와송(瓦松)이 만성 골수성 백혈병 세포주(K562)에서 세포사멸에 미치는 영향 (Effect of Orostschys japonicus A. Berger on Apoptosis in K562 Cell Lines)

  • Objectives : This study was performed to determine if Orostschys japonicus A. Berger has protective effects against CML in K562 cell lines. Materials and Methods : MTT assay, cell proliferation assay, Reverse transcription-polymerase reaction chain, RT-PCR, DNA fragmentation assay, Quantitative PCR were studied. Results : Orostschys japonicus A. Berger had no effects on Bax gene in K562 cell lines, but decreased Bcl-2 gene, and increased the Caspases-3 gene. This is indicate of induced apoptosis in K562 cell lines by Orostschys japonicus A. Berger. Conclusion : These results suggest that Orostschys japonicus A. Berger has effects on apatosis in K562 cell lines.

Inhibitory Effect of Scutellaria barbata Don Water-extracts on Growth and DNA Incorporation of Human Cancer Cells

  • The water-extracts of Scutellaria barbata Don (SBDE) were isolated from Chinese medicinal plant sources. The extracts showed strong growth-inhibitory activity and cancer chemopreventive activity on the growth and DNA incorporation of MG63 human osteosarcoma and K562 human leukemia cell lines. The growth of human cancer cells was inhibited in the presence of the extracts (20, 50 and 100 ${\mu}$g/ml), and the effects were concentration-dependent and incubation time-dependent up to 8 days. When 50 ${\mu}$g/ml of the extracts was added to the media of MG63 and K562, cell growth after 8 days or 6 days of incubation was retarded by 93.2 to 97.3% of the control group. Morphological changes of MG63 and K562 cell lines were observed. As the concentration of the extracts increased up to 50 ${\mu}$g/ml, degree of cell aggregation decreased. Moreover, the DNA incorporation of the cells which were labeled with [3H] thymidine was significantly reduced after 3 days of incubation at $37^{\circ}C$ with the extract. Therefore, it is suggested that the extract is highly effective on inhibition of cancer cell growth. The extract also inhibited gene expression of IGF-II in transcriptional level. Since IGF-II works as a mitogenic effector on MG63 and K562 cell lines, these results suggest that the growth inhibition is in part mediated through the inhibition of IGF-II gene expression.

비기환(?氣丸)이 백혈병(白血病)과 임파종(淋巴腫) 환자(患者)에서 추출(抽出)한 암세포(癌細胞)에 미치는 항암효과(抗癌效果) (Antitumor Effects of Bigihwan on Tumor Cells derived from Leukemia and Lymphoma Patients)

  • Bigihwan which has been widely used in Oh-jug in oriental Medicine was investigated on its antitumor effect employing blood cancer cell lines. K 562 derived from human erytholeukemia, Raji from lymphoma and $MO_4$ from hlastogenic cancer were used in this study to see the analytical evaluation of Bigihwan' s antitumor effect using three different kinds of methods such as $^{3}H-thymidine$ up take assay. MTT assay and live cell counts by Trypan blue assay. The result obtained are as follows. 1. When higher than 10% Bigihwan was treated. inhibitory effect of tumor killing action was observed showing the increasing order of $MO_4$, K 562 and Raji(Fig. 3). 2. When 1 to 5% of Bigi-hwan was treated, 4 to 30% of tumor cell survival was observed according to various blood tumor cell lines suggesting that antitumor effect of Bigi-hwan was different as the characteristics of tumor cells showing 70 to 95% cell killing effent(Fig. 4). 3. Compared the survivals of cells by relative scales though the initial cpm was variable because of different cell growth rate. Raji was most effective being killed 95% by the treatment of 1% Bigihwan while Raji and K562 showed 93% by 5% Bigihwan.(Fig. 5) 4. The survival rate of Raji derived from Burkitt lymphoma was rather increased to 2.3 times when Bigihwan concentration was increased from 1 to 10% lmplying of refraining from over use of this anticancer drug. specially to lymphoma patients(Fig. 5). 5. Bigihwan was most effective to K 562 and then $MO_4$ showing 95% tumor cell death by using 1% of this anticancer drug while it was least effective to Raji showing only 68% of tumor cell death(Fig.7). 6. Judging from the all the analytical methods used in this study, through all different three tumor cell lines. Bigihwan was most effective to K 562 derived from human erythroleukemia.

