In order to develop new physiologically active polysaccharides from persimmon leaves, two different crude polysaccharides were prepared using hot water (PLW-0) and pectinase digestion (PLE-0) and their immuno-stimulating activities were estimated. PLW-0 and PLE-0 showed similar sugar compositions with 15 different sugars, including rarely observed sugars in general polysaccharides such as 2-O-methyl-fucose, 2-O-methyl-xylose, apiose, aceric acid, 3-deoxy-D-manno-2-octulosonic acid, and 3-deoxy-D-lyxo-2-heptulosaric acid, but the uronic acid content of PLE-0 was lower than that of PLW-0 caused by pectinase treatment. Both PLW-0 and PLE-0 showed potent anti-complementary activity in a dose-dependent manner which was similar to a known immuno-stimulating polysaccharide, PSK, from Coriolus versicolor. The activity of PLE-0 at a low concentration ($100{\mu}g/m{\ell}$) was higher than that of PLW-0. In an in vitro cytotoxicity analysis, PLW-0 and PLE-0 (up to $1,000{\mu}g/m{\ell}$) did not affect the growth of peritoneal macrophages and Colon 26-M3.1 carcinoma cells. In contrast, they enhanced lymphocyte proliferation activity. Peritoneal macrophages stimulated with PLW-0 and PLE-0 produced various cytokines, such as IL-6 and IL-12. However, PLE-0 was more effective on the cytokine production. Intravenous administration of PLW-0 and PLE-0 significantly augmented natural killer (NK) cell cytotoxicity against Yac-1 tumor cells 3 days after the treatment of polysaccharide fractions. But NK cells obtained from the PLE-treated group showed higher tumoricidal activity even at a low dose of $40{\mu}g$/mouse. In experimental lung metastasis of Colon 26-M3.1 carcinoma cells, prophylactic administration of PLW-0 and PLE-0 significantly inhibited lung metastasis in a dose-dependent manner and PLE-0 was more effective on the inhibition of cancer metasasis. The results lead us to conclude that the pectinase-treated process is indispensable to preparing polysaccharides with higher immune-stimulating activity from persimmon leaves.
Background: To perform the successful dendritic cell-based cancer immunotherapy one of the main issues to be solved is the source of antigen for DC pulsing. Limitations occur by using auto-tumor lysate due to the difficulties obtaining enough tumor tissue(s) quantitatively as well as qualitatively. In this study the possibility of allogeneic tumor cell lysate as a DC pulsing antigen has been tested in mouse melanoma pulmonary me tastasis model. Methods: B16F10 melanoma cells $(1{\timeS}10^5/mouse)$ were inoculated intra venously into the C57BL/6 mouse. Therapeutic DCs were cultured from the bone marrow myeloid lineage cells with GM-CSF and IL-4 (1,000 U/ml each) for 7 days and pulsed with lysate of either autologous B16F10 (B-DC), allogeneic K1735 (C3H/He origin; K-DC) or CloneM3 (DBA2 origin; C-DC) melanoma cells for 18 hrs. Pulsed-DCs $(1{\times}10^6/mouse)_{[CGP1]}$ were injected i.p. twice with one week interval starting from the day 1 after tumor cell inoculation. Results: Without observable toxicity, allogeneic tumor cell lysate pulsed-DC induced the significantly better anti-tumor response (tumor scale: $2.7{\pm}0.3,\;0.7{\pm}0.3\;and\;0.3{\pm}0.2$ for saline, B-DC and C-DC treated group, respectively). Along with increased tumor specific lymphocyte proliferations, induction of IFN-${\gamma}$ secretion against both auto- and allo-tumor cell lysates was observed from the DC treated mice. (w/B16F10-lysate: $44.97{\pm}10.31,\;1787.94{\pm}131.18,\;1257.15{\pm}48.27$, w/CloneM3 lysate: 0, $1591.13{\pm}1.83,\;1460.47{\pm}86.05pg/ml$ for saline, B-DC and C-DC treated group, respectively) Natural killer cell activity was also increased in the mice treated with tumor cell lysate pulsed-DC ($8.9{\pm}_{[CGP2]}0.1,\;11.6{\pm}0.8\;and\;12.6{\pm}0.7%$ specific NK activity for saline, B-DC and C-DC treated group, respectively). Conclusion: Conclusively, promising data were obtained that allogeneic-tumor cell lysate can be used as a tumor antigen for DC-based cancer immunotherapy.
