ACE, α-Glucosidase and Cancer Cell Growth Inhibitory Activities of Extracts and Fractions from Marine Microalgae, Nannochloropsis oculata (해양 미세조류 Nannochloropsis oculata 추출.분획물의 ACE, α-glucosidase 및 암세포 저해 활성)

  • Extracts of the marine microalgae Nannochloropsis oculata were obtained using 80% methanol (MeOH) and water. The 80% MeOH extract was further fractionated with n-hexane, chloroform, ethyl acetate (EtOAc), n-butanol (n-BuOH), and water to isolate the active fraction. Seven samples were prepared and their angiotensin converting enzyme (ACE), $\alpha$-glucosidase, and cancer cell growth inhibitory activities in vitro were determined. The most profound ACE inhibitory activity was observed in the chloroform fraction, while the others had moderate effects. By contrast, greater $\alpha$-glucosidase inhibitory activity was found in the EtOAc fraction, n-hexane fraction, and water extract of N. oculata. The antiproliferative effects of the extracts and fractions against HL-60, U937, CT-26, and SK-Hep1 cancer cells were also determined. The n-BuOH fraction had the strongest antiproliferative effects on CT-26 cells in a time-dependant manner (P<0.05). These results suggest that the extracts and fractions from N. oculata could be used as a potential functional food or as pharmaceutical ingredients.

Cross-sectional TEM Specimen Preparation of GaN-based Thinfilm Materials Using Alumina Dummy Filler (Alumina dummy 충전재를 이용한 GaN 기반 박막재료의 단면 TEM 시편준비)

  • Practical difficulties for preparing a good crosssectional specimen of GaN-based materials for transmission electron microscopy have arisen due to large difference of mechanical properties between hard ceramic substrate and soft GaN-layered materials. Uneven polishing, sudden cracking, delamination, and selective sputtering during the conventional wedge polishing technique are often encountered as experimental hindrances. The preparation technique based on Strecker's method can be applied to overcome these difficulties, which eventually leads to mechanically stable TEM samples independent of the mechanical properties of materials. The basic idea is to use hard ceramic dummy filler for embedding the sample of interest into the dummy frame. In this study, we applied this technique into preparing cross-sectional TEM specimen of the GaN-based materials with mechanical instability and demonstrated usefulness of this hard dummy filler method in which the possible modifications of the sample of interest during the preparation must be avoidable. In addition, practical precautions during the preparation were discussed.

Development of Remote Data Analysis System for the Joint Use of Equipments (분석기기지원을 위한 원격 데이터 분석 시스템 개발)

  • In Korea Basic Science Institute(KBSI) the remote data analysis system is developed for the joint use of advanced equipments. This system enables the researchers to access the datas which are produced at KBSI and analyse them by Java program on the Web,. Except Web browser such as Internet Explorer or Netscape Navigator no additional softwares are required for analysing data. We have developed remote data analysis systems for five major equipments which KBSI supports for the researchers, The systems which are developed are those for NMR spectrometer High Reso-lution Tandem mass Spectrometer Microscopic Imaging System DNA Sequencer and Natural Ra-dioactivity Measruement System, These programs work on any computer platform and any operat-ing system only if the internet is available. This remote data analysis system will be served as a part of Collaboratory the remote collaborative system.

Strategic Management Audit to Government-sponsored Research Institutes : A Case of Korea Basic Science Institute (KBSI) (정부출연연구기관의 전략적 경영감사 : 한국기초과학지원연구원의 사례를 중심으로)

  • Government-sponsored Research Institutes (GRIs) have made a great contribution to the development of Korea's Science and Technology (S&T). However, recently, GRIs are facing the challenge of many problems, for instance, absence of reasonable strategies, insufficient resources, and so on. Therefore, many GRIs are required for solving these problems by enhancing their strategic management capabilities. For this purpose, this paper suggests a Strategic Management Audit Model for Korean GRIs. The concept of Strategic Management Audit Model has not been widely adopted in Korean GRIs. In this paper, we suggest GRI-specific Strategic Management Model and apply it to Korea Basic Science Institute (KBSI), which is under the Korea Research Council of Fundamental Science and Technology (KRCF). Based on our strategic management audit, we propose several potential strategies for Korean Basic Research Institutes (KBSI). Our model and its implications will also be helpful for other Korean GRIs.

  • Collagen, which accounts for one-third of human protein, is reduced due to human aging, and much attention is focused on making collagen into food to prevent such aging. Gel permeation chromatography with Reflective Index (RI) detection (GPC/RI) was chosen as the most suitable instrument to confirm molecular weight distribution, and we explored the use of this technique for analysis of collagen peptide molecular sizes and distributions. Data reliability was verified by matrix-assisted laser desorption/ionization coupled to time-of-flight (MALDI-TOF) mass spectrometric analysis. The data were considered meaningful for comparative analysis of molecular weight distribution patterns.

  • Interest in aspects of industrialization relating to human health has increased. Accordingly, the use of labels such as 'natural foods' and 'organic ingredients' has become more widespread, and greater emphasis is being placed on improving quality of life. Water is an essential element for human life, and water quality has a significant impact on human health. However, technology that can precisely determine the substances present in water is still lacking. This study was conducted to establish a complete mass spectrometry process, from pretreatment to analysis, to measure and characterize natural organic matter (NOM) in Korean spring water samples. Salts and other matrices were removed from the samples using solid-phase extraction (SPE) with two different columns (PPL and C18). After establishing an accurate analysis method, the experimental results were evaluated based on Van Krevelen diagrams and analysis of molar O/C and H/C ratios. The method for characterizing NOM introduced herein should facilitate evaluation of water quality.
