Identification of 'Chunpoong' among Panax ginseng Cultivars Using Real Time PCR and SNP Marker

  • The common DNA extraction methods are indispensable for genotyping by molecular marker analysis. However, genotyping a large number of plants is painstaking. A modified 'NaOH-Tris' method used in this study reduces the extraction time while keeping the cost low and avoiding the use of hazardous chemicals. The endpoint analysis by realtime PCR tends to be fast and effective for the development of SNP markers linked to the 'Chunpoong' cultivar of Panax ginseng. The 'Chunpoong' marker was developed by a major latex-like protein gene sequence. From our results, we suggest that this method is successful in distinguishing 'Chunpoong' from a large number of ginseng cultivars.

  • Sesquiterpenes are found naturally in plants and insects as defensive agents or pheromones. They are produced in the cytosolic acetate/mevalonate pathway for isoprenoid biosynthesis. The inducible sesquiterpene synthases (STS), which are responsible for the transformation of the precursor farnesyl diphosphate, appear to generate very few olefinic products that are converted to biologically active metabolites. In this study, we isolated the STS gene from Panax ginseng C.A. Meyer, designated PgSTS, and investigated the correlation between its expression and various abiotic stresses using real-time PCR. PgSTS cDNA was observed to be 1,883 nucleotides long with an open reading frame of 1,707 bp, encoding a protein of 568 amino acids. The molecular mass of the mature protein was determined to be 65.5 kDa, with a predicted isoelectric point of 5.98. A GenBank BlastX search revealed the deduced amino acid sequence of PgSTS to be homologous to STS from other plants, with the highest similarity to an STS from Lycopersicon hirsutum (55% identity, 51% similarity). Real-time PCR analysis showed that different abiotic stresses triggered significant induction of PgSTS expression at different time points.

Measurement of Short Reverberation Times at Low Frequencies Using Wavelet Filter Bank

  • In room acoustics, reverberation time is an important acoustic parameter. However it is often difficult to measure short reverberation times at low frequencies with a traditional band pass filter bank if the product of filter bandwidth (B) and reverberation time (T) is small. It it well known that the minimum permissible product of bandwidth and reverberation time of the traditional band pass filter is at least 16. This strict requirement makes it difficult to measure short reverberation times of an acoustic room at low frequencies exactly. In order to reduce this strict requirement, in the previous paper, the wavelet filter bank was developed and the minimum permissible product of bandwidth and reverberation time was replaced with 4. In the present paper it is demonstrated how the short reverberation times of an practical room at low frequencies are successfully measured by using the wavelet filter bank and the results are compared with the traditional method using a band past filer bank.

  • 타임뱅크(Time bank)는 '모든 사람의 시간과 모든 노동의 가치는 동일하다'는 원칙하에 지역 커뮤니티 내의 구성원 사이에 남을 도운 시간을 적립하고 남으로부터 도움을 받을 때 그 시간만큼 찾아 쓸 수 있는 상호호혜적인 봉사 시스템이다. 타임뱅크가 가능하기 위해서는 도움을 받는 사람과 주는 사람을 매칭시키고 시간 화폐를 계좌에 적립하고 관리하기 위한 웹 플랫폼이 필수적이지만, 국내에서는 관련 기술개발이 부진하다. 본 연구에서는 클라우드 서버 내에 Django 기반 데이터베이스를 구축하여 봉사 거래 및 타임뱅크 계좌가 관리될 수 있는 한국형 타임뱅크 웹 플랫폼을 제안한다.

Isolation and Characterization of Glycolate Oxidase Gene from Panax ginseng C. A. Meyer

  • The oxidation of glycolate to glyoxylate, a key step in plant photorespiration, is carried out by the peroxisomal flavoprotein glycolate oxidase (EC 1.1.3.15). To investigate the altered gene expression and the role of GOX in ginseng plant defense system, a cDNA clone containing a GOX gene designated as PgGOX was isolated and sequenced from Panax ginseng. The cDNA was 692 nucleotides long and have an open reading frame of 552 bp with a deduced amino acid sequence of 183 residues. A GenBank BlastX search revealed that the deduced amino acid of PgGOX shares a high degree homology with the Glycine max (95% identity). In the present study we analyzed the expression of PgGOX under various environmental stresses at different times using real time-PCR. The results showed that the expressions of PgGOX increased after various treatments involving salt, light, cold, ABA, SA, and copper treatment.

