Study on CO2-Coal Gasification Reaction Using Natural Mineral Catalysts (천연 광물질을 이용한 CO2 석탄 촉매 가스화 반응 특성 연구)

  • In this study, the effect of natural minerals on the reaction kinetics for lignite-$CO_2$ gasification was investigated. After physical mixing of lignite from Meng Tai area with 5 wt% of each natural mineral catalysts among Dolomite, Silica sand, Olivine and Kaolin, $CO_2$ gasification was performed using TGA at each 800, $850^{\circ}C$ and $900^{\circ}C$. The experimental data was analyzed with volumetric reaction model (VRM), shrinking core model (SCM) and modified volumetric reaction model (MVRM). MVRM was the most suitable among three models. As increasing the reaction temperature, the reaction rate constant became higher. With natural mineral catalysts, the reaction rate constant was higher and activation energy was lower than that of without catalysts. The lowest activation energy, 114.90 kJ/mol was obtained with silica sand. The highest reaction rate constant at $850^{\circ}C$ and $900^{\circ}C$ and lower reaction rate constant at $800^{\circ}C$ were obtained with Kaolin. Conclusively, the better catalytic performance could be observed with Kaolin than that of using other catalysts when the reaction temperature increased.

Functional Significance of Cytochrome P450 1A2 Allelic Variants, P450 1A2*8, *15, and *16 (R456H, P42R, and R377Q)

  • P450 1A2 is responsible for the metabolism of clinically important drugs and the metabolic activation of environmental chemicals. Genetic variations of P450 1A2 can influence its ability to perform these functions, and thus, this study aimed to characterize the functional significance of three P450 1A2 allelic variants containing nonsynonymous single nucleotide polymorphisms (P450 $1A2^*8$, R456H; $^*15$, P42R; $^*16$, R377Q). Variants containing these SNPs were constructed and the recombinant enzymes were expressed and purified in Escherichia coli. Only the P42R variant displayed the typical CO-binding spectrum indicating a P450 holoenzyme with an expression level of ~ 170 nmol per liter culture, but no P450 spectra were observed for the two other variants. Western blot analysis revealed that the level of expression for the P42R variant was lower than that of the wild type, however the expression of variants R456H and R377Q was not detected. Enzyme kinetic analyses indicated that the P42R mutation in P450 1A2 resulted in significant changes in catalytic activities. The P42R variant displayed an increased catalytic turnover numbers ($k_{cat}$) in both of methoxyresorufin O-demethylation and phenacetin O-deethylation. In the case of phenacetin O-deethylation analysis, the overall catalytic efficiency ($k_{cat}/K_m$) increased up to 2.5 fold with a slight increase of its $K_m$ value. This study indicated that the substitution P42R in the N-terminal proline-rich region of P450 contributed to the improvement of catalytic activity albeit the reduction of P450 structural stability or the decrease of substrate affinity. Characterization of these polymorphisms should be carefully examined in terms of the metabolism of many clinical drugs and environmental chemicals.

The Relationship between health related physical fitness and self-perceived health status (건강관련 체력과 주관적 건강인식에 관한 연구)

  • Modern technology has lessened the physical demand of every activities. Thus, physical inactivity has led to a rise in ‘Hypokinetic diseases’. The prefix, hypo, means lack of and kinetic refers to movement. Individuals who do not exercise regularly are at greater risk of developing hypokinetic diseases such as coronary heart disease, hypertension, hyperlipidemia, obesity, musculoskeletal disease. Systematic exercise program defenses development of hypokinetic diseases. Exercise program bases on basic physical fitness test. The physical fitness evaluation is used to degree of Wellness of the individuals. It includes the evaluation of Health-Related Physical Fitness -musclular strength, muscular endurance, flexibility, cardiorespiratory endurance, body composition - and skill or sports related fitness -agility, balance, coordination, reaction time, speed. In present study, the authors investigated health-related physical fitness test result and questionnaire for 133 male and 71 female from Jannuary 16 to March 15, 1997. The purpose of this study is to observe relationship health related physical fitness and self-perceived health status. The results are as follows. 1. There were statistically significant differences in BMI, grip strength, back strength, muscular endurance, cardiorespiratory endurance, and flexibility between male and female subjects. Flexibility is higher in female than male subject, but Others are higher in male than female subjects. 2. There were statistically significant differences in grip strength, muscular endurance between 30s, 40s, and 50s group In both gender subjects, and In cardiorespiratoryendurance between the groups especially in Female subjects. 3. For male subjects, flexibilitywas measured lower in group who classified low level in self-perceived health status than group of others. Also, for female, subjects, Muscular endurance & flexibility were measured. 4. It was shown that grip strength, back strength, and muscular endurance were significantly lower in group who have chronic diseases than normal group for Male subjects. But, For female subjects, all the component were statistically insignificant results between normal and disease group. 5. Both male and female subjects, there were Positive correlation among grip strength, back strength, muscular endurance, cardiorespiratory endurance, and flexibility but, negative correlation for age. Therefore, health-related physical fitness is very important component for Heath Promotion & Wellness. Physical Fitness test is valuable test in health evaluation, health management and health promotion, so available for hospital, sports center, community health center, industrial field, school etc.

