HPLC-UV에 의한 진득찰과 털진득찰의 Kirenol 정량분석 (Quantitative Analysis of Kirenol in Siegesbeckia glabrescens and S. pubescens by HPLC-UV)

  • Many diterpenoids from Siegesbeckia species (Compositae) and their anti-inflammatory actions have been examined. In this research, high-performance liquid chromatography-ultraviolet spectrophotometer (HPLC-UV) method was used to compare the quantitative level of kirenol (ent-pimarane-type diterpenoid) in the aerial parts of Korean S. glabrescens and S. pubescens and the Chinese Siegesbeckiae Herba. Fingerprints of the two HPLC chromatograms of Korean S. glabrescens and S. pubescens were similar, but considerably different from Chinese Siegesbeckiae Herba. The content of kirenol in S. pubescens ($16.51{\pm}0.10$ mg/ml dry weight as mean${\pm}$RSD) was higher than S. glabrescens ($13.48{\pm}0.12$ mg/g dry weight). These values were considerably higher than the Chinese Siegesbeckiae Herba ($1.55{\pm}0.74$ mg/g dry weight). Thin layer chromatography (TLC) analysis demonstrated the containing of kirenol in the three plant materials, but the presence of siegeskaurolic acid (entkaurane-type diterpenoid) only in the Chinese Siegesbeckiae Herba.

Protective Effects of Standardized Siegesbeckia glabrescens Extract and Its Active Compound Kirenol against UVB-Induced Photoaging through Inhibition of MAPK/NF-κB Pathways

  • Anti-photoaging effects of standardized Siegesbeckia glabrescens extract (SGE) and its major active compound kirenol were investigated using Hs68 human dermal fibroblasts and hairless mice, respectively. UVB-irradiated hairless mice that received oral SGE (600 mg/kg/day) showed reduced wrinkle formation and skinfold thickness compared with the UVB-irradiated control. Furthermore, SGE treatment increased the mRNA levels of collagen synthesis genes (COL1A1, COL3A1, COL4A1, and COL7A1) and activated antioxidant enzyme (catalase), while suppressing matrix metalloproteinase (MMP-2, -3, -9, and -13) expression. In Hs68 fibroblasts, kirenol also significantly suppressed MMP expression while increasing the expression of COL1A1, COL3A1, and COL7A1. Collectively, our data demonstrate that both SGE and kirenol attenuated UVB-induced photoaging in hairless mice and fibroblasts through inhibition of the mitogen-activated protein kinases and nuclear factor kappa B pathways, suggesting that SGE has potential to serve as a natural anti-photoaging nutraceutical.

털진득찰에서 분리한 Kirenol의 혈관 이완효과 (Vasodilatation effect of Kirenol isolated from Sigesbeckia pubescens)

  • 고혈압은 성인의 수축기 혈압이 140 mmHg 이상이거나 이완기 혈압이 90 mmHg 이상일 때를 의미하며 고혈압은 뇌졸중, 신부전 및 관상동맥질환 등 인체 전반에 걸쳐 다양한 합병증을 일으키며 고혈압 유병자의 생명과 건강을 직접적으로 위협한다(Kim and Kim, 2018). 그러나 고혈압 유병자들은 증상이 거의 나타나지 않으므로 혈압을 측정하기 전까지는 진단되지 않는다. 진득찰 즉 한방에서 희렴은 고혈압을 치료하는 처방 제제로 주로 사용됐기에 이에 착안하여 털진득찰을 이용하여 엔도델린(Endothelin)로 강제 수축 시킨 토끼 기저동맥의 이완 효과를 평가하였다. 본 연구에서는 털진득찰(Sigesbeckia pubescens)의 전초를 이용하여 키레놀 화합물을 분리 후 혈관 이완 효과를 평가하였다. 인체 내 가장 강력한 혈관수축 물질로 알려진 엔도델린은 혈관 평활근 세포막에 존재하는 엔도델린 수용체 아형 A (ETAR)와 혈관 내피세포막에 존재하는 수용체 아형 B2(ETB2R)에 작용하여 혈관의 긴장도를 높이고, ETB1R에 작용하여 긴장도를 낮추는 조절자 임무를 수행한다(Lucchelli et al., 1999). 일반적으로 엔도델린 펩타이드의 경우 염증, 당뇨, 및 심혈관계 질환에서 혈중 농도가 증가하며, 엔도델린 체계의 비정상적 항진은 만성신부전 및 사구체 경화증과 같은 신질환, 특발성 폐섬유화증 및 만성폐쇄성 폐 질환 등의 호흡기계 질환, 대사질환의 중요한 관심거리가 되는 당뇨병성 신경병증 및 망막증, 수족괴사 등의 질환과 전립선 및 대장 등의 암질환 등을 초래한다(Lavoie et al., 1997; Pancrazio et al., 1998). 본 연구에서 엔도델린 유도 뇌 기저동맥의 수축을 억제하는 키레놀의 농도(EC50)를 관찰한 결과 10 ㎍/mL의 농도에서 48% 이상의 유효한 혈관 이완 효능이 관찰되었다. 따라서 키레놀을 이용하여 엔도델린 활성조절 신소재로의 구조적·기능적 도출 연구가 추가로 진행된다면건강 기능성 식품 소재 또는 의약학 소재로의 개발이 가시화될 수 있을 것으로 사료된다.

ent-Kaurane- and ent-Pimarane-Type Diterpenoids from Siegesbeckia pubescens and Their Cytotoxicity in Caki Cells

  • ent-Kaurane- and ent-pimarane-type diterpenoids were isolated from the methanol extract of Siegesbeckia pubescens by column chromatography. Their structures were elucidated as ent-16$\alpha$H,17-hydroxy-kau­ran-19-oic acid (1), ent-4,17-dihydroxy-16$\alpha$-methyl-kau­ran-19-oic acid (2), ent-16$\beta$,17-dihydroxy-kauran-19-oic acid (3), kirenol (4) and ent-16$\beta$,17,18-trihydroxy-kauran­19-oic acid (5) by spectral analysis. The cytotoxicity of these compounds in Caki cells was assayed by a cell counting kit. Only one group treated with kirenol (4), an entpimarane-type diterpenoid, showed the inhibition of the cell growth in Caki cells.

The Constituents of Siegesbeckia orientalis

  • Two new diterpenoids, orientalin A (1), and B (2), have been isolated together with six known compounds, kirenol (3), $ent-16{\beta},17-dihydroxykauran-19-oic$ acid (4), $ent-16{\beta},17-dihydroxykauran-19-oic$ $acid-16{\beta},l7-acetonide$ (5), $3,7-dimethylquercetin$ (6), ${\beta}-sitosterol$ (7), and daucosterol (8) from the ethanol extract of Siegesbeckia orientalis (Compositae). Their chemical structures have been elucidated as $ent-15-acetoxy-2{\alpha},16,19-trihydroxypimar-8(14)-ene$ (1), $ent-16-acetoxy-2{\alpha},15,19-trihydroxypimar-8(14)-ene$ (2), respectively, on the basis of chemical and spectral evidence.
