Effects of Cryopreservation on Ca2+ Signals Induced by Membrane Depolarization, Caffeine, Thapsigargin and Progesterone in Boar Spermatozoa

  • Although the fertilizing ability of spermatozoa is greatly reduced after freezing, complete understanding of alterations induced by cryopreservation has not been elucidated. The present study evaluates the effects of cryopreservation on the $Ca^{2+}$ handling of boar spermatozoa using several sperm activators. Intracellular $Ca^{2+}$ signals from single spermatozoa were measured using confocal $Ca^{2+}$ imaging of unfrozen samples and of other spermatozoa after having been frozen. Elevation of the external $K^{2+}$ concentration elicited a three times larger $Ca^{2+}$ increase in fresh spermatozoa than in cryopreserved spermatozoa. Caffeine elicited $Ca^{2+}$ transients with some oscillations in the fresh spermatozoa, but not in the thawed spermatozoa. Depletion of the $Ca^{2+}$ store with thapsigargin induced a rapid rise in $Ca^{2+}$ in the control but generated a smaller increase of $Ca^{2+}$ after thawing. Exposure to progesterone induced a biphasic rise of the $Ca^{2+}$ level in the fresh spermatozoa only. Sperm viability was reduced by cryopreservation. Resting $Ca^{2+}$ levels in fresh and cryopreserved spermatozoa were similar. Longer incubation (2.5 h) of thawed spermatozoa partly recovered the $Ca^{2+}$ response to the interventions. These results suggest that cryopreservation reduces the responsiveness of spermatozoa to depolarization, modulators of the internal $Ca^{2+}$ store and progesterone in terms of the $Ca^{2+}$ signal, thus providing a possible mechanism for reduced fertility observed in cryopreserved boar spermatozoa.

  • The activity of $Ca^{2+}$calmodulin (CaM)-dependent protein kinase Ia (CaM kinase Ia) is shown to be regulated through direct phosphorylation by CaM kinase I kinase (CaMK IK). In the present study, three distinct CaMKIK peaks were separated from Q-Sepharose colunm chromatography of pig brain homogenate using a Waters 650 Protein Purification System. The purified CaMKIK from the major peak potently and rapidly enhanced CaM kinase Ia activity, reaching a maximal stimulation within 2min at the concentrations of 12-15nM. The activated state of CaM kinase Ia is characterized by a markedly enhanced $V_{max}4 as well as significantly decreased $K_m\;and\;K_a$ values toward peptide substrate and CaM, respectively. These observations suggest the activation process of CaM kinase Ia. The phosphorylation of CaM kinase Ia by CaMKIK may induce its conformational change responsible for the alterations in the kinetic properties, which ultimately leads to the rapid enzyme activation.

식이 중 칼슘수준과 톳 첨가가 난소절제한 흰쥐의 골지표와 혈청 지질 함량에 미치는 영향 (Effects of Dietary Calcium Level and Hijikia fusiforme Supplementation on Bone Indices and Serum Lipid Levels in Ovariectomized Rats)

  • This study explored the effects of dietary calcium level and Hijikia fusiforme supplementation on bone indices and serum lipid levels using 36 female Sprague-Dawley rats as a model. Rats received low Ca diet for 3 weeks after ovariectomy. The rats were then divided into six dietary groups and fed low (0.1% Ca), normal (0.5% Ca) and high (1.5% Ca) Ca diets (CaL, CaN, CaH) and low, normal, high Ca diets with Hijikia fusiforme supplementation (CaLH, CaNH, CaHH) for 3 weeks. After each experimental periods, 24 hour urine and/or blood samples, left and right femurs were collected for analysis. Serum Ca concentration showed no significant difference by dietary Ca levels and Hijikia fusiforme supplementation. Alkaline phosphatase activity was significantly higher in normal and high Ca group compared to low Ca group. Serum total cholesterol, triglyceride and total lipid were not significantly different among groups. HDL-cholesterol showed no significant difference by Hijikia fusiforme supplementation. However, the normal and high Ca groups showed significantly higher HDL-cholesterol compared to the low Ca group. Urinary hydroxyproline and hydroxyproline/creatinine ratio were not significantly different among groups. The wet weight of the femur was significantly higher in low Ca group compared to normal or high Ca group. The dry weight, wet weight/body weight, length and breaking force of the femur were not significantly different among groups. Ash contents/wet weight of the femur was significantly increased as dietary Ca levels up and significantly higher in Hijikia fusiforme supplementation groups. The Ca content of the femur were significantly higher in the normal and high Ca groups than the low Ca group. However, there was no significant difference in Ca content by Hijikia fusiforme supplementation.

