Effects of Thiabendazole on Green Mold, Trichoderma spp. during Cultivation of Oyster Mushroom, Pleurotus spp. (느타리버섯 푸른곰팡이병에 대한 Thiabendazole의 방제효과)

  • This study was conducted to find out the effects of Thiabendazole on controlling green mold causing serious damage to oyster mushroom, Pleurotus spp. during the cultivation. In vitro, the strains of oyster mushroom such as ASI 2018, 2072 and 2016 were inhibited by 500 ppm of the fungicide, but the strain of ASI 2001 and ASI 2070 was inhibited by 100 and 500 ppm on oatmeal agar, respectively. The mycelial growth of the oyster mushroom started to be inhibition by soak treatment at a 0.2g/1000 ml aqueous solution of the fungicide. When the oyster mushroom and green mold inoculated both or separately on the substrates of soak treatment, the green mold did not grow at all, but the oyster mushroom grown well. The maximum control effect of the green mold showed when $2g/m^2\;and\;5g/m^2$ of the fungicide was sprayed on the surface of substrates before pasteurization. The highest yield of the sporophores of oyster mushroom was obtained from $5g/m^2$ treatment.

  • This experiment was undertaken to investigate proper conditions for protoplast formation from Pleurotus spodoleucus. Novozym 234 was the most effective enzyme and a high yield of protoplasts was obtained. Combination of enzymes did not improve this result. Sucrose gave the best result to support the release and maintain the stability of protoplasts. The optimal reaction time of mycelium with the lytic mixture was 3 hrs. in shaking condition at 120 strokes $min-^1$. When mycelium of P. spodoleucus was cultured for 3 days on mushroom complete agar medium at $27^{\circ}C$, the formation of protoplasts was effective. The mushroom complete agar medium stabilized with 0.6 M sucrose was the most effective for reversion of protoplasts.

  • The transfer of the isolated nuclei from P. florida into protoplasts of P. ostreatus was induced with polyethylene glycol and $CaCl_2$. Three types of transfer products of nuclei were obtained when transferred to MMM. Type 1 colonies were more vigrously growing mycelium and stable on MCM. One of the type 1 colonies, appeared segregation on MCM plus benomyl. The mycelium did not form clamp connection. These results suggest that type 1 colonies were nuclear hybrids or allodiploids. Type 2 was main products of nuclear transfer. The mycelium formed clamp connection and fertile on sawdust media. Type 3 was very slow growing or non-viable colonies after debris of nuclei or chromosomes transfer into recipient protoplasts. Isozyme pattern of esterase in type 1 produced a new band. Type 2 and type 3 could be characterized by parental bands.

Transformation of Pleurotus florida with Neurospora pyr 4 Gene (Neurospora pyr 4 유전자를 이용한 사철 느타리버섯의 형질전환)

  • Transformation of an auxotrophic requirement for uracil in Pleurotus florida P101 has been achieved using chimeric vector containing Aspergillus nidulans ans 1, and Neurospora crassa pyr 4 DNA. Protoplasts of $Ura^-$strains of P. florida were incubated with plasmid pDJB3 containing the cloned pyr 4 gene in the presence of polyethylene glycol and $CaCl_2$. Transformants could grow on MMM showing mitotical stability. Southern hybridization analysis of DNA isolated from transformants showed that the Neurospora pyr 4 gene and vector sequence might be integrated into the P. florida chromosomes. As the transformants were monokaryon, each transformant was mated with the other monokaryon. Fruitbody shape of untransformant was eroded type but those of transformants were eroded type, funnel type, plane type and ungrowing cap type.

Selection of Highly Bitter taste Strains on Ganoderma sp. (영지버섯 고미성 균주 선발에 관한 연구)

  • A study was conducted on the characteristics of 11 strains of Ganoderma sp. to select the strains with more bitterness. Among the flat type strains, ASI 7071 and 7091 showed higher bitterness, while among the branched type strains ASI 7074 and 7094 were found to more bitter than other strains. The growth of ASI 7091 was best on Ganoderma complete media(GCM), while ASI 7010, 7048 and 7075 performed best in oak saw dust media. Among the branched type strains the esterase isozyme band patterns were similar. On the other hand among the flat type strains, the esterase isozyme band patterns differed from each other. Out of 11 strains, ASI 7004 was found to have the heaviest fruiting body, ASI 7071 the thickest cap and ASI 7094 the biggest cap.

