Uronic Acid Composition, Block Structure and Some Related Properties of Alginic Acid (3) On Alginic Acid Prepared from Sargassum ringgoldianum (알긴산의 화학적 조성 및 그 물성에 관한 연구 (3) 큰잎모자반의 알긴산)

  • In the previous papers (Kim and Park, 1984 a, b), we have reported on alginic acid from Ecklonia cava. The seasonal and portional variation in the composition of uronic acids and their block structures of alginic acid from Sargassum ringgoldianum (collected from Ichon-ri at the coast of Ilgwang-myon, Yangsan-gun, Kyongnam, Korea, in the period of January to December in 1982) are investigated, and their relationship between the chemical composition and some rotated properties is discussed in this study. The results are as follows: 1. One year average contents of alginic acid were $21.4\%$ in the stipe and $19.7\%$ in the frond, one-year average values of M/G ratios were 2.38 in the frond and 1.85 in the stipe. The value of frond was largest in the period of Jauuary to April and smallest in May and October to December. The value of stipe was largest January to April and smallest in May. In general, the proportions of M block in the both of frond and stipe were higher than those of G block. 2. The viscosity of frond alginic acid showed higher values of 31.1 cP in November, and lower (below 7.0 cP) in the stipe alginic acid. Furthermore, the dependence on temperature of frond alginic acid was also larger in November and others were lower. Ion exchange ability of frond alginic acid was highest in July and the exchange amounts were $Pb^{2+}\;5.2,\;Cu^{2+}\;3.1,\;Zn^{2+}\;1.7,\;Co^{2+}\;1.5$ meq/g, Na-Alg., and the ability of stipe alginic acid was highest in May and the amounts were $Pb^{2+}\;4.6,\;Cu^{2+}\;3.3,\;Zn^{2+}\;2.5,\;Co^{2+}\;1.4$ meq/g. Na-Alg.. The affinity with metallic ions appeared higher in order of $Pb^{2+}>Cu^{2+}>Zn^{2+}>Co^{2+}$, and the exchange ability assumed to relate with the block ratio of uronic acid.

  • This experiment was carried out to investigate the minerals content and post-mortem biochemical change of broiler leg and breast muscle during hot and cold storage period. The leg and breast muscle samples were stored at $2^{\circ}C$and $30^{\circ}C$ for 32 hours, and analyzed 0, 4, 8, 32 hours after slaughter, the results obtained as follows (1) Minerals composition were different between breast and leg muscle. (2) Total nitrogen content changed according to the storage period and temperature in the breast (P<0.005) and the leg (P<0.005) (3) The Content of phosphate (P<0.005), magnesium (P<0.005), sodium (P<0.025) in breast muscle was higher than in the leg mucle, and did not change according to the storage period and temperature. (4) Calcium and iron contents were not different between breast and leg muscle but significantly changed according to the storage period and temperature. (5) Sodium content was not different among the various factors, and pH was different between leg and breast (P<0.005) muscle, and ultimate pH was appeared at 8 hours of slaughter, but 20 hours of slaughter in hot stored breast muscle. (6) pH decline in cold stored breast muscle was faster than in the hot stored. (7) Penetration value was higher in the breast muscle than in the leg muscle (P<0.005) (8) Regormortis were not occurred in cold stored breast muscle and hot stored leg muscle, but occurred in cold stored leg muscle and hot stored breast muscle at 4 hours after slaughter.

The Changes of Dietary Reference Intakes for Koreans and Its Application to the New Text Book (한국인 영양섭취기준에 대한 이해 및 새 교과서에의 적용 방안)

