To compare the respiratory burst activity potential of the phagocytes isolated from head kidney, spleen, and peripheral blood 1ll cultured rockfish (Sebastes schlegeli), chemiluminescent (CL) response analysis was performed. The phagocytes isolated from peripheral blood showed greater and faster CL response to the opsonized zymosan compared to that of the phagocytes isolated from kidney or spleen. This may imply a significant role of the blood phagocytes in defence mechanism of rockfish. The different responses found in the CL analysis among the phagocytes isolated from peripheral blood, kidney, and spleen may reflect differences in activation state or activity of phagocytes.
Effects of Korean mistletoe, Viscum album coloratum on the non-specific immune responses of Japanese flounder, Paralichthys olivaceus were examined. Flounder were inoculated with mistletoe, Freunds complete adjuvant (FCA), or phosphate-buffered saline (PBS) as a control into their peritoneal cavities. Reactive oxygen intermediate (ROI) products were more enhanced in mistletoe-injected fish kidney phagocytes than in FCA-injected ones. The level of lysozyme activity detected in the serum of fish 4 d after injection with mistletoe was also significantly higher than that found in the serum of the control fish. The appropriate concentration of mistletoe in eliciting the highest level of serum lysozyme activity was 500 $\mu$m/300 g of fish. In phagocytic activity assays, mistletoe-sen-sitized flounder kidney phagocytes captured more yeasts than those of the control fish. Korean mistletoe appeared to be a good activator of the non-specific immune responses of Japanese flounder.
Effects of Escherichia coli genomic DNA on the production of phagocyte activating supernatants by the head kidney leucocytes isolated from olive flounder (Paralichthys olivaceus) were investigated. Phagocyte activating activity of the supernatants was estimated by. measuring reactive oxygen species (ROS) production in target head kidney phagocytes. All supernatants from olive flounder head kidney leucocytes-stimulated with E. coli DNA induced significantly (P<0.01) higher ROS production from target pagocytes than the unstimulated control supernatant. Maximum enhancement of chemiluminescent response was observed $5.0-10.0{\mu}g\;mL^{-1}$ of bacterial DNA while the increment ability was decreased significantly (P<0.01) at the concentration of $20.0{\mu}mL^{-1}$. The results demonstrate that olive flounder head-kidney leucocytes stimulated with bacterial DNA release a soluble phagocyte activating cytokines capable of enhancing the respiratory burst activity from target phagocytes.
To examine effects of cadmium on the respiratory burst of kidney phagocytes and antioxidant defense in liver, juvenile red seabream Pagrus major were fed a cadmium-incorporated diet $(1g\;CdC1_2/kg\;diet)$. The respiratory burst activity measured by chemiluminescence (CL) was significantly reduced by oral intake of cadmium. Lipid peroxidation in liver expressed as thiobarbituric acid reactive substances (TBARS) was significantly higher in the fish fed a cadmium-incorporated diet than that of the fish fed a control diet both on Day 3 and Day 9. Liver Glutathione S-transferase (GST) activitiy was significantly increased both on Day 3 and Day 9 by feeding a cadmium-incorporated diet, when compared with the controls. From the present results, it can be concluded that oral intake of cadmium in red seabream is associated with marked reduction of respiratory burst capacity of kidney phagocytes which can elevate susceptibility of fish against infecting pathogens. Cadmium administration also elicits significant increment of lipid peroxidation in liver, and fish try to detoxify cadmium by increasing GST activity.
식물성 생약재의 항균력과 활성산소(superoxide anion, $O_2^-$)의 생산에 미치는 in vitro 효과를 조사하여 금후 in vivo 실험에 적용할 때, 어류 체내 자연 저항성 증진을 꾀할 수 있는 식물성 생약재를 발굴하기 위한 기초자료를 얻고자 하였다. 식물성 생약재는 고삼, 귀전우, 구절초, 박하, 방풍, 연교, 약쑥, 지유, 삼백초, 삼지구엽초, 상백피, 소회향, 편축, 팔각 등 14종류로 이들을 열수추출하여 실험에 사용하였다. 항균력 시험에는 어병세균 Listonella anguillarum(Vibrio anguillarum), Vibrio sp., Vibrio alginolyticus, Edwardsiella tarda, Streptococcus sp., Lactcoccus garvieae를 대상으로 disk법을 사용하였으며, 활성산소의 생산능은 넙치 신장 대식세포를 이용한 nitroblue tetrazolium(NBT) 반응으로써 조사하였다. 그 결과,약쑥이 다른 생약재에 비하여 시험에 사용된 어병 세균에 대한 항균활성이 월등히 뛰어났으며, 또한 넙치 대식세포의 활성산소 생산능을 크게 자극하는 효과가 있음을 확인할 수 있었다.

