Expression and cDNA Cloning of klp-12 Gene Encoding an Ortholog of the Chicken Chromokinesin, Mediating Chromosome Segregation in Caenorhabditis elegans

  • In eukaryotes, chromosomes undergo a series of complex and coordinated movements during cell division. The kinesin motor proteins, such as the chicken Chromokinesin, are known to bind DNA and transport chromosomes on spindle microtubles. We previously cloned a family of retrograde C-terminus kinesins in Caenorhabditis elegans that mediate chromosomal movement during embryonic development. Here we report the cloning of a C. elegans klp-12 cDNA, encoding an ortholog of chicken Chromokinesin and mouse KIF4. The KLP-12 protein contains 1609 amino acid and harbors two leucine zipper motifs. The insitu RNA hybridization in embryonic stages shows that the klp-12 gene is expressed during the entire embryonic development. The RNA interference assay reveals that, similar to the role of Chromokinesin, klp-12 functions in chromosome segregation. These results support the notion that during mitosis both types, the anterograde N-terminus kinesins such as KLP-12 and the retrograde C-terminus kinesins, such as KLP-3, KLP-15, KLP-16, and KLP-17, may coordinate chromosome assembly at the metaphase plate and chromosomal segregation towards the spindle poles in C. elegans.

Antimicrobial Activity of Culture Filtrates from Pseudomonas aeruginosa KLP-2 Isolated from Cooling Tower-Water against Legionella pneumophila (냉각탑수에서 분리한 Pseudomonas aeruginosa KLP-2 배양여액의 Legionella Pneumophila에 대한 항균활성)

  • Pseudomonas aeruginosa KLP-2 possessing antimicro- bial activity against Legionella pneumophila was isolated from cooling tower-waters. The culture filtrates of P. aeruginosa KLP-2 showed antimicrobial activity against L. pneumophila Vibro cholerae non-Ol. Bacillus cereus, Bacillus subtilis and staphylococcus aureus. The culture filtrates of P. aeruginosa KLP-2 showed the highest anti-microbial activity against. L pneumophila among micoorganisms tested in this study. The optimal conditions of temperature, pH carbon source and nitogen source to obtain maximal antimicrobial activity from culture filtrates of P. aeruginosa KLP-2 were determined to be 35$^{\circ}C$ pH 7.0 % of glycerol and 0.6% of proteose peptone respec- tively. The antimicrobial activity of culture filtrates from P. seruginosa KLP-2 was highest against L. pneumophila when cultivated with shaking for 24 h and without shaking for 4 day at 35$^{\circ}C$.

  • Kirchhoff-Love plate (KLP) equation is a well established theory for a description of a deformation of a thin plate under certain outer source. Meanwhile, analysis of a vibrating plate in a frequency domain is important in terms of obtaining the main frequency/eigenfunctions and predicting the vibration of plate. Among various modal analysis methods, dynamic mode decomposition (DMD) is one of the efficient data-driven methods. In this work, we carry out DMD based modal analysis for KLP where thin plate is under effects of sine-type outer force. We first construct discrete time series of KLP solutions based on a finite difference method (FDM). Over 720,000 number of FDM-generated solutions, we select only 500 number of solutions for the DMD implementation. We report the resulting DMD-modes for KLP. Also, we show how DMD can be used to predict KLP solutions in an efficient way.

Energy-Efficient and High Performance CGRA-based Multi-Core Architecture

  • Coarse-grained reconfigurable architecture (CGRA)-based multi-core architecture aims at achieving high performance by kernel level parallelism (KLP). However, the existing CGRA-based multi-core architectures suffer from much energy and performance bottleneck when trying to exploit the KLP because of poor resource utilization caused by insufficient flexibility. In this work, we propose a new ring-based sharing fabric (RSF) to boost their flexibility level for the efficient resource utilization focusing on the kernel-stream type of the KLP. In addition, based on the RSF, we introduce a novel inter-CGRA reconfiguration technique for the efficient pipelining of kernel-stream on CGRA-based multi-core architectures. Experimental results show that the proposed approaches improve performance by up to 50.62 times and reduce energy by up to 50.16% when compared with the conventional CGRA-based multi-core architectures.

Physico-Chemical Properties and Antimicrobial Activity of Pyocyanine Produced by Pseudomonase aeruginosa KLP-2 (Pseudomonas aeruginosa KLP-2가 생산한 Pyocyanine의 항균활성 및 생리화학적 성상)