  • The ever increasing demand for energy and the shortage of resources all over the world have generated interest in recycling renewable sources such as lignocelluloses which otherwise would go to waste and cause environmental pollution. Lignin is the incrustation material for cellulose and hemicellulose, therefore, cellulose and hemicellulose are not easily degraded. Recycling lignocellulosic wastes as agricultural material are benefit to everybody and everything. In order to improve ligninase and laccase production of Phanerochaete chrysosporium, BKM-F-1767 and Ceriporiopsis subvermispora, FP 90031-SP, were compared. The ligninase activity of P. chrysosporium was maximum on day 4.5 of shaking culture at 150 rpm 2.5 cm in a back and forth cycle. The laccase activity of C. subvermispora was maximum on day 5.5 for 2% malt extract+0.1% yeast extract+0.1% Tween 20+6 mM Benzyl alcohol culture medium at stationary state.

Kretzschmaria quercicola sp. nov., an Undescribed Fungus from Living Oak in Mt. Daeryong, Korea

  • We encountered an unfamiliar ascomycete fruiting body, fitting characteristics of the genus Kretzschmaria, which features in a stipitate ascigerous stroma with carbonaceous interior and disintegrating perithecia. In this study, we report and characterize a new species of the decaying fungus. Compared to other species, one of the notable features of this specimen (TPML150908-046) is its stromatal size (up to 15 cm). Although TPML150908-046 is morphologically similar to K. milleri and K. sandvicensis, it differs sharply from both species in apical ring size (TPML150908-046, $6.5{\sim}10.5{\mu}m$; K. milleri, $11{\sim}16{\mu}m$) and ascospore width (TPML150908-046, $10.5{\sim}17{\mu}m$; K. sandvicensis, $8.5~11.5{\mu}m$). Phylogenetic trees based on ${\beta}$-tubulin, ITS, and RPB2 sequences showed that our collection clustered with K. sandvicensis, with the respective similarities for these sequences being 95.6%, 91.3%, and 97.7%, signifying it as another species. With these results, we report it as a new species, which we call Kretzschmaria quercicola sp. nov.

  • The transfer of isolated nuclei from Agrocybe aegerita mycelia of wild type into Pleurotus florida protoplasts of auxotroph was induced with polyethylene glycol. The type 1 of nuclear transfer products was spontaneous segregants of both parental morphology of colony. Hyphae of A. aegerita type had clamp connections while that of P. florida type lacked. Type 2 was main products of nuclear transfer which formed true clamp connections. Type 3 was clampless products. They all produced primordia and developed basidiocarps similar to Agrocybe aegerita. A comparison of nuclei transfer products was made using isozyme analyses of alcohol dehydrogenase, esterase, lactate dehydrogenase and peroxidase. Isozyme band patterns of some type 2 strains produced a new band. Band patterns from mycelial extracts of the other strains could be characterized by parental bands.

Interorder Protoplast Fusion between Pleurotus ostreatus and Ganoderma applanatum (느타리버섯과 잔나비걸상버섯과의 이목간(異目間) 원형질체(原形質體) 융합(融合))

  • Interorder heterokaryons were obtained by polyethylene glycol induced fusion of protoplasts from auxotrophic mutants of Pleurotus ostreatus in agaricales and Ganoderma applanaturm in aphyllophorales. When transferred to MMM plates, all fusion colonies exhibited an extremely growth rate. During three times subcultivation on MCM the growth rate of fusants showed faster little by little. Seventy-five % fusion products of potoplasts showed mixed morphologies between those of P. ostreatus and G. applannatum in the first subcultivation on MCM and MGM. The phenotype of these fusants changed similar those of P. osteatus type after three times subcultivation on MCM. However, phenotype of 25% stable strains did not change on subcultivation. Hyphae of all fusion products did not form true clamp connection. All these types did not produce primordia. A comparrison of interorder somatic hybrids was made using isozyme analysis of esterase, malate dehydrogenase and peroxidase. In most cases the enzyme patterns of G. applanatum were not distinct, however, fusant showed non-parental bands.

  • After treatment of basidiospores of P. cornucopiae with ultraviolet light, 84 putative mutants from 4671 isolates were obtained. The highest proportion of auxotrophic mutants was obtained from the isolates irradiated to give $0.4{\sim}1.0%$ survival. Fourteen auxotrophs were selected for protoplast fusion and each genetic marker was identified.
