The Whitening Effect by Water Extracts of Bombyx Mori Linne (백강잠(Bombyx Mori Linne) 물추출물의 미백 효능)

  • Numerous novel ingredients have been introduced for the hight functionality of whitening cosmetics. Tough the preliminary research, we have found water extracts Bombyx mori have high whitening efficacy. The results of the research for the whitening effect of Bombyx Mori L. are as follow 1. Bombyx Mori L. inhibited concentration dependently the generation of melanin increased by the stimulation of $\alpha$-MSH and protoporphyrin IX, and $IC_{50}$ value was 8.3, 9.2 ${\mu}M$ respectively. 2. Melanin increased by the stimulation of $\alpha$-MSH and protoporphyrin IX was five to seven times superior in the inhibiting effect, compared with kojic acid used as positive control group. 3. Bombyx Mori L. did not have a decolorizing effect on melanin already generated. 3. Bombyx Mori L. was observed to have toxicity of over 100 ${\mu}M$ for the mouse melanoma B16 cells. Therefore, These results suggest that water extracts of Bombyx mori have inhibitory activity against mushroom tyrosinase and inhibitory activity of melanin synthesis in B16 melanoma cells in vitro.

Di-acetyl-nor-aporphines: Novel molecules and a novel mechanism to inhibit melanogenesis

  • Nor-aporphine derivatives have been discovered which interfere with the flux of Calcium into and out of the cell interior. It has been shown that adrenergic antagonists that block the Calcium exchange lead to an inhibition of Protein kinase C activity, thus blocking tyrosinase activation. Di-acetyl-dimethoxy-methyl-nor-aporphine is a semi-synthetic molecule of natural origin with very high potency. On B16 melanocytes as well as in normal human melanocytes the decrease in melanin synthesis reaches -50% at a level of 40 ppm in the culture medium. On a molar concentration basis, this is 50 to 70 times stronger than Kojic acid inhibition. Yet, the cell viability is not affected. Reversibility studies show that after washing out of the active compound, melanogenesis returns to normal levels. Possible mechanisms of the activity are discussed. Tests carried out on SkinEthic(R) three-dimensional models of the epidermis and in vivo clinical studies on Asian population confirm the strong inhibition of melanogenesis. Safety evaluation of these molecules, on the other hand, demonstrates good skin tolerance and absence of toxicity.

  • Tyrosinase is one of the important enzymes in the mammalian melanin synthesis. In the process of melanin synthesis, tyrosine is oxidized to DOPA (3,4-dihydroxyphenylalanine), and DOPA is further oxidized to dopaquinone. Tyrosinase is an enzyme catalyzing this oxidation of tyrosine, so chemicals that inhibit the activity of tyrosinase can be used as skin whitening agents. In this study, we isolated five constituents from the 80% MeOH extract of Morus bombycis cortex by bioactivity-guided fractionation. We performed mushroom tyrosinase inhibition assay. As a result, 7,2',4'-trihydroxyflavanone (1), 2',4',2,4,-tetrahydroxychalcone (2), and oxyresveratrol (3) showed the more potent inhibitory effect compared to kojic acid, a well-known skin whitening agent with antityrosinase effect. Moracinoside M (4) and moracin M-3'-O-${\beta}$-D-glucopyranoside (5) also showed the moderate tyrosinase inhibitory activities.

Inhibitory Effect of Fructus Ligustri Lucidi on Tyrosinase and MITF Expressions (여정자 추출물의 Tyrosinase 및 MITF 발현 억제 효과)

  • The purpose of this study was to investigate the mechanism of Hexane extract of Fructus Ligustri Lucidi (HFLL)-induced regulation of melanogenesis. An apparent down-regulatory effect of tyrosinase activity was observed when B16F10 cells were incubated with HFLL. Interestingly, HFLL did not inhibit the catalytic activity of cell-free tyrosinase from B16F10 cells, whereas kojic acid directly inhibited tyrosinase activity. Regarding protein levels of melanogenic enzymes, the amounts of tyrosinase and tyrosinase-related protein 1 (TRP-1) were decreased by HFLL, while the amount of tyrosinase-related protein 2 (TRP-2) slightly was reduced after incubation with HFLL. Treatment with HFLL was found to down-regulate microphthalmia-associated transcription factor (MITF). These results suggest that HFLL is an effective inhibitor of pigmentation caused by down regulation via MITF, tyrosinase, and TRP-1 expressions.

Study of Inhibitory Effect of Melanogenesis and Antioxidant Activity of Agrimonia pilosa Ledeb (선학초 추출물의 멜라닌합성 억제 및 항산화효과)

  • The purpose of this study was to investigate the mechanism of ethanol extract of Agrimonia pilosa Ledeb. (EAP)-reduced melanogenesis and diphenyl-picryl-hydrazyl (DPPH) radical scavenging activity. Agrimonia pilosa Ledeb., a perennial herbaceous plant, has been used as an antihemorrhagic, anthelminntic, and antiinflammatory agents in Chinese herbal medicine. In the present study, we observed that melanin synthesis and tyrosinase activity of B16F10 cells were significantly decreased by EAP. However, EAP could not suppress tyrosinase activity in the cell-free system, whereas kojic acid directly inhibited tyrosinase activity. Furthermore, EAP decreased the protein expression of tyrosinase, tyrosinase-related prootein 1 (TRP-1), and tyrosinase-related prootein 2 (TRP-2). EAP scavenged DPPH radical up to 41% with 100 ${\mu}g/m{\ell}$ concentration. These results suggest that the hypopigmentary effect of EPA was due to regulation of tyrosinase protein.

