Effect of Growth Conditions on Saponin Content and Ginsenoside Pattern of Panax ginseng

  • For the elucidation of significance of saponin as quality criterion of ginseng ginsenoside content(GC) and ginsenoside pattern similarity(GPS) by simple correlation were investigated in relation to red ginseng quality factors, age, plant part, harvest season, mineral nutrition, soil physical characteristics, growth light and temperature, shading material, growth location, physiological disease and crop stand through survey of ginseng plantstions, field experiments, water culture and phytotron experiments. Effect of tissue culture was also reviewed. GC was negatively correlated with good quality of red ·ginseng and positively with bad quality. Age did not show any consistency with GC but GPS was less with the increase of age difference. GPS was less or not significant between taproot that is lowest in GC and epidermis highest, and significant between leaf and taproot. Harvest season marked with the lowest GC and Pattern was also different. Nutrient imbalance, the increase of hazardous soil nutrient and physical condition to growth increased GC, but GPS was little different. The higher the growth lights intensity and temperature the higher the GC but GPS was little changed. Root rust increased GC, but root scab decreased it. Sponge-like and inside cavity phenomena increased GC. Ginsenoside pattern of cultured tissues and rootlet showed great variation. These results strongly indicate that there are optimum saponin content and ginsenoside pattern and that these are accomplished under the optimum growth condition.

  • This study was conducted to elucidate the effects of light intensity using polyethylene (p.E) net shading on the specific gravity, red ginseng quality, sugar and saponin contents of ginseng root. The specific gravity significantly increased in the ginseng roots grown under the P.E net shading as compared with that of common straw shading. The red ginseng quality under the P.E net shading was improved in order of 10, 5, 15, 20, 30% of light intensity and the inside cavity and inside white part decreased remarkably as compared with those of common straw shading. The ginseng roots grown under the P.E net shading at 10% and 15% light intensity showed a significant increase in the total sugar content but a significant decrease in the reducing sugar content at 15% light intensity as compared with those of common straw shading. The amount of total saponin of ginseng roots was increased under the P.E net shading at high light intensity as compared with that of common straw shading and the ginseng roots grown under the P.E net shading at 10% light intensity showed an increase in the diol group saponin but the ratio of PT/PD was decreased. Extract contents of ginseng root under the P.E net shading was higher than those of common straw shading and the roots grown under the P.E net shading at 15% and 20% light intensity resulted in a remarkable increase in extract contents.

Antiviral Effect of Korean Red Ginseng Extract and Ginsenosides on Murine Norovirus and Feline Calicivirus as Surrogates for Human Norovirus

  • Korean red ginseng has been studied various biological activities such as immune, anti-oxidative, anti-microbial, and anticancer activities but antiviral mechanism needs further studies. In this study, we aimed to examine the antiviral effects of Korea red ginseng extract and ginsenosides on norovirus surrogate, including murine norovirus (MNV) and feline calicivirus (FCV). We evaluated the pre-, co-, and post-treatment effects of Korean red ginseng (KRG), ginsenosides $Rb_1$ and $Rg_1$. To measure the antiviral effect and cytotoxicity of KRG extract, and ginsenosides $Rb_1$ and $Rg_1$, we treated Crandell-Reese Feline Kidney for FCV or RAW264.7 cells for MNV with concentrations of 0, 5, 6.7, 10, 20 ug/mL total saponin. There was cytotoxic effect in the highest concentration 20 ug/mL of KRG extract so this concentration was excluded in this study. The FCV titer was significantly reduced to 0.23-0.83 $log_{10}$ 50% tissue culture infectious dose ($TCID_{50}$)/mL in groups pre-treated with red ginseng extract or ginsenosides. The titer of MNV was significantly reduced to 0.37-1.48 $log_{10}$ $TCID_{50}$/mL in groups pre-treated with red ginseng extract or ginsenosides. However, there was no observed antiviral effect in groups co-treated or post-treated with KRG and its constituents. Our data suggest that KRG extract has an antiviral effect against norovirus surrogates. The antiviral mechanisms of KRG and ginsenosides should be addressed in future studies.

Analysis of major ginsenosides in various ginseng samples

  • The contents of major ginsenosides (ginsenosides Rb1, ginsenoside Rc, ginsenoside Rd, ginsenoside Re, ginsenoside Rf, and ginsenoside Rg1) in ginseng cultivated in different areas in Korea, ginseng that underwent different cultivation processes and ages, and ginseng cultivated in different countries were determined using high-performance liquid chromatography equipped with UV/VIS detector. Ginsenoside Rc was the most abundant ginsenoside in all different ginseng samples. The highest total concentration of major ginsenosides was found in the ginseng cultivated in Jinan (0.931 mg/g) and 4-year grown red ginseng (1.785 mg/g). Major ginsenosides were the most abundant in Korean ginseng (1.264 mg/g), compared to those in Chinese and American ginseng. The results of this study showed the different contents of major ginsenosides in the ginseng samples tested and emphasized which sample could contain high yield of ginsenosides.

