방선균 KF923이 생산하는 Phospholipase D의 정제 및 특성 (Purification and Characterization of Phospholipase D from Actionmycetes KF923)

  • 국내 6개 지역에 걸쳐 채취한 토양 63점으로부터 1,000여 종의 방선균주를 분리하였다. 이들 균주의 배양 상징액 중 PLD의 생산이 양호한 균주를 탐색한 결과, 분해활성이 0.3U/$m\ell$이상인 131 균주를 일차로 선발하였다. 이중 전이 활성이 20% 이상인 균주는 23개였으며, 최종적으로 분해 및 전이활성이 가장 우수한 균주(방선균 KF923)를 선발하였다. 방선균 KF923 균주는 P 배지하에서 발효조 배양시 배양 48시간 후에 최고의 분해활성을 나타내었고(13U/$m\ell$) 전이활성은 95%로 나타났다. 방선균 KF923 균주가 생산한 PLD를 정제하여 비활성이 567U/mg의 PLD를 얻었다. 정제된 PLD는 분자량이 약 55kD으로 나타났고, 효소반응의 최적 pH는 6.0, 최적온도는 $60^{\circ}C$였으며, 효소의 안정성에 미지는 pH 및 온도의 영향은 pH 8.0부근 및 $40^{\circ}C$ 이하에서 가장 안정하였다. 또한, 특별한 금속이온의 요구성은 없는 것으로 나타났다. 방선균 KF923 균주가 생산한 PLD는 산업화에 충분히 활용 가능하며, 이를 위하여 효소의 대량생산 및 효소반응을 위한 공정개발 등의 연구가 필요하다고 생각한다.

보건의료종사자의 안면부여과식 마스크의 사용과 밀착도검사의 중요성 (Usage of Filtering-facepiece Masks for Healthcare Workers and Importance of Fit Testing)

  • Objectives: One aim of the study is to compare filtering facepiece masks for healthcare workers between Korea and other countries. The other is to emphasize the importance of fit testing for these masks using an analysis of previous research. Materials: An extensive literature review was performed by searching a number of websites and existing studies. Results: KF94 and KF99 masks certified by the Korean CDC are suitable for healthcare workers as filtering facepiece masks. The standards for these respirators are similar to FFP2 and FFP3 of EN 143 and 149. The performance, such as filtering efficiency, is almost the same between KP94 and N95. It was found that fit testing of respirators for healthcare workers was important to reduce infection risk. Conclusions: KF94 should be emphasized as filtering facepiece masks for healthcare workers rather than N95. Even though Korea has no fit testing regulations, implementing fit testing in healthcare settings is strongly recommended to decrease infection risk.

Plant Terpenes Enhance Survivability of Polychlorinated Biphenyl (PCB) Degrading Pseudomonas pseudoalcaligenes KF707 Labeled with gfp in Microcosms Contaminated with PCB

  • Polychlorinated biphenyl are toxic pollutants and their degradation is quite slow in the environment. Recently, interest if bioremediation using PCB-degrading bacteria has increaset,. In a previous report, plant terpenes (p-cymene, (S)-(-)-limonene, ${\alpha}-pynene$, and ${\alpha}-terpinene$) have been found to be utilized by a PCB degrader and to induce the biphenyl dioxygenase gene in pure culture. In this study, Pseudomonas pseudoalcaligenes KF707, a PCB-degrading Gram-negative soil bacterium, was used to determine whether the terpene stimulation of PCB degrader occurred in the natural environment. First, P. pseudoalcaligenes KF707 was genetically tagged using a transposon with gfp (green fluorescent protein) as a reporter gone. The population dynamics of P. pseudoalcaligenes KF707 harboring gfp gene in a PCB-contaminated environment was examined with or without terpenoids added to the microcosm. About 10-100-fold increase was found in the population of PCB degraders when terpene was added, compared with control (non-terpenes samples and biphenyl added samples). It was proposed that the gfp-monitoring system is very useful and terpenes enhance the survivability of PCB degraders in PCB-contaminated environments.

  • 국내 건축물의 다양한 거주목적과 특성에 따른 재실자가 거주하게 됨에 발생되는 내부 인테리어 공사 등을 포함한 건축변경으로 인해 증대되는 화재위험성에 대한 분석과 대응은 미비하다. 건축피난성능이 저하된 기존 건축물이 늘어갈수록 화재사고가 발생할 가능성이 높아짐에 따라 체계적인 화재위험도평가를 수행하여, 화재위험을 저감하고 관리할 수 있는 제도적 장치와 평가기법을 개발이 필요하다. 본 연구는 그 첫 번째 과정으로서 기존 건축물 3개소에 대해서 1단계는 현장조사, 2단계는 화재피난성능평가를 진행하여 문제점을 분석하고 개선방안의 제안을 목적으로 진행하였다.

