Evaluation and Comparison of Contrast to Noise Ratio and Signal to Noise Ratio According to Change of Reconstruction on Breast PET/CT (Breast PET CT 영상 재구성 변화에 따른 대조도 대 잡음비와 신호 대 잡음비의 비교평가)

  • The purpose of this study was to measure contrast to noise ratio (CNR) and signal to noise ratio (SNR) according to change of reconstruction from region of interest (ROI) in breast positron emission tomography-computed tomography (PET-CT), and to analyze the CNR and SNR statically. We examined images of breast PET-CT of 100 patients in a University-affiliated hospital, Seoul, Korea. Each patient's image of breast PET-CT were calculated by using ImageJ. Differences of CNR and SNR among four reconstruction algorithms were tested by SPSS Statistics21 ANOVA test for there was statistical significance (p<0.05). We have analysis socio-demographical variables, CNR and SNR according to reconstruction images, 95% confidence according to CNR and SNR of reconstruction and difference in a mean of CNR and SNR. SNR results, with the quality of distributions in the order of PSF_TOF, Iterative and Iterative-TOF, FBP-TOF. CNR, with the quality of distributions in the order of PSF_TOF, Iterative and Iterative-TOF, FBP-TOF. CNR and SNR of PET-CT reconstruction methods of the breast would be useful to evaluate breast diseases.

  • Recently, it has been a big issue to distinguish the dried roots of Cynanchum wilfordii and C. auriculatum in health functional food market. The original plant species of Cynanchi Wilfordii Radix belong to the Asclepiadaceae family is differentially described in the national pharmacopoeia of Korea, China and Japan. Owing to the morphological similarities of the dried roots of this plant to those of C. auriculatum, which is often misidentified in Korean herbal medicine marketplace, distinguishing these two species is exceedingly difficult. The purpose of this study was to compare the conventional-PCR with the real-time PCR for detection of C. wilfordii and C. auriculatum DNA. We also tried to realize a quantitative real-time PCR assay using species-specific matK primers, which allowed us to estimate the ratio of C. willfordii and C. auriculatum using varying ratios of mixed genomic DNA template from the two species. The differentiation of intentional and unintentional mixture in this study would be applied to food safety management and can be helpful for protection of consumer's right and cultivators.

Investigation of the Transfer Rate of Zearalenone in Herbal Medicines to Their Decoction (한약재 중 제랄레논의 탕액으로 이행률 조사)

  • The objective of this study was to establish an analytical method to detect zearalenone (ZEA) in herbal medicines and their decoctions and investigate the ZEA transfer rate from raw materials of herbal medicines to their decoctions. Herbal medicines (Trichosanthis Semenm, Eucommiae Cortex, Rubi Fructus) spiked with a known concentration of ZEA were presoaked or unsoaked (as a pretreatment) and boiled for 3 h at $100^{\circ}C$ or autoclaved for 1 h at $121^{\circ}C$. The decoction and the remnants were separated, cleaned up with an immunoaffinity column, and analyzed using HPLC. Recoveries for decoctions and remnants were 68.39-83.68% and 72.91-80.25%, respectively. ZEA was not detected in the decoction, whereas it was found in the remnants. Although ZEA in the raw material of herbal medicines was not transferred into the decoction during heating and autoclaving, the continuous monitoring for ZEA in raw herbal medicines should be carried out for the safe ingestion and utilization of herbal medicines.