托裡消毒散이 抗腫瘍 및 免疫作用에 미치는 效果 (The effect of TakliSodoksan extract on anti-tumor action and immune-function)

  • The purpose of this Study was to investigate effect of TakliSodokSan(TSS) on the anti-tumor, immunocytes and nitric oxide(NO) production from mice peritoneal macrophages. This Study estimated the proliferation of L1210 cell lines, A431 cell lines, Hep-G2 cell lines, K562 cell lines, 3T3 cell lines, mouse thymocytes and mouse splenocytes and NO production from pcritoneal macrophages in vitro, and estimated the proliferation of L1210 cells, thymocytes and splenocytcs, NO production from peritoneal macrophages and body weight in L1210 cells-transplanted mice in vivo. The results were obtained as follows; 1. TSS inhibited significantly the proliferation of L1210, A431, Hep-G2, K562 cell lines in vitro. 2. TSS accelerated the proliferation of mice thymocytes and splenocytes in vitro. 3. TSS was not increased the nitric oxide production from mice peritoneal macrophages in vitro. 4. TSS inhibited significantly the proliferation of L1210 cells in Ll210 cells∼transplanted mice. 5. TSS accelerated the proliferation of mice thymocytes and splenocytes In L1210 cells-transplanted mice. 6. TSS was increased significantly the nitric oxide production from mice peritoneal macrophages in L1210 cells-transplanted mice. 7. TSS was increased the body weight as comparing with control group in Ll210 cells-transplanted mice.

Antitumor activity of 2(S)-5,$2^{I}$,$5^{I}$-trihydroxy-7,8-dimenthoxyflavanone and its analogues

  • In an effort to increase of the antitumor activity of 2(S)-$2^{I}$,$5^{I}$-trihydroxy-7, 8-dimethoxyflavanone isolated from Scutellaria indica, we synthesized its analogues, II, III and IV. They showed potent cytotoxicity in vitro against cancer cell lines, L1210, K562 and A549. On the basis of $ED_50$ values against the cancer cell lines, III exhibited about 2-7 times stronger activity than I against various cell lines. We tested the antitumor activity of the analogues against Sarcoma 180 cells in vivo and evaluated the structure-activity relationship. The antitumor activity appeared to be related to the hydrogen bond between carbonyl group at C-4 and hydroxyl group at C-5, in contrast to cytotoxic action.

Antioxidant value and Antiproliferative Efficacy of Mitragynine and a Silane Reduced Analogue

  • Background: To investigate the antioxidant value and anticancer functions of mitragynine (MTG) and its silane-reduced analogues (SRM) in vitro. Materials and Methods: MTG and SRM was analyzed for their reducing power ability, ABTS radical inhibition and 1,1-diphenyl-2-picryl hydrazylfree radicals scavenging activities. Furthermore, the antiproliferation efficacy was evaluated using MTT assay on K 562 and HCT116 cancer cell lines versus NIH/3T3 and CCD18-Co normal cell lines respectively. Results: SRM and MTG demonstrate moderate antioxidant value with ABTS assay (Trolox equivalent antioxidant capacity (TEAC): $2.25{\pm}0.02$ mmol trolox / mmol and $1.96{\pm}0.04$ mmol trolox / mmol respectively) and DPPH ($IC_{50}=3.75{\pm}0.04mg/mL$ and $IC_{50}=2.28{\pm}0.02mg/mL$ respectively). Both MTG and SRM demonstrate equal potency ($IC_{50}=25.20{\pm}1.53$ and $IC_{50}=22.19{\pm}1.06$ respectively) towards K 562 cell lines, comparable to control, betulinic acid (BA) ($IC_{50}24.40{\pm}1.26$). Both compounds showed concentration-dependent cytototoxicity effects and exert profound antiproliferative efficacy at concentration > $100{\mu}M$ towards HCT 116 and K 562 cancer cell lines, comparable to those of BA and 5-FU (5-Fluorouracil). Furthermore, both MTG and SRM exhibit high selectivity towards HCT 116 cell lines with selective indexes of 3.14 and 2.93 respectively compared to 5-FU (SI=0.60). Conclusions: These findings revealed that the medicinal and nutitional values of mitragynine obtained from ketum leaves that growth in tropical forest of Southeast Asia and its analogues does not limited to analgesic properties but could be promising antioxidant and anticancer or chemopreventive compounds.

In Vitro Anticancer Activities of Anogeissus latifolia, Terminalia bellerica, Acacia catechu and Moringa oleiferna Indian Plants

  • The present study was designed to evaluate in vitro anti-proliferative potential of extracts from four Indian medicinal plants, namely Anogeissus latifolia, Terminalia bellerica, Acacia catechu and Moringa oleiferna. Their cytotoxicity was tested in nine human cancer cell lines, including cancers of lung (A549), prostate (PC-3), breast (T47D and MCF-7), colon (HCT-16 and Colo-205) and leukemia (THP-1, HL-60 and K562) by using SRB and MTT assays. The findings showed that the selected plant extracts inhibited the cell proliferation of nine human cancer cell lines in a concentration dependent manner. The extracts inhibited cell viability of leukemia HL-60 and K562 cells by blocking G0/G1 phase of the cell cycle. Interestingly, A. catechu extract at $100{\mu}g/mL$ induced G2/M arrest in K562 cells. DNA fragmentation analysis displayed the appearance of a smear pattern of cell necrosis upon agarose gel electrophoresis after incubation of HL-60 cells with these extracts for 24h.

