitating MCWG (mountain cultivated wild ginseng). Pesticide analysis of pot cultivation was carried out and the contents of bioactive components such as ginsenoside were also analyzed. Results: FCGP ginsenoside content was higher than that of FCG (field cultivated ginseng) and MCWG. FCGP has been shown to have a relatively high antioxidant effect compared with cultivated ginseng. Conclusion: It was confirmed that ginseng can be grown for 6 years without resorting to use of pesticides. In addition, it was confirmed that effective accumulation of physiologically active ingredients such as ginsenoside is possible. Our result represents FCGP is a novel method of pesticide-free ginseng cultivation

Ginseng Conservation Program in Russian Primorye:Genetic Structure of Wild and Cultivated Populations

  • “The Regional complex long-term program of restoration (reintroduction) of Primoryes ginseng population up to 2005” elaborated by Primorye governor administration, Regional Committee of Natural Resources and Russian Academy of Sciences operates in Russian Primorye. The Institute of Biology and Soil Science (IBSS) provides the scientific implementation of the program including the genetic analysis of extant ginseng populations, plant reproduction and off-spring identification. According to our investigations, the genetic resource of P. ginseng in Primorye is represented by three populations of wild-growing ginseng and a few pritate plantations. The results obtained by RAPD allowed concluding that the resource is dispersed among the wild and cultivated ginseng sub-populations in such a way that each of sub-populations studied has to be represented as a stock material to maintain species genetic variability. The allozyme analyses also showed that the small sub-populations of wild ginseng are characterized by unique genetic diversity and, therefore, they all need to be represented in reintroduction centers. Additionally the allozyme analysis discovered that the Blue Mountain and Khasan populations possess the most genetic diversity. So, at least one more reproductive ginseng unit has to be created besides two already existing reintroduction centers representing the Sikhote-Alin and the Blue Mountain populations.

산삼약침이 자궁내막증 유발 흰쥐에 미치는 영향 (Effects of Cultivated Wild Ginseng Pharmacopuncture on Surgically Induced Endometriosis in Rats)

  • Purpose : This study was carried out to identified the effects of Cultivated Wild Ginseng Pharmacopuncture(CWG Pharmacopuncture) to the rats with surgically induced endometriosis. Methods : Endometrial tissue was implanted in the serosal wall of the small intestine in rats. The rats were divided randomly into an experimental and control group. The experimental group was treated with CWG Pharmacopuncture intravenous injection to the vein of tail and the control group was given same dose of normal saline for 4 weeks. And the size of the ectopic uterine tissue was estimated, and the serum progesterone, estradiol and cytokine($TNF-{\alpha}$, IL-2, IL-4, IL-6, IL-10) concentrations were analyzed. Results : 1. The size of the ectopic uterine implants in the experimental group was much smaller than that in the control group. 2. In analysis of serum hormone, the concentration of progesterone showed higher value than that of control group and estradiol showed lower value. 3. In analysis of cytokine, $TNF-{\alpha}$, IL-2 and IL-6 concentrations showed lower value than that of control group. 4. In analysis of cytokine, IL-4 concentration showed no significant difference and IL-10 concentrations showed higher value than that of control group. Conclusion : These results indicated that CWG Pharmacopuncture is useful treatment for endometriosis.

미국 화기삼의 종류별 생산방법과 경제성분석 (Production Procedures and Economics of the American Ginseng)

  • The purpose of this study is classifying types of American ginseng and estimating their production cost and revenue by the types. Usually, the American ginseng can be classified as 4 different types; wild ginseng(WG), wild simulated ginseng(WSG), woods grown ginseng(WGG), and field cultivated ginseng(FCG). This paper estimates costs and benefits for FCG, WGG, and WSG per acre. The WGG & WSG are produced under the tree at mountain while the FCG is produced at large scale farm with machinery. Annual profit for the FCG is $2,222 while that of the WGG and the WSG are $2,759 and $3,799 per acre. Although quantity produced per acre for the WGG and WSG(600lbs and 160lbs) are much smaller than that of the FCG(3,000lbs), prices per pound for the WGG and WSG($125, 375$) are higher than that of the FCG($24). In addition, production costs for the WGG and WSG are lower than that of the FCG because of the costs for seeds, shadow facility, and chemicals are different by the types of production.

