Quality Characteristic of Hwangki(Astragalus membranaceus) Chungkukjang during Fermentation (황기청국장의 발효 중 품질특성)

  • This study investigated the effects of a Hwangki (Astragalus membranaceus) extract on the quality of Chungkukjang fermented by Bacillus subtilis KCCM 12148, at 30, 40, and $50^{\circ}C$, for 4 days. Changes in moisture contents, protein levels, pH values, ammonia-type nitrogen levels, color, angiotensin-converting-enzyme (ACE) inhibition rates, and fibrinolytic activities, were all determined. For both control and test, the moisture contents decreased gradually with time and the protein levels increased slightly. The pH values fell at the beginning of fermentation and then rose. The content of ammonia-type nitrogen was higher in Hwangki with Chungkukjang than in control, until 24 hr after fermentation commenced. After that time, the content of ammonia-type nitrogen control was higher in the control than in the Hwangki with Chungkukjang sample. Color features, such as lightness, redness, and yellowness, all decreased during fermentation, in both control and test. The highest ACE inhibition rates during fermentation at $40^{\circ}C$ were 90.9% in the control (48 hr after fermentation commenced) and 95.3% in Hwangki with Chungkukjang(24 hr). Fibrinolytic activities of Chungkukjang and Hwangki Chungkukjang were 100.7 and 74.4% respectively. The content of 2,6-dimethyl pyrazine in the control was higher than that in Hwangki with Chungkukjang. Sensory evaluationtests showed that the addition of Hwangki significantly improved the overall palatability of Chungkukjang.

Component changes in Commercial Salt-Fermented Anchovy Sauce by Long Fermentation (장기 숙성에 따른 시판 멸치액젓의 성분변화)

  • To investigate changes of components in salt-fermented anchovy sauce by long fermentation, various chemical properties were examined. The moisture, crude protein, crude lipid, and crude ash contents of the anchovy sauce by fermentation were 67.48~69.35%, 5.47~7.40%, 0.27~0.47% and 23.82~24.12%. The moisture and pH was decreased slightly, but the contents of crude protein, crude lipid, acidity and amino-N degree were increased gradually by long fermentation. And contents of crude ash, VBN and salinity showed almost no change. Total free amino acid contents of 1, 3, 6, 9 and 12 year fermented samples were 8,762.2 mg%, 9,484.6 mg%, 10,085.8 mg%, 10,650.7 mg% and 10,123.7 mg% respectively. Major free amino acid of samples were glutamic acid, leucine, lysine, alanine, valine, isoleucine in ordor. The samples were caused by their composition of the free amino acids rations, in which were umami, sweet and bitter taste in the extracts of anchovy during long fermentation. In Hunter values, fermented samples were generally lower in L, b values whereas higher in a, ${\Delta}E$ values. And absorbance at 453 nm was increased gradually by fermentation. Sensory evaluation result of samples, 6 years sample was the highest than the others in overall acceptance.

Correlation of Proliferating Cell Nuclear Antigen (PCNA) Expression and S-phase Fraction, Survival Rate in Primary Non-Small Cell Lung Cancer (원발성 비소세포 폐암에서 PCNA의 발현정도와 암세포의 분열능 및 생존률과의 관계)

  • Background : To study the prognosis of patients with lung cancer, many investigators have reported the methods to detect cell proliferation in tissues including PCNA, thymidine autoradiography, flow cytometry and Ki-67. PCNA, also known as cyclin, is a cell related nuclear protein with 36KD intranuclear polypeptide that is maximally elevated in S phase of proliferating cells. In this study, PCNA was identified by paraffin-embedding tissue using immunohistochemistry which has an advantage of simplicity and maintenance of tissue architecture. The variation of PCNA expression is known to be related with proliferating fraction, histologic type, anatomic(TNM) stage, degree of cell differentiation, S-phase fraction and survival rate. We analyzed the correlation between PCNA expression and S-phase fraction, survival. Method : To investigate expression of PCNA in primary lung cancer, we used immunohistochemical stain to paraffin-embedded sections of 57 resected primary non-small cell lung cancer specimen and the results were analyzed according to the cell type, cell differentiation, TNM stage, S-phase fraction and survival. Results : PCNA expression was divided into five group according to degree of staging(-, +, ++, +++, ++++). Squamous cell type showed high positivity than in adenocarcinoma. Nonsignificant difference related to TNM stage was noticed. Nonsignificant difference related to degree of cell differentiation was noticed. S-phase fraction was increased with advance of PCNA positivity, but it could not reach the statistic significance. The 2 year survival rate and median survival time were -50% 13 months, +75% 41.3 months, ++73% 33.6 months, +++67% 29.0 months, ++++25% 9 months with statistic significance (P<0.05, Kaplan-Meier, generalized Wilcox). Conclusion : From this study, PCNA expression was high positive in squamous cell cancer. And, there was no relationship between PCNA positivity and TNM stage, cellular differentiation or S-phase fraction. But, the patients with high positive PCNA staining showed poor survival rate than the patients with lower positive PCNA staining (p<0.05). It was concluded that PCNA immunostaining is a simple and useful method for survival prediction in paraffin embedded tissue of non-small cell lung cancer.