Treatment of Patients with Cancer in a Secondary Hospital in Korea (국내 일개 2차 병원의 암환자 치료 실태)

  • Purpose: This study aims to investigate treatment of cancer patients at a secondary hospital. Methods: A retrospective analysis was performed with electronic medical records of cancer patients admitted to a secondary hospital from January 1, 2009 through September 31, 2017. Results: A total of 223 patients were studied. Sixty-nine patients were hospitalized for supportive care after receiving a surgery, chemotherapy, and radiotherapy at a tertiary hospital, 58 patients for other supportive care, 53 patients for symptom control, 16 patients with a decision not to take active cancer treatment, and 27 patients for treatment of cancer that was diagnosed during their hospital stay. Among 75 patients who were discharged to other institutions, 50 were transferred to tertiary hospitals, 10 to long-term care hospitals, eight to hospice hospitals, four to nursing homes and two to secondary hospitals. Comorbidities were found in 120 patients (53.8%). For patients who consulted with more than one department, more consultations were for non-cancer diseases than cancer. Seventy-three patients had a do-not-resuscitate order. Conclusion: For treatment of cancer patients, it is needed to establish a cooperation system among medical institutions and provide comprehensive management including treatment of comorbidities.

Anti-oxidant, Anti-inflammatory and Anti-cancer Effect of Methanol Extract of Pogostemon cablin (광곽향 메탄올 추출물의 항산화, 항염증 및 암세포 증식 억제 효과)

  • In the present study, the substance that show anti-proliferation of cancer cells as well as anti-oxidant and anti-inflammatory effect was searched. As a results, the methanol extract of Pogostemon cablin (P. cablin), is a well-known herb for traditional medicine in Korea and China for treating the digestive disorders, less of appetite, vomiting and diarrhea, inhibited the growth of various cancer cells such as A549, HepG2, MCF7 and HT29 cells. Cytotoxic effect of methanol extraction of P. cablin was excellent in A549 cells. P. cablin extract induced cell cycle arrest at G1 phase of A549 in a dose dependent manner. And it induced phosphorylation of p38 and Cdc25A and reduced expression of Cdc25A, Cdks, Cyclins and phospho-Retinoblastoma (Rb) proteins. Therefore, P. cablin extract seems to act through the p38 - Cdc25A - Cdk - Cyclin - Rb pathway in A549 cells. In addition, P. cablin extract showed anti-oxidant effect by DPPH free radical scavenging assay and anti-inflammation effect by inhibition of lipopolysaccharide (LPS)-induced nitric oxide (NO) in RAW 264.7 cells in a dose-dependent manner. Taken together, these results suggest that P. cablin may be used as not only candidate materials for anti-cancer, anti-inflammatory and anti-oxidant, moreover, it would be possible utilized in various health functional food materials.

Inhibitory Effect of Ethanol Extract of Monascus-fermented Red Yeast Rice on Proinflammatory iNOS and COX-2 Protein Expression in LPS-stimulated RAW 264.7 Macrophage Cells (Monascus sp. BHN-MK로 발효생산한 홍국 에탄올 추출물의 Raw 264.7 대식세포에 있어 친-염증성 iNOS와 COX-2 단백질 발현 억제 효과)

  • Red yeast rice has been extensively used as a food and traditional medicine for thousands of years in Korea. Monascus produces many secondary metabolites during its growth, including pigments, monacolins, and γ-aminobutyric acid. Some metabolites, specifically monacolin K, γ-aminobutyric acid, and dimerumic acid, have been reported to lower cholesterol and blood pressure because of certain antioxidant effects. This study investigated the total phenolic content of ethanol extract from red yeast rice fermented with Monascus sp. BHN-MK and its anti-inflammatory effect on LPS-stimulated RAW 264.7 macrophage cells. To assess its anti-inflammatory effect, the inhibitory activity of the ethanol extract on LPS-induced NO production and expression levels of iNOS and COX-2 proteins in macrophage cells were measured. Its total polyphenol content was higher than that of ordinary non-fermented rice. Its NO production inhibition activity was comparable to that of the negative control group treated with LPS at a concentration of 400 ㎍/ml. Western blot revealed a significant decrease in the inhibition of iNOS and COX-2 protein expression at concentrations of 400 and 800 ㎍/ml, respectively. Red yeast rice ethanol extracts exerted the strongest anti-inflammatory effects. The results indicate that red yeast rice could be used as a functional cosmetic and anti-inflammatory material.

