Evaluation of Swab in Maintaining Survival Efficiency according to the CLSI M40-A2 Standard

  • Transporting clinical samples for microbiological testing requires a proper transport medium that guarantees the survival of microorganisms. Therefore, the aim of the study was to determine the ability of Amies Transport Medium (ATM) to maintain the viability of microorganisms in clinical specimens and its suitability as a transport medium for microbiological testing. This study evaluated the performance of swab provided by KS Co., Ltd. for three groups of bacteria comprising aerobic and facultative anaerobic bacteria, anaerobic bacteria, and fastidious bacteria, according to the Clinical and Laboratory Standard Institute (CLSI) 8.11.2. The ATM stability test was conducted by dividing the medium into two groups based on the product expiration date of use. All tested media, A and B (the date of manufacture and expiration date are different) showed ≥5 CFU, and there was no significant difference in the result values of Category A and Category B with different serial numbers for each test. The results of this experiment when cross-checked with the guidelines suggest that ATM is a suitable transport medium for microbiological testing, as it maintains the viability of microorganisms and is suitable for overgrowth trials. In addition, compared to the number of CFUs at the origin, the number of CFUs did not increase by more than 1 log after storage. These results have important implications for the development of transport media that can guarantee the survival of microorganisms in clinical specimens.

  • The main challenges for the development of agricultural bio-degradable mulching film concern the degradation during the lifetime of cultivated crops. A set of rice by-product (rice-hull and rice-bran) based bio-degradable mulching films was developed and tested, following the measurement of standard bio-degradability rate and adaptability in a large scale field experiment. The standard bio-degradability of bio-film passed the KS (Korea standard) regulation. The result of mechanical analysis of bio-degradable mulching film presented a higher mechanical strength and elongation rate compared with polyethylene film. In addition, bio-film could be degraded into fragments within 4 months under the field condition of several upland crops. Bio-degradable mulching film indicated great potential as a new source of agricultural bio-degradable material.

  • Mineral carbonation is a technology in which carbonates are synthesized from minerals including serpentine and olivine, and industrial wastes such as slag and cement, of which all contain calcium or magnesium when reacted with carbon dioxide. This study aims to develop the mineral carbonation technology for commercialization, which can reduce environmental burden and process cost through the reduction of carbon dioxide using steel slag and the slag reuse after calcium extraction. Calcium extraction was conducted using NH4Cl solution for air-cooled slag and convert slag, and ${\geq}98%$ purity calcium carbonate was synthesized by reaction with calcium-extracted solution and carbon dioxide. And we conducted experimentally to minimize the quantity of by-product, the slag residue after calcium extraction, which has occupied large amount of weight ratio (about 80-90%) at the point of mineral carbonation process using slag. The slag residue was used to replace silica sand in the manufacture of cement panel, and physical properties including compressive strength and flexible strength of panel using the slag residue and normal cement panel, respectively, were analyzed. The calcium concentration in extraction solution was analyzed by inductively coupled plasma optical emission spectrometer (ICP-OES). Field-emission scanning electron microscope (FE-SEM) was also used to identify the surface morphology of calcium carbonate, and XRD was used to analyze the crystallinity and the quantitative analysis of calcium carbonate. In addition, the cement panel evaluation was carried out according to KS L ISO 679, and the compressive strength and flexural strength of the panels were measured.

  • The Effects of Co-substitution in the nanocrystalline Nd-Fe-B-Mo-Cu alloys were investigated. $\alpha$-Fe based nanocrystalline Nd-Fe-B-Mo-Cu alloys were prepared by crystallization process of amorphous Nd-Fe-B-Mo-Cu alloy produced by rapid solidification process. The substitution of Co resulted in the decrease of grain size and improves the hard magnetic properties. The remanence, coercivity, and Curie temperature of nanocrystalline N $d_4$(F $e_{0.85}$ $Co_{0.15}$)$_{82}$ $B_{10}$M $o_3$Cu alloy showed more improved magnetic properties than those of Co-free alloy. The grain size was measured to be about 15 nm. The coercivity, remanence and maximum energy product were 239 kA/m, 1.41, and 103.5 kJ/ $m^3$, respectively, for the nanocrystalline N $d_4$(F $e_{0.85}$ $Co_{0.15}$)$_{82}$ $B_{10}$M $o_3$Cu alloy annealed for 0.6 ks at 640 $^{\circ}C$.