본 실험은 cyclosporin A(CsA)에 의한 시간의 경과에 따른 림프절에서의 세포성 면역억제를 조사하기 위해서 시행된 것으로 BALB/C계 생쥐에 10일동안 CsA(45mg/kg/day) 투여 후 림프절에서의 T 림프구, IL-2 수용기 그리고 자연살해(NK)세포의 분포 변화를 관찰하였다. 대조군의 림프절에서는 L3T4(CD4)에 양성반응을 보이는 도움 T림프구, Ly2(CD8)에 양성반응을 보이는 세포독성 T 림프구 그리고 CD25R에 양성반응을 보이는 IL-2 수용기를 가진 세포는 곁피질(paracortex)과 수질동(medullary sinus)에서 분포하였다. CsA 투여 후 처음 3일까지는 이들 양성반응세포의 분포 변화는 없었으며 양성반응성의 변화도 없었다. 그러나 CsA 투여 7일부터 양성반응 세포수의 감소와 양성반응성의 약화가 관찰되기 시작하였으며 이러한 변화는 시간이 경과하여 14일에 이르렀을 때 가장 큰 감소추세로 나타났다. 한편 NK1.1(CD56)에 양성반응을 보이는 자연살해세포는 피질과 수질에 분포하였으며 CsA 투여 후 시간의 경과에 따라 양성반응 세포수가 감소하였으며, 이러한 감소는 14일에서 가장 큰 것으로 나타났다. 이상의 결과로 미루어보아 CsA 투여는 림프절에서의 IL-2 분비저해를 통해 T 림프구와 NK세포의 활성을 차단하여 선택적이면서 효과적인 세포성 면역억제작용을 하고 있는 것으로 사료된다.
Recombination activating gene-2 (RAG-2) plays a crucial role in the development of lymphocytes by mediating recombination of T cell receptors and immunoglobulins, and loss of RAG-2 causes severe combined immunodeficiency (SCID) in humans. Rag-2 knockout mice created using homologous recombination in ES cells have served as a valuable immunodeficient platform, but concerns have persisted on the specificity of Rag-2-related phenotypes in these animals due to the limitations associated with the genome engineering method used. To precisely investigate the function of Rag-2, we recently established a new Rag-2 knockout FVB mouse line ($Rag-2^{-/-}$) manifesting lymphopenia by employing a CRISPR/Cas9 system at Center for Mouse Models of Human Disease. In this study, we further characterized their phenotypes focusing on histopathological analysis of lymphoid organs. $Rag-2^{-/-}$ mice showed no abnormality in development compared to their WT littermates for 26 weeks. At necropsy, gross examination revealed significantly smaller spleens and thymuses in $Rag-2^{-/-}$ mice, while histopathological investigation revealed hypoplastic white pulps with intact red pulps in the spleen, severe atrophy of the thymic cortex and disappearance of follicles in lymph nodes. However, no perceivable change was observed in the bone marrow. Moreover, our analyses showed a specific reduction of lymphocytes with a complete loss of mature T cells and B cells in the lymphoid organs, while natural killer cells and splenic megakaryocytes were increased in $Rag-2^{-/-}$ mice. These findings indicate that our $Rag-2^{-/-}$ mice show systemic lymphopenia with the relevant histopathological changes in the lymphoid organs, suggesting them as an improved Rag-2-related immunodeficient model.
