Transrectal Ultrasound Scanning for Detecting the Early Pregnancy in Korean Black Goats

  • This study was performed to establish the early pregnancy diagnosis with evaluation of size of embryonic vesicles and change of crown-rump length (CRL) by transrectal ultrasonography in the Korean black goats. Early pregnancy diagnosis was performed in the 114 goats from 19 to 35 days after coitus by transrectal 5.0 MHz transducer. Embryonic vesicles and CRL in the 142 goats from 19 to 42 days was measured to establish the correlation between gestational age and these parameters. The early pregnancy diagnosis achieved 80% accuracy after days 25 and 100% accuracy after days 28 after breeding. Diameters of embryonic vesicle and CRL were significantly increased with the gestational age. In these facts, early pregnancy diagnosis is possible on Days 25 after breeding and measurement of embryonic vesicles and CRL were useful to predict the gestational age in Korean black goat.

  • 2-Methylbutanoic, 3-methylbutanoic, 4-methylpentanoic, 2-ethylhexanoic, 4-methylhexanoic, 4-methyloctanoic, 6-methyloctanoic, 4-ethylhexanoic. 4-methylhexanoic 및 2-ethylhexanoic acids를 포함하는 다수의 volatile branched-chain fatty acids(VBCFAs)가 쇠고기, 돼지고기, 염소고기(미국산 흰염소고기, 한국산 흑염소고기), 양고기로부터 분리, 동정되었다. 쇠고기 향은 amino acid, carbohydrate, 그리고 lipid의 반응에 의해 생성된 향기성분에 의해 이루어지는 기본 meaty flavor로 결정되어 지는 반면, 염소고기가 양고기의 경우는 쇠고기의 기본향에 4-ethyloctanoic acid와 4-methyloctanoic acid의 독특한 노린내가 더해지므로 염소고기나 양고기 특유의 향을 내었다. 돼지고기는 3-methylbutanoic acid 함량이 다른 세종류에 비해 높고 3-methylbutanoic acid 특유의 꼬린냄새는 돼지고기의 기름층으로부터 나는 unclean flavor 에 중요한 영향을 미치는 것으로 생각된다. 염소고기는 C5-C7 chain length의 volatile branched 및 n-chain fatty acids를 적게 함유하고 있는 반면 4-ethyloctanoc acid는 염소고기의 노린내를 내는 결정적인 화합물이었다. 4-Methyloctanoic acid는 양고기의 sweaty-muttony flavor를 내는 중요한 화합물이었으며 염소고기와는 달리 C5, C6, 그리고 C7 branched-chain fatty acids 도 상당량 함유되어 있었다. 한국산 흑염소고기와 미국산 흰염소고기 중에 들어있는 VBCFAs의 종류는 같으나, 한국산 흑염소고기가 미국산 흰염소고기 보다 노린내가 강하며 4-ethyloctanoic 의 함량도 높았다.

SNP 정보를 활용한 재래흑염소와 교잡종 염소의 유전적 다양성 및 유연관계 분석 (SNP-based Genetic Diversity and Relationships Analysis of the Korean Native Black Goat and Crossbred Goat)

  • 본 연구는 국내 재래흑염소 집단 (당진 계통, 장수 계통, 통영 계통 및 경상대 계통)과 교잡종 염소 집단의 유전적 다양성 및 유전적 유연관계를 조사하기 위해 수행되었다. 각 집단에 존재하는 공통 SNP 45,658개를 이용하여 분석에 이용하였다. 유전적 다양성의 지표가 될 수 있는 기대, 관측 이형접합도는 교잡종, 경상대, 장수, 통영 계통 순으로 나타났다. 집단 사이의 유전적 다양성 정도를 나타내는 분산 성분은 당진과 경상대 계통 사이에서 19.98%로 가장 높게 나타났으며, 장수와 통영 계통 사이에서 8.87%로 가장 낮게 나타났다. 또한, 집단 사이의 유전적 거리는 장수, 통영 계통에서 하나의 분지를 형성하였으며, 당진, 경상대 계통이 하나의 분지로 나타냈다. 또한, 교잡종 집단은 당진, 경상대 계통과 하나의 분지를 이루는 것으로 나타났다. 따라서 본 연구 결과는 국내 계통 간의 불필요한 근친교배와 유전자원 흐름을 줄이기 위한 기초자료 및 국내 재래흑염소 유전자원의 고유성을 나타내는 기초자료로 활용이 가능할 것으로 사료된다.