Properties of Foamed Concrete According to Types and Concentrations of Foam Agent (기포제 종류 및 희석 농도에 따른 기포 콘크리트의 특성)

  • Recently, the government has been working feverously to save energy and reduce greenhouse gas emission by enacting Basic Act on Low Carbon Green Growth at the national level. Improving the insulation performance of building exterior and insulator can reduce the energy in the building sector. This study is about developing light-weight foamed concrete insulation panel that can be applied to buildings to save energy and to find the optimal condition for the development of insulation materials that can save energy by enhancing its physical, kinetic and thermal characteristics. Various experimental factors and conditions were considered in the study such as foam agent types (AES=Alcohol Ethoxy Sulfate, AOS=Alpha-Olefin Sulfonate, VS=Vegetable Soap, FP=Fe-Protein), foam agent dilution concentration (1, 3, 5%), and foam percentage (30, 50, 70%). Experiment results indicated that the surface tension of aqueous solution including foam agent, was lower when AOS was used over other foam agents. FP produced relatively stable foams in 3% or more, which produced unstable foams containing high water content and low surface tension when diluted at low concentration. Depending on foam agent types, compressive strength and thermal conductivity were similar at low density range but showed some differences at high concentration range. In addition, when concentrations of foam agent and foaming ratio increased, pore size increased and open pores are formed. In all types of foam agent, thermal conductivity were excellent, satisfying KS standards. The most outstanding performance for insulation panel was obtained when FP 3% was used.

Treatment of Mixed Fluoride Wastewater Using Cement Paste (시멘트 페이스트를 이용한 혼합 불산폐수 처리)

  • Fluorine compounds are the essential chemicals for wet processes of semiconductor and LCD production line. Problems of conventional treatments for fluoride wastewater are their high operation costs and low fluoride removal capacity. In this study, cement paste containing various Ca-bearing hydrates such as portlandite, calcium silicate hydrate(CSH), and ettringite was investigated for fluoride removal. The objectives of this study are to assess the feasibility of using cement paste cured mixture of cement and water as an alternative agent for treatment of fluoride wastewater and to investigate fluoride removal capacity of the cement paste. The performance of cement paste was comparable to that of lime in the kinetic test. In column experiment where the effluent fluoride concentrations were below 0.5 mg/L. Then the leached calcium reached the maximum level of 800 mg/L. The nitrate reduced to the level of less than 10 mg/L. Nitrate in the wastewater was exchanged with interlayer sulfate of these cement hydrate LDHs. Phosphate concentration could be reduced to 10 mg/L by forming calcium phosphate. These results indicate that the cement paste generally has advantageous characteristics as an economical and viable substitute for lime to remove fluoride.

A Study on Safety Estimation of Railroad Wheel (컨테이너 철도차륜의 안전성 평가에 관한 연구)

  • Recently, high speed of container freight cars is causing fatigue damage of wheel. Sudden failure accidents cause a lot of physical and human damages. Therefore, damage analysis for wheel prevents failure accident of container freight car. Wheel receives mechanical and thermal loads at the same time while rolling stocks are run. The mechanical loads applied to wheel are classified by the horizontal load from contact of wheel and rail in curve line section and by the vertical force from rolling stocks weight. Also, braking and deceleration of rolling stocks cause repeated thermal load by wheel tread braking. Specially, braking of rolling stocks is frictional braking method that brake shoe is contacted in wheel tread by high breaking pressure. Frictional heat energy occurs on the contact surface between wheel tread and brake shoe. This braking converts kinetic energy of rolling stocks into heat energy by friction. This raises temperature rapidly and generates thermal loads in wheel and brake shoe. There mechanical and thermal loads generate crack and residual stress in wheel. Wetenkamp estimated temperature distribution of brake shoe experimentally. Donzella proposed fatigue life using thermal stress and residual stress. However, the load applied to wheel in aforementioned most researches considered thermal load and mechanical vertical load. Exact horizontal load is not considered as the load applied to wheel. Therefore, above-mentioned loading methods could not be applied to estimate actual stress applied to wheel. Therefore, this study proposed safety estimation on wheel of freight car using heat-structural coupled analysis on the basis of loading condition and stress intensity factor.

Study on the prediction of the stopping probabilities in case of train fire in tunnel by Monte Carlo simulation method (몬테카를로 시뮬레이션에 의한 화재열차의 터널 내 정차확률 예측에 관한 연구)

  • The safety of tunnels is quantified by quantitative risk assessment when planning the disaster prevention facilities of railway tunnels, and it is decided whether they are appropriate. The purpose of this study is to estimate the probability of the train stopping in the tunnels at train fire, which has a significant effect on the results of quantitative risk assessment for tunnel fires. For this purpose, a model was developed to calculate the coasting distance of the train considering the coefficient of train running resistance. The probability of stopping in case of train fire in the tunnel is predicted by the Monte Carlo simulation method with the coasting distance and the emergency braking distance as parameters of the tunnel lengths and slopes, train initial driving speeds. The kinetic equations for predicting the coasting distance were analyzed by reflecting the coefficient train running resistance of KTX II. In the case of KTX II trains, the coasting distance is reduced as the slope increases in a tunnel with an upward slope, but it is possible to continue driving without stopping in a slope downward. The probability of the train stopping in the case of train fire in tunnel decreases as the train speed increases and the slope of the tunnel decreases. If human error is not taken into account, the probability that a high-speed train traveling at a speed of 250 km/h or above will stop in a tunnel due to a fire is 0% when the slope of the tunnel is 0.5% or less, and the probability of stopping increases rapidly as the tunnel slope increases and the tunnel length increases.

