Optimization of microwave-assisted extraction process for blue honeysuckle (Lonicera coerulea L.) using response surface methodology (반응표면분석법을 이용한 댕댕이 기능성성분의 마이크로웨이브추출조건 최적화)

  • Functional compounds including flavonoids, anthocyanins, polyphneols and antioxidants were extracted from blue honeysuckle (Lonicera caerulea L.) using highly efficient microwave-assisted extraction. And extraction process was modeled and optimized according to response surface methodology (RSM). The independent variables ($X_n$) were ethanol concentration ($X_1$: 0, 25, 50, 75, 100%), irradiation time ($X_2$: 1, 3, 5, 7, 9 min), and microwave power ($X_3$: 60, 120, 180, 240, 300 W). Dependent variables ($Y_n$) were total flavonoid contents ($Y_1$), total anthocyanin contents ($Y_2$), total polyphenol contents ($Y_3$) and antioxidant activity ($Y_4$). Four-dimensional response surface plots were generated based on the fitted second-order polynomial models to get optimal conditions. Estimated optimal conditions for 4 responses were ethanol concentration of 54-72%, irradiation time of 7.1-7.6 min, and microwave power of 243-251 W. Ridge analysis predicted the maximal responses of total flavonoid content, total anthocyanin content, total polyphenol content and antioxidant activity were 38.00 mg RE/g, 6.80 mg CGE/g, 14.90 mg GAE/g, 89.10%, respectively. Verification experiment was carried out at predicted optimal conditions and experimental values for total flavonoid content, total anthocyanin content, total polyphenol content and antioxidant activity were 38.10 mg RE/g, 6.72 mg CGE/g, 14.91 mg GAE/g and 89.13%, respectively. No significant difference was observed between predicted and experimental values, indicating good fitness of fitted model and successful application of RSM.

Standard Chemotherapy with Excluding Isoniazid in a Murine Model of Tuberculosis (마우스 결핵 모델에서 Isoniazid를 제외한 표준치료의 예비 연구)

  • Background: Isoniazid (INH, H) is a key drug of the standard first-line regimen for the treatment of tuberculosis (TB), yet some reports have suggested that treatment efficacy was maintained even though INH was omitted from the treatment regimen. Methods: One hundred forty C57BL/6 mice were infected with the H37Rv strain of M. tuberculosis with using a Glas-Col aerosol generation device, and this resulted in depositing about 100 bacilli in the lung. Four weeks after infection, anti-TB treatment was initiated with varying regimens for 4-8 weeks; Group 1: no treatment (control), Group 2 (4HREZ): 4 weeks of INH, rifampicin (R), pyrazinamide (Z) and ethambutol (E), Group 3: 1HREZ/3REZ, Group 4: 4REZ, Group 5: 4HREZ/4HRE, Group 6: 1HREZ/3REZ/4RE, and Group 7: 4REZ/4RE. The lungs and spleens were harvested at several time points until 28 weeks after infection, and the colony-forming unit (CFU) counts were determined. Results: The CFU counts increased steadily after infection in the control group. In the 4-week treatment groups (Group 2-4), even though the culture was negative at treatment completion, the bacilli grew again at the 12-week and 20-week time points after completion of treatment. In the 8-week treatment groups (Groups 5-7), the bacilli did not grow in the lung at 4 weeks after treatment initiation and thereafter. In the spleens of Group 7 in which INH was omitted from the treatment regimen, the culture was negative at 4-weeks after treatment initiation and thereafter. However, in Groups 5 and 6 in which INH was taken continuously or intermittently, the bacilli grew in the spleen at some time points after completion of treatment. Conclusion: TThe exclusion of INH from the standard first-line regimen did not affect the treatment outcome in a murine model of TB in the early stage of disease. Further studies using a murine model of chronic TB are necessary to clarify the role of INH in the standard first-line regimen for treating TB.

