Adaptive Characteristics of the Longtooth Grouper Epinephelus bruneus according to Stocking Density and Water Temperature (사육밀도와 수온에 따른 자바리(Epinephelus bruneus)의 적응특성)

  • The aim of this study was to investigate the effects of stocking density and water temperature in the rearing of the longtooth grouper Epinephelus bruneus. Juvenile fish [mean body weight (BW)=$6.9{\pm}1.1g$] were raised for 6 weeks, in four density groups of 0.25, 0.5, 0.75, and 1 individuals/L. There were no significant differences in weight gain (WG) or specific growth rate (SGR) among the density groups; however, both WG and SGR tended to be higher in the low density group. After 6 weeks of rearing in temperature-controlled water ($23.6{\pm}0.8^{\circ}C$) and ambient temperature water ($19.6{\pm}1.8^{\circ}C$), there were no significant differences in WG or SGR values, but both tended to be higher in the temperature controlled water. The feed efficiency (FE) and daily feed intake (DFI) and WG values were also higher in the temperature-controlled water than in the ambient temperature water. WG values were lower at lower water temperatures. The same pattern was observed for SGR, FE, and FI, all of which exhibited lower values at lower temperatures. In particular, WG, SGR, and FE values all tended to decrease at the $16^{\circ}C$ water temperature. The effects of water temperature on 1-year fish (mean BW = $387.6{\pm}30.2g$) and 3-year fish (mean BW = $1,338.3{\pm}73.8g$) were similar to those of the juvenile fish (0-year fish).

  • Seed production of the cherry salmon, Oncorhynchus masou (Brevoort) were studied in terms of egg development, hatching rate, juvenile growth, smolt duration, smolt rate, and adult growth rates. Fork length and body weight of $0^+$ juvenile were $9.32{\pm}1.19\;cm$ and $9.36{\pm}3.50\;g$ for females and $9.07{\pm}1.02\;cm$ and $8.57{\pm}3.04\;g$ for males, respectively. Body weights of $1^+$ smolt were $84.09{\pm}18.1\;g$ and $86.33{\pm}41.2\;g$ for females and males, while body weights of $1^+$ parr were $101.88{\pm}60.9\;g$ and $98.38{\pm}39.6\;g$ for females and males, respectively. Monitoring of gonadosomatic index (GSI) confirmed that maturations of both sexes were not synchronous; males achieved highest GSI in September, while females achieved it in October.

Survival and Physiological Response of Hybrid Grouper (Epinephleus bruneus♀ × Epinephleus lanceolatus♂) Exposed to Different Salinity Water (교잡바리(대왕자바리)의 생존과 생리학적 반응에 미치는 염분의 영향)

  • The profer salinity range for aquaculture of hybrid grouper (Epinephleus bruneus♀ × Epinephleus lanceolatus♂) were evaluated in terms of their survival, growth, feed intake and physiological responses after 30 days of exposure to different salinities (0 psu, 3 psu, 5 psu, 10 psu, 15 psu, 20 psu, 25 psu, 30 psu). Hybrid grouper all died at 0 psu salinity on the 4th day of exposure but all hybrid survived at a salinity of 5 psu or higher during the exposure period. Body length and weight increased above 5 psu salinity and the growth rate was the highest at 30 psu. Hybrid exposed to 3 psu did not eat the feed pellet. Feed intake did not show a significant difference between salinity 20 psu, 25 psu and 30 psu. Plasma osmolality of the juvenile hybrids exposed to different salinities was 341~368 mg Osmol/kg above 5 psu. The oxygen consumption rate was the highest at 30 psu with 163.6±22.3 mg O2/kg fish/h. SOD, CAT and GSH-PX were significantly higher at 15 psu. Therefore, the critical salinity for survive of the juvenile hybrid was 5 psu, and the proper salinity for optimal culture was analyzed with 20~30 psu.

  • Changes of feeding habitat and feeding organ of a land-locked sweetfish, Plecoglossus altivelis related to their growth process were investigated in the Lake Okjeong, Sannae-myeon, Jeongeup-si, Jeollabuk-do, Korea from 2004 to 2005. In the lake, the post-larvae from 25 to 40 mm in standard length fed mainly on animal organisms such as Copepoda, Branchiopoda and Rotatoria. Also, the juveniles from 50 to 60 mm (SL) fed on Chironomidae of Diptera, Copepoda and Branchiopoda. However, the juvenile from 60 to 70 mm (SL) in the Churyeong Stream fed on animal organisms and aquatic algae, the immature fishes more than 70 mm (SL) fed on only periphyton such as Synedra, Cymbella, Navicula and Pinnularia. Also, the feeding organs of the tooth and stomach were changed according to its growth and food change.

