The water-extracts of Scutellaria barbata Don (SBDE) were isolated from Chinese medicinal plant sources. The extracts showed strong growth-inhibitory activity and cancer chemopreventive activity on the growth and DNA incorporation of MG63 human osteosarcoma and K562 human leukemia cell lines. The growth of human cancer cells was inhibited in the presence of the extracts (20, 50 and 100 ${\mu}$g/ml), and the effects were concentration-dependent and incubation time-dependent up to 8 days. When 50 ${\mu}$g/ml of the extracts was added to the media of MG63 and K562, cell growth after 8 days or 6 days of incubation was retarded by 93.2 to 97.3% of the control group. Morphological changes of MG63 and K562 cell lines were observed. As the concentration of the extracts increased up to 50 ${\mu}$g/ml, degree of cell aggregation decreased. Moreover, the DNA incorporation of the cells which were labeled with [3H] thymidine was significantly reduced after 3 days of incubation at $37^{\circ}C$ with the extract. Therefore, it is suggested that the extract is highly effective on inhibition of cancer cell growth. The extract also inhibited gene expression of IGF-II in transcriptional level. Since IGF-II works as a mitogenic effector on MG63 and K562 cell lines, these results suggest that the growth inhibition is in part mediated through the inhibition of IGF-II gene expression.
Quercetin is one of the most abundant dietary flavonoids widely present in many fruits and vegetables. Previous in vitro studies has shown that quercetin acts as an antioxidant and anti-inflammatory agent and it has potent anticarcinogenic properties as an apoptosis inducer. In this study we examined apoptotic effects of quercetin on the K562 erythroleukemia cell line. K562 cells were induced to undergo apoptosis by hydrogen peroxide. Cell viability and apoptosis level were assessed by annexin V and PI staining methods using flow cytometry. Viability of K562 cells was increased by low dose of quercetin (5-100 ${\mu}M$) for 3 hours. High doses of quercetin proved toxic (100-500 ${\mu}M$, 24 hours) and resulted in decrease of K562 cell viability as expected (p<0.01). As to results, 100 ${\mu}M$ quercetin was defined as a protective dose. Also, K562 cell apoptosis due to hydrogen peroxide was decreased in a dose dependent manner. As indicated in previous studies, reduction of superoxides by free radical scavengers like quercetin could be beneficial for prevention of cancer but consumption of such flavonoids during cancer treatment may weaken effects of chemotherapeutics and radiotherapy. Especially cancer patients should be carefully considered for traditional phytotherapy during cancer treatment, which can lead to controversial results.
Leukemia is a clonal disorder with blocked normal differentiation and cell death of hematopoietic progenitor cells. Traditional modalities with most used radiation and chemotherapy are nonspecific and toxic which cause adverse effects on normal cells. Differentiation inducing therapy forcing malignant cells to undergo terminal differentiation has been proven to be a promising strategy. However, there is still scarce of potent differentiation inducing agents. We show here that Angelica sinensis polysaccharide (ASP), a major active component in Dong quai (Chinese Angelica sinensis), has potential differentiation inducing activity in human chronic erythro-megakaryoblastic leukemia K562 cells. MTT assays and flow cytometric analysis demonstrated that ASP inhibited K562 cell proliferation and arrested the cell cycle at the G0/G1 phase. ASP also triggered K562 cells to undergo erythroid differentiaton as revealed by morphological changes, intensive benzidine staining and hemoglobin colorimetric reaction, as well as increased expression of glycophorin A (GPA) protein. ASP induced redistribution of STAT5 protein from the cytoplasm to the nucleus. Western blotting analysis further identified that ASP markedly sensitized K562 cells to exogenous erythropoietin (EPO) by activating EPO-induced JAK2/STAT5 tyrosine phosphorylation, thus augmenting the EPO-mediated JAK2/STAT5 signaling pathway. On the basis of these findings, we propose that ASP might be developed as a potential candidate for chronic myelogenous leukemia inducing differentiation treatment.
