Kinetic Mechanism in the Absence of Metal of Hafnia alvei Aspartase in the Amination Direction

  • The kinetic mechanism of Hafnia alvei aspartase in the amination direction has been determined in the absence of metal. The initial velocity pattern obtained by varying the concentration of fumarate at several fixed concentrations of NH4+ , shows an intersection on the left of the ordinate at pH 7.0, indicating that the kinetic mechanism is a sequential mechanism in which substrate inhibition by fumarate is observed. The dead-end inhibition pattern by varying the concentration of NH4+ at several fixed concentration of succinate shows an intersection on the left of the ordinate. These data are consistent with random addition of NH4+, or fumarate. The Haldane relationship gives a Keq of 1.18 ${\times}$10-3 M at pH 7.0, which is in agreement with the values obtained from the direct determination of reaction concentrations at equilibrium (6.0 $\pm0.2$ ${\times}$10-3 M).

Determination of Critical Generator Group Using Accelerating Power and Synchronizing Power Coefficient in the Transient Energy Function Method

  • This paper proposes an algorithm for determining critical generator lists using accelerating power and synchronizing power coefficient (SPC), and critical generator group (CGG) from CGG candidates, which is a combination of critical generators. The accurate determination of CGG provides a more accurate energy margin while providing system operator with information of possible unstable generator group. Classical transient energy function (TEF) method selects the critical generators with big corrected kinetic energy of each generator at the moment of fault removal. However, the generator with small acceleration after fault, that is, the generator with small corrected kinetic energy, is also likely to belong to CGG if the generator has small synchronizing power. The proposed algorithm has been verified to be effective compared with the classical TEF method. We utilized the power system of Korean Electric Power Corporation(KEPCO) as a test system.

  • The present paper is focused on sorption concentration of silver (I) on some complex-forming ion exchangers in the initial thiocyanate form and subsequent determination of Ag(I) in the phase of anion exchanger AN-25 by diffuse reflectance spectroscopy. The sorption and kinetic characteristics of the sorbents were investigated. The apparent stability constants of thiocyanate silver complexes in the ion exchanger phase were calculated. The sorption-spectroscopic method is proposed for Ag(I) determination in aqueous solutions. The calibration curve is linear in the concentration range of 10-200 mg/L (sample volume is 10.0 mL) and the detection limit is 2 $\mu$g/mL. The presence of $Na^+,\;K^+,\;Mg^{2+}$ (macrocomponents) as well as of Ni (II), Co (II), Cu (II) do not hinder the determination of silver (I).

Kinetic modeling of organic and nitrogen removal from domestic wastewater in a down-flow hanging sponge bioreactor

  • A down-flow hanging sponge (DHS) bioreactor was operated for the treatment of domestic wastewater. The Stover-Kincannon model was applied for kinetic evaluation of the reactor performance during the operational period. As a result, the coefficient of determination (R2) for straight lines of effluent concentration from the experimental data and from the predictive data of BOD5; NH4+-N; and TN were 0.9727; 0.9883; and 0.9934, respectively. The calculation of saturation value constant (Umax - g L-1 d-1) and maximum utilization rate constant (KB - g L-1 d-1) were 56.818 and 75.034 for BOD5; 2.960 and 4.713 for NH4+-N; 2.810 and 8.37 for TN, respectively. The study suggests that Stover-Kincannon model can be used for effective evaluation of kinetic removal of BOD5; NH4+-N; and TN from domestic wastewater treated in a DHS bioreactor.

  • Corynebacterium glutamicum의 NADPH-specific glutamate dehydrogenase를 이용하여 NADPH, NH$_4$Cl, $\alpha$-ketoglutarate의 기질에 대한 kinetics를 고찰하였다. 이들의 kinetic constants를 측정함으로서 정반응에로의 효소반응 기작은 첫번째 효소와 반응하는 기질이 NADPH 임을 확인할 수 있었다. Glutamate dehydrogenase 활성의 조절을 위한 metabolites의 효과를 고찰하여 본 결과 malate와 citrate 만이 효소에 억제 효과를 나타내었으며, potassium chloride는 효소활성에 가장 많은 영향을 주었다.

