Syntheses and Central Nervous Depressant Activity of Piperine Derivatives(III) N-Piperoyl Amino Acids (Piperine유도체의 합성 및 중추억제작용에 관한 연구(III) N-Piperoylamino Acid 유도체)

  • Twelve new N-piperoyl amino acids were synthesized by acylation of amino acids with piperoylchloride in order to develop the new central nervous depressants. Silylation of the carboxylic group of amino acids not only precluded side reactions but-also facilitated acylation at room temperature without catalysts. The acute toxicity of all the synthesized compounds were lower than that of piperine itself, whereas they showed strong protective effect against the strychnine-induced mortality.

  • The structure of the title compound has been determined by single-crystal X-ray diffraction work. The crystals are orthorhombic, space group $Pna2_1$ with a=11.095(3), b=21.834(7), c=12.574(4) $\AA$, and R1=0.0667. The average carbon bond length in aromaticity In the molecule is 1.386(1) $\AA$ and the average single bond length linking the central benEene ring and peripheral phenyl rings is 1.491(3) $\AA$. The average dihedral angle between the central benzene ring and each of six peripheral phenyl rings is $67.1(1)^{\circ}$ and the average dihedral angle between neighboring two phenyl rings is $55.0(1)^{\circ}$. Thus the molecule adopts a quasi-propeller configuration with approximate six-fold rotation symmetry.

  • This study was made on the taxa Chrysanthemum zawadskii complex that grow in South Korea on the basis of chromosomes, epidermis, pollens and gross morphology. I have found four types of chromosome numbers, 36, 45, 54, and 72 as a polyploidal series. Even though the gross morphology was quite similar almost the same gross morphology, chromosome number was different among the taxa. The taxa of 36 chromosomes present broad and fine lobed leaves which grow separately, broad leafed taxon in the mainland of Korea and the other's in Ullungdo Island which is isolated form the mainland in the East Sea. The taxa of 54 chromosomes are also present in the broad and in the fine lobed leaves. The fine lobed leave taxon grows in central to northern Korea and in the high altitude of mountains. Broad leafed taxon grows in central to southern Korea and comparatively lower altitude of the mountains. The taxon of 72 chromosomes is grown in the high altitude of Mt. Hallasan which is isolated from the mainland of Korea. According to this study of Chrysanthemum zawadskii complex, I have arranged the scientific names, as Chrysanthemum zawadskii subsp. latilobum, subsp. acutilobum, subsp. naktongenese, subsp. lucidum, subsp. coreanum and hybrid between subsp. acutilobum X subsp. latilobum.

The Effect of 6-Hydroxydopamine on the Hypotensive Action and Contractile Force of Isolated Vas Deferens Smooth Muscle by Clonidine (Clonidine의 혈압강하및 적출정관 평활근수축에 미치는 6-Hydroxydopamine의 영향)

  • The effect of neurotoxic compound 6-hydroxydopamine (6-OHDA) on the change in blood pressure and contractile response of Vas deference by centrally acting agents has been studied in normal and DOCA-salt induced hypertensive rats. The treatment of neonatal rats with 6-OHDA (2$\times$100mg, 250mg Kg$^{-1}$s.c) significantly inhibited the antihypertensive and relaxant effects of Vas deference of clonidine(100$\mu\textrm{g}$ Kg$^{-1}$iv.). The simultaneous administration of desipramine with clonidine into neonatal rats decreased the antihypertensive response of clonidine although treated did not affect the relaxative response of Vas deference. Furthermore, the antihypertensive and relaxant responses of clonidine were reduced by the neonatal rats with 6-OHDA regardless of the administration of desipramine. When neonatal rats were administered with 6-OHDA, the development of DOCA-salt hypertension was prevented. These results suggest that 6-OHDA, clonidine and desipramine hada significant effect on the development and the inhibition of central hypertension mediating the central adrenergic neuron due to their affinity to the central nervous system.

