Studies on Some Parasites from Aquarium Rockfishes, Sebastes spp.

  • The results of study of the parasites of rockfishes, Sebastes spp. collected at Oregon coast aquarium during April 6 to June 30, 2002 are presented. Six species of parasites were recovered from examination of 19 rockfishes with an incidence of infection of 42.1 %. This study investigated parasitic infestation of 7 black rock-fish, Sebastes melanops ; 3 quillback rockfish, S. maliger ; 2 yellowtail rockfish, S. flavidus ; 3 canary rockfish, S. pinniger 2 yelloweye rockfish, S. ruberrimus ; 2 tiger rockfish, S. nigrocinctus. Sebastes spp. from aquarium were found to be infested with parasites: Mycrocotyle sebastis, Megalocotyle trituba, Clavella uncinata, Anisakis simplex larvae, Contracaecum sp. larvae, Pseudoterranova decipiens larvae. Prevalence and mean intensity of rockfishes were infected respectively as follows: 21.1%, 4.0 of M. sebastis, 10.5%, 55.5 of M. trituba, 21.1%, 34.3 of A. simplex larvae, 10.5%, 15.0 of Contracaecum sp. larvae, 10.5%, 8.5 of Pseudoterranova decipiens larvae and 10.5%, 5.0 of Clavella uncinata. This study reveled that gills and gastrointestinal tracts of S. pinniger were heavily infested with M. trituba and A. simplex larvae.

Cloning and Characterization of cDNA for Korean Rockfish (Sebastes schlegeli ) Insulin-like Growth Factor-I

  • 어류의 insulin-like growth factor-I (IGF-I)의 생화학적 작용기작을 연구하기 위하여 한국산 조피볼락의 IGF-I cDNA 유전자 cloning을 행하였다. 완전한 cDNA 유전자 염기서열은 PCR과 RACE 방법을 통하여 얻어진 DNA로부터 결과를 얻을수 있었다. 결정된 IGF-I의 염기서열은 flounder, chinook salmon, human IGF-I의 염기서열과 비교한 결과 각각 93.6%, 90.7%, 85.4%의 높은 상동성을 보였다. 생화학적으로 활성이 있는 재조합 IGF-I을 얻기 위하여 IGF-I의 B-C-A-D domain 부분을 PCR로 얻은 뒤 E. coli BL21(DE3)에 넣어 overexpression 시켰다. Ni-NTA colummn을 사용하여 순수한 재조합 단백질을 정제할수 있었다. 정제된 단백질은 SDS-PAGE 상에서 7 kDa의 단일 band를 보여 주었으며 [$^3H$]-thymidine 결합정도를 측정하는 방법으로 활성을 가지고 있음을 확인할수 있었다.

Comparative Analysis of Morphometric Characteristics of Scorpaenidae and Gobioninae

  • Measurements of closely related sets of classical and truss dimensions were analyzed to discriminate species of scorpaenidae including the dark banded rockfish, Sebastes inermis, the black rockfish, S. schlegeli, and gobioninae including the striped shiner, Pungtungia herzi, and the slender shiner, Pseudopungtungia tenuicorpa. The measurements of the dimensions were arc sin square root transformed, and compared as a function of the standard length of each species for statistical analysis. For values of the classical dimensions of the rockfish, 6 were greater for the dark banded rockfish than for the black rockfish, 1 value was smaller for the former, and for 2 values there was no statistically significant difference (P > 0.05). For values of the classical dimensions of the shiners, 9 values were greater for the striped shiner than for the slender shiner, 2 values were smaller for the former, and for 1 value there was no statistically significant difference (P > 0.01). For values of the truss dimensions of the rockfish, 6 were greater for the dark banded rockfish than for the black rockfish, 1 was smaller for the former, and for 4 values there was no statistically significant difference (P > 0.05). For values of the truss dimensions of the shiners, 13 values were greater for the striped shiner than for the slender shiner, 3 values were smaller for the former, and for 6 values there was no statistically significant difference (P > 0.01). The dimension sets used in this study may be useful as taxonomic indicators for discriminating among fish species in Korea.