Activity Determination, Kinetic Analyses and Isoenzyme Identification of Gamma Glutamyltransferase in Human Neutrophils

  • Gamma-glutamyltransferase (GGT, EC 2.3.2.2) which hydrolyzes glutathione (GSH), is required for the maintenance of normal intracellular GSH concentration. GGT is a membrane enzyme present in leukocytes and platelets. Its activity has also been observed in human neutrophils. In this study, GGT was purified from Triton X-100 solubilized neutrophils and its kinetic parameters were determined. For kinetic analyses of transpeptidation reaction, $\gamma$-glutamyl p-nitroanilide was used as the substrate and glycylglycine as the acceptor. Apparent $K_m$ values were determined as 1.8 mM for $\gamma$-glutamyl p-nitroanilide and 16.9 mM for glycylglycine. The optimum pH of GGT activity was 8.2 and the optimum temperature was $37^{\circ}C$. It had thermal stability with 58% relative activity at $56^{\circ}C$ for 30 min incubation. L-serine, in the presence of borate, was detected as the competetive inhibitor. Bromcresol green inhibited neutrophil GGT activity as a noncompetetive inhibitor. The neutrophils seem to contain only the isoenzyme that is present in platelets. We characterized the kinetic properties and compared the type of the isoenzyme of neutrophil GGT with platelet GGT via polyacrylamide gel electrophoresis (PAGE) under a standart set of conditions.

저장유통 조건에 따른 오징어 품질예측 모델링 (Kinetic Modeling for Predicting the Quality of Squid (Todarodes pacificus) during Storage and Distribution)

  • 오징어의 품질을 평가하기 위한 여러 항목 들이 있으나 현장에서 소비자 및 유통업자들의 경우 주관적인 관능평가에 의존하고 있으며 소비자의 어류 구매의사 결정에 부패취가 직접적인 영향을 미칠 수 있다. 따라서 관능적 평가를 입증 할 수 있는 과학적인 품질인자를 제시하고자 하였으며, 온도와 저장시간에 따른 상호관계를 분석하였다. 과학적 품질인자로는 TVC, Pseudomonas spp., pH, VBN 으로 선정하였으며 세 가지 온도 조건에서 보관하였다. 이취발생검지시점은 R-index를 이용해 측정하고 kinetic 모델링을 진행하였다. 모델링 결과 활성화 에너지는 이취발생검지 시간이 51.210 kJ/mol, TVC는 42.888 kJ/mol, Pseudomonas spp.는 50.283 kJ/mol, VBN은 72.594 kJ/mol, pH는 41.990kJ/mol 으로 나타났다. Pseudomonas spp., TVC, pH, VBN 순으로 이취발생검지 시간의 활성화 에너지에 근사하였으며, 특히 Pseudomonas spp.는 이취발생검지 시간의 활성화 에너지와 가장 유사하였다. 따라서 오징어의 이취발생 indicator는 이취발생시간과 가장 유사한 양상을 나타내는 Pseudomonas spp.로 판단 할 수 있었다.

고구마 조직의 가열변화에 대한 반응속도론적 상수 결정 (Determination of Kinetic Parameters for Texture Changes of Sweet Potatoes during Heating)

  • 가열에 의한 품종별 고구마 조직의 변화에 대한 반응속도론적 상수를 biphasic model과 fractional conversion method를 사용하여 결정하였다. Biphasic model에 따라 고구마의 가열에 의한 조직의 변화는 2단계의 연속적인 1차 반응식으로 표시할 수 있었으며, 활성화에너지는 고구마의 품종에 따라 $71.0{\sim}75.1\;kJ/mol$$48.4{\sim}59.6\;kJ/mol$이었다. 또한 고구마의 가열에 의한 조직변화를 fractional conversion method를 사용하여 1개의 1차 반응식으로 표시할 수 있었는데, 이 때의 활성화에너지는 $67.5{\sim}75.3\;kJ/mol$로서 biphasic model에 의한 제 1단계의 반응에 대한 값과 유사한 값을 나타냈다. 이들 방법으로 결정된 반응속도론적 상수들 사이에는 kinetic compensation 효과가 있었으며, 두 방법 모두 고구마의 가열에 의한 조직변화에 대한 반응속도론적 상수들을 결정하는데 적용할 수 있음을 알 수 있었다.