CaSO4 기반 산소전달입자의 환원과 분해특성 (Reduction and Decomposition Characteristics of CaSO4 Based Oxygen Carrier Particles)

  • As a candidate for cheap oxygen carrier, $CaSO_4$ based oxygen carriers have been developing. However, research on reaction characteristics and side reaction of $CaSO_4$ based oxygen carrier is very limited. There are many possible reactions for main components of syngas from coal. In this study, we prepared three $CaSO_4$ based oxygen carriers ($CaSO_4$-$Fe_2O_3$/bentonite, $CaSO_4$-$K_2CO_3$/bentonite, $CaSO_4$-CaO/bentonite) and performed reduction tests by hydrogen. Cyclic reduction-oxidation tests up to $5^{th}$ cycle are also conducted using hydrogen as fuel. Reduction reactivity of those $CaSO_4$ based oxygen carriers were compared with that of NiO based oxygen carrier (OCN703-1100). Real weight change fractions of $CaSO_4$ based oxygen carriers were higher than theoretical oxyen transfer capacity and reactivity of these particles decreased with the number of cycle increased. To check possible side reaction of $CaSO_4$ based oxygen carriers, $CaSO_4$ decomposition tests were carried out and $SO_2$ was detected even at $700^{\circ}C$. Consequently, we could conclude that $CaSO_4$ based oxygen carriers decompose and release $SO_2$ and this reaction lead reactivity decay of $CaSO_4$ based oxygen carries.