조피볼락의 종묘생산에 미치는 Rotifer와 Artemia의 영양강화 효과 (Effects of Enrichment of Rotifers and Artemia on Larvae Mass Productin of Rockfish, Sebastes schlegeli)

  • 조피볼락 종묘생산시 발생하는 대량폐사를 막기 위하여 자어의 다양한 먹이생물을 공급하여 성장을 비교하였고 여러 가지 방법으로 먹이생물을 영양강화시켜 공급하여 그에 따른 조피볼락 자어의 생존율과 성장률을 비교하였다. 성게 유생은 쉽게 구할 수 있으나 조피볼락 자어의 생존율을 저하시켰고 유생의 지방산 분석 결과를 볼 때 그다지 적합한 먹이로 생각되지 않는다. Rotifer나 Artemia nauplii를 단독으로 조피볼락의 자어를 먹이로 공급하는 것보다 혼합공급하는 것이 더 효과저거인 것으로 나타났다. 그리고 rotifer나 Artemia nauplii를 ${\omega}$-yeast, Spirulina platensis나 Super Selco로 영양강화시킨후에 먹이로 공급할 경우 자어의 생존율이나 성장률에서 많은 개선을 보였다. Rotifer를 Super Selco로 영양강화를 시켰을 경우 $\omega$-yeast로 영양강화를 시킨 경우보다 높은 생존율과 성장률의 결과를 보였다. 산출 후 10일 경에 대량폐사가 발생하였으며 초기에는 rotifer가 자어의 먹이로 우수한 것으로 나타났으나 10일 경 이후부터는 Artemia nauplii가 우수한 먹이로 판단된다. 그러나 rotifer와 Artemia nauplii 혼합 공급하였을 경우 rotifer나 Artemia nauplii를 단독 공급하는 경우보다 우수한 결과를 보였다.

Distillers Dried Grain as a Partial Replacement for Wheat Flour in the Diet of Juvenile Rockfish Sebastes schlegeli

  • A 9-week feeding experiment was designed to test the effects of the dietary inclusion of distillers dried grain (DDG) on the growth performance, body composition, and antioxidant activity of juvenile rockfish Sebastes schlegeli. Five isonitrogenous and isocaloric diets were formulated to contain 0% DDG (DDG0), 7% and 14% DDG from rice (diets DDG-R7 and DDG-R14, respectively), as well as 7% and 14% DDG from rice and wheat flour (diets DDG-RW7 and DDG-RW14, respectively). Three replicate groups of juvenile rockfish averaging $68.0{\pm}0.4g$ were fed one of the diets to apparent satiation twice a day. Survival, weight gain, feed efficiency, and the protein efficiency ratio of fish were not affected by dietary DDG (P > 0.05). Proximate and amino acid compositions of the whole body were not affected by dietary DDG (P > 0.05). Plasma total protein, glucose, total cholesterol, glutamate oxaloacetate transaminase, glutamate pyruvate transaminase, phospholipid, and triglyceride were not affected by dietary DDG (P > 0.05). Radical scavenging activity on 1,1-diphenyl-2-picryl-hydrazyl (DPPH) in the plasma of rockfish fed the DDG-RW14 diet was higher than that of fish fed the DDG0 diet (P < 0.05). Hydroxyl radical scavenging activity in the liver of rockfish fed diets containing DDG was higher than that of fish fed the DDG0 diet (P < 0.05). Alkyl radical scavenging activity in the liver of rockfish fed the DDG-R7 diet was higher than that of fish fed the DDG0 diet (P < 0.05). These results suggest that DDG is a suitable ingredient for the partial replacement of wheat flour and can be used at a rate of up to 14% in the diet without incurring negative effects on the growth performance or body chemical composition of juvenile rockfish.

Molecular cloning and expression of black rockfish Sebastes schlegelii p47-phox (neutrophil cytosolic factor 1)

  • The black rockfish Sebastes schlegelii neutrophil cytosolic factor components p47 phox (phagocyte oxidase) cDNA was cloned. The sequence of the cDNA showed that rockfish p47 phox cDNA consisted of 1,952 bp contained open reading frame encoding predicted polypeptide of 420 amino acids. Additionally analysis of the p47 phox amino acid sequence showed two potential SH3 domains. The functional domains are highly conserved in many animals, though the sequence of the components of the black rockfish showed low homology with that of mammals. The deduced amino acid sequence of the black rockfish p47 phox was similar to those of the carp (60.4%), zebrafish (59,2%), rainbow trout (68.5%), xenopus (55.2%), mouse (54.2%), rabbit (54.5%), rat (53.7%), and chicken (50.9%). The expression of the rockfish p47 phox molecule was induced in peripheral blood leukocytes (PBLs) from 1 to 12 h following LPS stimulation, with a peak at 6 h after the stimulation, and which increased at 1, 3, and 12 h after treated with Poly I:C compared with the control. The rockfish p47 phox gene was expressed in various tissues of healthy fish. The level of p47 phox expression was high in the PBLs, kidney and spleen.