Steaming and Ultrasonic extraction conditions for enhancing the ginsenoside contents and anti-aging efficacy of the Ginseng sprout leaf/stem (새싹삼 잎 줄기의 진세노사이드 함량 및 항노화 효능 증진을 위한 증숙 및 추출조건)

  • 인삼은 우리나라에서 오랜 역사동안 많은 연구가 진행되어 왔으며, 현재는 다양한 방법으로 홍삼과 흑삼으로 만들어 식품, 화장품, 의약품 등 다양한 방면으로 사용하고 있다. 본 연구에서 시중에서 구매한 새싹삼(인삼새싹) 잎/줄기에 함유된 진세노사이드(Re, Rg1, Rb1, Rg3, Rh1) 함량을 높이기 위하여 증숙과 초음파 추출조건에 관한 연구를 수행하여 우수한 항노화 소재를 개발하기 위하여 실시하였다. 실험은 새싹삼 잎/줄기를 증숙 온도와 시간의 조건에서 진세노사이드 함량이 가장 높은 조건을 선정하였으며, 선정된 조건의 새싹삼 잎/줄기에 파장과 출력에 대한 조건으로 초음파 추출을 진행하여 진세노사이드가 가장 높은 함량을 보이는 조건을 선정하였다. 그 결과 새싹삼 잎/줄기추출물(GSE; Ginseng Sprout Extract)의 진세노사이드 함량은 4.8 mg/g으로 확인되었으나 증숙공정을 통해 8.82 mg/g으로 함량이 증가되었으며, 상기 증숙된 새싹삼 잎/줄기에 초음파공정을 적용하여 추출한 새싹삼 잎/줄기초음파추출물(SU-GSE; Steaming & dry Ultrasonication-Ginseng Sprout Extract)에서는 최대 10.65 mg/g으로 함량이 증가되었다. 반면, 새싹삼 뿌리의 진세노사이드는 2.30 mg/g으로 확인되었으나 증숙공정을 통해 4.95 mg/g으로 함량이 증가되었으며, 초음파추출공정을 통해 최대 5.82 mg/g으로 함량이 증가된 것을 확인할 수 있었으나, 새싹삼 잎/줄기에 비해 진세노사이드 함량이 낮은 것을 확인하였다. 항노화 소재로의 활용가능성을 평가하기 위하여 새싹삼 잎/줄기추출물 GSE와 SU-GSE에 대한 세포생존률, 항산화 및 항노화에 대한 효능평가를 진행하였으며 GSE의 경우 $100{\mu}g/ml$에서 세포생존률이 82.4%를 보인 반면 SU-GSE에서는 $1,000{\mu}g/ml$의 농도에서 101.8%의 세포 생존률을 보였다. 항산화 활성의 경우 GSE와 SU-GSE $100{\mu}g/ml$ 농도에서 각각 52%와 81%의 항산화 활성을 나타냄으로써 SU-GES의 조건에서 항산화 활성이 우수한 것으로 확인되었다. 또한, 항노화 활성에 대한 실험결과 MMP-1 유전자 발현에 대한 억제율을 비교한 결과 GSE와 SU-GES $100{\mu}g/ml$의 농도에서 각각 18%와 29%의 억제율을 보임에 항노화 소재로의 활용가능성을 확인하였다.

Validation on the Analytical Method of Ginsenosides in Red Ginseng

  • A cross-examination between KT&G Central Research Institute and Guangzhou Institute for Drug Control was carried out in order to select optimum conditions for extraction, separation and determination of ginsenosides in red ginseng and to propose a better method for the quantitative analysis of ginsenosides. The optimum extraction conditions of ginsenosides from red ginseng were as follows: the extraction solvent, $70\%$ methanol; the extraction temperature, $100^{\circ}C;$ the extraction time, 1 hour for once; and the repetition of extraction, twice. The optimum separation conditions of ginsenosides on the SepPak $C_{18}$ cartridge were as follows: the loaded amount, 0.4 g of methanol extract; the washing solvents, distilled water of 25 ml at first and then $30\%$ methanol of 25 ml; the elution solvent, $90\%$ methanol of 5 ml. The optimum HPLC conditions for the determination of ginsenosides were as follows: column, Lichrosorb $NH_2(25{\times}0.4cm,$ 5${\mu}m$, Merck Co.); mobile phase, a mixture of acetonitrile/water/isopropanol (80/5/15) and acetonitrile/water/isopropanol (80/20/15) with gradient system; and the detector, ELSD. On the basis of the optimum conditions a method for the quantitative analysis of ginsenosides were proposed and another cross-examination was carried out for the validation of the selected analytical method conditions. The coefficient of variances (CVs) on the contents of ginsenoside-$Rg_{1}$, -Re and $-Rb_1$ were lower than $3\%$ and the recovery rates of ginsenosides were $89.4\~95.7\%,$ which suggests that the above extraction and separation conditions may be reproducible and reasonable. For the selected HPLC/ELSD conditions, the CVs on the detector responses of ginsenoside-Rg, -Re and $-Rb_1$) were also lower than $3\%$, the regression coefficients for the calibration curves of ginsenosides were higher than 0.99 and two adjacent ginsenoside peaks were well separated, which suggests that the above HPLC/ELSD conditions may be good enough for the determination of ginsenosides.