  • The purposes of this paper are to describe the newly established reference values of nutrient intakes: to apply the changed dietary reference intakes to the new text book based on the revised curriculum: and to contrive substantial contents in the domain of dietary life(foods & nutrition) of new text book. Dietary Reference Intakes for Koreans(KDRIs) is newly established reference values of nutrient intakes that are considered necessary to maintain the health of Koreans at the optimal state and to prevent chronic diseases and overnutrition. Unlike previously used Recommended Dietary Allowances for Koreas(KRDA), which presented a single reference value for intake of each nutrient, multiple values are set at levels for nutrients to reduce risk of chronic diseases and toxicity as well as prevention of nutrient deficiency. The new KDRIs include the Estimated Average Requirement(EAR), Recommended Intake(RI), Adequate Intake(AI), and Tolerable Upper Intake Level(UL). The EAR is the daily nutrient intake estimated to meet the requirement of the half of the apparently healthy individuals in a target group and thus is set at the median of the distribution of requirements. The RI is set at two standard deviations above the EAR. The AI is established for nutrients for which existing body of knowledge are inadequate to establish the EAR and RI. The UL is the highest level of daily nutrient intake which is not likely to cause adverse effects for the human health. Age and gender subgroups are established in consideration of physiological characteristics and developmental stages: infancy, toddler, childhood, adolescence, adulthood and old age. Pregnancy and lactation periods were considered separately and gender is divided after early childhood. Reference heights and weights are from the Korean Agency for Technology and Standards, Ministry of Commerce, Industry and Energy. The practical application of DRIs to the new books based on the revision in the 7th curriculum is to assess the dietary and nutrient intake as well as to plan a meal. It can be utilized to set an appropriate nutrient goal for the diet as usually eaten and to develop a plan that the individual will consume using a nutrient based food guidance system in the new books based on the revision in the 7th curriculum.

Formation of Secondary Amines in Soysauce and Soy-paste -Part 2. Periodical changes of sceondary amines during Korean soysauce aging- (장류(醬類)에 있어서 2급(級) Amine의 형성(形咸) -제2보(第2報) 재래식(在來式) 간장숙성중(熟成中) 2급(級) amine의 동태(動態)-)

  • To clarify the formation of secondary amines in Korean native soysauce, periodical changes during 90 days aging were investigated. The results were as follows; 1. The average content of secondary amines in Korean native meju was 20.86 ppm $(7.25{\sim}50.76{\;}ppm)$ as dimethylamine(DMA), higher than modified meju(artifcial lyinoculated Aspergillus oryzae). Especially, the high amount was detected in severely deteriorated meju. 2. Cooked soybean, wheat, barley and rice contained 1.28, 0.57, 0.34 and 0.35 ppm of secondary amines, respectively. The more amounts were detected in koji, the respective contents in soybean, wheat, barley and rice koji were 2.63, 1.09, 0.64 and 0.54 ppm. 3. The new formation of secondary amines was not recognized in normally fermented Korean native soysauce during 90 days aging. 4. Secondary amines were formed below 18% sodium chloride under dark condition during soysauce aging, but not formed at 14% under sun-light condition. 5. The modified soysauce, prepared with modified meju, did not contain the newly synthesized secondary amines at 14% sodium chloride under sun-light condition during 30 days aging. 6, Drying of meju and boiling of soysauce did not affect the evaporation of secondary amines.

Change of Fragrant Components by Flowering Stages in Lilium Oriental Hybrid 'Casa Blanca' (오리엔탈 나리 '카사블랑카'의 개화단계별 향기성분 변화)

  • This experiment was conducted to find out suitable extraction time of available fragrant component in Lilium Oriental Hybrida 'Casa Blanca' based on qualitative and quantitative variation of its fragrant component in its flowering stages. The content of essential oil in its flowering stages increased with the progress of flowering stages except flower bud stage and it had the most oil content in its stage after full bloom. The analysis of the essential oil in Lilium Oriental Hybrida 'Casa Blanca' in its flowering stages shows that its main constituents were farnesol (18.96%), benzyl salicylate (13.81%), butyl-hydroxy toluene (12.87%), geranyl linalool isomer (7.46%), isoeugenol (7.17%) in its each stage. Benzenoids had much content at half bloom stage and full bloom stage while fatty acid derivatives had much content at initial flowering stage and after flowering stage. Most benzenoids such as butyl-hydroxy toluene and isoeugenol, which are some of main constituents of Lilium Oriental Hybrida 'Casa Blanca' had strong antioxidant effect. So they were expected to be used as antioxidant agents for food, feed, vegetable oil. The content of monoterpene compounds like geranyl linalool isomer increased at its later stage. Sesquiterpene such as farnesol, which is main component of lily existed only at full bloom stage. Therefore farnesol and geranyl linalool isomer of Lilium Oriental Hybrida 'Casa Blanca' are expected to be used for important spices with good fragrance. The suitable extraction time for the usable main constituents of Lilium Oriental Hybrida 'Casa Blanca' is as follows; farnesol and benzyl salicylate: at full bloom stage, butyl-hydroxy toluene: at half bloom stage, geranyl linalool isomer and isoeugenol: at stage after full bloom. Finally there was variation in essential oil components and contents of Lilium Oriental Hybrida 'Casa Blanca' in its flowering stages and they are expected to be used usefully for flavor industry, food industry, aromatherapy, when they are extracted at their suitable extraction time.