Effects of Hypobaric Conditions on Apoptosis Signalling Pathways in HeLa Cells

  • Nowadays increasing effectiveness in cancer therapy and investigation of formation of new strategies that enhance antiproliferative activity against target organs has become a subject of interest. Although the molecular mechanisms of apoptosis can not be fully explained, it is known that cell suicide program existing in their memory genetically is activated by pathophysiological conditions and events such as oxidative stress. Low pressure (hypobaric) conditions that create hypoxia promote apoptosis by inhibiting cell cycling. In this study, determination of the effects of fractional hypobaric applications at different times on HeLa cells at cellular and molecular levels were targeted. Experiments were carried out under hypobaric conditions (35.2 kPa) in a specially designed hypobaric cabin including 2% $O_2$ and 98% N. Application of fractional hypobaric conditions was repeated two times for 3 hours with an interval of 24 hours. At the end of the implementation period cells were allowed to incubate for 24 hours for activation of repair mechanisms. Cell kinetic parameters such as growth rate (MTT) and apoptotic index were used in determination of the effect of hypobaric conditions on HeLa cells. Also in our study expression levels of the Bcl-2 gene family that have regulatory roles in apoptosis were determined by the RT-PCR technique to evaluate molecular mechanisms. The results showed that antiproliferative effect of hypobaric conditions on HeLa cells started three hours from the time of application and increased depending on the period of exposure. While there was a significant decrease in growth rate values, there was a significant increase in apoptotic index values (p<0.01). Also molecular studies showed that hypobaric conditions caused a significant increase in expression level of proapoptotic gene Bax and significant decrease in antiapoptotic Bfl-1. Consequently fractional application of hypobaric conditions on HeLa cell cultures increased both antiproliferative and apoptotic effects and these effects were triggered by the Bax gene.

The Inhibitory Effect of Cornus walteri Extract Against ${\alpha}-amylase$ (말채나무 추출물의 ${\alpha}-amylase$ 저해 활성)

  • ${\alpha}-Amylase$ inhibitor is used to control blood glucose level by inhibiting starch digestion in the small intestine and delaying the absorption of glucose. In this study, we investigated the effect of the ethanol extracts from more than 1400 species of plants against ${\alpha}-amylase$ with the aim of developing a new ${\alpha}-amylase$ inhibitor. In the results, Cornus walteri extracts showed the highest inhibition activity. The inhibitory effect of Cornus walteri extract on the carbohydrate hydrolysis enzymes has different sensitivities against ${\alpha}-amylase$ from salivary and pancreatin and against ${\alpha}-glucosidase$ from yeast and porcine small intestine. In the study of inhibition kinetics of ${\alpha}-amylase$ and ${\alpha}-glucosidase$, Cornus walteri extract showed competitive inhibition against salivary and pancreatin while showing the combination of uncompetitive and noncompetitive inhibition against ${\alpha}-glucosidase$. The Cornus walteri extract was stable at acidic and thermal conditions. As for the blood glucose and body weight levels of Cornus walteri extract, we confirmed anti-hyperglycemic and anti-obesity effects. Also, in the investigation of the mRNA lever, Cornus walteri extract upregulated the level of GLUT4 mRNA in the quadriceps muscle.

A Study on Traveling Characteristics and Evaluation about Noise of Hydraulic Turbine Dynamo in Dam (댐 수차 발전기 소음의 전달특성과 평가에 관한 연구)

  • In case of the domestic condition, as the initiating from Sumjin River Dam, total 14 units of multipurpose dams had been constructed in 1965 for the roles of flood control, waterpower generation, irrigation, water supply, industrial water supply. In the case of such multipurpose dam, it produces electric energy by converting the potential energy utilizing its head and quantity of the water into kinetic energy. However, in this process, since during the time when the turbine connected to the hydraulic turbine dynamo revolves and there occurs a loud noise, it brings the physical, mental bad influences to those people also a decline of an effective working efficiency. On such point of view, after selection of various 16 measurement points, this study has measured and analyzed the travelling characteristics of noise generated at the hydraulic turbine dynamo in Daechung Dam, and also has evaluated the degree of indoor noise using the evaluation index such as PSIL, NC. As the result of noise-evaluation, in case of Daechung Dam, since the noise damage grade appears very seriously at various spaces, it is considering that its soundproof measure would be necessitated keenly. Also, it is considered that such data could be utilized as the valuable material hereafter for establishment of an efficient noise-reduction countermeasure and a comfortable working environment for the hydraulic turbine dynamo plant.