Soft Magnetic Properties of CoFeHfO Thin Films (CoFeHfO 박막의 자기적 특성)

  • Amorphous alloys of Co-rich magnetic amorphous films are well known as thpical soft magnetic alloys. They are used for many kinds of electric and electronic parts such as magnetic recording heads, transformers and inductors. CoFeHfO thin films were prepared by RF magnetron reactive sputtering. The films were deposited onto Si(100) substrates with a power of 300 W at room temperature. The reactive gas was introduced up to 10% ($O_2$/(Ar + $O_2$)) during deposition, and the $Co_{39}Fe_{34}Hf_{9.5}O_{17.5}$ thin film exhibit excellent soft magnetic properties : saturation magnetization ($4{\pi}M_s$) of 19kG, magnetic coercivity ($H_c$) of 0.37 Oe, anisotropy field ($H_k$) of 48.62 Oe, and an electrical property is also shown to be as high as 300 ${\mu}{\Omega}cm$. It is assumed that the good soft magnetic properties of $Co_{39}Fe_{34}Hf_{9.5}O_{17.5}$ thin film results from high electrical resistivity and large anisotropy field.

Comparison of Monitor Units Obtained from Measurements and ADAC Planning System for High Energy Electrons (측정과 ADAC 치료계획 시스템에서 계산된 고에너지 전자선의 Monitor Unit Value 비교)

  • The purpose of this study is to evaluate the monitor unit obtained from various methods for the treatment of superficial cancers using electron beams. Thirty-three breast cancer patients who were treated in our institution with 6, 9, and 12 MeV electron beams, were selected for this study. For each patient, irregularly shaped treatment blocks were drawn on simulation film and constructed. Using the irregular blocks, monitor units to deliver 100 cGy to the dose maximum (dmax) were calculated from measurement and three-dimensional radiation treatment planning (3D RTP) system (PINNACLE 6.0, ADAC Laboratories, Milpitas CA) Measurements were made in solid water phantom with plane parallel (PP) chamber (Roos, OTW Germany) at 100 cm source-to surface distances. CT data was used to investigate the effect of heterogeneity. Monitor units were calculated by overriding CT values with 1 g/㎤ and in the presence of heterogeneity. The monitor unit values obtained by the above methods were compared. The dose, obtained from measurement in solid water phantom was higher than that of RTP values for irregularly shaped blocks. The maximum differences between monitor unit calculated in flat water phantom at gantry zero position were 4% for 6 MeV and 2% for 9 and 12 MeV electrons. When CT data was used at a various gantry angle the agreement between the TPS data with and without density correction was within 3% for all energies. These results indicate that there are no significant difference in terms of monitor unit when density is corrected for the treatment of breast cancer patients with electrons.

Physicochemical characteristics of Sengmaksan added with Liriope platyphylla roasted for different times (덖음 처리 시간을 달리한 맥문동을 첨가한 생맥산의 이화학적 특성)

  • This study investigates, the physicochemical characteristics of Sengmaksan (SM) prepared with Liriope platyphylla (LP) that had been roasted for different times (0, 30, 60, and 90 min, denoted as S-0, S-30, S-60, and S-90, respectively) The Hunter's color values such as lightness (L), redness (a), and yellowness (b) were the highest in S-0, while the lowest was found in S-90. The amount of soluble solid and reducing sugar content of S-60 were higher than the others. None of the samples exhibit significant differences in, their pH and acidity. The total content of phenolic compounds increased with the LP roasting time, but the total flavonoid and total anthocyanin contents of the SM decreased at the same time. The total ginsenoside (Ro, Rb2, Re, Rf, Rg1, Rg2, Rg3, Rh1, and Rh2) content did not show significant differences. The DPPH and ABTS radical scavenging activities increased according to the concentration, as well as with the LP roasting time. The ferric reducing antioxidant power (FRAP) showed trends similar to the radical scavenging activity, but it was more sensitive to the LP roasting time. From these results, the active ingredient in S-60 was higher, and the antioxidant activities of SM increased along with the roasting time of LP.