RRP Loading Patterns and Standard Dimensions for Block Pattern in Membership Wholesale Clubs (Membership Wholesale Club에서의 RRP 적재패턴 및 블록패턴 표준규격에 관한 연구)

  • Purpose - This study analyzes loading efficiency by loading pattern for package standardization and reduction of logistics costs, along with the creation of revenue for the revenue review panel (RRP) of Membership Wholesale Clubs (MWC). The study aims to identify standard dimensions that can help improve the compatibility of the pallets related to display patterns preferred by the MWC and thereby explore ways to enhance logistics efficiency between manufacturers and retailers through standardization. Research design, data, and methodology - The study investigates and analyzes the current status based on actual case examples, i.e., manufacturer A and Korea's MWC (A company, B company, and C company), and thus devises improvement measures. To achieve this, the case of manufacturer A delivering to MWC was examined, and the actual pallet display patterns for each MWC were investigated by visiting each distribution site. In this study, TOPS (Total Optimization Packaging Software, USA) was used as the tool for pallet loading efficiency simulations the maximum allowable dimension was set to 0.0mm to prevent the pallet from falling outside the parameters, and the loading efficiency was analyzed with the pallet area. In other words, the study focused on dimensions (length x width x height) according to the research purpose and thereby deduced results. Results - The analysis of pallet loading patterns showed that the most preferred loading patterns for loading efficiency according to product specification, such as pinwheel, brick, and block patterns, were used in the case of the general distribution products, but the products were configured with block patterns in most cases when delivered to MWCs. The loading efficiency by loading pattern was analyzed with respect to 104 nationally listed standard dimensions. Meanwhile, No.51 (330 × 220mm) of KS T 1002 (1,100 × 1,100mm) was found to be the dimension that could bring about an improved loading efficiency, over 90.0% simultaneously in both the T-11 and T-12 pallet systems in a loading pattern configuration with the block pattern only, and the loading efficiency simulation results also confirmed this as the standard dimension that can be commonly applied to both the T-11 pallet (90.0%) and the T-12 pallet (90.7%) systems. Conclusions - The loading efficiency simulation results by loading pattern were analyzed: For the T-11 pallet system, 17 standard dimension sizes displayed the loading efficiency of 90.0% or more as block patterns, and the loading capacity was an average of 99.0%. For the T-12 pallet system, 35 standard dimension sizes displayed the loading efficiency of more than 90% as block patterns (the average loading efficiency of 98.6%). Accordingly, this study proposes that the standard dimensions of 17 sizes with the average loading efficiency of 99.0% should be applied in the use of the T-11 pallet system, and those of 35 sizes with the average loading efficiency of 98.6% should be reviewed and applied in the use of the T-12 pallet system.

Racking Property of Light-framed Shear Wall with Hold-down Connector (홀드다운을 적용한 경골목조 벽체의 전단성능)

  • As the height of the light-framed building increases, the lateral load and overturn-moment are increased and the possibility of the building overturn becomes larger. Because the shear wall resists lateral load in light-framed building, the reinforcement of shear wall is required. In order to reinforce the light-framed shear wall, using lag screw fastener type (B-HD) and using bolt type (S-HD) hold-down connectors were applied for test. And domestic larch lumbers, $38{\times}140mm$ and $89{\times}140mm$, KS 2nd grade, were used for the stud. The North American OSB panels were used for sheathing panel. Static loads, load speed 6 mm/min, were applied on top of the wall. As a result, shear strength of the wall that using hold-down connector was improved sufficiently. And when applying the S-HD type hold-down connector, stud should be reinforced against weakening by drilled hole. As increasing the number of lag screw, the number of bolt and the product allowable strength, the strength of shear wall that using hold-down connector was also increased. When applying hold-down connector to light-framed building using 38 mm stud, it must be reinforced by enlarging the thickness of stud like as 38 mm doubled column.

A Study on the Safety Estimation of Wiring Connection Connector Manufactured by Housing Type (하우징 형태(Housing Type)로 제작된 배선 연결 커넥터의 안전성 평가에 관한 연구)

  • The purpose of this study is to evaluate the safety of a wire connector fabricated for the effective installation of a lighting fixture including its contact resistance, insulation resistance, withstanding voltage characteristics, etc., and to provide the basis for the analysis and judgment of PL(Product Liability) dispute by presenting a damage pattern due to a general flame and overcurrent. This study applied the Korean Standard (KS) for the incombustibility test of the connector using a general flame and performed an overcurrent characteristics test of the connector using PCITS (Primary Current Injection Test System). The contact resistance of the housing connector was measured using a high resistance meter and the insulation resistance was measured using a multimeter. In addition, a supply voltage of AC 1,500V for testing the withstanding voltage characteristics was applied to both ends of the connector. Measurement was performed on 5 specimens and the measured values were used as a basis for judgment. Since the connector is fabricated in the form of a housing, it can be connected and separated easily and has a structure that allows no foreign material to enter. In addition, since it has a structure that allows wires to be connected only when their polarity is identical, any misconnection that may occur during installation can be prevented. When the incombustibility test was performed by applying a general flame to the connector, it showed outstanding incombustibility characteristics and the blade and blade holder connected to the housing remained firmly secured even after the insulation sheath (PVC) was completely destroyed by fire. In addition, the mechanism of the damaged connecting wire showed a comparatively uniform carbonization pattern and it was found that some residual melted insulation material was attached to both ends. In the accelerated life test (ALT) to which approximately 500% of the rated current was applied, the connector damage proceeded in the order of white smoke generation, wire separation, spark occurrence and carbonization. That is, it could be seen that the connector damaged by overcurrent lost its own metallic color with traces of discoloration and carbonization. The contact resistance of the connector at a normal state was 2.164mV/A on average. The contact resistance measured after the high temperature test was 3.258mV/A. In addition, the insulation resistance after the temperature test was completed was greater than $10G\Omega$ and the withstanding voltage test result showed that no insulation breakdown occurred to all specimens showing stable withstanding voltage and insulation resistance characteristics.