  • $Ca^{2+}$ and calmodulin (CaM), a key $Ca^{2+}$ sensor in all eukaryotes, have been implicated for defense responses of plants. Eukaryotic CaM contains four structurally and functionally similar $Ca^{2+}$ domains named I, II, III and IV. Each $Ca^{2+}$ binding loop consists of 12 amino acid residues with ligands arranged spatially to satisfy the octahedral symmetry of $Ca^{2+}$ binding. To investigate the altered gene expression and the role of CaM in ginseng plant defense system, cDNA clone containing a CaM gene, designated PgCaM was isolated and sequenced from Panax ginseng. PgCaM, which has open reading frame of 450 nucleotides predicted to encode a precursor protein of 150 amino acid residues. Its sequence shows high homologies with a number of other CaMs, with more similarity to CaM of Daucus carota (AAQ63461). The expression of PgCaM in different P. ginseng organs was analyzed using real time PCR. The results showed that PgCaM expressed at different levels in young leaves, shoots, and roots of 3-week-old P. ginseng. In addition, the expressions of PgCaM under different abiotic stresses were analyzed at different time intervals.

  • To decide on the queuing system of the optimum-sized bank window, data by means of simulation was reckoned. That is, by linking the average arrival rate and the average service rate with the exponential random number, customers' arrival time and service time was reckoned and simulation size optionally decided. By so doing, this paper is aimed at predicting the conditions of a bank, average arrival time, average waiting time, aveerage service time, average queuing length, servers' idle time, etd, and at preparing for a simulation model of the queuing system that can apply not only to the bank window box but also to all system under which queuing phenomena may arise.

The Effect of the Bilateral Expansion of Korean & Chinese Banks on Bank Performance (한·중은행의 상호진출이 경영성과에 미치는 영향)

  • This paper studies the influence of the bilateral expansion of Korean and Chinese banks on performance. To conduct the empirical analysis, we select eleven Banks in South Korea and thirteen Banks in China over the 2000-2014 time frame. The main regression results are as followed. First, the sample bank's overseas subsidiaries have a significant effect on profitability and the profitability is somewhat deteriorates. Second, the sample bank's overseas branch and subsidiaries have a significant effect on the bank's risk. That is, the loan loss provision ratio is slightly increased but impaired loan ratio is highly decreased. Third, the sample banks' overseas branch and subsidiaries also have a significant effect on bank efficiency. The ratio of customer deposits per employee worsens somewhat but the gross loan per employee are increased. In conclusion, as time passes, bank profitability or efficiency from mutual expansion of Korean banks and Chinese banks seem to be improving somewhat, and this phenomenon is more prominent at Korean banks than at Chinese banks.

  • This paper is aiming at analyzing case law of India in relation with reasonable time to make decision whether to accept or to refuse the documents received from the presenter in credit transactions. As specified in UCP, the failure to refuse to accept the documents within a reasonable time precludes the Issuing Bank, Confirming Bank (if any) and Nominated Bank from asserting that they are discrepant. Compliance of the stipulated documents on their face with the terms and conditions of the credit shall be determined by international standard banking practice as reflected in this Articles of UCP 500. The Issuing bank is only to be held responsible for honoring the documents presented by beneficiary through the nominated banks if they are strictly in compliance with terms and conditions of the Credit. As any well experienced banker knows, however, a word-by-word, letter-by-letter correspondence between the documents and the credit terms means a practical impossibility. Thus the notion of reasonable care in conjunction with the doctrine of strict compliance mixed with International Standard Banking Practices has not played a right functional standard for checking the documents as stipulated in the credit and UCP 500. And so the rejection rate is highly estimated at approximately 50% in EU and 40 to 70% according to their geographical locations in the USA. As a result, it can possibly be inferred from this fact that the credit industry would be facing the functional failure as the international trade credit facility, if not supported with motive power as a relevant scheme in UCP 500. It is quite important to note that UCP 500 Article 13(b) which specify the time limit for the banks to notify the presenter their decision not to accept the documents within a reasonable time not to exceed seven banking days following the day of receipt of documents would be the motive engine to improve the negotiability of documents in international trade financial facility.

Isolation and Characterization of Pathogenesis-Related Protein 5 (PgPR5) Gene from Panax ginseng

  • A pathogenesis-related protein (PgPR5) gene that isolated from the leaf of Panax ginseng was characterized. The ORF is 756 bp with a deduced amino acid sequence of 251 residues. The calculated molecular mass of the matured protein is approximately 27.5 kDa with a predicated isoelectric point of 7.80. A GenBank BlastX search revealed that the deduced amino acid of PgPR5 shares highest sequence similarity to PR5 of Actinidia deliciosa (80% identity, 87% similarity). PgPR5 has a C-terminal and N-terminal signal peptide, suggesting that it is a vacuolar secreted protein. The expression of PgPR5 under various environmental stresses was analyzed at different time points using real-time PCR. Our results reveal that PgPR5 is induced by salt stress, chilling stress, heavy metal, UV, and pathogen infection. These results suggest that the PgPR5 could play a role in the molecular defence response of ginseng to abiotic and pathogen attack. This is the first report of the isolation of PR5 gene from the P. ginseng.