전통발효식초에 속하는 사과식초로부터 에탄올 및 증류수를 이용, 다당체를 분리하여 새로운 면역활성 물질을 확인하고자 하였다. 사과식초를 농축하고, 80% 에탄올을 가하여 침전물을 얻었으며 소량에 물에 용해하여 투석 및 동결건조를 거쳐 조다당 KAV-0를 얻었다. 구성당 분석 결과, KAV-0는 주로 mannose 38.2%, galactose 19.1% 및 glucose 14.3%로 구성되어있음을 확인하였고, 정상세포주(RAW 264.7 세포 및 대식세포) 및 종양세포주(B16-BL6 melanoma 세포)에 대해 세포독성 측정 결과, 직접적인 세포독성을 나타내지 않았다. 대식세포 유래 세포주인 RAW 264.7 세포 자극에 대한 사이토카인 및 산화질소 생산능을 측정한 결과, IL-6, $TNF-{\alpha}$ 및 산화질소 모두 농도 의존적으로 분비능이 증가하고 저농도에서도 우수한 활성을 나타냈으며, 마우스 복강유래 대식세포 자극에 대한 사이토카인 및 산화질소 생산능에서도 동일한 양상의 결과를 보여 사과식초 유래 조다당 KAV-0가 높은 대식세포 활성능을 소유함을 확인할 수 있었다. 한편, KAV-0의 정맥투여에 따른 NK 세포 활성화를 측정한 결과(ex vivo), 시료에 의해 활성화된 NK 세포는 Yac-1 종양 세포주에 대한 치사활성을 농도 의존적으로 증진시켰으며, $1,000{\mu}g/mouse$에서 월등히 우수한 NK 세포 활성능을 유도하였다. 또한, B16-BL6 melanoma를 이용한 폐암전이모델에서 KAV-0를 정맥투여한 뒤 항전이 활성을 측정한 결과, 농도 의존적으로 암 저해 효과가 나타났으며, 특히 $1,000{\mu}g/mouse$에서는 약 53%의 우수한 암전이 억제 활성을 나타냈다. 이상의 결과로부터 사과식초 유래 조다당 KAV-0는 인체의 면역계를 자극하여 우수한 면역활성 및 항전이 효과를 나타낼 수 있음이 확인되었고 건강 기능성 소재로의 활용 가능성이 충분할 것으로 최종 판단되었다.
적양배추 유래물의 면역 활성 및 항전이 활성을 평가하기 위하여 적양배추 유래 열수추출물 RCW 및 조다당 RCP를 분리하였다. RCW는 중성당이 60%의 높은 비율로 검출되었으며, 이중 Glc가 28.2%로 구성되어 있었으며, RCP는 중성당과 산성당이 각각 40.0% 및 58.9%로 이루어져 있으며, 구성당 분석 결과, Ara(27.5%)와 Gal (17.2%)가 높은 비율로 검출되었다. RCW와 RCP는 정상세포 및 암 세포에 대한 어떠한 독성도 나타나지 않았으며, 오히려 정상세포에서는 약간의 증식능이 확인되었다. 또한 대식세포 자극을 통한 cytokine 분비능을 측정한 결과, RCW는 IL-6, IL-12 및 $TNF-{\alpha}$ 생산능은 어떠한 농도에서도 확인되지 않았으며, $1000{\mu}g/mL$ 고농도에서 NO 생산능이 확인되었다. 반면에 RCP는 $8{\mu}g/mL$의 저농도에서도 IL-6, IL-12, $TNF-{\alpha}$ 및 NO의 생산능이 확인되었다. 높은 cytokine 생산능을 나타낸 RCP의 NK 세포 활성 및 항전이 활성을 측정한 결과, RCP를 $1000{\mu}g/mouse$농도로 정맥투여할 경우, NK 세포를 무처리군 대비 최대 12배 활성화 시키는 것으로 확인되었으며, 47.3%의 항전이 활성이 확인되었다. 적양배추 유래 면역 활성 다당의 구조를 추정하기 위하여 ${\beta}-Glucosyl$ Yariv reagent와의 반응성을 검토한 결과, RCP $1000{\mu}g/mL$에는 약 9.7%의 $arabino-{\beta}-3,6-galactan$이 검출되었다. 본 결과로부터 적양배추에는 $arabino-{\beta}-3,6-galactan$을 일부 함유하는 RG-I 형태의 다당으로 존재하며, 이들에 의해 강력한 면역 증진 활성이 나타나는 것이라 결론지을 수 있었다.
허혈/재관류 손상은 급성신부전의 주요 원인이며 이식된 신장의 기능지연을 유발하여 거부반응의 발생을 증가시킨다. 본 연구에서는 쥐의 허혈/재관류 손상모델에서 허혈시간에 따른 면역세포의 변화를 평가하였다. 8주령 수컷 SD rat의 좌신을 각각 30, 45, 60분간 허혈/재관류 동안에 우신을 적출 하였고, 대조군은 우신만 적출하였다. 신장기능은 0, 1, 2, 3, 5, 7일에 각각 평가하였으며, 허혈/재관류 후 1일과 7일에 신장조직을 채취하여 면역형광염색(DCs, NK cells, macrophages, B cells, CD4+ and CD8+ T cells)과 H&E 염색을 실시하였다. 허혈 시간이 증가할수록 신장기능이 감소되었으나, 신장 세뇨관괴사와 염증세포의 침윤이 증가하였다. 허혈/재관류 후 신장조직에서 DCs, NK cells, macrophages, CD4+ T cells, CD8+ T cells의 침윤 증가와 재관류 후 7일째 B cells의 침윤이 관찰되었다. 면역세포는 허혈시간 뿐 아니라 재관류 시간이 증가에 따라 뚜렷하게 관찰되었다. 이 결과는 허혈시간이 증가됨에 따라 면역반응이 증가될 수 있으며, 허혈재관류 손상이 비항원성 면역반응을 유도할 수도 있다는 것을 의미한다.