흑염소와 약용식물 복합 증탕추출액 및 증류액이 조골세포 증식과 파골세포 형성에 미치는 영향 (Effect of water extract and distillate from the mixture of black goat meat and medicinal herb on osteoblast proliferation and osteoclast formation)

  • 골기능 개선용 흑염소 액상제품을 개발하기 기초연구로서 흑염소 및 약용식물 복합추출물이 MG-63 조골세포 및 마우스 골수세포 유래 파골세포의 분화에 미치는 영향을 분석하였다. 흑염소 원료육의 일반성분, 휘발성 염기질소, 무기질함량, 유리아미노산 조성 및 지방산 조성 등의 영양성분을 분석하여 기초자료를 마련하였다. 흑염소에 첨가할 약용식물의 종류와 배합을 달리한 두 그룹의 한약재 첨가군에 대해 증탕추출액과 증류액을 제조하여 총 6개 시료군 (흑염소육 (BG-E, BG-D), 6종 한약재 첨가군 (BG-E6, BG-D6) 및 8종 한약재 첨가군 (BG-E8, BG-D8)을 대상으로 골강화 활성을 분석하였다. 식품공전상 식품의 원료로 사용이 가능한 원료 중 황기, 홍화씨, 당귀, 황정, 속단, 우슬 각각 2/2/2/2/1/1의 배합비를 지닌 한약재 6종 첨가군 및 동일배합에 녹용과 녹각을 각각 0.3/1.2 로 추가배합한 한약재 8종 첨가군을 사용하였다. 조골세포 MG-63의 증식 촉진 활성에 대해 시료별 및 농도별로 유의적인 차이가 나타났으며 한약재 무첨가 흑염소군보다 6종 및 8종 등 한약재 첨가량이 많을수록 활성이 증가하였다. 증탕추출액과 달리 증류액은 모두 유의한 효과가 없었다. 조골세포의 골석회화 촉진활성시험 결과 대조군에 비해 모든 시료 처리군에서 칼슘함량이 유의적으로 증가하여 MG-63 세포의 석회화 결절 형성을 농도 의존적으로 유의하게 증가시켰다. BG-E6는 대조군에 비해 석회화 형성을 170.3% 증가시켰고, 증류액인 BG-D와 BG-D6는 각각 168.5% 및 159.8%의 증가를 나타내었다. 시료별 차이에 있어 한약재 첨가군이 무첨가군보다 높았고, 증탕추출액이 증류액보다 높은 활성을 보였다. 세포의 칼슘 흡수량을 측정한 결과 모든 증탕추출액에서 활성이 증가하였고 특히 BG-E6와 BG-E8은 각각 615.5%와 628.1%로 유의적으로 가장 높은 증가율을 보였다. 증류액은 BG-D6의 1/10 농도군외에 효과가 없었다. 마우스 골수세포 유래 파골세포의 증식억제 실험결과 TNF-${\alpha}$ 만을 처리한 대조군에 비해 모든 시료군이 TRAP활성을 억제하는 경향을 나타내었다. 특히 BG-D 및 BG-E6, BG-E8은 유의하게 파골세포로의 분화를 억제하였다. 종합적으로 흑염소육을 비롯하여 황기, 홍화씨, 당귀, 황정, 속단, 우슬, 녹용 및 녹각 등 한약재의 복합추출물은 골 기능 강화에 매우 효과적인 기능성 원료가 될 수 있을 것으로 사료된다.