  • A rotting bacterium was isolated from decayed root of ginseng (Panax ginseng Meyer), cultured purely, and its pathogenicity was confirmed by reinoculation test. The strain causing ginseng root rot was identified as Pseudomonas fluoresens biotype II. The strain was somewhat different from P.marginalis and P.talaasii, considering the number of flagella, pathotype and ability of indole production. The strain did not exhibit pathogenicity to other plants tested, such as red kidney bean(Phasolus vulgaris L.), soy bean (Glycine max Merr.), cucumber (Cucumis sativus L.) lettuce (Lactuca sativa L.) and cowpea bean (Vigna sinensis Savi.).

고려인삼의 유래 및 효능의 서지학적 고찰 (Bibliographic consideration on the efficacy and the origin of Korean ginseng)

  • 고려인삼은 대한민국의 대표적인 특산물로서 가장 인기있는 건강식품중의 하나로 한반도(북위 33~48도)가 원산지이다. 인삼은 중국 전한 시대 '사유'에 의해 쓰여진 "급취장" (BC 48~BC 33)에 최초로 언급되어 있으며, 1145년 고려시대 김부식이 지은 "삼국사기"를 보면 고구려, 백제, 신라가 의약적 치료용으로 중국에 인삼을 수출하기 시작하였다고 기록되어 있다. 또한 인삼의 인공재배에 대한 기록은 1596년 중국 명나라 '이시진'의 "본초강목"에 처음으로 나타난다. 우리나라에서는 조선 중기이후 야생삼이 감소하는 시기와 맞물려 숙종 13년(1687년) "승정원일기"에 '묘삼'의 기록이 나타난다. 묘삼은 벼의 이앙법을 활용한 독창적인 발명으로 1600년대 초반에 인삼의 인공재배법이 확립되었다는 것을 암시해준다. 한편 홍삼에 관해서는 1123년 중국 송나라 '서긍'이 지은 "고려도경"에 '숙삼'(인삼을 쪄서 건조한 삼)이라는 용어가 처음으로 등장하며 이것이 홍삼에 대한 최초 기록이다. 조선 중종 17년(1552년) "조선왕조실록"에 '판삼'(삼을 찌고 압착하여 얇은 판처럼 만든 후 풀로 붙인 삼)이라는 용어가 나오고 선조 35년(1602년)에는 '파삼'(삼을 찌고 압착하여 얇은 판처럼 만든 후 실로 꿰어 붙인 삼)이라는 용어가 등장하는 것으로 보아 조선 중종(1506~1545) 시기부터 선조(1567~1608) 시기에 홍삼의 제조방법이 개발되었다는 것을 알 수 있다. 물론 홍삼의 제조법은 그 전부터 이루어졌다고 추정되지만 조선 중종~선조 시기에 더욱 발전하였다고 보는 것이 타당하다. '홍삼'이란 용어는 정조 21년(1797) "조선왕조실록(정조실록)"에 처음 등장하며, 고종 36년(1899) "조선왕조실록(고종실록)"에는 대한제국 궁내부 내장원 삼정과에서 홍삼을 제조하였다는 기록이 나타난다. 아울러 순조 10년(1810년)에는 증포소를 개성으로 이전한다는 기록이 있는 것으로 볼 때 이시기에 대규모 홍삼제조가 이루어졌다고 볼 수 있다. 요약하면 조선 숙종시대에 확립된 인삼 인공재배기술과 정조시대의 홍삼제조법, 그리고 순조시대의 증포기술 등이 결합하여 대량생산 체계를 갖추기 시작하는 것이다. 이후 1916년 '김택영'이 지은 "소호당집"에는 홍삼의 제조방법이 상세히 기록되어 있는데 이것이 전통적 한국홍삼 제조법의 모태가 되는 것이다. 한편 인삼의 효능에 대한 최초의 기록은 BC 83-96년 중국에서 저술된 "신농본초경"으로 이 효능은 그 이후 여러 의학서에서 지속적으로 계승되어 오고 있다. 앞으로 고려인삼에 대한 역사적 기록물은 지속적으로 발굴되고 보존되어야 하며 이러한 서지학적 노력은 과학적 연구결과와 더불어 세계속에 고려인삼의 독창성을 알리고 유지하는 가장 좋은 방법으로 생각된다.