Detection of Anticancer Activity from the Root of Angelica gigas In Vitro

  • Anticancer activity of a fraction of the ethanol extract from the root of Korean angelica (Angelica gigas Nakai) was recognized in human cancer cell lines HeLa $S_3$, K-562, and Hep $G_2$. The extract blocked the phorbol ester-inducing megakaryocytic differentiation of K-562 cells, which indicated the modification of protein kinase C (PKC) activity. In vitro assay showed the activation of PKC by the extract. An effective fraction of the Angelica gigas extract, of which $R_f$ value was 0.64 in a thin layer chromatography, was a different component from those of European angelicas. The $ED_50$ value of the fraction was 8, 9, and $16\;\mu\textrm{m}/ml$ against HeLa $S_3\;Hep\;G_2$, and K-562 cells, respectively, while the fraction showed higher $ED_50$ values against normal cell lines.

  • The purpose of this study was to investigate effect of water extract of Carthami Flos on the proliferation of human cancer cell-lines. The effects of Carthami Flos on the proliferation of A431, HeLa, MOLT-4, K562 cells, Balb/c 3T3 cells, mouse thymocytes, splenocytes and human lymphocytes were estimated by MTT colorimetric assay. The results were as follows; 1. Carthami Flos did not effect A431, HeLa, MOLT-4, K562 cells. 2. The cytotoxicity of mitomycin C on K562 cells was increased by the combination of Carthami Flos. 3. Carthami Flos inhibited the proliferation of Balb/c 3T3 cells. 4. Carthami Flos stimulated the proliferation of thymocytes. 5. Carthami Flos stimulated the proliferation of splenocytes. 6. Carthami Flos stimulated the proliferation of human lymphocytes.

다약제내성 발현 암세포에서 $^{99m}Tc-sestamibi$$^{99m}Tc-tetrofosmin$ 섭취의 비교 (Comparison of the Uptakes of $^{99m}Tc-sestamibi\;and\;^{99m}Tc-tetrofosmin$ in Cancer Cell Lines Expressing Multidrug Resistance)

  • 목적: 다약제내성이 유발된 암세포에서 $^{99m}Tc-sestamibi$$^{99m}Tc-tetrofosmin$의 암세포 내 섭취정도를 비교하고 다약제내성 극복제로 잘 알려진 verapamil 과 cyclosporin A 처리에 의한 두 방사성 의약품의 암세포 내 섭취정도를 비교해 보았다. 재료 및 방법: Doxorubicin으로 다약제내성이 유발된 HCT15/CL02 대장암 세포와 doxorubicin과 vincristine으로 다약제내성을 유발시킨 K562(Adr)과 K562(Vcr) 백혈병 세포를 사용하였다. 다약제내성의 발현은 RT-PCR로 증명하였으며, verapamil은 1, 10, 50, 100, 200 ${\mu}M$의 농도로, cyclosporin A는 0.1, 10, 50, 100 ${\mu}M$의 농도로 각각 사용하였다. MIBI와 tetrofosmin의 암세포내 섭취는 $37^{\circ}C$에서 $1{\times}10^{6}cells/ml$ 농도의 단일세포 부유상태에서 1, 15, 30, 45, 60분 간격으로 배양하여 각 시간대별로 상층액과 침전물을 분리하여 각각의 방사능을 감마 계수기로 측정하였다. 결과: 다약제내성이 발현된 암세포에서는 모세포에 비하여 MIBI와 tetrofosmin의 섭취가 감소되었다. 두 방사성약품의 섭취정도는 HCT15/CL02세포와 K562(Adr)세포에서는 유의한 차이가 없었으나, K562(Vcr)세포에서는 MIBI가 tetrofosmin보다 다소 높았다. Verapamil과 cyclosporin A를 처리하였을 때 MIBI와 tetrofosmin의 섭취율은 기저치보다 모두 증가하였고, verapamil 에 의한 MIBI와 tetrofosmin의 섭취율(30분)을 기저치(30분)와 비교해 본 결과 HCT15/CL02 세포에서($100{\mu}M$)는 각각 11.9배와 6.8배, K562(Adr) 세포에서($50{\mu}M$)는 각각 14.3배와 8배, K562(Vcr) 세포에서($10{\mu}M$)는 각각 7배와 5.7배 증가하였다. Cyclosporin A에 의한 MIBI와 tetrofosmin의 섭취율(30분)을 기저치(30분)와 비교해 본 결과 HCT15/CL02세포에서($50{\mu}M$)는 각각 10배와 2.4배, K562(Adr)세포에서($50{\mu}M$)는 각각 44배와 13배, K562(Vcr)세포에서($10{\mu}M$)는 각각 18.8배와 11.8배 증가하여, MIBI의 섭취율이 tetrofosmin보다 1.2배에서 4배정도 높게 나타났다. 결론: 이러한 결과로 보아 MIBI와 tetrofosmin은 다약제내성의 발현을 평가할 수 있는 방사성의약품으로 판단되며, 다약제내성 극복제의 효능평가에는 MIBI가 tetrofosmin보다 더 우수할 것으로 사료되나, 세포추에 따른 차이가 있을 수 있으므로 보다 많은 세포주에서의 추가적인 연구가 필요할 것이다.