P53 Overexpression and Outcome of Radiation Therapy in Head & Neck Cancers (두경부종양 환자에서 p53의 과발현과 방사선치료결과)

  • Purpose : Experimental studies have implicated the wild type p53 In cellular response to radiation. Whether altered p53 function can lead to changes in clinical radiocurability remains an area of ongoing study. This study was performed to investigate whether any correlation between change of p53 and outcome of curative radiation therapy in patients with head and neck cancels. Methods : Immunohistochemical analysis with a mouse monoclonal antibody (DO-7) specific for human p53 was used to detect to overexpression of protein in formalin fixed, paraffin-embedded tumor sample from 55 head and neck cancer patients treated with curative radiation therapy (median dose of 7020 cGy) from February 1988 to March 1996 at 51. Mary's Hospital. Overexpression of p53 was correlated with locoregional control and survival using Kaplan-Meier method. A Cox regression multi-variate analysis was peformed that included all clinical variables and status of p53 expression. Results : Thirty-seven (67.2$\%$) patients showed overexpression of p53 by immunohistochemical staining in their tumor. One hundred percent of oral cavity, 70$\%$ of laryngeal, 66.7$\%$ of oropharyngeal, 66.7$\%$ of hypopharyngeal cancer showed p53 overexpression (P=0.05). The status of p53 had significant relationship with stage of disease (P=0.03) and history of smoking (P=0.001). The overexpression of p53 was not predictive of response rate to radiation therapy. The locoregional control was not significantly affected by p53 status. Overexpression of p53 didn't have any prognostic implication for disease free survival and overall survival. Primary site and stage of disease were significant prognostic factors for survival. Conclusions : The p53 overexpression as detected by immunohistochemical staining had significant correlation with stage, primary site of disease and smoking habit of patients. The p53 overexpression didn't have any predictive value for outcome of curative radiation therapy in a group of head and neck cancers.

Comparison of In Vitro Lipid Deposition and Change of Optical Characteristics on Daily Disposable Lenses (1-day) and 3-days Lenses Over 3 days (3-days lenses와 daily disposable lenses(1-day)의 착용 시간 별 지방 침착량 및 광학적 특성 변화의 비교)

  • The study aimed to investigate in vitro lipid deposition of oleic acid, oleic acid methyl ester and cholesterol on a daily disposable (1-day lenses) and 3-days lenses over 3 days and changes of optical characteristics is also investigated. Artificial tear solutions were prepared to simulate actual tear compositions. Two types of contact lenses (1-day lenses (Senofilcon A) and 3-days lenses (silicone tripolymer)) were soaked in the artificial tear solutions within an incubator at 37 ℃ with 150 rpm for 8, 16, 24 hours. Lipid deposition (oleic acid, oleic acid methyl ester and cholesterol) were measured using high performance liquid chromatography (HPLC) instrument. In addition, measurements of oxygen transmissibility, light transmittance and observation of lens surface were conducted. The amount of lipid deposition on the 1-day lenses were 127.55 ㎍/lens for Day 1, 302.96 ㎍/lens, for Day 2, and 353.30 ㎍/lens for Day 3. The 3-days lenses were 46.22 ㎍/lens for Day 1, 66.07 ㎍/lens for Day 2, and 67.45 ㎍/lens for Day 3. Oxygen transmissibility were 81×10-9(cm/sec)(ml O2/ml×mmHg)(Baseline) and 48×10-9(cm/sec)(ml O2/ml×mmHg) (Day 3) for the 1-day lenses, it were 13.23×10-9(cm/sec)(ml O2/ml×mmHg)(Baseline) and 9.6×10-9(cm/sec)(ml O2/ml×mmHg) (Day 3) for the 3-days lenses. Transmittance of each lenses were 97.21% (Baseline) and 94.25% (Day 3) for the 1-day lenses, 97.65% (Baseline) and 95.15% (Day 3) for the 3-days lenses. Observation of surface deposition indicated greatest deposition for the 3-days lenses type on Day 3. Lipid deposition for both lens types increased by day and was greater for the 1-day lenses type. Surface deposition appeared to differ as it was greatest for the 3 days lens type, which may suggest other deposits such as protein may be present.