  • Growth, feed efficiency and condition factor of the olive flounder fed supplemented diets containing different levels of Obosan were significantly (P<0.05), especialluy at the optimum level of 0.6% Obosan. The hematological values (RBC count, hemoglobin, hematocrit, MCHC, MCV and MCH) were not significantly different among the flounder fed diets containing different levels of Obosan (P> 0.05). However, serum concentrations of total protein and glucose in the flounder, feds Obosan supplemented diets, were significantly higher, while the levels of total cholesterol, COT and GPT were lower than those of control fish (P<0.05)i the values for COT and GPT were only about half that of the control (P<0.05).

A Study on the Dry Matter Yield and Nutritive Values of Wild Korean Lespedeza (Lespedeza stipulacea Maxim.)

  • This study was conducted to evaluate the dry matter (DM) yield, quality and utilization of nutrients in Korean lespedeza (Lespedeza stipulacea Maxim.) swards as a roughage source. DM yield in various stages, chemical composition, and DM digestibility were observed, and DM intake, digestibility and utilization of nitrogen and energy by Korean native goat were determined. Experimental diets include three treatments: mixture hay (MH) 100%, MH (80%)+wild Korean lespedeza hay (LH) 20% and MH (60%)+LH (40%). As growth stage advanced, the DM yield of Korean lespedeza tended to increase significantly (p<0.05). The mean DM yield of all growing stages was observed as 7,336 kg/ha. Crude protein (CP) content of Korean lespedeza tended to decrease, but fibrous contents tended to increase when the growth stage advanced. The tannin content of Korean lespedeza was the highest at the bud stage (64.7 mg/g). However, it showed a tendency to decrease as the growth stage advanced, and thus it was the lowest at the ripe seed stage (26.8 mg/g) (p<0.05). With increasing level of LH, voluntary DM intake by Korean native goats slightly increased, but no differences were observed between diets. The digestibility of DM, cellular constituents and NDF was slightly higher in LH containing diets than that of MH 100% diet (p<0.05). There was no significant difference in apparently digested N% among all diets, but retained N% and retained N% of the absorbed were higher in MH (60%)+LH (40%) than those of other diets (p<0.05). A difference in the utilization of energy was not detected. In conclusion, Juvenile LH improved the digestibility of nutrients, presumably due to its high CP content and low fibrous compound. Especially, tannin in LH did not affect in DM intake, but increased the nitrogen utilization of Korean native goats. Accordingly, it could be suggested that Korean lespedeza has a potential to be provided as a roughage source for Korean native goats.

Growth, Feed Utilization and Nutrient Retention of Juvenile Olive Flounder (Paralichthys olivaceus) Fed Moist, Semi-moist and Extruded Diets

  • In an attempt to develop an artificial diet for growing olive flounder (Paralichthys olivaceus), weight gain, feed utilization and nutrient retention were investigated in fish fed moist (MP), semi-moist (SMP) and extruded pellets (EP). Excretion of nitrogen and phosphorus was also estimated based on their whole body gain and intake. EP and MP composed of raw fish and SMP made of formulated powder feed with water were prepared to have the same energy contents on a dry matter basis. A total of 240 fish with an average initial weight of 120 g were randomly distributed to each (20 fish/tank) of 12 circular plastic tanks (4 tanks/treatment) and fed experimental diets for 8 weeks. Fish groups fed EP (209 g) or SMP (209 g) recorded higher final weight than those fed MP (176 g), while dry feed consumption was highest in SMP groups (106 g), followed by MP (71 g) and EP groups (62 g). As a consequence, fish groups fed EP showed much lower feed conversion ratio than the other two groups. Protein efficiency ratio was also significantly higher in fish groups fed EP (2.55) than in those fed MP (1.44) and SMP (1.31). Fish groups fed EP, which showed the highest nitrogen retention of 43.9%, resulted in the lowest excretion of nitrogen of 35.5 g per kg gain. Also, the lowest phosphorus excretion of 6.0 g per kg gain was found in the EP groups with the highest P retention (37.0%) among treatments. Although the EP groups had the lowest dietary energy intake, they retained the highest energy in the whole body among treatments. The present results showed that EP could be more advantageous than MP or SMP in terms of growth, feed utilization and excretion of nitrogen and phosphorus for olive flounder.

