The precipitation polymerization of acrylonitrile (AN) was carried out in a mixture solution of dimethyl sulfoxide (DMSO) and water at $50{\sim}65^{\circ}C$ using ${\alpha},{\alpha}'$-azobisisobutyronitrile (AIBN) as an initiator. The increased molecular weight polyacrylonitrile (PAN) was prepared with increasing the $H_2O$/DMSO ratio from 10/90 to 80/20. The viscosity average molecular weight of $H_2O$/DMSO solvent was 4.4 times larger than that of $H_2O$/DMF solvent, and precipitation polymerization was accelerlated due to the far decreased chain transfer effect of DMSO. Based on the experimental results, the increased PAN molecular weight was regarded as the summation of two mechanisms: i) particle-particle aggregation and ii) particle-radical attachment. The theoretical equation derived from the mechanisms was well coincided with the experimental results showing the linear relationship between the viscosity average molecular weight and the H2O/DMSO ratio.

Chinese named entity recognition (NER) is a challenging work that seeks to find, recognize and classify various types of information elements in unstructured text. Due to the Chinese text has no natural boundary like the spaces in the English text, Chinese named entity identification is much more difficult. At present, most deep learning based NER models are developed using a bidirectional long short-term memory network (BiLSTM), yet the performance still has some space to improve. To further improve their performance in Chinese NER tasks, we propose a new NER model, IDCNN-BiLSTM-Highway, which is a combination of the BiLSTM, the iterated dilated convolutional neural network (IDCNN) and the highway network. In our model, IDCNN is used to achieve multiscale context aggregation from a long sequence of words. Highway network is used to effectively connect different layers of networks, allowing information to pass through network layers smoothly without attenuation. Finally, the global optimum tag result is obtained by introducing conditional random field (CRF). The experimental results show that compared with other popular deep learning-based NER models, our model shows superior performance on two Chinese NER data sets: Resume and Yidu-S4k, The F1-scores are 94.98 and 77.59, respectively.
The term "Big Data" has been defined to encapsulate a broad spectrum of data sources and data formats. It is often described to be unstructured data due to its properties of variety in data formats. Even though the traditional methods of structuring data in rows and columns have been reinvented into column families, key-value or completely replaced with JSON documents in document-based databases, the fact still remains that data have to be reshaped to conform to certain structure in order to persistently store the data on disc. ETL processes are key in restructuring data. However, ETL processes incur additional processing overhead and also require that data sources are maintained in predefined formats. Consequently, data in certain formats are completely ignored because designing ETL processes to cater for all possible data formats is almost impossible. Potentially, these unconsidered data sources can provide useful insights when incorporated into big data analytics. In this project, using big data solution, Apache Spark, we tapped into other sources of data stored in their raw formats such as various text files, compressed files etc and incorporated the data with persistently stored enterprise data in MongoDB for overall data analytics using MongoDB Aggregation Framework and MapReduce. This significantly differs from the traditional ETL systems in the sense that it is compactible regardless of the data formats at source.
Melanosomes are specific melanin-containing intracellular organelles of epidermal melanocytes. In epidermal melanocytes, there are three kinds of key player proteins. Rab27a, melanophilin or Slac2-a and Myosin 5a form a tripartite complex connects the melanosome. Mature melanosomes make movements through the tripartite protein complex along actin filaments. In this study, we found that the haplamine (6-Methoxyflindersine) induced melanosome aggregation around the nucleus in epidermal melanocyte. In an attempt to elucidate the inhibitory effect of haplamine on melanosome transport, effect of haplamineon the expression level of Rab27a, melanophilin and Myosin 5a was measured. The results indicated that haplamine up to 5M effectively suppressed mRNA and protein expression level of melanophilin. To determine the upstream regulator of melanophilin regulated by haplamine, we checked the level of MITF, c-JUN and USF1. Those are possible transcription factor of melanophilin. Among them, treatment of USF1 siRNA decreased mRNA and protein expression level of USF1 as well as melanophilin. Also, treatment of haplamine decreased mRNA and protein expression level of melanophilin as well as USF1 in a dose-dependent manner. Consequently, we found the inhibitory effect of haplamine on melanosome transport in melan-a melanocyte. Treatment of haplamine reduced melanophilin expression level which is a key protein of melanosome transport. We identified that USF1 could be a major transcription factor of melanophilin regulated by haplamine.
Due to the high incidence of malignant melanoma, the establishment of in vitro models that recapitulate the tumor microenvironment is of great biological and clinical importance for tumor treatment and drug research. In this study, 3D printing technology was used to prepare GelMA/PEGDA composite scaffolds that mimic the microenvironment of human malignant melanoma cell (A375) growth and construct in vitro melanoma micro-models. The GelMA/PEGDA hybrid scaffold was tested by the mechanical property, cell live/dead assay, cell proliferation assay, cytoskeleton staining and drug loading assay. The growth of tumor cells in two- and three-dimensional culture systems and the anti-cancer effect of luteolin were evaluated using the live/dead staining method and the Cell Counting Kit-8 (CCK-8) method. The results showed a high aggregation of tumor cells on the 3D scaffold, which was suitable for long-term culture. Cytoskeleton staining and immunofluorescent protein staining were used to evaluate the degree of differentiation of tumor cells under 2D and 3D culture systems. The results indicated that 3D bioprinted scaffolds were more suitable for tumor cell expansion and differentiation, and the tumor cells were more aggressive. In addition, luteolin was time- and dose-dependent on tumor cells, and tumor cells in the 3D culture system were more resistant to the drug.
P19 cells are pluripotent embryonal carcinoma cells and can be differentiated into neuronal cell type by treatment with retinoic acid (RA) and aggregation culture. Cannabinoids are the active components of Cannabis sativa and they have diverse pharmacologic activities, such as pain control, anti-inflammatory effects, neuro-protection effects and tumor regression. Cannabinoids also involved in neuronal proliferation, migration, differentiation and survival in developing brain. Here, we studied the role of cannabinoids on neuronal differentiation of P19 cells. Treatment with cannabinoids increased the neuronal differentiation induced by RA and also promoted transcriptional activity of neurogenin 1, key transcription factor for neuronal differentiation of P19 cells. These results suggest that the cannabinoids can accelerate neuronal differentiation of P19 cells.