Background: Saponins are a major active component for the traditional Chinese medicine, Rubus parvifolius L., which has shown clear antitumor activities. However, the specific effects and mechanisms of saponins of Rubus parvifolius L. (SRP) remain unclear with regard to human chronic myeloid leukemia cells. The aim of this study was to investigate inhibition of proliferation and apoptosis induction effects of SRP in K562 cells and further elucidate its regulatory mechanisms. Materials and Methods: K562 cells were treated with different concentrations of SRP and MTT assays were performed to determine cell viability. Apoptosis induction by SRP was determined with FACS and DAPI staining analysis. Western blotting was used to detect expression of apoptosis and survival related genes. Specific inhibitors were added to confirm roles of STAT3 and AMPK pathways in SRP induction of apoptosis. Results: Our results indicated that SRP exhibited obvious inhibitory effects on the growth of K562 cells, and significantly induced apoptosis. Cleavage of pro-apoptotic proteins was dramatically increased after SRP exposure. SRP treatment also increased the activities of AMPK and JNK pathways, and inhibited the phosphorylation expression level of STAT3 in K562 cells. Inhibition of the AMPK pathway blocked the activation of JNK by SRP, indicating that SRP regulated the expression of JNK dependent oon the AMPK pathway. Furthermore, inhibition of the latter significantly conferred resistance to SRP pro-apoptotic activity, suggesting involvement of the AMPK pathway in induction of apoptosis. Pretreatment with a STAT3 inhibitor also augmented SRP induced growth inhibition and cell apoptosis, further confirming roles of the STAT3 pathway after SRP treatment. Conclusions: Our results demonstrated that SRP induce cell apoptosis through AMPK activation and STAT3 inhibition in K562 cells. This suggests the possibility of further developing SRP as an alternative treatment option, or perhaps using it as adjuvant chemotherapeutic agent for chronic myeloid leukemia therapy.
TNF-related apoptosis-inducing ligand (TRAIL) 는 암세포에만 작용하고 정상세포에는 영향을 주지 않는 항암제로서 알려져 있지만, TRAIL에 내성을 나타내는 암세포의 출현이 문제점으로 지적되고 있다. 사람 백혈병세포인 K562 및 CEM 세포는 TRAIL에 내성을 나타낸다. 본 연구에서는 이러한 백혈병 세포의 TRAIL 내성에 대한 새로운 표적 분자의 발굴과 이를 토대로 한 새로운 내성극복 방법을 연구하였다. 새로운 TRAIL sensitizer로서 quercetin을 발굴하고, 이를 K562 세포에 TRAIL과 병용 투여하므로서 TRAIL의 효과 증강에 의한 내성극복을 시도하였다. Quercetin은 DNA-PK/Akt 신호전달경로를 억제하므로서, caspases 활성 증강과 PARP cleavage, 이에 따른 Bax의 발현을 증강시키는 기전으로 K562 세포의 TRAIL에 의한 apoptosis를 증대시키는 활성이 있음을 밝혔다. 이러한 quercetin 병용 처리에 의한 TRAIL의 활성 증강으로 TRAIL 내성이 극복됨을 CEM 세포에서도 확인하였다. 이러한 연구 결과는 DNA-PK 발현 증강에 의한 Akt의 활성화가 TRAIL 내성을 유발하는 기전을 토대로 함을 밝힘으로써, DNA-PK 활성 억제제를 TRAIL과 병용하므로서 TRAIL 내성을 나타내는 암세포에 내성 극복 효과를 얻을 수 있는 새로운 약제 병용 방법을 제시하였다.
60종의 생약의 methanol 추출물을 L1210세포에 대하여 1차스크리닝실험을 수행하여 항암활성물질을 검색하고, 그 중에서 비교적 세포독성이 강하게 나타나는 산국(Chrysanthemum boreale M.)에서 항암활성물질을 분리정제하였다. 산국의 용매분획물의 L1210, K562, A549세포에 대한 세포독성실험에서는 chloroform 분획에서 $ED_{50}$값이 각각 3.98, 4.28, 3.84 (${\mu}g/ml$)로 나타났다. 이 chloroform 분획에서 유효세포독성물질을 정제하여 Compound I과 Compound II를 각각 얻었으며, 이 중에서 Compound I은 L1210, K562, A549세포에 대하여 각각 $ED_{50}$값이 0.55, 0.0003, 0.001 (${\mu}g/ml$)로 강한 세포독성을 나타내었으나, Compound II는 K562에 대해서만 $ED_{50}$ 값이 4.79 (${\mu}g/ml$)의 세포독성을 나타내었다.