Conceptus-related measurements at early pregnancy in Black Bengal goat: an abattoir study

  • The present study aimed to investigate the conceptus-related changes during early pregnancy in the Black Bengal breed of goat. A total of 22 gravid genitalia of the Black Bengal goats were collected from local slaughterhouses. The crown-rump lengths (CRL) of the conceptuses were determined to estimate the gestational age (GA). The length and diameter of uterine horn and amniotic sac were measured, and volume of amniotic and allantoic fluid formed by individual conceptus were recorded. The results reveal that the CRL is positively correlated with GA of the conceptus (R2 = 0.89, p < 0.05); however, CRL was not influenced by number of conceptus. Both the left and the right uterine horn gradually increased in size with the advancement of pregnancy irrespective of conceptus number present in the horn. The size of the amniotic sac of conceptus gradually increased with the conceptus age but maintained spherical shape from 5 to 7 weeks of pregnancy. The amniotic fluid formed by individual conceptus rapidly increased from 5 weeks (3.4 ± 0.3 mL) to 7 weeks (21.0 ± 2.0 mL) and 9 weeks (111.5 ± 4.0 mL). The volume of allantoic fluid formed by individual conceptus was steadily increased until 7 weeks (60.0 ± 5.0 mL) and began to decline slowly thereafter (50.0 ± 5.0 mL at 9 weeks). Notably, there was no effect of conceptus number per pregnancy on individual amniotic and allantoic fluid volume. The cotyledons have first appeared on the allanto-chorionic surface from 4 to 5 weeks of pregnancy. The closed eye, nostril and hooves of the conceptus became visible at 7 weeks of pregnancy. The present study has shown the basic information on conceptus-related developmental changes during early pregnancy up to 9 weeks in Black Bengal goat.

조사료의 종류와 사료 내 영양소 수준이 흑염소의 반추위 in vitro 발효성상과 메탄생성에 미치는 영향 (Effect of Different Forage Source and Nutrient Levels in Diet on in vitro Goat Rumen Fermentation and Methane Production)

  • The present study investigated the effect of forage sources and their inclusion levels in diet on the rumen fermentation of Korea traditional goat. Timothy and alfalfa were used as forage sources. Forages were mixed with concentrate diet in different ratios. The ratios of forage to concentrate diets were varied to 1:9, 5:5 and 9:1. The rumen fluid of goat was gathered from slaughter house. Dry matter digestibility was decreased and methane production was increased as forage levels in diet was increased. When forage sources, timothy and alfalfa, were compared, groups with timothy showed greater methane production than the groups of alfalfa. Molar ratio of produced acetate and valerate were increased when forage level in diet was increased. In the case of propionate and butyrate, they were decreased as elevated forage levels in diet. The result of this study provided a basic information for rumen fermentation of Korean traditional goat and these information could be applied in the development of nutritional and feeding strategy.

Laparoscopic Transabdominal Transfer of Blastocysts in Korean Black Goats

  • As a part of the effort to improve post-transfer survival rate of embryos in Korean black goats, a technique for laparoscopic uterine transfer of blastocysts was carried out. A total of 26 transferrable embryos (morula to expanded blastocysts) were transferred to 13 recipient goats via transabdominal laparoscopic method. In consequence of our hormone protocol, 65% of the recipients (13/20) were found to have synchronized estrus. After confirmation of corpus luteum in each recipient goat, a Babcock laparoscopic forceps was inserted into the lower abdominal cavity to hold a uterine horn and fasten it near the peritoneum without causing injury. Then 7.5cm long 16G IV catheter was inserted directly into the uterine lumen through the abdominal wall. After removal of the stylet of the IV catheter, the embryo transfer tube (identical in size to the stylet and loaded with blastocysts) was inserted into the uterine lumen through the catheter to unload the embryos. Of the 13 estrus synchronized recipients, 9 were transferred blastocysts and 4 were transferred molurae (2 embryos in each recipient) in uterine ipsilateral to the ovary with corpus luteum. Four of the 9 recipients which blastocysts were transferred using this method has been confirmed pregnant (44.4% pregnancy rate).

