Chemical Imaging Analysis of the Micropatterns of Proteins and Cells Using Cluster Ion Beam-based Time-of-Flight Secondary Ion Mass Spectrometry and Principal Component Analysis

  • Micropatterns of streptavidin and human epidermal carcinoma A431 cells were successfully imaged, as received and without any labeling, using cluster $Au_3{^+}$ ion beam-based time-of-flight secondary ion mass spectrometry (TOF-SIMS) together with a principal component analysis (PCA). Three different analysis ion beams ($Ga^+$, $Au^+$ and $Au_3{^+}$) were compared to obtain label-free TOF-SIMS chemical images of micropatterns of streptavidin, which were subsequently used for generating cell patterns. The image of the total positive ions obtained by the $Au_3{^+}$ primary ion beam corresponded to the actual image of micropatterns of streptavidin, whereas the total positive-ion images by $Ga^+$ or $Au^+$ primary ion beams did not. A PCA of the TOF-SIMS spectra was initially performed to identify characteristic secondary ions of streptavidin. Chemical images of each characteristic ion were reconstructed from the raw data and used in the second PCA run, which resulted in a contrasted - and corrected - image of the micropatterns of streptavidin by the $Ga^+$ and $Au^+$ ion beams. The findings herein suggest that using cluster-ion analysis beams and multivariate data analysis for TOF-SIMS chemical imaging would be an effectual method for producing label-free chemical images of micropatterns of biomolecules, including proteins and cells.

Flight Model Development of the MIRIS, the Main Payload of STSAT-3

  • MIRIS (Multipurpose Infra-Red Imaging System) is the first Korean Infrared Space Telescope developed by KASI (Korea Astronomy and Space Science Institute), and is the main payload of STSAT-3 (Science and Technology Satellite-3). The FM (fight model) of MIRIS has been recently completed, and various performance tests have been made to measure system parameters such as readout noise, system gain, linearity, and dark current. Final thermal-vacumm test of the MIRIS and the vibration test of the electronics box have been performed. Band response tests showed good agreement with the initial design requirements. No significant dark difference was measured within the expected temperature variation range during observation in orbit. Using Pa-alpha band from a uniform source, the readout noise and system gain were measured by mean variance test. To obtain uniform flat image, flat fielding tests were made for each band, and the data will be compared to that obtained in orbit for calibration. The final version of MIRIS FM will be delivered in March, and it will be integrated into the satellite system for the AIT (Assembly Integration, Test) procedure. The launch of MIRIS is expected in November 2012.

  • 1997읜 이후 한국기초과학지원연구원에 K-Ar 연대 측정 시스템이 도입되어 현재 이용중이다. 이 시스템은 흑연전기로, 가스 전처리 장치, 질량분석기와 자료획득장치로 구성되어 있으며 K-Ar연대 측정은 $^{38}$ Ar을 스파이크로 이용하는 동위원소 희석법에 의한 정밀한 Ar 농도 측정과 원자흡광분석을 통한 K 정량분석에의해 이루어진다. 연대가 잘 알려진 K-Ar 연대측정용 표준물질을 이용하여 시스템의 정밀성과 정확성을 확인하였다. 백만년 이하의 연대를 갖는 시료의 경우 질량분석기의 감도와 질량차별지수의 미약한 변화에도 큰영향을 받기 때문에 정확한 연대 측정이 곤란하나 중생대 및 제 3기의 K-Ar 연대를 갖는 표준시료의 연대를 측정했을 때, 믿을 만한 결과를 내고 있다. $92.6\pm$0.6 Ma의 연대가 알려진 SORI93혹운모의 경우 92.1$\pm$1.1 Ma의 연대를 얻을 수 있었고, $18.5\pm$0.6 Ma의 Bern4M 백운모는 권고치와 유사한 $17.8\pm$0.2 Ma의 연대를 얻을 수 있었다. .

  • In preparation of the biological samples for electron microscopy, the chemical fixation by glutaraldehyde, paraformaldehyde, and OsO4 has been generally used for a long time. However, the chemical fixation method has some problems: the infiltration time is a little bit long and the ultrastructure of cell or tissue transforms before complete fixation of sample. So, recently, cryo-fixation is considered more often in biomedical field. In this study, we compared High Pressure Freezing (HPF) method with chemical fixation method using a algal sample (Ptilota filicina J. Agardh), which was difficult to fix using chemical fixation method. In chloroplast, the ultrastructure of thylakoid lamella and phycobilisome can not show clearly by chemical fixation. In this study we could observe the ultrastructure of thylakoid lamella and phycobilisome of chloroplast very clearly using HPF fixation. An improved images of ultrastructures of nucleus, mitochondrion and floridean starch could obtain. These results suggest that HPF method is very useful method in algal specimen for electron microscopy.

Iron Oxide Nanoparticle-incorporated Alginate Capsules as Magnetic Field-assisted Potential Delivery Platforms for Agriculture Pesticides and Biocontrol Agents

  • Purpose: Biocompatible capsules have recently been highlighted as a novel platform for delivering various components, such as drug, food, and agriculture pesticides, to overcome the current limitations of living systems, such as those in agriculture, biology, the environment, and foods. However, few active targeting systems using biocompatible capsules and physical forces simultaneously have been developed in the agricultural engineering field. Methods: Here, we developed an active targeting delivery platform that uses biocompatible alginate capsules and controls movements by magnetic forces for agricultural and biological engineering applications. We designed and fabricated large-scale biocompatible capsules, using custom-made nozzles ejecting alginate solutions for encapsulation. Results: To develop the active target delivery platforms, we incorporated iron oxide nanoparticles in the large-scale alginate capsules. The sizes of alginate capsules were controlled by regulating the working conditions, such as concentrations of alginate solutions and iron oxide nanoparticles. Conclusions: We confirmed that the iron oxide particle-incorporated large-scale alginate capsules moved actively in response to magnetic fields, which will be a good strategy for active targeted delivery platforms for agriculture and biological engineering applications, such as for the controlled delivery of agriculture pesticides and biocontrol agents.