Dietary Supplementation of Citrus and Fermented Citrus By-product for Juvenile Red Seabream Pagrus major at Low Water Temperature (저수온기(13-15℃) 치어기 참돔(Pagrus major) 사료 내 비타민 C 대체제로써의 감귤착즙박 및 발효감귤착즙박의 이용 가능성)

  • This study compared the effects of dietary supplementation of citrus by-product (CBP) and CBP fermented with Bacillus subtilis (F-CBP) on growth performance, feed utilization, intestinal histology and innate immunity of red seabream Pagrus major with three commercial immune-boosting products. The six experimental diets were supplemented with L-ascorbyl-2-polyphosphate (LAPP; the control diet), CBP or F-CBP at a concentration of 100 mg vitamin C equivalent/kg diet or one of three commercial immune boosters. The Experimental diets were fed to triplicate groups of 17 fish (initial body weight, 116 g) for 8 weeks. The water temperature during the feeding trial was maintained at $13-15^{\circ}C$. Growth and feed utilization did not differ significantly among the six dietary treatments, nor did the phagocytic activity, superoxide dismutase or total immunoglobulin concentrations. However, myeloperoxidase activity was significantly higher in the CBP groups. For the intestinal histology, the intestine diameter, villi and enterocyte heights and number of goblet cells did not differ significantly among groups. Therefore, CBP or F-CBP can be used as a valuable eco-friendly byproduct in diets for fishes including red seabream to maintain their normal growth and health.

Effects of Dietary Supplementation with Bacillus sp. on Growth Performance, Feed Utilization, Innate Immunity and Disease Resistance against Streptococcus iniae in Olive Flounder Paralichthys olivaceus (사료 내 Bacillus 균주의 첨가가 넙치(Paralichthys olivaceus) 치어의 성장, 사료효율, 선천성면역 및 연쇄구균 질병저항성에 미치는 영향)

  • We investigated the effect of dietary supplementation with probiotics on growth performance, feed utilization, innate immunity and disease resistance against Streptococcus iniae in juvenile olive flounder Paralichthys olivaceus. Four experimental diets were made by supplementation with three Bacillus sp. probiotics (B. subtilis, B. pumilus and B. licheniformis) into a basal diet (control) at 0.5% each. Three-hundred fish (average BW, 20 g) were randomly distributed (25 per tank) into twelve 150 L capacity polyvinyl circular tanks with three replicates per dietary treatment. Triplicate groups of fish were fed the experimental diets to apparent satiation (twice per day, 09:00 and 17:00 h) for nine weeks. After the feeding trial, a challenge test was directly conducted against S. iniae. The results indicated that dietary supplementation with the probiotic, Bacillus sp, seemed to improve growth performance of olive flounder. Hematological parameters and innate immunity as determined by nitroblue tetrazolium, myeloperoxidase or superoxide dismutase activities were not significantly affected by the probiotic supplementation. The result from the challenge test suggested that dietary supplementation with probiotics Bacillus sp. can improve the disease resistance of olive flounder against S. iniae.

Identification of 2-methylbutyric Acid as a Nematicidal Metabolite, and Biocontrol and Biofertilization Potentials of Bacillus pumilus L1

  • The present study described the isolation of 2-methylbutyric acid (2-MBA) produced from Bacillus pumilus L1, to subsequently investigate its nematicidal activity for the control of the root-knot nematode. The results showed that 2-MBA could be purified by chromatographic techniques and was identified using nuclear magnetic resonance and liquid chromatography-mass spectrometry. Crude extract and partially purified compounds had a significant effect on the inhibition of egg hatchability and second-stage juvenile (J2) mortality. A dose-dependent effect of 2-MBA was observed for J2 mortality and egg hatchability. Egg hatchability was 69.2%, 59.9%, 32.7%, and 0.0% at 125, 250, 500, and $1000{\mu}g\;mL^{-1}$ of 2-MBA after 4 d of incubation, respectively. Meanwhile, J2 mortality was in the range of 24.4%-100.0% after 2 d of incubation, depending on the concentrations of 2-MBA used. A pot experiment also demonstrated that treatment of B. pumilus L1 culture caused a significant reduction in the number of galls, egg masses, and J2 population than that of the tap water (TW) control. However, as the B. pumilus L1 culture concentration was decreased, the efficacy of nematode control by treatment of B. pumilus L1 culture was reduced compared to that of TW. B. pumilus L1 inoculation at different concentrations also promoted cucumber plant growth. Therefore, our study demonstrated the potential of 2-MBA from B. pumilus L1 as a biocontrol agent against the root-knot nematode and a plant growth promoter for cucumber plants.