한국흑염소 보습코기관의 형태학적 관찰 (A morphological study of vomeronasal organ of Korean black goat (Capra aegagrus hircus))

  • The vomeronasal organ (VNO) plays an important role in reproduction and social activities in ruminants including goats. A morphological study on the structure of VNO and its epithelial cells was carried out in Korean black goats. Grossly, the VNO of Korean goats opens into mouth through incisive ducts. Microscopically, the epithelium of VNO consisted of medial sensory epithelium and lateral non-sensory epithelium. Several blood vessels and nerve bundles were observed in the lamina propria encased by vomeronasal cartilage. Immunohistochemical staining showed that protein gene product (PGP) 9.5 was immunostained in the receptor cells of the sensory epithelium and in some cells of the non-sensory epithelium. Galectin-3 was mainly observed in the supporting cells of sensory and non-sensory epithelium. Lectins including wheat germ agglutinin, Ulex europaeus agglutinin, Bandeiraea simplicifolia lectin Isolectin B4, Dolichos biflorus agglutinin and soybean agglutinin used in this study were bound in VNO sensory, non-sensory epithelia as well as in the lamina propria with varying intensity. Collectively, this is a first descriptive morphological study of VNO of Korean black goat with special reference to lectin histochemistry.

Effects of Different Levels of Concentrate in the Diet on Physicochemical Traits of Korean Native Black Goat Meats

  • The effects of feeding of diets based on roughage supplemented with concentrate on the carcass and meat quality attributes of Korean black goats were investigated. The 40 male goats at 5 mon age were divided into four treatment groups; T1 (1.5%), T2 (2.0%), T3 (2.5%) and T4 (ad libitum) feeding of concentrate with ad libitum rice straw. Forty bucks at the end of the feeding were fasted for 12 h before slaughter and were then slaughtered. For carcass measurement, cold buck carcasses were examined after 24 h of chilling at $5^{\circ}C$. The carcasses were chilled in refrigerator at $2{\pm}2^{\circ}C$ for 24 h, then the longissimus dorsi muscle was removed from the carcasses. The samples were separately vacuum packaged, frozen and stored at $-20^{\circ}C$ for up to 1 mon, prior to physicochemical evaluations. Carcass yields were greatly affected by rate of concentrate feeding. T4 showed the heaviest live and carcass weights and the highest dressing and fat percentages (p<0.05). The moisture contents were lower in T4 compared with other treatments, while the crude protein and fat contents were higher (p<0.05). mono-unsaturated fatty acid in T4 was significantly higher than others, which showed the highest percentages of oleic acid (C18:1). For the sensory test, T4 was tenderer and produced better flavor scores than others. Increasing the level of concentrate in the diet resulted in an improvement in growth performance, and carcass and physicochemical characteristics of goat meats.

흑염소 간-조직과 Ferrocene 매개체를 사용한 과산화수소정량 전류법 바이오센서 (Amperometric Biosensor for Hydrogen Peroxide Determination Based on Black Goat Liver-Tissue and Ferrocene Mediation)

  • 흑염소의 간-조직과 ferrocene을 탄소반죽 전극에 고정하여 과산화수소를 정량할 수 있는 전극을 만들었다. Ag/AgCl 전극에 대하여 $-0.3{\sim}+0.0\;V$의 전위 범위에서 전극의 감도를 관찰하였으며 전극의 감응시간은 12 s로 나타났다. 전극의 검출한계는 2.25${\times)10^{-6}M$ (S/N=3)이었으며,1.0${\times}10^{-2 }$M 과산화수소를 이용해 15회 반복 측정한 결과, 상대표준편차는 1.87%이었다. 또한, 방해물질의 영향도 없는 것으로 나타났다. 효소전극의 감도는 19일 사용 후 50%로 감소하였다.