산양삼의 품질특성 및 항산화 활성에 미치는 영향 (Effects of Quality Characteristics and Antioxidant Activity of Korean Cultivated Wild Ginseng Extract)

  • 본 연구는 산양삼의 이용가치를 높이고, 기능성 식품소재 개발을 위하여 산양삼의 이화학적 특성과 추출용매를 달리하여 추출한 각각의 추출물의 항산화 활성을 비교하였다. 산양삼의 일반성분은 수분 7.56%, 탄수화물 73.01%, 단백질 12.58%, 지질 1.99%, 회분 5.54%를 나타내었고, 총아미노산 함량은 16.17 mg/100 g이었으며, 그중 필수아미노산은 1.42 mg/100 g을 나타내었다. 총 ginsenoside 함량은 15.98 mg/g을 나타내었고, 그중 major ginsenoside($Rb_1$, $Rb_2$, $Rb_3$, Rc, Rd, Re, Rf, $Rg_1$)의 함량은 15.94 mg/g, minor ginsenoside($Rg_3$, $Rh_1$, $Rh_2$) 함량은 0.04 mg/g을 나타내었다. 1 mg/mL 농도로 조정한 증류수를 이용한 산양삼 추출물(KGW), 70% 에탄올을 이용한 산양삼 추출물(KGE)의 항산화 활성을 측정한 결과 KGE가 모든 항목에서 가장 높게 활성을 나타냈으며, 각각 8.93 mg/g(총폴리페놀 함량), 3.96 mg/g(총플라보노이드 함량), 57.57%(DPPH 라디칼 소거능), 70.73%(ABTS 라디칼 소거능), 44.12%(아질산염 소거능), 78.05%(SOD 유사활성), $1.08O.D_{700nm}$(환원력), 55.33%(ferrous ion chelating activity)를 나타내었다. 또한, 각각의 산양삼 추출물의 elastase, collagenase 및 tyrosinase 저해활성을 측정한 결과 역시 KGE가 모든 항목에서 가장 높은 활성을 나타내었으며, 각각 81.96%, 78.96%, 30.96%를 나타내었다.

B16/F10세포를 이식한 C57BL/6 생쥐에서 산삼약침의 항암효과 및 Doxorubicin에 의한 생식독성 완화효과 (Anti-cancer Effects of Cultivated Wild Ginseng Herbal Acupuncture in C57BL/6 Mice Injected with B16/F10 Cells and Reproductive Toxicity by Doxorubicin)

  • To investigate anti-cancer effects of wild ginseng herbal acupuncture and mitigation of anti-cancer drug when taken concurrently, cancer cells from B16/F10 melanoma were injected intraperitoneally in C57BL/6. After inducing cancer, anti-cancer effects and mitigation of reproductive toxicity of Doxorubicin were evaluated. 1. For changes in weight, Doxorubicin treated group showed significant decrease, and administration of wild ginseng herbal acupuncture didn't cause any weight change. 2. Volume of tumor was significantly reduced in Doxorubicin teated group. Wild ginseng herbal acupuncture groups showed slight decrease but insignificant compared to the control group. 3. For hematological evaluation, Doxorubicin only group's reticulocytes were significantly decreased compared to the control group, and Platelet Count was significantly increased. Wild ginseng herbal acupuncture group showed significant increase of Neutrophils and significant decrease of Lymphocytes compared to the control group. 4. For histological evaluation of the tumor, necrosis occurred in a wide range in the Doxorubicin treated group. Wild ginseng herbal acupuncture didn't cause much histological changes. 5. For histological evaluation of the testis, seminiferous tubules of the control group suffered severe damage on epithelial cells. When wild ginseng herbal acupuncture was administered concurrently, damage on the seminiferous tubules was significantly inhibited compared to the Doxorubicin only group. 6. Diameter of seminiferous tubules and spermatogonia count were insignificant between the experiment groups. 7. For BrdU positive reaction of testicle tissue, Doxorubicin only group failed to show any reaction of spermatogonia, but spermatocytes and spermatids showed slight positive reaction. When wild ginseng herbal acupuncture was treated concurrently, much greater positive reaction was made but similar to that of the control and normal groups. 8. For observation of changes in BrdU spermatogonia count of the testicle tissue, Doxorubicin only group didn't show any positive reaction, and relative increase was shown in the group with concurrent administration of wild ginseng herbal acupuncture. 9. For observation of TUNEL positive reaction cells of the testicle tissue, no significant changes were witnessed in all the experiment groups.

