Solid Phase Extraction of Phospholipids from Brazil Nut (Bertholletia excelsa) and Their Characterization by Mass Spectrometry Analysis

  • The Brazil nut (Bertholletia excelsa - Lecythidaceae) is considered a product with high economic value, being a food widely appreciated for its nutritional qualities. Although previous studies have reported the biochemical composition of Brazil nut oil, the knowledge regarding the phospholipid composition exhibits a disagreement: the composition of fatty acids present in the structures of phospholipids is reported as being different from the composition of the free fatty acids present in the oil. In this work, solid phase extraction (SPE) was employed to provide a fast extraction of the phospholipids from Brazil nuts, in order to compare the phospholipid profile of the in nature nuts and their fatty acids precursor present in the oil. The major phospholipids were characterized by mass spectrometry approach. Their fragmentation pattern through direct infusion electrospray ionization ion-trap tandem mass spectrometry ($ESI-IT-MS^2$) proved to be useful to unequivocal characterization of these substances. High resolution (HR) experiments through ESI using a quadruple time of flight mass spectrometry (QTOF) system were performed to reinforce the identifications.

  • As a continuation of Part I, which developed a design formula of the minimum corner radius (Rmin) for restraining tribological failures, Part II investigates design parameters such as material properties and contact force. As design examples, Al 7075-T651, SST 304 and HT-9 are chosen for the materials and 1, 10 and 100 kN are used for the forces. The results show that the difference in Rmin decreases as either the elastic modulus increases or the contact force decreases. Given the same material and force, the permissible Rmin decreases as the flat region increases and vice versa. Because the Rmin values obtained from the examples are very small, the dimensions of the corner radius normally designed in engineering structures are regarded acceptable. The von Mises stress evaluated for a typical example, which is far below the yield strength, confirms this interpretation. Nevertheless, the present work can provide a design criterion as well as a guideline for quality control in the manufacturing of, in particular, contact corners, which has not been attempted before to the best of the author’s knowledge. In addition, this paper considers the problem of a step that may be formed in the contact contour by using a similar approach. The result shows that no size of the step is permissible.

Novel Fabrication of Designed Silica Structures Inspired by Silicatein-a

  • Silicatein-${\alpha}$, the enzyme extracted from silica spicules in glass sponges, has been studied extensively in the way of chemistry from 1999, in which the pioneering work by Morse, D. E. - the discovery of the enzymatic hydrolysis in Silicatein-${\alpha}$ - was published. Since its reaction conditions are physiologically favored, synthesis of various materials, such as gallium oxide, zirconium oxide, and silicon oxide, was achieved without any hazardous wastes. Although some groups synthesized oxide films and particles, they have not achieved yet controlled morphogenesis in the reaction conditions mentioned above. With the knowledge of catalytic triad involved in hydrolysis of silicone alkoxide and oligomerization of silicic acid, we designed the novel peptide amphiphiles to not only form self-assembled structure, but also display similar activities to silicatein-${\alpha}$. Designed templates were able to self-assemble into left-handed helices for the peptide amphiphiles with L-form amino acid, catalyzing polycondensation of silicic acids onto the surface of them. It led to the formation of silica helices with 30-50 nm diameters. These results were characterized by various techniques, including SEM, TEM, and STEM. Given the situation that nano-bio-technology, the bio-applicable technology in nanometer scale, has been attracting considerable attention; this result could be applied to the latest applications in biotechnology, such as biosensors, lab-on-a-chip, biocompatible nanodevices.

An Optimized Methodology to Observe Internal Microstructures of Aloe vera by Cryo-Scanning Electron Microscope

  • Aloe vera has been used in the pharmaceutical, food and cosmetic industry for its therapeutic properties. However, there are not many current studies on the microstructure of A. vera compared to studies on the chemical constituents and health efficacy of A. vera. Therefore, we compared the morphology of an A. vera leaf using an optical microscope, a conventional scanning electron microscope (SEM) and a cryo-SEM. Especially, this study focused on observing the gel in the inner leaf of A. vera, which is challenging using standard imaging techniques. We found that cryo-SEM is most suitable method for the observation of highly hydrated biomaterials such as A. vera without removing moisture in samples. In addition, we found the optimal analytical conditions of cryo-SEM. The sublimation conditions of $-100^{\circ}C$ and 10 minutes possibly enable the surface of the inner leaf of A. vera to be observed in their "near life-like" state with retaining moisture. The experiment was repeated with A. arborescens and A. saponaria to confirm the feasibility of the conditions. The results of this study can be applied towards the basic research of aloe and further extend previous knowledge about the surface structures of the various succulent plants.

  • Molecular chaperone의 발견은 생명과학자들에게 살아있는 세포 내에서 어떻게 생체활성단백질이 만들어지고 유지되는지에 대한 자극과 함께 그것을 증명하기 위한 실험동기를 부여하였다. 초기에는 Molecular chaperone이 nucleosomes의 assembly에 관여하는 단백질을 설명하기 위하여 사용되었으나, 지금은 기본적인 세포생리기능의 하나인 단백질의 folding과 assembly를 돕는 assistant protein으로 주로 사용된다. 단백질합성 뿐만 아니라 단백질수송, oligomeric structure의 assembly와 disassembly, heat shock을 포함한 각종 내, 외부스트래스에 의해서 변성된 단백질의 세로내분화와 회복에도 Molecular chaperone이 관여하고 있다. 그러나 아직까지는 Molecular chaperone들의 3차구조와 그들간의 상호작용에 관한 정보가 부족하여 크게 진전되지 못하고 있지만, 많은 연구자에 의한 정보축적으로 인하여 빠른 시일 내에 Molecular chaperone에 세포내역할이 분명하게밝혀질 것이다.