Protective Effect of Korean Red Ginseng against Aflatoxin B1-Induced Hepatotoxicity in Rat

  • Korean red ginseng (KRG), the steamed root of Panax ginseng Meyer, has a variety of biological properties, including anti-inflammatory, antioxidant and anticancer effects. Aflatoxin $B_1$ ($AFB_1$) produced by the Aspergillus spp. causes acute hepatotoxicity by lipid peroxidation and oxidative DNA damage, and induces liver carcinoma in humans and laboratory animals. This study was performed to examine the protective effects of KRG against hepatotoxicity induced by $AFB_1$ using liver-specific serum marker analysis, histopathology, and terminal deoxynucleotidyl transferase-mediated dUTP nick-end labeling. In addition, to elucidate the possible mechanism of hepatoprotective effects, superoxide dismutase, catalase, glutathione peroxidase, and malondialdehyde were analyzed. Rats were treated with 250 mg/kg of KRG (KRG group) or saline ($AFB_1$ group) for 4 weeks and then received 150 ${\mu}g/kg$ of $AFB_1$ intraperitoneally for 3 days. Rats were sacrificed at 12 h, 24 h, 48 h, 72 h, or 1 wk after $AFB_1$ treatment. In the KRG pre-treatment group, serum alanine aminotransferase, aspartate aminotransferase, and malondialdehyde levels were low, but superoxide dismutase, catalase, and glutathione peroxidase activities were high as compared to the $AFB_1$ alone group. Histopathologically, $AFB_1$ treatment induced necrosis and apoptosis in hepatocytes, and led to inflammatory cells infiltration in the liver. KRG pre-treatment ameliorated these changes. These results indicate that KRG may have protective effects against hepatotoxicity induced by $AFB_1$ that involve the antioxidant properties of KRG.

Comparative Study of Korean White, Red, and Black Ginseng Extract on Cholinesterase Inhibitory Activity and Cholinergic Function

  • This study evaluated cholineresterase inhibitory activity of Korean white ginseng extract (WGE), red ginseng extract (RGE), and black ginseng extract (BGE) and the cholinergic effect on scopolamine (SCOP)-induced amnesic mice. WGE, RGE, and BGE inhibited acetylcholineserase (AChE), as well as butyrylcholineserase (BuChE) in a concentration-dependent manner. BGE presented strong inhibition of AChE with an $IC_{50}$ value of 1.72 mg/mL, followed by WGE (5.89 mg/mL), RGE (6.30 mg/mL), respectively. The inhibitory activity of the three ginseng extracts on BuChE showed similar values among the groups. To better understand the mechanisms of the possible effect of ginseng extract on the cholinergic function, this study assessed the expression of the cholinergic markers of choline acetyltransferase (ChAT) and AChE using western blot and RT-PCR analysis in the brains of amnesic mice. Treatment with ginseng extracts led to inhibition of AChE expression and, the activation of ChAT expression in the hippocampus and the cerebral cortex of amnesic mice as induced by SCOP. The results suggest that ginseng extracts including BGE, appear to modulate the metabolism of acetylchoine (ACh), which would greatly increase synaptic ACh levels and most potently revert SCOP-induced amnesia.

홍삼농축액에서 Saccharomyces cerevisiae와 Saccharomyces carlsbergensis에 의한 Ginsenosides의 bioconversion (Bioconversion of Ginsenosides in Red Ginseng Extract by Saccharomycescerevisiae and Saccharomyces carlsbergensis)

  • Ginseng(Panax ginseng C.A. Meyer) is reported to have many pharmaceutical activities. The minor ginsenosides(Rd, Rg3, Rh2 and compound K) display pharmaceutical properties superior to those of the major ginsenosides. These minor ginsenosides, which contribute a very small percentage, are produced by hydrolysis of the sugar moieties of the major ginsenosides. The pH of red ginseng extracts fermented with S. cerevisiae and S. carlsbergensis decreased rapidly during 3 days of fermentation, with no further significant change thereafter. After 20 days of fermentation, a relatively small difference remained in the acidity of extracts fermented with S. cerevisiae (0.54%) and S. carlsbergensis (0.58%). Reducing sugar in the S. cerevisiae and S. carlsbergensis extracts decreased from 25.86 to 4.54 mg/ml and 4.32 mg/ml glucose equivalents, respectively; and ethanol contents increased from 1.5% at day 0 to 16.0 and 15.0%, respectively, at 20 days. Ginsenosides Rb1, Rb2, Rc, Re, Rf, and Rg1 decreased during the fermentation with S. cerevisiae, but Rd and Rg3 increased by 12 days. Ginsenosides Rb1, Rb2, Rc, Re and Rg1 decreased gradually in the extract with S. carlsbergensis, but Rd and Rg3 were increased at 6 days and 9 days.

Efficacy of Korean Red Ginseng in the Treatment of Alopecia Areata

  • Alopecia areata (AA) is an autoimmune disease that can affect any hair-bearing area. AA is known to be caused by immunological disorder but still, the pathogenesis is not fully understood. Many therapeutic modalities have been used to treat alopecia areata, with variable efficacy and safety profiles. Unfortunately, none of these agents is definitely curative or preventive alone. We studied hair growth efficacy and safety of Korean red ginseng (KRG) in AA comparing corticosteroid intra-lesional injection (ILI) alone patient group with ILI with KRG taking patient group using Folliscope 2.5 for 12 wk. Herein, we would like to report the efficacy of KRG in the treatment of AA and recommend KRG as a useful complimentary food for gaining efficacy of treatment for AA.