The Activity of Dopamine $\beta$-Hydroxylase of Central Nervous System in Genetically Epilepsy Prone Rats

  • Abnormality in the central noradrenergic system may be related to the seizure prone state in the genetically epilepsy prone rats (GEPR). The present work deals with the characterization of the deficit in noradrenergic system if susceptitibility and intensity of seizure are dependent on central noradrenregic activities by comparing the activities of dopamine $\beta$-hydroxylase (DBH) which hydroxylates dopamine into noradrenaline. DBH activities were measured in 5 areas of brain of normal rats, native GEPR and severe GEPR. The results suggest that lower DBH activities in the midbrain of GEPRs may positively be coupled to the susceptibility to seizure, whereas the same characteristics of the native or severe GEPR are not neccessarily in parallel with the intensity of seizure.

Radiosensitizing and Topoisomerase I Inhibitory Effects of Aloe vera, Formitella fraxinea, and Ulmus davidiana Extracts

  • Ulmus davidiana, Formitella fraxinea, and Aloe vera extracts were detected to have inhibitory effects against topoisomerase I at treatment of $5{\mu}g$. Ulmus davidiana and Aloe vera extracts were found to show inhibitory effect similar to camptothecin, Formitella fraxinea extract was found to have weak activity. We also found the potential use of those extracts as a radiation sensitizer. Radiosensitizing effect at combination treatment was increased more than 2 times at single treatment of radiation, Ulmus davidiana or Formitella fraxinea extracts. Ulmus davidiana and Formitella fraxinea extracts were found to have significant radiosensitizing effect on test tumor cell line. But, Aloe vera extract was not detected to have activity as a radiosensitizer. Ulmus davidiana and Formitella fraxinea extracts are potent radiosensitizers on tumor cell lines and should be considered for further study of active compounds.

  • The relationship between the maintenance of hypertension and the central noradrenergic nervous system activity in spontaneously hypertensive rats (SHR) was studied. The norepinephrine turnover rates in 5 brain areas; telencephalon, hypothalamus/thalamus, midbrain, pons/medulla, cerebellum as a measure of noradrenergic neuronal activity were measured at the ages of 14 weeks in SHR and normotensive Wistar rats. In 14-week old SHR, blood pressure was significantly higher than in normotensive rat, and central norepinephrine turnover rates were significantly greater in telencephalon, hypothalamus/thalamus, midbrain. There were no differences between norepinephrine turnover rates in pons/medulla, cerebellum of SHR and those of normotensive rats.

  • The hydrogen storage is one of the key technologies to achieve the successful hydrogen economy and a chain to connect hydrogen production to its utilization. In this paper, characteristics and strong candidates of hydrogen storage technologies were analyzed from the objective information of patents. Also, the hydrogen storage technology trends and gaps were assessed using statistical or qualitative analysis. In this study the patents applied in Korea, Japan, US and EU from 10 or 20 years ago to 2011 were analyzed. The result of patent analysis could be used for developing or searching for promising technology of the hydrogen storage.

  • A sensitive spectrophotofluorometric method was developed for the analysis of 2[[3-(trifluoromethyl)phenyl]amino]-3-phridine carboxylic acid (I) in urine. The method is based on the fluoroscence behavior of the I-aluminum complex in absolute ethanol. This fluorophore has activation and emission wavelengths of 355 and 450 nm, respectively. Optimum conditions for the reaction were investigated. The fluorescence was linear in the range of 0.25 3.0 ug of I/ml. Replicate studies of spiked urine samples, each containing 2.5 ug of I/ml showed good precision with a relative standard deviation of 0.019. Overall recovery percent from five spiked urine samples was 99.4 $\pm$1.32%.

  • Relationship between the maintenance of hypertension and central serotonergic nervous system activity in opontaneously hypertensive rats (SHR) was studied. Serotonin turnover-rates were measured in 5 brain areas as an index of serotonergic neuronal activity and compared at the ages of 14 weeks in two types of animals; (1) spontaneously hypertensive rats (SHR) (2) normotensive wistar kyoto rats (WKY). In 14-week old SHR, central serotonin turnover rate was significantly lower in telencephalon, hypothalamus/thalamus and midbrain than normotensive rat, but significantly higher in cerebellum. There were no significant differences between serotonin turnover rate in pons/medulla of SHR and that of normotensive rat. THese data suggest that the abnormally lower turnover rates of serotonin in telencephalon, hypothalamus/thalamus and midbrain may be one of the underlying neuronal factors for manifestation of hypertension in SHR.
