Antifungal Activity of 2-Hydroxy 4,4'6'Trimethoxy Chalcone

  • Antifungal activity of 2-hydroxy 4,4'6'trimethoxy chalcone individually was tested against spore germination of ten fungi of different genera. Efficacy of the chemical was also tested against conidial germination and other growth parameters of Erysiphe pisi on excised pea leaves. 2-Hydroxy 4,4'6'trimethoxy chalcone inhibited spore germination at all the concentrations. Maximum inhibition was observed at 2000 ppm where more than 78 per cent inhibition of spore germination was observed in Ustilago cynodontis, Alternaria brassicicola, A. solani and Aspergillus flavus. It also reduced conidial germination of E. pisi significantly, when applied as pre-inoculation treatment.

  • Phytoterpenoid isolated from Artabotrytis odoratissimus inhibited spore germination of some plant pathogenic as well as saprophytic fungi e.g. Alternaria alternata, A. solani, Cercospora sp., Curvularia maculans, C. pennisetti, Fusarium udum, Helminthosporium echinochlova, H. frumentacie, H. penniseti and Ustilago cynodontis. In Curvularia maculans and H. frumentacie, spore germination was completely inhibited at 2000 ppm. However, Curvularia maculans and C. pennisetti showed considerable sensitivity to this chemical even at 500 ppm.

  • Neohaplonts were obtained to improve mushroom strain from dikaryon strains of Flammulina Velutips, Ganoderma lucidum, Lyophyllum ulmarium, Pleurotus cornucopiae and Pleurotus spodoleucus by protoplast reversion. The noehaplont frequency from reversion colonies of protoplasts was 4.47-47.7%. Four more types of morphological characteristics were detected from neohaplonts of protoplast reversion in L. ulmarium, P. cornucopiae and P. spodoleucus, but one or two types were neohaplonts in F.velutipes and G. lucidum. Growth rate of neohaplonts was slow compared with dikaryon strains.

  • Factors influencing the fusion frequency of protoplasts were investigated with auxotrophic mutants of Pleurotus florida and Pleurotus ostreatus. Immediately after the polyethylene glycol (PEG) solution was added, the protoplasts adhered firmly and shrank. During the subsequent dilution with 0.6 M sucrose, the protoplasts regained their normal size and larger bodies were observed. Interspecific heterokaryons were obtained by fusion of the nutritionally complementing protoplasts. Hyphae of the heterokaryotic fusants formed true clamp connections. The optimum conditions were a total of 1 to 15 million protoplasts per ml, 30% polyethylene glycol 8000 solution with adjustment to pH 8.0 and 0.6 M sucrose stabilized regeneration medium. Other parameters such as $CA^{++}$, glycine, exposure time and temperature influenced mainly the viability of the protoplasts.

Effect of Sodium Chloride on Biology of Catenaria anguillulae

  • Growth studies of Catenaria anguillulae isolates in response to sodium chloride indicated that all the isolates grew in linseed oil-cake agar medium containing sodium chloride up to 1.0%. Medium with 1.5% sodium chloride, however, completely checked the growth of all the isolates. The size of zoosporangia greatly increased with abundant zoospore production in medium containing sodium chloride at 0.5%.

  • Phanerochaete chrysosporium is a white rot fungus which secrets a family of lignin-degrading enzymes under nutrient limitation. Ligninase was extracellularly produced in agitated submerged cultures of P. chrysosporium, SC 26. Addition of veratryl alcohol(4 mM), and benzyl alcohol(10 mM) with 0.1% Tween 20 to the culture medium stimulated ligninase production. However, ligninase was not detected when both treatments of veratryl alcohol and benzyl alcohol without Tween 20 were added to the medium. Addition of 0.1 % Tween 20 to the culture medium had little effect on ligninase activity. The ligninase activity was maximum on day 5-8 for veratryl alcohol, and benzyl alcohol with 0.1 % Tween 20 additive medium.

  • This experiment was undertaken to investigate considerable economy of labor for selection of auxotrophs from strains of Lentinus edodes. Various types of auxotrophs of this strain were isolated after treatment of basidiospores with ultraviolet light, and the highest proportion of putative mutants was also obtained from isolates irradiated to give 1.30 % survival.