Bioactive Cyclopentenone Derivatives from Marine Isolates of Fungi

  • As part of an effort to discover bioactive natural products from marine sources, we investigated the bioactive secondary metabolites from two marine isolates of the fungi, Trichoderma viride and Rhizopus stolonifer. Three cyclopentenones, myrothenones A (1) and B (2) and trichodenone A (3), were isolated from T. viride and two cyclopentenones, 2-bromomyrothenone B (4) and botrytinone (5), were isolated from R. stolonifer. The molecular structures and absolute stereochemistries of the cyclopentenones were determined from chemical and physicochemical evidence, including quantum chemistry calculations, X-ray analysis, and the circular dichroism (CD) exciton chirality method. Myrothenone A (1) displays tyrosinase inhibitory activity, with an I$C_{50}$ value of 6.6 ${\mu}M$, and is therefore more active than the positive control, kojic acid.

Biological Activity of Ethanol Extracts from Fermented Opuntia humifusa with 3 Different Mushroom Mycelia (3종의 버섯균사체로 발효한 천년초 에탄올 추출물의 생리활성)

  • The present study was carried out to investigate the biological activity of ethanol extracts from fermented Opuntia humifusa with 3 different mushroom mycelia: Phellinus linteus, Lentinula edodes, and Pleurotus ostreatus. Fermented Opuntia humifusa by Phellinus linteus (FOP) and Lentinula edodes (FOL) showed more DPPH and ABTS radical scavenging activities than non-fermented Opuntia humifusa (NFO) and fermented Opuntia humifusa by Pleurotus ostreatus (FOPO). At a concentration of 250 ppm, the ABTS radical scavenging activities of the FOP and FOL were similar to that of BHA, a synthetic antioxidant. The total polyphenol content had a similar tendency to that of the radical scavenging activity. However, the flavonoid content was increased in the order of NFO, FOL, FOPO, and FOP. At all concentrations, the tyrosinase inhibitory activity of FOP and FOPO were significantly higher than that of kojic acid. During adipocyte differentiation, NFO and FOL showed no significant difference in lipid accumulation in 3T3-L1 cells. FOP and FOPO showed a higher fat accumulation inhibitory effect than NFO and FOL. These results provide baseline data for Opuntia humifusa as a novel functional food.

Isolation of Inhibitory Components on Tyrosinase Activity from the Bark of Paeonia moutan (목단피로부터 멜라닌 생성 억제성분의 분리)

  • The MeOH extract of the bark of Paeonia moutan showed potent inhibitory effect on the mushroom tyrosinase activity in vitro. The activity-guided fractionation of t he MeOH extract resulted in the isolation of three active compounds. The chemical structures of these compounds were elucidated by chemical and spectroscopic evidence as catechin, 1,2,3,6-tetra-O-galloyl-${\beta}$-D-glucose and 1,2,3,4,6-penta-0-galloyl-${\beta}$-D-glucose, respectively. Among them, the inhibitory activity by 1,2,3,6-tetra-galloyl-${\beta}$-D-glucose on mushroom tyrosinase was more potent $(IC_{50}=3.5\;{\mu}M)$ than that of kojic acid $(IC_{50}=8.7\;{\mu}M)$ ,but catechin enhanced the mushroom tyrosinase activity 50% in the concentration of 34.5M.

Inhibitory Activity of Medicinal Plant Extracts against Tyrosinase (약용식물 추출물의 Tyrosinase 억제 활성)

  • (1) Objectives: To discover natural skin-lightening agents, we have evaluated the inhibitory activity of EtOH extracts from 20 medicinal plants against mushroom tyrosinase. (2) Methods: Tyrosinase activity was determined by the dopachrome method using L-tyrosine as the substrates. (3) Results: Of the plant extracts tested, the extracts of 4 plants, Albizzia julibrissin, Curcuma longa, Anethum graveolens and Sophora flavescens, exhibited potent inhibitory activity (> 50%) in mushroom tyrosinase assay. Four plant extract, extracts of Agrimonia pilosa, Paeonia moutan, Magnolia obovata and Eugenia caryophyllata also showed relatively strong inhibitory (> 40%) against mushroom tyrosinase. (4) Conclusion: These active medicinal plants may be useful for the development of skin-whitening agents. Since the active medicinal plants may contain effective tyrosinase inhibitors even more than kojic acid, further study to identify the active constituents from the plants is expected.

Tyrosinase Inhibitory Activity of Plant Extracts (III): Fifty Korean Indigenous Plants

  • The purpose of this study was to evaluate tyrosinase inhibitory activity of plant extracts, especially Korean indigenous plants, for the cosmetic use of skin whitening. When 50 plant extracts were tested, the methanol extracts of Agrimonia pilosa, Aster scaber; Dianthus sinensis, Fatsia japonica, Hemistepta lyrata, Lespedeza cuneata, Osmunda japonicum, Pyrvla japvnica, Rodgersia phodophylla and Veratrum grandiforum possessed the considerable tyrosinase inhibitory activity at 3-300 $\mu\textrm{g}$/mL. Especially, L. cuneata, aerial part of O. japonicum and V. gandiforum exhibited the strong inhibition (>50% inhibition at 300 $\mu\textrm{g}$/mL). In particular, the methanol extract of V. grandiforum and its ethylacetate fraction showed potent inhibition ($IC_{50}$/=30 and 13$\mu\textrm{g}$/mL, respectively), while the reference compound, kojic acid, showed $IC_{50}$/ value of 26$\mu\textrm{g}$/mL. These plant extracts may be used as tyrosinase inhibitors in cosmetics.