An Improvement of Estimation Method of Source Term to the Environment for Interfacing System LOCA for Typical PWR Using MELCOR code

  • Background: Interfacing-system loss-of-coolant-accident (ISLOCA) has been identified as the most hazardous accident scenario in the typical PWR plants. The present study as an effort to improve the knowledge of the source term to the environment during ISLOCA focuses on an improvement of the estimation method. Materials and Methods: The improvement was performed to take into account an effect of broken pipeline and auxiliary building structures relevant to ISLOCA. An estimation of the source term to the environment was for the OPR-1000 plants by MELOCR code version 1.8.6. Results and Discussion: The key features of the source term showed that the massive amount of fission products departed from the beginning of core degradation to the vessel breach. Conclusion: The release amount of fission products may be affected by the broken pipeline and the auxiliary building structure associated with release pathway.

Wind flow characteristics and their loading effects on flat roofs of low-rise buildings

  • Wind flow and pressure on the roof of the Texas Tech Experimental Building are studied along with the incident wind in an effort to understand the wind-structure interaction and the mechanisms of roof pressure generation. Two distinct flow phenomena, cornering vortices and separation bubble, are investigated. It is found for the cornering vortices that the incident wind angle that favors formation of strong vortices is bounded in a range of approximately 50 degrees symmetrical about the roof-corner bisector. Peak pressures on the roof corner are produced by wind gusts approaching at wind angles conducive to strong vortex formation. A simple analytical model is established to predict fluctuating pressure coefficients on the leading roof corner from the knowledge of the mean pressure coefficients and the incident wind. For the separation bubble situation, the mean structure of the separation bubble is established. The role of incident wind turbulence in pressure-generation mechanisms for the two flow phenomena is better understood.

원예시설용 망의 압력강하 특성에 대한 실험적 연구 (An Experimental Study on Characteristics of Pressure Drop of Screens Used in Horticultural Facilities)

  • This study was carried out to present the pressure drop for various wind speeds through nine types of screens used in horticultural facilities. The screens have been widely used to prevent harmful insects from being entered into agricultural facilities, to reduce strong wind and to shade a light as well. Whatever the usage of the screens was, it was necessary to have good knowledge of how much the screen caused a pressure drop for wind speeds when analyzing both the inner thermal-flow distribution in the facility and the effect of reducing wind speed by using CFD. Furthermore, as for wind screens, the pressure drop for wind speeds was needed as a design load in evaluating the structural stability of the structures supporting the screens. Therefore, the pressure drop through the screens for wind speeds of 5~30 $m{\cdot}s^{-1}$ at about 5 $m{\cdot}s^{-1}$ interval and inflow angles of $0{\sim}45^{\circ}$ at an interval of $15^{\circ}$ was respectively measured in a subsonic wind tunnel. The relation of the pressure drop for various screens was well fitted as a secondorder polynomial expression.

Elucidation of Antifungal Metabolites Produced by Pseudomonas aurantiaca IB5-10 with Broad-Spectrum Antifungal Activity

  • Antifungal metabolites were isolated from a culture of Pseudomonas aurantiaca IB5-10. Chemical structures of the metabolites were elucidated as phenazine-1-carboxylic acid (PCA; 1), 2-hydroxyphenazine (2-OH-PHZ; 2), and cyclo-(L-Pro-L-Val; 3), respectively, based on spectroscopic methods. Among them, 3 was isolated for the first time from this strain. The antifungal activities of 1-3 were evaluated against a variety of plant pathogens. To the best of our knowledge, the antifungal activities of 3 against plant fungal pathogens have been evaluated for the first time in this work. PCA (1) showed the most potent antifungal activities against Phytophthora capsici, Rhizoctonia solani AG-1(IA), and Pythium ultimum with MICs (${\mu}g/ml$) of less than 1.0, 1.3, and 2.0, respectively. On the other hand, 2-OH-PHZ (2) showed potent antifungal activity against R. solani AG-1(IA) with the MIC (${\mu}g/ml$) of 2.0, whereas it showed moderate antifungal activity against P. ultimum with the MIC (${\mu}g/ml$) of 50.0. In addition, 3 showed antifungal activity against only R. solani AG-1(IA).

Construction and Characterization of an Enhanced GFP-Tagged TIM-1 Fusion Protein

  • TIM-1 (also known as KIM-1 and HAVcr-1) is a type I transmembrane glycoprotein member of the TIM family that may play important roles in innate and adaptive immune responses. The overexpression of proteins associated with membrane proteins is a major obstacle to overcome in studies of membrane protein structures and functions. In this study, we successfully coupled the overexpression of the TIM-1 protein with a C-terminal enhanced green fluorescent protein (GFP) tag in Escherichia coli. To the best of our knowledge, this report is the first to describe the overexpression of human TIM-1 in E. coli. The purified TIM-1-EGFP fusion protein recognized and bound directly to apoptotic cells and did not to bind to viable cells. Furthermore, we confirmed that the interactions of TIM-1-EGFP with apoptotic cells were blocked by TIM-1-Fc fusion proteins. This fusion protein represents a readily obtainable source of biologically active TIM-1 that may prove useful in future studies of human TIM-1.