  • 수도삼요소(水稻三要素) 이용율(利用率)(1967 및 1968년(年))과 기상권(氣象圈), 답토양(畓土壤) 단면(斷面)의 추적양식(推積樣式) 배수등급(排水等級) 토성(土性) 및 작토(作土)의 토양화학적(土壤化學的) 특성(特性)과의 관계(關係)를 검토(檢討)한 결과(結果) 다음과 같은 결과(結果)를 얻었다. 1. N이용율(利用率)은 남부(南部)>중부(中部)>북부(北部)의 순(順)이고 K이용율(利用率)은 남부(南部)>북부(北部)>중부(中部)의 순(順)이며 P이용율(利用率)은 연도별(年度別)로 다르다. 2. P이용율(利用率)은 남부(南部)의 도(道)에서 연도별(年度別) 변동(變動)이 크고 추적(推積) 배수(排水) 토성별(土性別)에서도 연도별(年度別) 변동(變動)이 커서 토양(土壤)과 기상(氣象)의 교호작용(交互作用)에 크게 영향 받는것을 알 수 있다. 3. 추적양식별(推積樣式別) N이용율(利用率)은 Alluvial Colluvial (AC)>Alluvial (A)>Fluvomarine (FM)>Old Alluvial (OA)의 순(順)이고 K이용율(利用率)은 OA>AC>A>FM의 순(順)이다. 4. N이용율(利用率)은 토양단면(土壤斷面)의 배수(排水)가 양호(良好)할수록 적어지고 K와 P의 이용율(利用率)은 약간양호(若間良好)에서 높은 경향이고 불량(不良)이나 매우 양호(良好)의 편에서 낮은 경향이다. 5. N이용율(利用率)은 미사식양질(微砂埴壤質)에서 가장높고 사질(砂質)로 갈수록 떨어지며 식질(埴質)에서도 낮으나 K와 P 이용율(利用率)은 식양질(埴壤質) 이하(以下)의 토성(土性)에서 높은 경향(傾向)이다. 6. N이용율(利用率)은 지산 극락 신답통에서 크고 화동 규암 용지 화봉에서 적었다. 7. N이용율(利用率)은 토양중(土壤中) 유기물함량(有機物含量)(OM), Ca 및 CEC와 유의정상관(有意正相關)을 보이고 P이용율(利用率)이 높았던 해에만 P함량(含量)과 유의부상관(有意負相關)을 보였으며 P이용율(利用率)은 CEC, Mg 및 Ca와 유의정상관(有意正相關)을 보였다. 8. K이용율(利用率)은 그것이 적었던 해에는 K/(Ca+Mg), P, OM, K의 순(順)으로 부상관(負相關)을 Ca, Mg, CEC 순(順)으로 정상관(正相關)을 보였는데 K/(Ca+Mg)와만 유의성(有意性)이 있으며 K이용율(利用率)이 큰 해에는 P, OM, K/(Ca+Mg) K의 순(順)으로 정상관(正相關)을 CEC, Mg, Ca 순(順)으로 부상관(負相關)을 보였는네 P, OM, CEC 와만 유의성(有意性)을 갖는다. K이용율(利用率)이 컸던 해에만 토양중 K는 토양중 P 및 OM과 유의정상관(有意正相關)을 갖는다. 9. 삼요소이용율(三要素利用率)과 pH는 유의성(有意性)은 없으나 부상관(負相關)이다. 10. OM은 P K 및 K/(Ca+Mg)와 유의정상관(有意正相關)을 Mg와는 유의부상관(有意負相關)을, P는 K, K/Ca+Mg와 유의정상관(有意正相關)을 Mg 및 CEC와 유의(有意) 부상관(負相關)을 K는 K/(Ca+Mg), Mg 및 CEC와 유의정상관(有意正相關)을 K/(Ca+Mg)는 Ca와 유의정상관(有意正相關)을 Mg 및 CEC와는 유의부상관(有意負相關)을, Ca는 Mg CEC 및 pH와, Mg는 CEC와 유의(有意) 정상관(正相關)을 보였다. 11. 삼요소(三要素) 이용율(利用率) 및 토양(土壤)의 화학적(化學的) 특성(特性) 상호간(相互間)의 관계에서 OM이 시비(施肥)N의 흡수(吸收)를 돕고 K흡수(吸收)는 K보다 K/(Ca+Mg)에 의(依)하여, P흡수(吸收)는 Mg에 의(依)하여 크게 영향받는 것을 알수있고 P와 K이용율(利用率)이 해에 따라 다른 원인(原因)이 부분적(部分的)으로 설명(說明)될 수 있었다.

흰쥐의 뇌 기저동맥에서 CGRP에 의한 혈관 이완반응의 기전에 대한 연구 (Pharmacological Action Mechanism(s) of Vasodilator Effect of Calcitonin Gene-related Peptide in Rat Basilar Arteries)

