Gene Editing for Major Allergy Genes using Multiplex CRISPR-Cas9 System & Prime Editing in Peanuts (Arachis hypogaea L.)

  • Recently, food-induced allergies have emerged as major global concerns. In the past ten years, it has doubled in western nations, and it has also increased in Asia and Africa. In many cases of food allergy, peanut allergy is prevalent, typically permanent, and frequently life-threatening. Therefore, we utilized gene editing techniques on the three major allergen genes in peanuts, Ara h 1, Ara h 2, and Ara h 3. Using gibson assembly and golden gate assembly, we created two vectors, the gRNA-tRNA array CRISPR-Cas9 system and Prime-editing. Using LBA4404 strain and agrobacterium-mediated transformation, the vectors were transferred to two elite Korean peanut lines. After co-cultivation and tissue culture, we extracted the tissue cultured peanut DNA amplified the hygromycin resistance gene and Cas9 gene in the T-DNA region. The integration of the T-DNA region into the host genome was demonstrated by the presence of a specific band in some samples. There have only been a few reported peanut gene editing studies. So, this study will contribute to peanut allergy and gene editing research.

Characterization of the Effects of Relative Humidity and Bed-depth on $CO_2$ Capture for Maximizing the Utilization Rate of Soda Lime Sorbent

  • The advanced method for $CO_2$capture is currently one of the most important environmental issues in worldwide and it is therefore necessary to have available technologies, which minimize the discharge of $CO_2$ including Carbon-14 from nuclear facilities into the atmosphere. A key aspect of this work is to provide the technically principal data required to improve a $CO_2$ removal system for the utilization of regenerative sorbent use, specifically include suggestions regarding its modified column design (parallel dual-bed assembly), stop-restart operation and the economic feasibility of sorbent use. The removal performance of soda lime and the effects of relative humidity (RH) and packing bed-depth (BD) on $CO_2$ removal were investigated. In a single-bed, it revealed that the utilization of soda lime for $CO_2$ removal at line velocity of 13 cm/sec and bed depth of 12 cm increased with the increased relative humidity up to 85%. However, in the parallel dual-bed assembly applied with the stop-restart operation, a maximum utilization rate of soda lime for $CO_2$ removal was obtained even at 55% of RH and 8 cm of BD, specifically the utilization rate of soda lime by using this $CO_2$ removal assembly was about two-fold superior to that in a single-bed.

De novo gene set assembly of the transcriptome of diploid, oilseed-crop species Perilla citriodora

  • High-quality gene sets are necessary for functional research of genes. Although Perilla is a commonly cultivated oil crop and vegetable crop in Southeast Asia, the quality of its available gene set is insufficient. To construct a high-quality Perilla gene set, we sequenced mRNAs extracted from different tissues of Perilla citriodora, the wild species (2n = 20) of Perilla. To make a high-quality gene set for P. citriodora, we compared the quality of assemblies produced by Velvet and Trinity, the two well-known de novo assemblers, and improved the de novo assembly pipeline by optimizing k-mers and removing redundant sequences. We then selected representative transcripts for loci according to several criteria. The improved assembly yielded a total of 86,396 transcripts and 38,413 representative transcripts. We evaluated the assembled transcripts by comparing them to 638 homologous Arabidopsis genes involved in fatty acid and TAG biosynthesis pathways. High proportions of full-length genes and transcripts in the assembled transcripts matched known genes in other species, indicating that the P. citriodora gene set can be applied in future functional studies. Our study provides a reference P. citriodora gene set for further studies. It will serve as valuable genetic resource to elucidate the molecular basis of various metabolisms.

Detection of Potential Invalid Function Pointer Access Error based on Assembly Codes (어셈블리어 코드 기반의 Invalid Function Pointer Access Error 가능성 검출)

  • Though a compiler checks memory errors, it is difficult for the compiler to detect function pointer errors in code level. Thus, in this paper, we propose a method for effectively detecting Invalid function pointer access errors, by analyzing assembly codes that are obtained by disassembling an executable file. To detect the errors, assembly codes in disassembled files are checked out based on the instruction transition diagrams which are constructed through analyzing normal usage patterns of function pointer access. When applying the proposed method to various programs having no compilation error, a total of about 500 potential errors including the ones of well-known open source programs such as Apache web server and PHP script interpreter are detected among 1 million lines of assembly codes corresponding to a total of about 10 thousand functions.

  • This paper deals with the characteristics evaluations of PM magnetization using stator coil in a Post-Assembly Line Start Permanent Magnet Motor (LSPMM) using a coupled Finite Element Method (FEM) and Preisach modeling, which is presented to analyze the magnetic characteristics of permanent magnets. The focus of this paper is the characteristics analysis relative to magnetizing direction and quantity of permanent magnets due to the eddy current occurring in the rotor bar during magnetization of Nd-Fe-B.

  • This paper proposes a design methodology for customer, assembly and disassembly considering recycling. The disign process starts with the identification of customer needs, which are in turn converted into functional requirements. The concepts of Design for Customer(DFC), Design for Assembly(DFA), Design for Variety(DFV) and Design for Disassembly(DFD) are considered in the product design phases in order to decreas production variety and mass customization. And, a new module generation approach is developed for rearranging and clustering parts and subassemblies for disassembly and recycling. Based on the result of the module generation, a new configuration methodology is suggested to minimize the disassembly time or number of disassembly operations for recycling.

  • This paper deals with a case study about productivity improvement in medium and small-sized manufacturing factories. It is popular to improve the productivity of lines by shortening the production cycles and assigning multi jobs to workers. We analyze the production lines and improve the productivity in the case study through multi-functional workers and shortened production lines. Based on the case study, we propose some guidelines to improve the productivity of production lines.

  • The electron cyclotron resonance plasma source with a belt-type magnet assembly (BMA) is designed for effective plasma confinements. For characterizing the plasma source, the plasma parameters are measured by Langmuir probe. However, the plasma parameters and the motion of charged particles near the ECR zone are not easy to diagnostics, because of the high plasma density and temperature. Thus, as an alternative method, the electromagnetic simulation of the plasma source has been performed by using three-dimensional particle-in-cell and Monte Carlo collisional (PIC-MCC) simulation codes. For considering the limitation of simulation resources and time, the periodic boundary condition is applied and the coulomb collision is neglected. In this paper, we present the results of 3D PIC simulations of ECR plasmas with BMA and we compare them with the experimental results.