동양에 있어서의 인삼재배 역사 (The history of ginseng cultivation in Orient)

  • Ginseng has been recognized as a lifespan extending medicine which has been regarded as one of the medicines classified as top medicines, as the Boncho (medical herbs) study which is influenced by the idea of guidance's costume and food concept mainly in China is gaining its bona fide form. As the demand for ginseng has been expanded to other levels, the demand for ginseng has been increasing. Ginseng from the nature reached its supply chain limit due to its extinction and difficulty of picking, so it translated into ginseng cultivation of economy rather than harvesting in nature. After the start of ginseng cultivation, the ginseng cultivation was further enhanced by the rapid development of processing methods such as white-ginseng and red-ginseng, and the surge of consumption due to the traditional belief in ginseng drug efficacy and support of scientific research. In the Joseon Dynasty, the name Gasam (cultivated ginseng) had been created as ginseng was cultivated on farmland after the stage of SanYang (wild cultivated ginseng), the purpose of the new name Gasam is to differentiate from natural ginseng, and natural ginseng lost its firm position as the genuine ginseng as the Gasam replaced the genuine ginseng, and the natural ginseng got a new name of SanSam (wild ginseng). Because the real ginseng substance concept dissipated, and as Gasam is being called ginseng, the name Gasam was also disappeared. As a result, it was possible to grow large quantities according to the arrival of the Gasam era, and it was possible to supply the demand for ginseng, and it could become one agricultural industry. In this ginseng cultivation, in Japan where ginseng did not grow naturally, it was difficult to obtain ginseng from Joseon and faced with a shortage of ginseng at all times. Therefore, the shogun cultivated the Gasam systematically at the national level by the inside of the shogunate. However, since the natural ginseng is native to China and Korea, there is a concern about the deterioration of the quality of natural ginseng due to the incorporation of cultivated ginseng (Gasam). To protect the interests, the cultivation of ginseng was subject to control. For this reason, the lack of historical information on Gasam cultivation, which had to be started secretly, would be a natural result. In this paper, althouh not sufficient enough, the historical informations were used to summarize the history of ginseng cultivation in China, Japan and Korea.

Variation in the number of nucleoli and incomplete homogenization of 18S ribosomal DNA sequences in leaf cells of the cultivated Oriental ginseng (Panax ginseng Meyer)

  • Background: Wild ginseng, Panax ginseng Meyer, is an endangered species of medicinal plants. In the present study, we analyzed variations within the ribosomal DNA (rDNA) cluster to gain insight into the genetic diversity of the Oriental ginseng, P. ginseng, at artificial plant cultivation. Methods: The roots of wild P. ginseng plants were sampled from a nonprotected natural population of the Russian Far East. The slides were prepared from leaf tissues using the squash technique for cytogenetic analysis. The 18S rDNA sequences were cloned and sequenced. The distribution of nucleotide diversity, recombination events, and interspecific phylogenies for the total 18S rDNA sequence data set was also examined. Results: In mesophyll cells, mononucleolar nuclei were estimated to be dominant (75.7%), while the remaining nuclei contained two to four nucleoli. Among the analyzed 18S rDNA clones, 20% were identical to the 18S rDNA sequence of P. ginseng from Japan, and other clones differed in one to six substitutions. The nucleotide polymorphism was more expressed at the positions 440-640 bp, and distributed in variable regions, expansion segments, and conservative elements of core structure. The phylogenetic analysis confirmed conspecificity of ginseng plants cultivated in different regions, with two fixed mutations between P. ginseng and other species. Conclusion: This study identified the evidences of the intragenomic nucleotide polymorphism in the 18S rDNA sequences of P. ginseng. These data suggest that, in cultivated plants, the observed genome instability may influence the synthesis of biologically active compounds, which are widely used in traditional medicine.