  • 본 실험에서는 흰쥐의 뇌 기저동맥을 이용하여 $K^+$과 U46619에 의한 수축과 세포내 $Ca^{2+}$의 변동을 관찰하고, 이들 반응을 CGRP 전처치시 나타나는 반응과 비교하였다. CGRP (30과 100 nM)는 U46619에 의하여 야기된 수축반응과 세포내 $Ca^{2+}$의 증가반응을 억제시켰으나, $K^+$ (90 mM)에 의하여 나타나는 반응에는 영향을 미치지 아니하였다. 게다가, U46619에 의하여 야기되는 장력에 대하여 세포내 $Ca^{2+}$의 변동 $(F_{340}/F_{380})$을 도표화하여 세포내 $Ca^{2+}$ 농도와 장력의 발생과의 상관관계를 검토하고, 이들 결과를 CGRP 전처치시 나타나는 결과와 비교하였다. CGRP (30과 100 nM) 전처치군에서 얻어진 직선이 오른쪽으로 치우치지는 않으면서 아래쪽으로 이동하는 점으로 볼 때, CGRP가 $Ca^{2+}$에 대한 수축기구의 감수성에는 영향을 미치지 않으면서 세포내 $Ca^{2+}$ 농도를 저하시킴에 의하여 U46619에 의한 근수축반응을 억제시키는 것으로 보여진다. 이러한 CGRP의 효과는 CGRP1 수용체 길항제인 CGRP$(8{\sim}37)$ 분획(100 nM)의 전처치시 현저히 억제되었다. CGRP에 의한 수축력과 세포내 $Ca^{2+}$의 저하는 large conductance $Ca^{2+}$에 의하여 활성화되는 $K^+$ 통로 봉쇄제인 charybdotoxin (100 nM)과 iberiotoxin (100 nM)의 전처치에 의하여 완전하게 역전되었으나, small conductance $Ca^{2+}$에 의하여 활성화되는 $K^+$ 통로 봉쇄제인 apamin (300 nM)과 ATP에 감수성이 높은 $K^+$ 통로 봉쇄제인 glibenclamide (1 ${\mu}M$)에 의해서는 영향을 받지 아니하였다. 이상의 결과로 볼 때 CGRP1 수용체는 $Ca^{2+}$에 의하여 활성화되는 $K^+$ 통로를 개방시킴으로 세포내 $Ca^{2+}$을 감소시켜 뇌 기저동맥의 이완반응을 매개하는 것으로 사료된다.

Oxidized Low-density Lipoprotein- and Lysophosphatidylcholine-induced $Ca^{2+}$ Mobilization in Human Endothelial Cells

  • The effects of oxidized low-density lipoprotein(OxLDL) and its major lipid constituent lysophosphatidylcholine(LPC) on $Ca^{2+}$ entry were investigated in cultured human umbilical endothelial cells(HUVECs) using fura-2 fluorescence and patch-clamp methods. OxLDL or LPC increased intracellular $Ca^{2+}$ concentration($[Ca^{2+}]_i$), and the increase of $[Ca^{2+}]_i$ by OxLDL or by LPC was inhibited by $La^{3+}$ or heparin. LPC failed to increase $[Ca^{2+}]_i$ in the presence of an antioxidant tempol. In addition, store-operated $Ca^{2+}$ entry(SOC), which was evoked by intracellular $Ca^{2+}$ store depletion in $Ca^{2+}$-free solution using the sarcoplasmic reticulum $Ca^{2+}$ pump blocker, 2, 5-di-t-butyl-l,4-benzohydroquinone(BHQ), was further enhanced by OxLDL or by LPC. Increased SOC by OxLDL or by LPC was inhibited by U73122. In voltage-clamped cells, OxLDL or LPC increased $[Ca^{2+}]_i$ and simultaneously activated non-selective cation(NSC) currents. LPC-induced NSC currents were inhibited by 2-APB, $La^{3+}$ or U73122, and NSC currents were not activated by LPC in the presence of tempol. Furthermore, in voltage-clamped HUVECs, OxLDL enhanced SOC and evoked outward currents simultaneously. Clamping intracellular $Ca^{2+}$ to 1 ${\mu}M$ activated large-conductance $Ca^{2+}$-activated $K^+(BK_{ca})$ current spontaneously, and this activated $BK_{ca}$ current was further enhanced by OxLDL or by LPC. From these results, we concluded that OxLDL or its main component LPC activates $Ca^{2+}$-permeable $Ca^{2+}$-activated NSC current and $BK_{ca}$ current simultaneously, thereby increasing SOC.