Studies on the major nutritional components of commercial dried lavers (Porphyra yezoensis) cultivated in Korea (시판 건조김의 주요 영양성분)

  • This study was conducted to provide basic date on the proximate composition and amino acid, fatty acid, sugar, and mineral contents of commercial dried lavers cultivated in Korea (in Seocheon, Wando, Goheung and Busan). The moisture, crude ash, crude fat, crude protein, and carbohydrate content ranges of the dried lavers were 5.67~7.43%, 8.01~8.95%, 1.54~2.25%, 37.77~39.98%, and 43.83~46.24%, respectively. The total amino acid, free amino acid, and essential amino acid concentrations were highest (25,982.15 mg/100 g, 4,545.44 mg/100 g, and 9,048.28 mg/100 g) in the dried lavers cultivated from Wando. The compositions of the fatty acids in four dried lavers were 16.30~22.15% saturated fatty acids, 6.04~7.73% monounsaturated fatty acids, and 70.33~76.03% polyunsaturated fatty acids. The sugar contents per 100 g of dried lavers were 3,678.84~4,052.52 mg of galactose, 2,112.30~2,473.86 mg of fructose, 1,103.74~1,648.39 mg of mannose, 361.67~590.21 mg of glucose, 8.63~10.38 mg of ribose, and 21.43~48.32 mg of xylose. The mineral concentrations in the dried lavers cultivated from Wando and Busan were low (Cu < Mn < Zn < Fe < Ca < Mg < Na < K), as were those in the dried lavers cultivated from Seocheon and Goheung (Cu < Mn < Zn < Fe < Mg < Ca < Na < K).

Effects of Feeding Whole-Crop Barley Silage and Corn Silage on Nutrients Availability, Growth Performance and Blood Parameters in Korean Black Goats (청보리사일리지 및 옥수수사일리지 급여가 육성기 재래 흑염소의 영양소 이용률, 성장 및 혈액성상에 미치는 영향)

  • This study examined the effects of feeding whole-crop barley silage (BS), corn silage (CS) and the mixture of BS and CS (CB) on feed consumption, daily gain, nutrients apparent digestibilities and blood parameters in order to investigate the feed evaluation in Korean black goats. BS, CS, and CB were fed to twelve Korean black goats, which were allocated into three groups in metabolic cages according to the latin-square design. As a result, dry and organic matter intakes for goats per day have shown a tendency to decrease in the order of CS, CB and BS, but there were no significant differences. We also found a similar tendency when the intakes of crude protein, crude fat, and non-fiber carbohydrates (NFC) were examined. However, ADF and NDF consumptions showed a decreasing tendency in the order of CB, BS and CS. Dry matter intakes per metabolic body weight showed the decreasing tendency in the order of CS (57.6g), CB (53.8g) and BS (52.7g), however with no statistical differences. Average daily gain of 64.2 g in CB was significantly higher than the other groups (42.0g to 46.0g). Nitrogen intakes and nitrogen retention showed a decreasing tendency in the order of CS, CB and BC. Nevertheless, nitrogen retention in CS group was higher by 30% than that of the BC group with p<0.05. Apparant digestibilities of dry and organic matters were 72.6 and 72.8% for the CS group respectively, which were significantly higher than those of the BS group (67.5 and 69.0%) and the CB group (66.0 and 67.1%). In conclusion, the results of this study demonstrated that the CS group showed more positive feed consumption, digestibility, and nitrogen retention in Korean black goats than those of BS group. The CB group, however, showed similar results to the results of the CS group.