Estimating Moisture Content of Cucumber Seedling Using Hyperspectral Imagery

  • Purpose: This experiment was conducted to detect water stress in terms of the moisture content of cucumber seedlings under water stress condition using a hyperspectral image acquisition system, linear regression analysis, and partial least square regression (PLSR) to achieve a non-destructive measurement procedure. Methods: Changes in the reflectance spectrum of cucumber seedlings under water stress were measured using hyperspectral imaging techniques. A model for estimating moisture content of cucumber seedlings was constructed through a linear regression analysis that used the moisture content of cucumber seedlings and a normalized difference vegetation index (NDVI). A model using PLSR that used the moisture content of cucumber seedlings and reflectance spectrum was also created. Results: In the early stages of water stress, cucumber seedlings recovered completely when sub-irrigation was applied. However, the seedlings suffering from initial wilting did not recover when more than 42 h passed without irrigation. The reflectance spectrum of seedlings under water stress decreased gradually, but increased when irrigation was provided, except for the seedlings that had permanently wilted. From the results of the linear regression analysis using the NDVI, the model excluding wilted seedlings with less than 20% (n=97) moisture content showed a precision ($R^2$ and $R^2_{\alpha}$) of 0.573 and 0.568, respectively, and accuracy (RE) of 4.138% and 4.138%, which was higher than that for models including all seedlings (n=100). For PLS regression analysis using the reflectance spectrum, both models were found to have strong precision ($R^2$) with a rating of 0.822, but accuracy (RMSE and RE) was higher in the model excluding wilted seedlings as 5.544% and 13.65% respectively. Conclusions: The estimation model of the moisture content of cucumber seedlings showed better results in the PLSR analysis using reflectance spectrum than the linear regression analysis using NDVI.

Optimization of Lipase-Catalyzed Interesterification for Production of Human Milk Fat Substitutes by Response Surface Methodology (반응표면분석에 의한 모유대체지의 효소적 합성조건 최적화)

  • 1,3-Dioleoyl-2-palmitoylglycerol (OPO)-rich human milk fat substitute (HMFS) was synthesized from tripalmitin (PPP)-rich fraction and oleic ethyl ester by a lipase-catalyzed interesterification. Response surface methodology (RSM) was employed to optimize the presence of palmitic acid at sn-2 position ($Y_1$, %) and of oleic acid at sn-1,3 ($Y_2$, %), with the reaction factors as substrate molar ratio of PPP-rich fraction to oleic ethyl ester ($X_1$, 1:4, 1:5 and 1:6), reaction temperature ($X_2$, 50, 55 and $60^{\circ}C$), and time ($X_3$, 3, 7.5 and 12 h). The optimal conditions for HMFS synthesis were predicted at the reaction combination of $55^{\circ}C$, 3 h and 1:6 substrate ratio. HMFS re-synthesized under the same conditions displayed 70.70% palmitic acid at the sn-2 position and 69.58% oleic acid at the sn-1,3 position. Reaction product was predominantly (90.35%) triacylglycerol (TAG) was observed in which the major TAG species, OPO, comprised 31.24%.

Bioactive Compounds and Antioxidant Activity of Jeju Camellia Mistletoe (Korthalsella japonica Engl.) (제주 동백나무 겨우살이의 용매별 기능성 성분 및 항산화 작용)

  • Mistletoes are hemi-parasitic plant growing on different host tree and shrubs. They are traditionally used in folkloric medicine for the treatment of diarrhea, cough, diabetes, hypertension, cancer and skin infection. The purpose of this study was to determine the contents of phenolics and antioxidant activity of 70% ethanol, 100% methanol and hot water extracts of Jeju camellia mistletoe (Korthalsella japonica Engl.). Ethanol was most effective in extracting total phenols (7,427 mg gallic acid equivalent (GAE)/100 g) and flavonoid (1,777 mg rutin equivalent (RE)/100 g). The free radical scavenging activity, 1,1-diphenyl-2-picryl hydrazyl (DPPH) (EC50 = 7.8 mg/ml) and hydrogen peroxide (H2O2) (EC50 = 1.4 mg/ml), and the capacity for chelating metal ions (EC50 = 8.0 mg/ml) and reducing power (EC50 = 14.9 mg/ml) of the samples also higher in ethanolic extracts. The strong correlation (r2 = −0.996~−0.881) between antioxidant capacities and the phenolic contents implied that phenolic compounds are a major contributor to the antioxidant activity of the ethanolic extracts of Jeju camellia mistletoe. As conclusions, Jeju camellia mistletoe contains bioactive substances with a potential for reducing the physiological as well as oxidative stress and this could explain the suggested cancer preventive effect of these plants as well as their protective role on other major diseases.