영지버섯, 여주, 쓴메밀 및 혼합 시료의 메탄올 추출물의 생리활성 (Biological Activity of Methanolic Extract from Ganoderma lucidum, Momordica charantia, Fagopyrum tataricum, and Their Mixtures)

  • 영지버섯, 여주, 쓴메밀 및 이들 혼합 시료의 70% 메탄올 추출물을 이용하여 생리활성물질(폴리페놀 화합물 및 플라보노이드) 농도와 항산화(DPPH radical 소거 활성, 생체막 지질과산화, Fe/Cu 환원력), ${\alpha}$-glucosidase 저해, tyrosinase 저해 및 항혈전 활성에 대하여 검토하였다. 추출 수율과 폴리페놀 화합물 및 플라보노이드의 함량은 혼합 추출물에서 가장 높았다. 또한 혼합 추출물에서 ${\alpha}$-glucosidase 저해, tyrosinase 저해, 항산화, 환원력 및 항혈전 활성은 혼합 추출물에서 가장 높았으며, 이러한 효과는 처리 농도 의존적 이였다. 이상의 실험 결과에서 영지버섯, 여주, 쓴메밀의 혼합 메탄올 추출물은 폴리페놀 화합물과 플라보노이드를 많이 함유함으로서 높은 ${\alpha}$-glucosidase 저해 및 항산화 활성과 관련성이 높아 보여 항당뇨 건강 기능성식품의 소재 개발에 활용될 가능성이 높아 보인다.

  • The MIRIS (Multi-purpose IR Imaging System), as the main payload of Science and Technology Satellite-3 (STSAT-3), is being developed by KASI in collaboration with several institutes for wide-field space observation in near IR wavelength. The Engineering Qualification Model (EQM) of MIRIS has been designed and fabricated in the laboratory. The system performance tests have been made including opto-mechanics, vibration test, thermal-vacuum environmental test and passive cooling test down to 200K. Most of the performance test results were satisfied with system requirements. The results of MIRIS performance tests have been presented at Critical Design Review (CDR) on September 2009. Several revisions were also recommended for Flight Model (FM) design, and detailed plan to develop FM of MIRIS is discussed in this paper.

One-pot Syntheses of Metallic Hollow Nanoparticles of Tin and Lead

  • Hollow Sn and Pb nanoparticles have been prepared by a rapid injection of an aqueous solution of $SnCl_2$- poly(vinylpyrrolidone) (PVP, surfactant) and $Pb(OAc)_2${\cdot}$3H_2O-PVP$ into an aqueous solution of sodium borohydride (reducing agent) in simple, one-pot reaction at room temperature under an argon atmosphere, respectively. The two hollow nanoparticles have been fully characterized by TEM, HRTEM, SAED, XRD, and EDX analyses. Upon exposure to air, the black Pb hollow nanoparticles are gradually transformed into a mixture of Pb, litharge (tetragonal PbO), massicot (orthorhombic PbO), and $Pb_5O_8$. The order and speed of mixing of the reactants between the metal precursor-PVP and the reductant solutions and stoichiometry of all the reactants are crucial factors for the formation of the two hollow nanocrystals. The Sn and Pb hollow nanoparticles were produced only when 1:(1.5-2) and 1:3 ratios of the Sn and Pb precursors to $NaBH_4$ were employed with a rapid injection, respectively.

First Light of the MIRIS, a Compact Wide-field Space IR Telescope

  • The MIRIS (Multi-purpose InfraRed Imaging System) is a compact IR space Telescope, which has been developed by KASI since 2008 as the main payload of Korean STSAT-3. It was launched successfully by a Dnepr Rocket at Yasny Launch site, Russia in November 2013. After the launch, the STSAT-3 successfully settled down at Sun synchronous orbit with altitude of ~ 600km. Communications were regularly made between the ground station and the MIRIS with other secondary payload. We made a series of tests of the MIRIS during the verification period and found that all functions including the passive cooling are working as expected. The MIRIS has a wide-field of view $3.67{\times}3.67$ degrees and wavelength coverage from 0.9 to 2.0 micro-meter with the angular resolution of 51.6 arcsec. The main science missions of the MIRIS are (1) mapping of the Galactic plane with Paschen-alpha line (1.88 micro-meter) for the study of warm interstellar medium and (2) the measurement of large angular fluctuations of cosmic near infrared background radiation with I (1.05 micro meter) and H (1.6 micro meter) bands to identify their origin. We present the results of MIRIS initial operation in this paper.

  • The Multi-purpose InfraRed Imaging System (MIRIS) is the main payload of Science and Technology Satellite 3 (STSAT-3), which was launched onboard Dnepr rocket from Russian Yasny Launch Base in November 2013. The MIRIS is an infrared (IR) camera, and the telescope has to be cooled down to below 200K in order to reduce thermal background noise. For the effective cooling and low-power consumption, we applied passive cooling method to the thermal design of the MIRIS. We also conducted thermal analysis and tested for the passive cooling before the launch of STSAT-3. After the launch, we have received State-of-Health (SOH) data from the satellite on orbit, including temperature monitoring results. It is important that the temperature of the telescope was shown to be cooled down to below 200K. In this paper, we present both the temperature data of the MIRIS on orbit and the thermal analysis results in the laboratory. We also compare these results and discuss the verification of the passive cooling.