  • The antimicrobial substance produced by Pseudomonas aeruginosa KLP-2 strain was purified and identified. The substance was identified as a pyocyanine by the fast atom bombardment mass(FAB-MS). In physic-chemical properties, the pyocyanine was dark blue needles, and was soluble in various organic solvents such as chlorogorm, methanol, ethanol and ethyl acetae. The pyocyanine possessed a ultraviolet absorbance spectrum in methanol, 0.1 M HCl, and chlorogorm. The maximum absorption peak of the pyocyanine showed at 318 mm in methanol. The molecular formula of the pyocyanine was determined to the $C_{13}$ H$_{10}$ N$_{2}$O and protonate molecular ion species (M+H)$^{+}$ was observed at m/z 211 by FAB-MS. The pyocyanine showed antimicrobial against Bacillus cereus, Bacillus subtilis, Micrococcus luteus, Rodococcus equi, Staphylococcus aureus, Streptococcus faecalis, E. col, Legionella pneumophila, Shigella flexneri Shigella boydii, shgella sonnei, NAG Vibrio cholerae, Vibrio parahaemolyticus, Vibro vulnificus, Yersinia enterocolitica, and Saccharomyces cerevisiae. However, Salmonella spp. Shigela dysenteriae, 3 strains of Pseudomonas aeruginosa, Klebsiela pneumoniae, and Aspergillus niger were resistant to the pyocyanine. The pyocyanine showed the highest antimicrobial activity aganist Legionella pneumophila based on the size of inhibition zone by the disk contained 0.5 $\mu\textrm{g}$ of the pyocyanine.e.

The Effect of Syllable Frequency, Syllable Type and Final Consonant on Hangeul Word and Pseudo-word Lexical Decision: An Analysis of the Korean Lexicon Project Database (한글 두 글자 단어와 비단어의 어휘판단에 글자 빈도, 글자 유형, 받침이 미치는 영향: KLP 자료의 분석)

  • This study attempted to find out how lexical decision of two-syllable words or pseudo-words is affected by syllabic information, such as syllable frequency, syllable (i.e. vowel) type, and presence of final consonant (i.e. batchim), through the analysis of the Korean Lexicon Project Database (KLP-DB). Hierarchical regression of RT data showed that lexical decision of words was influenced by the frequency of the first syllable, the syllable type of the first and second syllables, batchim for the first and second syllables, and also by the interaction of the two syllable types and the interaction of syllable frequency and batchim of the second syllable. For pseudo-words lexical decision was influenced by the frequency of the first and second syllables, syllable type of the first syllable, and batchim for the first and second syllables, and also by the interaction of the two syllable frequencies, the interaction of the two syllable types, and the interaction of syllable frequency and batchim of the first syllable. Word frequency had a strong effect on lexical decision of words, while syllabic information had a stable effect on the lexical decision of pseudo-words. These results indicate that syllabic information should be seriously considered in constructing word and pseudo-word lists and interpreting lexical decision time. Understanding the effect of syllabic information will also contribute to the understanding of word recognition process.

Complete Sequence of a Gene Encoding KAR3-Related Kinesin-like Protein in Candida albicans

  • In contrast to Saccharomyces cerevisiae, little is known about the kinesin-like protein (KLP) in Candida albicans. The motor domain of kinesin, or KLP, contains a subregion, which is well conserved from yeast to humans. A similarity search, with the murine ubiquitous kinesin heavy chain region as a query, revealed 6 contigs that contain putative KLPs in the genome of C. albicans. Of these, the length of an open reading (ORF) of 375 amino acids, temporarily designated CaKAR3, was noticeably short compared with the closely related S. cerevisiae KAR3 (ScKAR3) of 729 amino acids. This finding prompted us to isolate a ${\lambda}$ genomic clone containing the complete CaKAR3 ORF, and here the complete sequence of CaKAR3 is reported. CaKAR3 is a C-terminus motor protein, of 687 amino acids, encoded by a non-disrupting gene. When compared with ScKAR3, the amino terminal region of 112 amino acids was unique, with the middle part of the 306 amino acids exhibiting $25\%$ identity and $44\%$ similarity, while the remaining C-terminal motor domain exhibited $64\%$ identity and $78\%$ similarity, and have been submitted to GeneBank under the accession number AY182242.

  • 개체명 인식(Named Entity Recognition) 시스템은 문서에서 인명(PS), 지명(LC), 단체명(OG)과 같은 개체명을 가지는 단어나 어구를 해당 개체명으로 인식하는 시스템이다. 개체명 인식 시스템을 개발하기 위해 딥러닝 기반의 워드 임베딩(word embedding) 자질과 문장의 형태적 특징 및 기구축 사전(lexicon) 기반의 자질 구성 방법을 제안하고, bi-directional LSTM, CNN, CRF과 같은 모델을 이용하여 구성된 자질을 학습하는 방법을 제안한다. 실험 데이터는 2017 국어 정보시스템 경진대회에서 제공한 2016klpNER 데이터를 이용하였다. 실험은 전체 4258 문장 중 학습 데이터 3406 문장, 검증 데이터 426 문장, 테스트 데이터 426 문장으로 데이터를 나누어 실험을 진행하였다. 실험 결과 본 연구에서 제안하는 모델은 BIO 태깅 방식의 개체 청크 단위 성능 평가 결과 98.9%의 테스트 정확도(test accuracy)와 89.4%의 f1-score를 나타냈다.