조피볼락과 참돔 유어에 대한 암모니아 독성 비교 (Comparison of Acute Toxicity of Ammonia in Juvenile Rockfish and Red Sea Bream)

  • 조피볼락 초기 사육시 발생하는 대량 폐사 원인의 하나로 수질악화를 들 수가 있다. 수질악화의 주요원인이 되는 어류의 주요대사물인 암모니아에 대한 조피볼락의 내성을 참돔과 비교, 분석하였다. 사육수조 내에 통기를 시키지 않을 경우 산소 부족에서 오는 스트레스로 큰 어체가 작은 어체에 비해 암모니아 독성에 대한 내성이 강한 것으로 나타났다. 조피볼락의 경우, 용존산소량이 높을 때 (통기를 시킬 때), 6 시간이나 96 시간에서의 $NH_45^+$의 반수치사농도가 용존산소량이 낮을 때 (통기를 시키지 않을 때)보다 높아서 수중의 용존산소량이 암모니아 독성을 완화시킨다는 것을 보였다. 또한, 조피볼락과 참돔의 96 시간에서의 $NH_4^+$ 와 비이온화된 $NH_3$ 의 반수치사농도 결과를 비교해 볼 때, 조피볼락은 참돔에 비해 암모니아에 대한 내성이 약한 것으로 나타났다.

볼락류 Sebastes melanops 아가미에 단생 흡충류 Microcotyle sebastis의 부착에 관하여 (Attachment of the Microcotyle sebastis (Monogenea: Microcotylidae) to the Gills of Black Rockfish, Sebastes melanops)

  • 2001년 9월 19일부터 동년 10월 3일까지 Hartfield Marine Science Center Aquarium에서 구입한 볼락류 Sebastes melanops 7마리의 아가미에 부착한 단생 흡충류를 조사하였다. 파악기와 정소 수를 근거로 한 불락류(S. melanops)의 단생 흡충류는 Microcotyle sebastis였다.M. sebastis의 감염률은 42.9%이었고 감염강도는 1-11(평균 2.0)을 나타내었다. 단생 흡충류의 감염부위는 주로 감염어 아가미의 제 2차와 제 3차 새변이었다. 따라서, 아가미의 제 2차와 제 3차 새변 표면은 M. sebastis의 분포에 영향을 끼치고 있음을 알 수 있었다.

Critical Thermal Maximum (CTM) of Cultured Black Rockfish, Sebastes schlegeli

  • The critical thermal maximum (CTM) of black rockfish, Sebastes schlegeli, was evaluated. Black rockfish were acclimated at $24^{\circ}C$, and then exposed to temperatures from 24 to $33^{\circ}C$. Black rockfish were kept in constant darkness and subjected to a gradual temperature increase $(1 ^{\circ}C\;12^{-1})$. The oxygen consumption rate (OCR) was measured using an automatic intermittent­flow-respirometer (AIFR) during the exposure period (from 119.3 to 143.5 h). The OCR increased from 94.5 to 214.2mL $O_2 kg^{-1}\;ww\;h^{-1}$ as the temperature rose from 24 to $29.4-30.9^{\circ}C$. Subsequently, the OCR increased abruptly, reaching 245.8-412.7mL $O_2 kg^{-1}\;ww\;h^{-1}$ at $32^{\circ}C$. This study suggests that the CTM for black rockfish is $29.4-30.9^{\circ}C$ when temperature is increased at $1^{\circ}C\;12h^{-1}$ following acclimation at $24^{\circ}C$.

  • 조피볼락 Sebastes schlegeli 자어와 점농어 Lateolabrax sp. 자어를 대상으로 기아시 성장과 간세포 핵 크기 변화를 조사하였다. 전장과 습중량 성장에 있어 조피볼락과 점농어의 간세포 핵 크기는 첫 먹이 섭취 자어의 영양상태와 연관성을 보였다. 본 연구의 조피볼락과 점농어 자어에서의 기아시 간세포 핵 크기 조사 결과는 조피볼락과 점농어에서 초기 영양상태 판명의 지표가 될 수 있으며 초기 먹이 공급 공정시 내생적 영양상태에서 외생적 영양상태로의 효율적 전환을 가능케 할 것으로 사료된다.