Conclusion: Korean mistletoe appeared to be a good activator of the non-specific immune responses of eel.

The aim of this study was to investigate the in vivo toxicity of di-2-ethylhexyl phthalate (DEHP), on the immune system of the rock bream, Oplegnathus fasciatus. Fish were injected twice intraperitoneally with DEHP (200, 400, and 800 mg/kg BW), and the cellularity and functional activity of phagocytes in the spleen and head kidney were measured. The number of head kidney leukocyte cells was significantly greater in fish treated with 800-mg DEHP/kg BW. Nonspecific immunity, as determined by the phagocytic index, was significantly decreased at 800-mg DEHP/kg BW in the head kidney. A significant reduction in phagocytic capacity was observed in the head kidney at ${\geq}$400-mg DEHP/kg BW. Furthermore, significantly increased levels of serum glutamic oxaloacetate transaminase and glutamic pyruvate transaminase indicated a marked hepatic dysfunction in immunosuppressed fish. Total serum protein was significantly reduced at ${\geq}$400-mg DEHP/kg BW; however, there were no significant changes in lysozyme activity. These results demonstrate that immune responses in the rock bream, Oplegnathus fasciatus can predict immunotoxicity at doses ${\geq}$400-mg DEHP/kg BW.

Lucigenin (Lg)- and luminol (Lm)-dependent chemiluminescence (CL) was used to compare the respiratory burst of rockfish (Sebastes schlegeli) phagocytes after stimulation with phorbol myristate acetate (PMA). To establish which reactive oxygen species (ROS) contributes to the observed CL, the modulators of ROS metabolism, such as superoxide dismutase (SOD), catalase, and sodium azide $(NaN_3)$ were used. Although LgCL responses were inhibited significantly by the addition of either SOD or catalase, in comparison to the control, significantly lower LgCL responses were recorded by SOD than catalase. LmCL also showed significantly decreased responses by the addition of SOD and catalase. However, there were no statistical differences in CL responses between SOD and catalase additions. More profound and significant decrease of LmCL responses were recorded by simultaneous addition of SOD and catalase. Sodium azide markedly enhanced LgCL responses, while it significantly inhibited LmCL responses. These results indicate that LgCL and LmCL can be used to measure extracellular $O_2$ production and myeloperoxidase (MPO)-mediated ROS production in fish phagocytes, respectively. Furthermore, LmCL can be used for analyzing intracellular ROS production by simultaneous addition of both SOD and catalase.
In this study, the effcts of kelp (Ascophyllum nodosum) meal on growth and immune responses of parrot fish (Oplegnathus fasciatus) were studied. Fish were fed an experimental diet supplemented with $2{\%}\;and\;5{\%}$ kelp meal in a controled diet. Several factors such as weight gain, hematological parameters and nonspecific immune responses were evaluated far 0, 2, 4 and 8 weeks after the administration of the kelp meal supplemented diet. Weight gain in the fish fed the diet supplemented with $2{\%}\;and\;5{\%}$ kelp meal was not significant among the tested groups. The NBT reaction of the phagocytes in the head kidney and the phagocytic rate/index of phagocytes in the peripheral blood were significantly higher than the control group. But, there was no significant difference both in complement bactericidal activity, mucus Iysozyme activity and hematology among each group.
