In vitro Effects of the Complex of Korean Citrus junos and Medicinal Herbs on the Cell Growth in CCD-986sk Human Fibroblast Cell Line

  • In order to investigate whether or not CCD-980sk cell line can be affected by Korean Citrus junos and medicinal herbs, we examined the MTT assay when we treated Korean Citrus junos and medicinal herbs in CCD-986sk human fibroblast cell line. The samples that added complex of grinded extracts of Korean Citrus junos and boiling-water extracts from Korean medicinal herbs were tested toy cell proliferation activity by means of a modification of the MTT assay. Among mixture of Citron 3 (Citron 3, less mellowed citron which was ripened for three months) and boiling-water extracts, the group Citron 3+Phellinus linteus showed significantly strong cell proliferation activity. And among mixture of Citron 4 (Citron 4, completely mellowed citron which was ripened for four months) and boiling-water extracts, the group Citron 4+Cordyceps militaris and Citron 4+Phellinus linteus showed significantly strong cell proliferation activity, respectively. These results suggest that complex of Korean Citrus junos and medicinal herbs could be an excellent candidate toy protection of human skin aging.

저장온도 및 Blanching 처리가 유자의 저장성에 미치는 영향 (Effects of Storage Temperature and Blanching Treatment on the Storage Stability of Citron (Citrus junos))

  • 저장온도 및 blanching 처리가 유자의 저장성에 미치는 영향을 밝혀 유자의 적정 저장조건을 설정함으로서 유자의 저장성을 향상시키고자 하였다. 유자의 저온장해 현상은 $0^{\circ}C{\sim}5^{\circ}C$ 범위에서 나타나지 않았고, 유자의 호흡은 저장시간에 따라 증가하는 climacteric 특성으로 분류될 수 있었고 유자 호흡량의 $Q_{10}$값은 $2.3{\sim}2.4$의 범위였다. 유자의 부패율은 blanching 처리보다는 저장 온도가 유자의 저장성에 영향이 컸다. 유자의 중량감소율은 처리구에 따라서 중량감소율의 변화는 크지 않았으나 온도에 따라서는 그 영향이 지대하였다. 유자 저장중 과실의 맛을 결정하는 당산비는 $3^{\circ}C$에서 완만하게 증가하고 $20^{\circ}C$에서 급격히 증가하다 감소하였는데, 이는 산함량의 증가보다는 당도의 증가에 의한 영향이 큰 것으로 판단되었다. 유자저장중 갈변도는 과육보다 껍질부분의 갈변정도에 따라서 유자전체의 갈변에 크게 영향을 미치고 있으므로 껍질부분이 갈변의 지표가 됨을 확인할 수 있었다.

유자 과피를 첨가하여 제조한 진양주의 품질 및 항산화 특성 변화 (Effect of Citrus junos Peel on the Quality and Antioxidant Activity of Traditional Rice Wine, Jinyangju)

  • 찹쌀에 유자 과피를 각각 0, 10, 20, 30, 40% 첨가하여 제조한 진양주의 품질 및 생리활성 특성의 변화를 조사하였다. 유자 과피 첨가량의 증가에 따라 pH는 감소되었고 총산의 함량은 증가된 것으로 나타났다. 총당의 함량은 유자 과피의 첨가량의 증가에 따라 증가하였고 알코올 함량은 감소하였다. 색도 중 L값은 유자 과피의 첨가량이 증가에 따라 증가되었으나 a와 b값은 감소한 것으로 나타났다. DPPH 자유라디칼 소거 활성은 유자 과피의 첨가량의 증가에 따라 증가된 것으로 나타났다. 관능검사 결과 전체적 기호도에서 20% 유자 과피를 첨가하여 제조한 진양주가 제일 우수한 것으로 나타났다. 이화학적 특성, 항산화 특성, 관능적 특성으로 보아 20% 유자 과피를 첨가하여 진양주를 제조하는 것이 적합하다고 생각된다.

