일당귀 n-hexane분획이 에탄올을 투여한 흰쥐의 항산화계 및 지질과산화에 미치는 영향 (Effects of n-Hexane Fraction of Angelica acutiloba on Antioxidative System and Lipid peroxidation in Ethanol-Induced Hepatotoxicity of rats)

  • 일당귀의 n-hexane분획이 횐쥐에서 에탄올투여로 생성된oxygen free radical대사와 지질과산화작용에 미치는 영향을검토하기 위하여 실험동물을 정상군, 일당귀 n-hexane 분획70 mg/kg투여군, 알코올투여군(35$\%$ ethanol 10 mL/kg, b.w./day), 알코올(35$\%$ ethanol 10 mL/kg, b.w./day) 및 일당귀 n-hexane 분획 70 mg/kg 병합투여군과 알코올(35$\%$ ethanol 10mL/kg, b.w./day) 및 일당귀 n-hexane 분획 140 mL/kg 병합투여군의 5군으로 나누어 6주간 사육하여 체중증가율, 식이효율 및 혈청중 ALT, AST 측정과 간손상 억제효과를 검토하기 위하여 간조직중 유리기 생성계 효소인 XO활성, 유리기해독계 효소인 SOD, calaase 및 GSH-Px활성과 지질과산화물인 TBARS 및 GSH함량을 측정한 결과는 다음과 같다. 각 분획의 항산화력은 n-hexane, ethylacetate, chloroform,n-butanol 및 water 분획순으로, n-hexane 분획의 항산화력이가장 우수하였다. 6주간의 체중증가율과 식이효율은 알코올투여로 정상군에 비하여 유의적으로 저하되었으나 일당귀 n-hexane 분획투여로 정상군에 근접하게 상승되었고, 알코올투여로 상승된ALT 및 AST활성도 유의하게 저하되었다. 간 조직중 XO활성은 알코올투여로 정상군보다 상승되었으며 일당귀 n-hexane 분획 투여로 저하는 되었지만 유의성 있는 변화는 아니었다. 알코을 투여로 상승된 SOD활성은일당귀 n-hexane 분획 투여로 정상군에 비하여 유의하게 저하되었으나 catalase와 GSH-Px활성은 유의적인 변화를 나타내지 않았다. TBA 반응성 산물량은 정상군과 일당귀 n-hexane 분획 투여군간에 차이는 없었고 알코올투여로 정상군보다 많은 증가를 나타냈으나 일당귀 n-hexane 분획투여로 정상군에 근접하게 감소되었다. 알코올투여로 감소된 GSH 함량은 일당귀 n-hexane분획을 투여하여 용량의존적으로 증가되었다. 이상의 실험결과에서 일당귀의 항산화작용은 유리기 해독계 효소인 SOD활성억제와 비효소적 항산화작용을 나타내는GSH량을 증가시킴으로서 지질과산화물에 대한 방어력이 증강되어 나타난 결과로 판단되어지며, 또한 혈청중 ALT 및AST활성을 유의성있게 감소시키므로써 일당귀가 알코올성 간질환에서 손상된 간기능을 회복시킬 수 있을 것으로 추정되었다.

Acetylcholinesterase Inhibitors from the Roots of Angelica dahurica

  • In the course of finding Korean natural products for acetylcholinesterase (AChE) inhibitory activity, we found that a methanolic extract of the roots of Angelica dahurica showed significant inhibitory effects on AChE. Bioassay-guided fractionation of the methanolic extract resulted in the isolation of three furanocoumarins, isoimperatorin (1), imperatorin (2) and oxypeucedanin (3), as active principles. These compounds inhibited AChE activity in a dosedependent manner, and the $IC_{50}$ values of compounds 1-3 were 74.6, 63.7 and 89.1 uM, respectively.

Bacterial Black Stem Rot on Angelica acutiloba Caused by Xanthomonas campestris

  • Soaked black rot symptom was observed on the stem of Angelica acutiloba from July to August 2000 at Kumsan, Chungnam in Korea. This disease usually occurred under humid and high temperature conditions. The lesions on the stem appeared as soft rot with brown elliptical spots, which developed into large black spots at a later stage. When the bacterial isolates from the diseased plants were inoculated onto healthy plants by artificial needle prick method, symptoms similar to that observed in the fields developed. According to the cultural characteristics and pathogenicity of the isolates on the host plant the causal bacterium was identified as Xanthomonas campestris. This study proposed that the disease be named "bacterial black stem rot of A. acutiloba"loba".

Determination of Biosynthetic Pathway of Decursin in Hairy Root Culture of Angelica gigas

  • To establish the biosynthetic pathway of decursin in Angelica gigas Nakai, feeding experiment with stable isotope-labeled precursors were conducted. Umbelliferone and decursin were labeled with deuterium at C-3. The umbelliferone, the decursin, and other commercially available putative precursors, L-phenylalanine-ring-$d_5$ and trans-cinnamic acid-$d_7$, were fed to the hairy root culture of A. gigas. Each deuterated compound was incorporated into decursinol, decursinol angelate, and decursin as determined by mass spectrometric analysis. These findings confirmed the coumarin biosynthesis pathway sequence is composed of phenylalanine, cinnamic acid, umbelliferone, decursinol, and decursin.