비타민 E 유도체인 $RRR-{\alpha}-tocopheryl$ succinate (vitamin E succinate, VES)는 만성골수성 백혈병 세포인 K562세포에서 apoptosis를 유도하였다. VES의 처리에 의해 apoptosis가 유도되는 과정에서 K562 세포 내의 ROS의 생성이 증가되었으며, ROS와 관련된$NF-{\kappa}B$, COX-2 그리고 $p21^{WAF1/CIP1}$등의 유전자가 활성화되었다. 뿐만 아니라, apoptosis의 과정 중 중요한 역할을 하는 Bax의 발현증가 및 손상된 DNA의 회복에 중심적 기능을 하는 PARP의 분열이 야기되었다. VES를 처리한 세포의 세포주기 분석에서는 apoptotic phase인 sub-Gl phase에서 세포사멸이 증가되고, 형태적으로는 염색질의 응축이 일어나는 결과로 미루어볼 때 VES는 K562세포의 apoptosis를 유도한 것을 알 수 있다. C57BL/C의 림프종 이종이식을 통한 VES의 항암활성 실험 결과, C57BL/C의 대조군에 비하여 종양의 성장억제를 확인하였으며 높은 생존율을 확인하였다. 이러한 결과는 백혈병 치료에 대한 분자적 기초를 제공하였으며 동물실험을 통하여 보다 실질적인 백혈병 치료의 가능성을 보여주었다.
목적: 다약제내성(MDR) 극복제 verapamil은 MDR이 발현된 암세포에서 Tc-99m MIBI(MIBI)와 tetrofosmin(TF)의 섭취를 증가시키는 것으로 알려져 있으나, 세포의 종류에 따라서는 MIBI와 TF의 섭취를 감소시킬 수 있다는 보고가 있다. 본 연구는 암세포의 종류에 따라 verpamile이 MIBI와 TF의 섭취에 미치는 영향이 세포에 따른 차이가 있는지를 알아보고, 이러한 차이가 세포독성이나 PKC효소의 발현에 따른 차이인가를 알아보았다. 방법: 백혈병세포 K562세포와 유방암세포 MCF7, 난소암세포 SK-OV-3 세포 및 MDR이 유발된 K562(Adr)세포를 배양하였다. 시험관에서 $1{\times}10^6\;cells/ml$ 농도의 single-cell suspension 상태로 분주하고 verapamil을 1, 10, 50, 100, $200{\mu}M$의 농도로 처리한 후 MIBI와 TF를 배양한 후, $37^{\circ}C$에서 1, 15, 30, 45, 60분 동안 반응시킨 후 pellet과 supernatant의 방사능 치를 감마계수기로 측정하여 투여한 방사능 치에 대한 세포내 섭취백분율로 표시하였다. Verapamil의 세포독성은 MTT assay로 측정하였고, 세포내의 PKC isotypes의 변화는 western blotting analysis로 평가하였다. 결과: 4종류의 세포 모두에서 MIBI와 TF의 섭취는 1, 15, 30, 45, 60분 배양 시간에 따라 증가하였다. verapamil을 처리시 다약제 내성이 유발된 K562(Adr)세포에서 $100{\mu}M$의 농도까지는 MIBI와 TF 섭취가 증가하였고, 최대 $10{\mu}M$에서 10배 증가하였다. 그러나 K562세포를 verapamil $1{\mu}M$로 처리하였을 때는 기저치와 유사하였으나, verapamil의 농도가 증가함에 따라 MIBI와 TF의 세포섭취는 모두 감소하였다. K562세포의 60분 MIBI 섭취율은 79%($10{\mu}M$), 47%($50{\mu}M$), 29%($100{\mu}M$), 1.5%($200{\mu}M$)로 verapamil의 용량이 증가함에 따라 감소하였으며, TF 섭취율도 84% ($10{\mu}M$), 60%($50{\mu}M$), 42%($100{\mu}M$), 2.7%($200{\mu}M$)로 감소하였다. MCF7, SK-OV-3세포에서는 verapamil $10{\mu}M$까지는 MIBI와 TF의 섭취가 기저치와 유사하거나 소량 증가하였으나 $50{\mu}M$이상의 용량에서는 감소하여 $100{\mu}M$에서는 각각 40%와 5%만 섭취되었다. MTT assay상 K562(Adr)세포에서는 verapamil $100{\mu}M$ 이상에서는 유의하게 낮았으나 다른 세포는 $200{\mu}M$까지에도 차이가 없었다. PKC 아형의 분석상 PKC $\varepsilon$이 K562(Adr)세포에서 많이 발현되었으나, K562와 K562(Adr)세포에서는 verapamil처리에 따른 PKC 아형의 변화는 없었다. 결론: Verapamil은 암세포의 종류에 따라 MIBI와 TF의 섭취를 감소시켰고, 고용량에는 MDR세포의 섭취도 감소시켰으며 이러한 현상은 세포독성 이나 PKC효소 아형과는 관련이 없었다. 그러므로 MDR의 진단시 verapamil을 처치에 따른 MIBI와 TF의 섭취 정도를 기준으로 하는 경우에는, verapamil의 농도와 세포의 종류에 따라 현저한 차이가 있을 수 있다는 점을 생각하여야 한다.