Preparation and Quality Characteristics of Japanese Spanish Mackerel Scombermorus niphonius Processed Products with Different Physical Properties as Senior-friendly Seafoods (물성을 달리한 삼치(Scombermorus niphonius) 활용 고령친화식품의 제조 및 품질특성)

  • This study was conducted to prepare braised product with soy sauce (BP), rose gratin (RG) and white mousse (WM) using Japanese Spanish mackerel Scombermorus niphonius as senior-friendly seafoods with different physical and nutritional properties and to examine their quality characteristics. The hardness of BP, RG and WM were 428.0×103, 46.9×103 and 16.0×103 N/m2, respectively. The nutritional values of BP, RG and WM are 17.8, 9.1 and 18.5 g, respectively, for protein; 11.86, 23.32 and 13.35 ㎍ RAE, respectively, for vitamin A; 1.27, 0.49 and 0.17 ㎍, respectively, for vitamin D; 45.21, 12.79, and 35.54 mg, respectively, for vitamin C; 0.17, 0.13 and 0.23 mg, respectively, for riboflavin; 4.28, 2.94 and 3.65 mgNE, respectively, for niacin; 19.6, 104.0 and 48.1 mg, respectively, for Ca; 441.6, 271.1 and 250.1 mg, respectively, for K; 0.20, 0.80 mg and undetection, respectively, for dietary fiber. Escherichia coli was undetected in all the products. These results indicated that the products be classified as steps 1 for BP, step 2 for RG and step 3 for WM of senior-friendly seafoods based on the KS, and as nutritional and physical properties-improved senior-friendly seafoods based on the Food Code.

Skin barrier and anti-inflammatory effect of petasites japonicus (머위(Petasites japonicus)의 피부장벽과 항염증 효과)

  • In this study, to investigate the possibility of using ethanol extract of Petasites japonicus (PJE) as a functional material, we investigated the activity of improving skin barrier and inflammation through UVB-induced human keratinocyte (HaCaT cell). As a result of confirming the antioxidant effect through DPPH radical scavenging activity, ABTS+ radical scavenging activity, and hydrogen peroxide scavenging activity, it was confirmed that it had an antioxidant effect similar to that of ascorbic acid, a control, at a concentration of 1 mg/ml. As a result of confirming the mRNA expression of the production ability of filaggrin and aquaporin-3 in HaCaT cells induced by UVB, it was confirmed that the reduced expression level by UVB stimulation increased in a concentration-dependent manner when the PJE was treated. It was confirmed that the mRNA expression of TNF-𝛼 and IL-1𝛽 were increased by UVB stimulation and decreased when the PJE was treated. As a result of the migration assay, it was confirmed that the proliferation of skin keratinocytes and the recovery rate of wounds were increased in a concentration-dependent manner. Based on the experimental results, it suggests that Petasites japonicus can be used as a functional cosmetic product that can improve skin moisturizing and skin barrier function.

Physicochemical Characteristics of Marinated Abalone Haliotis discus hannai in Seasoned Soy Sauce for the Elderly Using Texture Modification Technology (물성조절 기술을 활용한 고령친화식 전복(Haliotis discus hannai)장의 품질 특성)

  • This study aimed to develop and commercialize abalone Haliotis discus hannai marinated in seasoned soy sauce (AM-S) for the elderly by adjusting its texture (hardness) and strengthening its nutritional components. The number of pounding cycles [X1, 2-14 times] and heating time in a retort [X2, 5-19 min] were selected as independent variables, and hardness (Y1) and overall acceptance (Y2) were selected as dependent variables. The optimal conditions for X1 and X2 were 12 pounding cycles and heating in a retort for 17 min, respectively. The hardness values 209.2×103 N/m2 for the third method and 259.1×103 N/m2 for the first method. The nutritional value was 13.1 g/100 g crude protein and vitamins A, D, and C were not detected. The mineral content was 33.2 mg/100 g calcium and 258.7 mg/100 g potassium. Organisms from the coliform group and Escherichia coli were not detected. Therefore, AM-S for the elderly was classified as a hardness- and nutrient-controlled product based on the standards and specifications of senior-friendly foods provided in the MFDS and classified as the first step of senior-friendly foods according to the standards and specifications of senior-friendly foods provided in the KS.