산양산삼약침의 지질강하 및 항산화효과에 대한 생화학적 및 분자생물학적 검토 (Effects of cultivated wild ginseng pharmacopuncture on lowering lipid and oxidative capacity in biochemical and molecular biological study in obese rats)

  • Objectives : This study was carried out to identified the effects of distilled cultivated wild ginseng pharmacopuncture to the obesity. Methods : Cultivated wild ginseng pharmacopuncture was administered on the points of chung-wan(CV12), $Ch'{\breve{o}}nch'u$(ST25), and Chok-samni(ST36) on lowering lipid and oxidative capacity in biochemical and molecular biological aspects were investigated in obese rats fed with high fat diet. Results : 1. The contents of plasma ${\beta}-lipoprotein$ showed a tendency to decrease in the pharmacopuncture groups compared to the control group. In the pharmacopuncture groups, the values of ST25 and ST36 pharmacopuncture groups showed lower value. 2. The contents of plasma free fatty acids showed a tendency to decrease in pharmacopuncture groups compared to the control group. However, in the pharmacopuncture groups, the values were not significantly different. 3. Plasma triglyceride and glucose showed lower value in the ST25 pharmacopuncture groups compared with the other groups. 4. The activity of AST showed a tendency to decrease in the pharmacopuncture groups. However, the activity of ALT was not significantly different in all the treatment groups. 5. Plasma total cholesterol and LDL-cholesterol showed lower value in the ST25 and ST36 pharmacopuncture groups and HDL-cholesterol showed higher value in the CV12 pharmacopuncture groups than that of the other treatment groups. 6. Liver total cholesterol values didn't show significant difference in all the treatment groups, and triglyceride showed lower value in the pharmacopuncture groups. 7. The contents of plasma TEARS showed lower value in the ST25 pharmacopuncture group and contents of liver TBARS showed a tendency to decrease in the pharmacopuncture groups. However these values didn't show significant difference in the pharmaco puncture groups. 8. Liver super oxide dismutase activity showed higher value in the ST25 and ST36 pharmacopuncture groups, and the value of liver glutathione peroxidase and catalase activity showed a tendency to increase in the pharmacopuncture groups. However, these values showed no significant difference in the pharmacopuncture groups. 9. Expression of apo-B and E mRNA in liver cells was lower in the ST25 pharmacopuncture group than that of the other treatment groups. However, expression of $TNF-{\alpha}$ and leptin mRNA in adipose cell showed no difference among all the treatment groups. 10. ST25 pharmacopuncture group showed a good histological character of liver. It showed similar to that of normal group. However other treatment groups and control group showed slight vasodilation and slight fat accumulation. Conclusion : These results indicate that distilled cultivated wild ginseng pharmacopuncture suppressed adipose tissue mass and lipid peroxidation, and increased antioxidant capacity.

Soil properties of cultivation sites for mountain-cultivated ginseng at local level

  • Background: Identifying suitable site for growing mountain-cultivated ginseng is a concern for ginseng producers. This study was conducted to evaluate the soil properties of cultivation sites for mountain-cultivated ginseng in Hamyang-gun, which is one of the most well-known areas for mountain-cultivated ginseng in Korea. Methods: The sampling plots from 30 sites were randomly selected on or near the center of the ginseng growing sites in July and August 2009. Soil samples for the soil properties analysis were collected from the top 20 cm at five randomly selected points. Results: Mountain-cultivated ginseng was grown in soils that varied greatly in soil properties on coniferous, mixed, and deciduous broad-leaved stand sites of elevations between > 200mand < 1,000 m. The soil bulk density was higher in Pinus densiflora than in Larix leptolepis stand sites and higher in the < 700-m sites than in > 700-m sites. Soil pH was unaffected by the type of stand sites (pH 4.35-4.55), whereas the high-elevation sites of > 700mwere strongly acidified, with pH 4.19. The organic carbon and total nitrogen content were lower in the P. densiflora stand sites than in the deciduous broad-leaved stand sites. Available phosphorus was low in all of the stand sites. The exchangeable cationwas generally higher in the mixed and low-elevation sites than in the P. densiflora and high-elevation sites, respectively. Conclusion: These results indicate that mountain-cultivated ginseng in Korea is able to grow in very acidic, nutrient-depleted forest soils.