Effects of Dietary Mugwort Powder on the VBN, TBARS, and Fatty Acid Composition of Chicken Meat during Refrigerated Storage (쑥 분말의 급여가 계육의 저장기간 중 VBN, TBARS 및 지방산 조성에 미치는 영향)

  • The goal of this study was to investigate the effects of dietary mugwort on the proximate composition, volatile basic nitrogen (VBN), thiobarbituric acid reactive substance (TBARS) and fatty acid in chicken meats. One hundred sixty broiler chicks (1 d old) were assigned to one or four dietary groups: Control; commercial feed supplemented with 1% mugwort (T1); commercial feed with 3% mugwort (T2) and commercial feed with 5% mugwort (T3). After 42 d, broilers from each group were slaughtered and meat samples were vacuum packaged and stored at $4{\pm}1^{\circ}C$ over a period of 0, 1,2,3, and 4 wk. Chicken breast was not influenced by all treatments in moisture, crude protein and crude fiber, while crude fat was lowered (p<0.05) in chickens fed with the T2 and T3 diets compared to the control and T1 diets. All treatments with mugwort diets tended to have decreased VBN values for chicken breast and thigh compared to control. As storage time increased, VBN was increased for all chickens (p<0.05). No significant differences in TBARS were observed among all treatments at 0 wk. TBARS values were reduced with the T2 and T3 diets and initially increased from 0 through 3 wk, then abruptly decreased at 4 wk. Dietary mugwort supplementation resulted in increased stearic acid (excepted T2) and oleic acid and decreased linoleic acid. Stearic acid in thigh meat was decreased in the T1, T2 and T3, however linoleic acid levels tended to increase with mugwort powder supplementation. It is concluded that dietary mugwort has a positive effect on increasing unsaturated fatty acid contents and decreasing saturated fatty acids.

Effects of Iron Overload during Pregnancy on Oxidative Stress in Maternal Rats (임신 쥐의 철 과잉섭취가 조직의 산화적 스트레스에 미치는 영향)

  • Although iron is an essential mineral, excess iron intake during pregnancy may increase oxidative stress in tissues. This study was conducted to investigate the effects of iron overload during pregnancy on iron status and oxidative stress in maternal rats. Ten week-old female Sprague-Dawley rats were mated with male rats. Non-pregnant (control) and pregnant rats were fed diets containing normal Fe (35 mg/kg diet), high Fe (350 mg/kg diet), or excess Fe (1,050 mg/kg diet) during pregnancy. Rats were sacrificed on pregnancy day 19. No significant difference in weight gain, diet intake, or litter size was observed according to iron intake levels. Furthermore, serum iron, hemoglobin, and hematocrit were not different among the rats administered the three levels of Fe both in the control and pregnant groups. However, the iron levels were lower in pregnant rats than those in the control. The liver and spleen iron contents increased significantly in the excess Fe group. An increase in liver ferritin levels with increasing iron intake was observed. Protein carbonyl content, as a marker of oxidative stress, increased significantly in liver with increasing iron intake but not malondialdehyde. Glutathione peroxidase activity in the liver of pregnant rats fed excess iron decreased significantly. Bcl-2 protein expression in the liver declined remarkably with increasing maternal iron intake in pregnant rats. Taken together, iron overload during pregnancy had little effect on hematology. However, the deposits of iron in the liver and the decline in antioxidant enzyme activity implied increased oxidative stress in tissues of the excess Fe group. These results suggest that excess iron intake during pregnancy increases oxidative stress in maternal tissues and may also affect fetal tissues.