Free Radical Scavenging Activity of Butanol Fraction from the Fruit of Citrus junos

  • In this study, we investigated the free radical [1,1-diphenyl-2-picrylhydrazyl (DPPH), hydroxyl radical (${\cdot}OH$) and superoxide anion (${O_2}^-$)] scavenging activity of MeOH extract and 3 fractions of Citrus junos. Of the tested fractions, the BuOH fraction showed the strongest DPPH scavenging activity, showing the $IC_{50}$ values of 63.4 mg/mL. Therefore, we continuously carried out DPPH, ${\cdot}OH$ and ${O_2}^-$ scavenging activity tests of BuOH fraction of Citrus junos. The BuOH fraction of Citrus junos inhibited DPPH radical to 97.5% at a concentration of 1000 mg/mL and the scavenging activities were increased concentration-dependently. In addition, BuOH fraction from Citrus junos also scavenged ${\cdot}OH$ in a concentration dependent manner from 5 to 1000 mg/mL. Furthermore, BuOH fraction showed about 56% ${O_2}^-$-scavengimg activity at 25 mg/mL concentration but, the scavenging activities were not enhanced in a dose dependent manner. The present results suggest that BuOH fraction of Citrus junos would have the protective potential from oxidative stress induced by free radicals.

Effects of Korean Citrus junos and Medicinal Herbs on Liver Protection and Lipid Metabolism of Alcohol Fed Rats

  • In order to investigate whether or not the alcohol-treated rat liver cells can be protected by Korean Citrus junos and medicinal herbs, We compared the serum biochemistry of rats administered both alcohol and the complex of Korean Citrus junos and medicinal herbs to control rats treated with alcohol alone. The activities of Aspartate aminotransferase (AST) and Alanine aminotransferase (ALT) in the citron 3 (Citron 3, less mellowed citron which was ripened for three months)+Phellinus linteus, Alnus japonica, Dendropanax morbifera and citron 4 (Citron 4, completely mellowed citron which was ripened fer four months)+Phaseolus radiatus, Cordyceps militaris group were significantly low when compared with the negative control group (p<0.05). The levels of triglyceride (TG) in all experimental groups were significantly lower than the negative control group (p<0.05). The concentrations of total cholesterol in the citron 3+Phellinus linteus, Atnus japonica, Dendropanax morbifera and citron 4+Phaseolus radiatus, Cordyceps militaris, Phellinus linteus were lower than the negative control group (p<0.05). The activities of alcohol dehydyogenase (ADH) in all experimental groups were significantly high when compared with the normal control group (p<0.05). These results suggest that the complex of Korean Citrus junos and medecinal herbs could be an excellent candidate for protecting vat liver cell damage induced by alcohol.

Korean Medicinal Herb Extracts Inhibit Melanin Formation in Clone M-3 Mouse Melanocyte Cell Lines

  • In order to search for anti -melanin formation agents from Korean medicinal herbs, we selected 21 Korean medicinal herbs, based on a review of Korean traditional medicine books and the recommendations of Korean traditional medical doctors. We tested for inhibition effect of melanin pigmentation of Clone M-3 mouse melanocyte cell lines when we treated the extracts of 21 medicinal herbs in the mouse melanocyte cell lines, respectively. Among 21 medicinal herb extracts, 5 extracts showed a inhibition effect of melanin formation. The sample Phaseolus radiatus L, Cordyceps militaris, Pinellia ternata, Phellinus linteus and Citrus junos Tanaka showed a significantly little formation of melanin pigments compared with control groups. Especially extract of Citrus junos Tanaka was more potent than the others. These results suggest that extract of Korean Citrus junos may represents an excellent candidate for inhibition of melanin pigmentation at in vitro level.

Citrus junos Fractions Decrease Alcohol-induced Liver Damage and Influence Lipid Metabolism in Alcohol-fed Rats

  • The effect of treatment with Citrus junos fractions (citron 3W, citron 3H, citron 4W and citron 4H) upon rat hepatocytes exposed to alcohol was investigated. We compared the serum biochemistry of rats administered both alcohol and Citrus junos fractions to control rats treated with alcohol alone. The effects of Alanine amino transferase (ALT) were significantly lower in the citron 3H extract group compared with the negative control group (p<0.05) and other experimental groups were not significantly low but a little low compared to negative control group. The levels of triglyceride (TG) were significantly low in all experimental groups compared with negative control group. Especially triglyceride level of citron 3H was lowest near to normal control group. The concentration of total cholesterol was significantly high in negative groups compared with normal control group but in all experimental groups, the concentration of total cholesterol was similar to that of negative control group. Total cholesterol of the citron 4W group was somewhat low compared with negative control group. In contrast, activities of alcohol dehydrogenase (ADH) were significantly higher in all experimental groups compared with the negative control (p<0.05) group. These data suggest that Citrus junos fractions may represents an excellent candidate for protection of rat hepatocytes from alcohol-mediated damage.