The Effect of Long-term Administration of Green Juice of Angelica Keiskei Koidz on the Contents of Several Selected Elements of the Mouse Organs

  • In order to study the content of selected elements in the organs of the mice, mice were fed with green juice of Angelica keiskei Koidz for 16 weeks. The results obtained from the experiments are summarized as follows : 1) The element contents(cr, Mn, Ni, Cu, Zn, Cd, Ba, Pb, and Fe) in the whole plant are generally the same as those the previous reports. 2) The element contents(Cr , Mn, Ni, Cu, Zn, Cd, Ba, Pb and Fe) in the green juice did not exceed those of the edible water standard. 3) The element contents (Cr, Mn, Ni, Cu, Zn, Cd, Ba, Pb and Fe) in the brain is slightly higher than those in the whole plants. 4) The element contents(Cr, Ni, Pb and Fe) in the organs of the mice fed green juice for 16 weeks are the same as those of the previous reports.

Decursin from Angelica gigas Nakai Blocks hKv1.5 Channel

  • Decursin was purified from Angelica gigas Nakai, and its effects on the human Kv1.5 (hKv1.5) currents were recorded in mouse fibroblasts ($Ltk^-$ cells) by whole-cell patch-clamp technique. Decursin inhibited hKv1.5 current in a concentration-dependent manner, with an $IC_{50}$ value of $2.7\;{\mu}M$ at +60 mV. Decursin accelerated the inactivation kinetics of the hKv1.5 channel, and slowed the deactivation kinetics of the hKv1.5 current, resulting in a tail crossover phenomenon. Also, decursin inhibited the hKv1.5 current in a use-dependent manner. These results strongly suggest that decursin is a kind of open-channel blocker of the hKv1.5 channel.

  • The anti-oxidant activities of decursinol angelate (1) and decursin (2) isolated from Angelica gigas were investigated. These two coumarins exhibited decrease in serum transaminase activities elevated by hepatic damage induced by $CCl_4-intoxication$ in rats. They also showed increase in anti-oxidant enzyme such as hepatic cytosolic superoxide dismutase (SOD), catalase, and glutathione peroxidase (GSH-px) in $CCl_4-intoxicated$ rats. These results suggest that decursinol angelate (1) and decursin (2) from A. gigas possess not only the anti-oxidant, but also the hepatoprotective activities in rats.

Neuroprotective Effect of Scopoletin from Angelica dahurica on Oxygen and Glucose Deprivation-exposed Rat Organotypic Hippocampal Slice Culture

  • This study examined the neuroprotective effect of scopoletin from Angelica dahurica against oxygen and glucose deprivation-induced neurotoxicity in a rat organotypic hippocampal slice culture. Scopoletin reduced the propidium iodide (PI) uptake, which is an indication of impaired cell membrane integrity. In addition, it inhibited the loss of NeuN, which represents the viability of neuronal cells. The results suggests that scopoletin from A. dahurica protects neuronal cells from the damage caused by oxygen and glucose deprivation.

  • From the roots of Angelica dahurica Bentham et Hooker (Umbelliferae), three known coumarin derivatives have been isolated and identified as 8-(2-hydroxy-3-methoxy-3-methylbutyloxy) psoralen, 5,8-di(2,3-dihydroxy-3-methylbutyloxy) psoralen, 9-[3-($\beta$-D-glucopyranosyloxy)-2-hydroxy-3-methylbutoxy]-7H-furo[3,2-g][1]benzopyran-7-one. This is the first report of the occurrence of these compounds in this plant. These three compounds were tested for activity in septic shock model. Among these compounds, 2 showed relatively strong preventive activity against septic shock.

  • Angelica decursiva Fr. et Sav. (Umbelliferae) has traditionally been used to treat different diseases due to its antitussive, analgesic, and antipyretic activities. It is also a remedy for thick phlegm, asthma, and upper respiratory infections. Recently, the leaf of A. decursiva has been consumed as salad without showing any toxicity. This plant is a rich in different types of coumarin derivatives, including dihydroxanthyletin, psoralen, dihydropsoralen, hydroxycoumarin, and dihydropyran. Its crude extracts and pure constituents possess anti-inflammatory, anti-diabetic, anti-Alzheimer disease, anti-hypertension, anti-cancer, antioxidant, anthelmintic, preventing cerebral stroke, and neuroprotective activities. This valuable herb needs to be further studied and developed not only to treat these human diseases, but also to improve human health. This review provides an overview of current knowledge of A. decursiva metabolites and their biological activities to prioritize future studies.