Effect of Cecectomy on Nitrogen Utilization ]Rate and Uric Acid Excretion in Growing Chicks (성장중인 닭에 있어서 맹장절제수술이 질소의 이용률과 요산의 배설량에 미치는 영향)

  • A 2 ${\times}$ 2 factorial experiment was conducted to determine the effect of cecectomy and urea supplementation on nitrogen utilization In growing chicks. Birds were either cecectomized or shamoperated and fed a 19% protein diet with or without 1% urea supplementation. All chicks were provided ad libitum access to feed and water throughout the six-day experimental period. In the cecectomized chicks, fred efficiency was significantly decreased when fed a 19% protein diet supplemented with 1% urea (P<0.05). When chicks were fed a 19% protein diet, cecectomy significantly shortened gastrointestinal passage time (GPT) of food (P<0.05). However, the GPT was significantly lengthened in chicks fed a 19% protein diet supplemented with urea (P<0.05). Regardless of diets, cecectomized chicks increased the moisture contents of the excreta (P<0.05) and feeding a 1% dietary urea also significantly increased (P<0.05). Cecectomy significantly decreased uric acid N excretion by about 30 mg per 100 g body weight per day (P<0.05) and uric acid N/ total N excretion when fed both diets (P<0.05). The present study suggests that cecectomy improves nitrogen utilization rate by decreasing uric acid excretion in growing chicks. These results are in good agreement with those obtained previously in a cecectomy study of adult chickens.

  • The study is aimed to recognize the meaning of foundation study through understanding history and flow of foundation study in Korea. The analysis period is from 1998 when there was first boom of venture foundation in Korea after IMF to 2013 September when the latest thesis was made, and the analysis intends total 382 dissertations the online. The analysis was conducted in the fields of foundation study, foundation support, foundation steps, founding company forms, founder's demographic features, and from the analysis, the following conclusion could be drawn out. First, the field actively carrying out foundation studies is the business management & economics field, but the foundation is dealt as an object of study across the all industries. It could be identified that of the all industries, especially IT industry, food service industry, and contents industry are the fields showing much interest about the foundation. Second, the field attracting most keen attention among the fields of foundation support was the field related with the founders. Third, the main problem relevant to foundation study was that the foundation was judged in comprehensive viewpoint, not being judged with strategies, in detail. Fourth, investing from the company's form aspect, it was found that during the starting period, the main foundation trend was the venture firm foundation, but recently the foundation-relevant attention is focused on the small business foundation and the most dominant founder type is the youth including college students, and the women's foundation is paid continual attention and senior foundation is being actively discussed. According to these analyzed results, future foundation studies should actively promote the general foundation studies, the specified foundation studies reflecting each industry's specialities, the fusion studies.

Development of High Purity Purification Method of Chondroitin Sulfate Extracted from Skate Cartilage (홍어 연골로부터 추출된 콘드로이틴 황산의 고순도 정제방법 개발)

  • A purification method was established for high-purity chondroitin sulfate from skate cartilage. Hydrolytic extraction of skate backbone cartilage was investigated with the proteases alcalase and protamex, and the extraction contents of chondroitin sulfate were measured with several physicochemical processes. The yield of extract from skate cartilage with $40^{\circ}Brix$ concentration was 23.3% with 2% alcalase hydrolysis, which was decreased to 8.47% and 3.37% with the first and second additional ethanol purifications, respectively. The yield was 16.62% with one ethanol purification after hydrolysis with a mixture of 1% alcalase and 1% protamex. The content of chondroitin sulfate was measured as 39.88-45.08% with different ratios of ethanol solvent. The content was 42.92% at a solvent ratio of 1:1 with alcalase protease and 45.08% with a ratio of 1:2 using a protease mixture of alcalase and protamex. The molecular weight range of chondroitin sulfate was about 110-310 thousand Da, and the purity of chondroitin sulfate was 24.87-49.92% with a mixture of alcalase and protamex in GPC analysis. The maximum purity of chondroitin sulfate was 53.93% after ultrafiltration. The odor strength of chondroitin sulfate was decreased by 33% and 38% after ethanol purification and additional filtration with activated carbon, respectively. The odor concentration of ammonia and TMA from chondroitin sulfate was decreased by 52.1% and 37.89% with activated carbon filtration and two ethanol purifications, respectively, but it was necessary to eliminate the odor components efficiently using additional physicochemical processes.