담배, 토마토 및 고추에서 분리한 TMV 계통의 추가적인 생물학적 특성 (Additional Biological Characteristics of TMV Strains Isolated from Tobacco, Tomato and Pepper Plants)

  • Tobacco (Nicotiana tabacum) cultivars including NC 82 and KF 114, and Datura stramonium, Physalis floridana, Gomphrena globosa, and Chenopodium spp. were added to the previous host plants tested for the further examination on the biological characteristics of tobacco mosaic virus (TMV) strains isolated from tobacco (TMV-Common), tomato (TMV-Tomato), and pepper (TMV-Pepper), In TMV-Tomato and TMV-Pepper, different symptoms were noted in P. floridana (no symptom development), and NC 82 (local lesion production on the inoculated leaves) from TMV-Common with which systemic mosaic symptoms were developed. Local lesions were developed in KF 114, D. stramonium, G. globosa, and Chenopodium spp by TMV-Common and TVM-Tomato, while no symptom was observed in KF114 and G. globosa. Also the number and size of local lesions were smaller in KF 114 than Xanthi-nc tobacco (local lesion host) infected with TMV-Tomato. Systemic necrosis was induced in Xanthi-nc and KF 114 when infected with TMV-C at high temperature, but not with the other strains.

Development of Tobacco Mosaic Virus (TMV) resistant Flue-cured $\textrm{F}_1$ Hybrid, KF118

  • Tobacco mosaic virus (TMV), TMV-common strain, causes severe damage at growth, yield and leaf quality on flue-cured tobacco in Korea. Controlling TMV outbreak with cultivating practices and chemicals are limited, thus, the best way would be developing TMV resistant variety. A new flue-cured hybrid, KF118, was developed from a cross of maternal parent MSTC175 and TC853 at the Agro-tech. Research Group, KT&G Central Research Institute. KF118 exhibited more vigorous growth characteristics in early stage than that of NC82, standard variety in Korea, while other characters were favorable to those of NC82, It is highly resistant to TMV and bacterial wilt(Ral-stonia solanacerum), and black shank(phytophthora parasitica var. nicotianae) is comparable to that of NC82. It should adapt well to the flue-cured production area, and can reduce premature flowering under unfavorable weather conditions. Yield of KF118 is 1 to 2% higher, and leaf quality is 4% higher than NC82. Fl hybrid, KF118, met acceptable standards for chemical and physical characteristics of cured leaf, and ranked high in good smoking taste by panel members in KT&G.

  • Tobacco mosaic virus (TMV), TMV-common strain, causes severe damage on flue-cured tobacco in Korea. As the results, breeding programs were conducted to transfer the TMV resistance from NC567 to other cultivars. Advantage of F1, hybrid may not lie only in their heterotic value but also in directly combining of valuable dominant traits, e.g. for disease resistance, either in pairs or as groups already assembled in the two parents. A new TMV resistant flue-cured tobacco variety KF114 was developed from a cross as maternal parent MSNC567 and NC82 at the Suwon Experiment Station. KF114 was more vigorous growth characteristics in early stage than that of NC82, standard variety in Korea, and other characters were similar to those of NC82. It os high resistance to TMV, but bacterial wilt(Pseudomonas solanacerum) and black shank (Phytophthora parasitica var. nicotianae) is comparable to NC82. It should adapt well to the flue-cured production area and can reduce premature flowering under unfavorable weather conditions. Yield of KF114 is 1% to 5% higher, and nearly equal in value per KG compared with NC82. This variety, F1 hybrid, met acceptable standards for chemical and physical characteristics of cured leaf and high ranked good smoking taste by panel members in Korea Ginseng and Tobacco Research Institute.

An improved Kalman filter for joint estimation of structural states and unknown loadings

  • The classical Kalman filter (KF) provides a practical and efficient way for state estimation. It is, however, not applicable when the external excitations applied to the structures are unknown. Moreover, it is known the classical KF is only suitable for linear systems and can't handle the nonlinear cases. The aim of this paper is to extend the classical KF approach to circumvent the aforementioned limitations for the joint estimation of structural states and the unknown inputs. On the basis of the scheme of the classical KF, analytical recursive solution of an improved KF approach is derived and presented. A revised form of observation equation is obtained basing on a projection matrix. The structural states and the unknown inputs are then simultaneously estimated with limited measurements in linear or nonlinear systems. The efficiency and accuracy of the proposed approach is verified via a five-story shear building, a simply supported beam, and three sorts of nonlinear hysteretic structures. The shaking table tests of a five-story building structure are also employed for the validation of the robustness of the proposed approach. Numerical and experimental results show that the proposed approach can not only satisfactorily estimate structural states, but also identify unknown loadings with acceptable accuracy for both linear and nonlinear systems.