Component of oxytetracycline on exfoliation of abalone, Haliotis discus hannai (옥시테트라사이클린의 전복박리 성분)

  • Exfoliation of fouling abalone, Haliotis discus hannai from shelters by commercial oxytetracycline (OTC) was observed in culture farms. In the present study, different components of OTC for exfoliation of abalone were investigated to understand how to work. Abalone were detached from shelter in 14,000 ppm of commercial OTC (main ingredients of OTC: OTC-hydrogen chloride (HCl), 50%; glucose, 49%; blue pigment, <1%), but not below 8,000 ppm. A 95% of exfoliation rate was observed in OTC-HCl (7,000 ppm, pH 2.8) but no exfoliation in OTC-HCl (7,000 ppm, pH 5.0), glucose (7,000 ppm) or blue pigment (140 ppm). Moreover 100% exfoliation rate was observed in HCl-seawater of pH 2.8. These results indicate that HCl is the component resulting in exfoliation of the fouling abalone. Abalone was detached in HCl solution (pH 2.5-3.2) within 2 min. However, a lower pH and longer treatment resulted in delayed recovery of the detached abalone. Thus, exfoliation of fouling abalone can be achieved by a low pH treatment with cautious handling.

Monitoring of viruses in cultured walleye pollock Gadus chalcogrammmus (양성 중인 명태(Gadus chalcogrammus)의 바이러스 모니터링)

  • This study was conducted to monitor the prevalence of viral hemorrhagic septicemia virus (VHSV), nervous necrosis virus (NNV) and marine birnavirus (MABV) in cultured walleye pollock Gadus chalcogrammus by RT-PCR. All of the viruses tested were not detected by one-step PCR in 62 spleen sample sets, except for NNV in one brain sample set (1/55). By two-step PCR, VHSV was detected in 51.6%(32/62) and NNV was detected in 1.6%(1/62) spleen sample set, but MABV was not detected. In the brain sample sets, the detection rate of NNV was 3.6%(2/55). VHSV and NNV were detected for the first time in cultured walleye pollock in this study. However, the titers of viruses in these sample sets are thought to be very low, because most of the positive sample sets were detected by two-step PCR and none of the fish showed any clinical symptoms of each virus. Continuous monitoring, subsequent virus isolation and validation of carrier fish will be necessary.

  • Two steroidal saponins were isolated from MeOH extract of the rhizomes of Asparagus oligoclonos. Using the spectroscopic techniques of $^1H,\;^{13}C$ NMR and 2D NMR, they were identified as $3-O-[{\beta}-D-glucopyranosyl-(1{\rightarrow}2)-{\beta}-D-glucopyranosyl]-(25S)-spirostan-3{\beta}-ol$ and $3-O-{{\beta}-D-glucopyranosyl-(1{\rightarrow}2)-[{\beta}-D-xylopyranosyl-(1{\rightarrow}4)]-{\beta}-D-glucopyranosyl}-(25S)-spirostan-3{\beta}-ol$, respectively. They were first isolated from A. oligoclonos. The in vitro antibacterial activities as well as antibacterial spectrums against 20 bacteria strains were investigated with MIC test. Both saponins inhibited the growth of 10 bacteria strains at the concentration of$100\;{\mu}g/ml$.

  • The repeated silica gel colum chromatographies of EtOAc fraction, showing anticonvulsant activity, obtained from MeOH extracts of Schizandra chinensis B. fruits led to isolation of a sesquiterpenoid, four lignans and a sterol glycoside. Their chemical structures were determined to be chamigrenal, gomisin A, gomisin H, gomisin N. schizandrin and daucosterol. Among them, schizandrin and daucosterol inhibited GABA degrative enzymes, succinic semialdehyde dehydrogenase and succinic semialdehyde reductase, respectively. It is postulated that the schizandrin and daucosterol are able to elevate the neurotransmitter GABA levels in central nervous system by inhibitory action on GABA degrative enzymes and act as anticonvulsant drugs.