Objectives: This study aimed to evaluate the effects of low-level exposure to toluene on T lymphocytes subpopulations.s. Methods: The study lasted from April to October 2010. The subjects were 390 male workers, among whom 137 were chronically exposed to toluene in video-tape manufacturing factories and 253 were controls had never been occupationally exposed to hazardous chemicals. The subpoupulations of CD4+, CD8+, CD16+ (natural killer cells) and total (CD3+) T lymphocytes were examined by two-color staining using monoclonal antibodies. The general and job characteristics of subjects were assessed through a self-administered questionnaire. Results: There was no significant difference in general and job characteristics between both groups. No significant difference in lipid peroxide level was observed between the control and exposed workers, but the concentration of hydrogen peroxide was significantly higher in the exposed workers. The numbers of CD16+ T lymphocytes in controls were significantly higher than those in exposed workers, but no significant differences were found in CD4+, CD8+ and CD3+ T lymphocytes. Hydrogen peroxide levels showed a significantly negative correlation with CD8+ (r = -0.29, p < 0.01), CD16+ (r = -0.56, p < 0.01) and CD3+(r = -0.22. p < 0.01), and toluene levels was significantly negative correlated with CD3+ (r = -0.29, p < 0.05). Conclusions: Our results suggest that chronic low-level exposure to toluene affects cell-mediated immunity and the effects might mediate through ROSs (Reactive Oxygen Species) such as hydrogen peroxide.

Purpose: The objective of this prospective study was to evaluate the relationship between the circulating lymphocyte subpopulation counts during preoperative chemoradiotherapy (CRT) and tumor response in locally advanced rectal cancer. Materials and Methods: From August 2015 to June 2016, 10 patients treated with preoperative CRT followed by surgery were enrolled. Patients received conventional fractionated radiotherapy (50.4 Gy) with fluorouracil-based chemotherapy. Surgical resection was performed at 4 to 8 weeks after the completion of preoperative CRT. The absolute blood lymphocyte subpopulation was obtained prior to and after 4 weeks of CRT. We analyzed the association between a tumor response and change in the lymphocyte subpopulation during CRT. Results: Among 10 patients, 2 (20%) had evidence of pathologic complete response. In 8 patients with clinically node positive, 4 (50%) had nodal tumor response. All lymphocyte subpopulation counts at 4 weeks after CRT were significantly lower than those observed during pretreatment (p < 0.01). A high decrease in natural killer (NK) cell, count during CRT (baseline cell count - cell count at 4 weeks) was associated with node down staging (p = 0.034). Conclusion: Our results suggest that the change of lymphocyte subset to preoperative CRT may be a predictive factor for tumor response in rectal cancer.
This study was purposed to investigate the effect of Gamgung-tang(GGT) on immune responses induced by glucocorticoid in mice. GGT solution was treated by intraperitoneal injection for 7 days after glucocorticoid treatment(80㎎/㎏). And then B and T cell proliferation and cytolytic activity of natural killer(NK) cells were measured. There was 25% inhibition in B cell proliferation with treatment of glucocorticoid. However, B cell proliferation was not influenced by GGT treatment. T cell proliferation was also inhibited by 18.4% with treatment of glucocorticoid. On the other hand, T cell proliferation was increased dose-dependent manner in GGT treated group. Furthermore in purified T cell, the proliferation was furtherly increased than non-purified T cell. At concentration of 18㎎/mouse GGT, purified T cell proliferation was increase to above level of normal group. The cytotoxic activity of NK cell was decreased by 35.3% with treatment of glucocorticoid. In GGT treated group, the cytotoxic activity of NK cell was increased to the normal level. In purified NK cell, the cytolytic activity of NK cell was further increased than non-purifed NK cell. These results suggest that GGT may proliferate T cell that is suppressed by glucocorticoid, and activate NK cell activity.

회원이 이용계약을 해지하고자 하는 때에는 [가입해지] 메뉴를 이용해 직접 해지해야 합니다.
제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.

1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