신육성 왜성 한국유자 (Citrus junos)의 밀식재배 및 조기 다수확을 위한 생리적 연구 (Physiological Study for Dense Planting and Early High Yielding Potential of New Korean Dwarf Yuzu (Citrus junos Sieb. Ex Tanaka))

  • 유자(Citrus junos)에 있어서 수관내 위치별 엽, 엽면적, 엽중의 특성차이와 정아의 발생상태, 화아분화와 과실 품질에 미치는 기초자료들을 얻고져 실험을 실시하였던 바 결과는 다음과 같다. 1. 동, 서, 남, 북의 방향별 엽수, 엽면적, 엽중에 있어서는 유의차가 없었다. 2. 수관의 상하간에는 엽수와 엽면적은 유의차가 었었다. 3. 정아의 횡경과 종경은 방향별로는 차이가 없었지만, 상하간에는 유의한 차이를 나타내었다. 4. 투과광도와 엽 및 엽면적과의 상관관계는 광도 감소율에 따라 엽의 엽비중이 가장 큰 영향을 받았으며, 단과지당 전체엽중, 평균 엽중, 단과지 정아의 횡경, 종경도 높은 상과관계를 나타내었다. 5. 화아분화에 차광은 비효과적이었으며, GA처리도 같은 경향을 나타내었다. 6. 적과는 1과당엽수를 40잎 이상으로 하는 것이 화아분화율과 과실의 품질을 향상시킬 수 있었다.

유자의 항 Influenza 바이러스 A형 활성에 관한 연구 (Study on the Anti-influenza Virus A type Activity of Citrus junos)

  • To evaluate anti-influenza virus activity of 113 specimens of Korean traditional medicine both water and methanol extracts were examined using haemagglutination inhibition test. The water extract from Citrus junos was found to inhibit influenza virus A/Taiwan/l/86(H1N1). The survival rates of virus were determined by in situ cellular enzyme-linked immunosorbent assay. The water extract of Citrus junos was fractionated by chromatographic separating using Amberlite XAD-4, 40% MeOH and 60% MeOH layer had antiviral activity. The half inhibition concentration $(IC_{50})$ of 40% MeOH layer on survival of influenza virus was $MIC>361.5{\mu}g/ml$ and $IC_{50}$ value of fr. 40-4 fractionated from 40% MeOH layer was $677.19{\mu}g/ml$. These results suggested that the fractions of Citus junos have potent anti-influenza A virus activity.

유자 및 매실추출물 첨가가 천연건조 한우 육포의 품질특성에 미치는 영향 (Effect of natural antioxidant extracted from Citrus junos seib. or Prunus mume. on the quality traits of sun-dried Hanwoo beef jerky)

  • The objective of the study was to evaluate the quality characteristics of sun-dried Hanwoo beef jerky added with the extract of Citrus junos seib. or Prunus mume. Hanwoo beef shank muscles were sliced, marinated, and sun-dried at $25^{\circ}C$, relative humidity of 35%. The physicochemical quality and microbiological safety of the Hanwoo beef jerky aerobically packaged were analyzed during the storage of $25^{\circ}C$. The moisture content of beef jerky with Citrus junos seib. was the lowest among the treatments after 20 d. Citrus junos seib. and Prunus mume jerky after 10 d had significantly lower aw than those after 0 and 20 d (p<0.05). The pH values of jerky generally ranged from 5.76 to 5.84. The pH value of Prunus mume jerky was significantly higher than those of other jerky samples (p<0.05). Prunus mume jerky showed significantly lower TBARS value than the others after 20 d (p<0.05). Citrus junos seib. jerky showed a significantly lower lightness ($L^*$), redness ($a^*$) and yellowness ($b^*$) than the others during the storage (p<0.05). Prunus mume samples after 10 d had significantly higher $L^*$, $a^*$, and $b^*$ values than the others (p<0.05). With regard to sensory properties, Citrus junos seib. and Prunus mume jerky showed significantly higher flavor and overall acceptability scores than the control (p<0.05). The extracts of Prunus mume will be used in sun-dried Hanwoo beef jerky as a natural agent to retard lipid oxidation and to improve meat color.