DTNB oxidation effects on T-type $Ca^{2+}$ channel isoforms

  • Redox regulation is one of the ubiquitous mechanisms to modulate ion channels. We here investigated how 5,5'-dithio-bis (2-nitrobenzoic acid), a cysteine specific oxidizing reagent, modulates $Ca_v3.1$ and $Ca_v3.2$ T-type $Ca^{2+}$ channels expressed in Xenopus oocytes. Application of the reagent inhibited $Ca_v3.1$ and $Ca_v3.2$ currents in a dose-dependent manner. The oxidizing reagent (1 mM) reduced the peak amplitude of $Ca_v3.1$ and $Ca_v3.2$ currents by ~50% over 2-3 minutes and the decreased currents were fully recovered upon washout of it. The reagent slowed the activation and inactivation kinetics of $Ca_v3.1$, $Ca_v3.2$, and $Ca_v3.3$ channel currents. Notably, the reagent positively shifted both activation and steady-state inactivation curves of $Ca_v3.1$, while it did not those of $Ca_v3.2$. Utilizing chimeric channels from $Ca_v3.1$ and $Ca_v3.2$, we localized the domains III and IV of $Ca_v3.1$ responsible for the positive shifts of channel activation and steady-state inactivation. These findings provide hints relevant to the electrophysiological and molecular mechanisms accounting for the oxidative regulation of T-type channels.

규산질다공체와 무기첨가물의 수중 인 제거 효과 (Effects of CellCaSi and Inorganic Additives on Phosphorus Removal in Water)

  • 녹조 제어를 위하여 수중의 인 제거에 관여하는 CellCaSi의 여러 가지 조건별 인 제거효과를 조사하였다. CellCaSi의 입도에 따른 인 제거효과는 직경 1, 2 그리고 4mm 이하의 3가지 중 1mm 이하의 작은 입자를 사용했을 때 가장 우수하였다. CellCaSi의 인 제거효과는 처리량의 증가에 비례하였다. 수중의 pH 5, 7, 9에서 초기 인 농도 0.20mg/l는 각각 0.09, 0.08및 0.08mg/l로 감소되어 상이한 pH조건에 따른 CellCaSi의 인 제거효과는 큰 차이를 보이지 않았다. CellCaSi에서 용출되는 양이온은 Ca이며, Al과 Fe양이온은 검출되지 않았다. 초기에 급격히 용출된 Ca양이온은 수중의 인과 반응하는 것으로 확인되었다. 초기 인 농도 0.10, 1.0mg/1는 CellCaSi와 Ca, Fe 화합물을 첨가한 처리구에서 8일 후 각각 0.03,0.47mg/l를 기록하며 가장 많이 감소하였다. 이에 따라 인 제거효과는 CellCaSi와 Ca, Fe 화합물을 동시에 처리하였을 때 가장 좋은 것으로 조사되었다. CellCaSi의 처리에 따라 pH가 약간 증가하였고, 전기전도도도 증가되지만 상수원수 기준 허용치 (500${\mu}$S/cm) 이내였다.

고지방식을 섭취한 본태성 고혈압쥐에서 칼슘과 소디움 섭취가 혈압과 무기질 대사에 미치는 영향 (Effect of Dietary Ca and Na Levels on Blood Pressure and Mineral Metabolism in Spontaneously Hypertensive Rats Fed High Fat Diet)

  • This study explored the effect of dietary levels of Na and Ca on spontaneously hypertensive rats (SHR). SHR were randomly divided into 5 groups and fed a high fat/cholesterol diet containing three levels of Na (0.05, 0.1, 1.5%) and Ca (0.1, 0.5, 1.5%) for 9 weeks. Body weight gain was not influenced by dietary intake but water intake significantly increased in high Na supplementation. Systolic blood pressure was not influenced by dietary Na and Ca levels but was decreased by dietary low Na/high Ca levels at 9 weeks. Angiotensin-II level was affected by dietary Na level but not by Ca levels. Plasma Ca, Mg, K and Na levels were in the normal range regardless of dietary Na and Ca levels. Weight, and K and Na contents of the heart and kidney were not significantly different among those with different dietary Na and Ca levels. Ca and Mg contents of the heart and kidney were significantly higher in the normal Na/normal Ca group. Ca and Mg in the feces were higher in those with high Ca intake. Na in the feces was higher in those with high Na intake. Therefore, Na and Ca had different mechanisms in the hypertension/hyperlipidemia models, respectively. And we suggested that Mg must be supplemented when Ca intake was high because Mg excretion was increased by Ca supplementation.