Catch up growth in children born small for gestational age by corrected growth curve (부당 경량아로 출생한 소아들에서 교정성장곡선을 이용한 따라잡기 성장에 대한 연구)

  • Purpose : Being small for gestational age (SGA) is a risk factor of short stature in children. Genetic background such as mid-parental height (MPH) is known to influence growth of children born SGA. We studied the relationship between growth of children born SGA and MPH and studied the effects of insulin-like growth factor (IGF-I) and insulin-like growth factor binding protein 3 (IGFBP-3) on postnatal growth in children born SGA according to MPH. Methods : Forty-nine neonates born SGA were included in this study. We defined corrected height standard deviation score (cHtSDS) by modified height SDS (HtSDS) based on their MPH. We categorized subjects into group 1 consisting of children with cHtSDS ${\geq}0$ (n=35) and group 2 consisting of children with cHtSDS <0 (n=14), and compared IGF-I and IGFBP-3 between the two groups. Results : The HtSDSs and cHtSDSs in groups 1 and 2 were $0.06{\pm}1.05$ vs. $-0.95{\pm}0.85$ (P=0.000) and $0.78{\pm}0.93$ vs. $-0.46{\pm}0.67$ (P=0.000), respectively. IGF-I SDS was higher in group 1 than in group 2 ($2.82{\pm}3.69$ vs. $0.23{\pm}2.42$, P=0.012). Total cHtSDS ($0.42{\pm}1.03$) was significantly higher than HtSDS ($-0.22{\pm}1.10$) (P=0.000). Conclusion : Our results show that cHtSDS differs significantly from HtSDS. Growth assessment by standardized growth curve does not uniformly show effects of genetic factors. A more accurate assessment of growth uses a personalized corrected growth curve that considers the genetic factor measured by MPH.

"Youhan", New Whole Crop Barley Cultivar of Hooded Spike and Fine Growth Ability in Spring (초기생육이 빠른 삼차망 청보리 신품종 '유한')

  • "Youhan" (Hordeum vulgare L.), a new whole-crop barley cultivar, was developed by the breeding team at National Institute of Crop Science, RDA in 2012. Youhan has the growth habit of III, a light green and mid-sized leaf, hooded and lax-type spikes. The cultivar showed 107 cm of culm length, 641 spikes per $m^2$. The heading date of Youhan was May 1, which is one day later than that of the check cultivar "Yuyeon" in upland, and 2 days earlier than that of Yuyeon in paddy field. The maturation time was similar to check cultivar Yuyeon on June 4 in upland and May 31 in paddy field. In terms of winter hardiness and resistance to lodging and disease, Youhan also performed better than the check cultivar. The average forage dry matter (DM) yield in the regional yield trial was approximately 12.6 ton $ha^{-1}$ and 12.0 ton $ha^{-1}$ in upland and paddy field, respectively, which were 6% and 5% higher than that of the check. The yield also showed 7.3% of crude protein, 26.8% of ADF (acid detergent fiber), 47.8% of NDF (neutral detergent fiber), 67.7% of TDN (total digestible nutrients), and a higher grade of silage quality for the whole-crop barley. Fall sowing cropping of Youhan is recommended only in those areas where the average daily minimum-mean temperatures in January are higher than $-8^{\circ}C$, and it should not be cultivated in mountainous areas of Korea.

Effect of Cooking methods with Various Heating Apparatus on the Quality Characteristics of Beef (가열기구에 따른 조리방법이 소고기의 품질특성에 미치는 영향)

  • This study showed the chemico-physical characteristics and sensory evaluation of beef loin and ribs with various kinds of cooking apparatus and methods like pan heating, boiling, grilling, steaming, cooking heating, charcoal heating and double layer pan heating. Double layer pan filled with Phase Change Materials(PCM) heating was also tested. Heating time of beef loin and ribs by oven heating was the longest as 55.5 and 25.7 min. Cooking loss of beef loin and ribs were 40.49%, 27.98% each and showed biggest in charcoal heating(p<0.05). In the chemical analysis, grilling resulted in the highest protein contents of 29.3% in the loin and double layer pan heating had 28.6% in the ribs. In the loin, 20.3% of crude fat was the highest in the double layer pan heating, and 21.9% of crude fat of ribs in the PCM heating(p<0.05). In the test of shearing forces, double layer pan heating had the lowest result of $9.14kg/cm^2$ in the loin and $7.03kg/cm^2$ in the ribs. In the 9 point-scale sensory evaluation, PCM treatment showed the best result of 7.67 in the juiciness of loin and single layer pan heating and grilling treatment had the highest score of 7.00 in the ribs. Loin in the PCM heating had 7.11 and ribs in the grilling had 7.13, showing the best in overall acceptability. With this experiments, we could find the chemico-physical and sensory characteristics of beef loin and ribs were affected by various cooking methods.
