Sesquiterpenoids from the heartwood of Juniperu s chinensis

  • A new sesquiterpenoid, 11-hydroxy-valenc-1(10),3(4)-dien-2-one (3), two chemically synthesized but first isolate from nature, $3-oxocedran-8{\beta}-ol$ (1) and valenc-1(10),3(4),11(12)-trien-2-one (2) along with four known compounds, sugiol (4), (+)-nootkatone (5), 11-hydroxy-valenc-1(10)-en-2-one (6), and clovandiol (7), were isolated from the heartwood of Juniperus chinensis. All chemical structures were elucidated using extensive spectroscopic analysis including 1D and 2D NMR spectroscopy. Valenc-1(10),3(4),11(12)-trien-2-one (2) exhibited significant inhibitory activity against butyrylcholinesterase with an $IC_{50}$ value of $68.45{\mu}M$.

Fungal Endophytes Isolated from Needle Leaves of Three Coniferous Species on Mt. Seodae of Korea

  • We investigated endophytic fungi diversity of two Pinaceae species (Pinus densiflora, Pinus rigida) and one Cupressaceae species (Juniperus rigida) on Mt. Seodae, Korea. In total, 34 isolates were obtained from 19 host plants and identified according to internal transcribed spacer (ITS) region. As a result, they were placed in 13 taxa. Among them, 52.9% belonged to Leotiomycetes, 32.3% belonged to Sordariomycetes, 5.8% belonged to Agaricomycetes, and 3.0% for isolates belonged to either Eurotiomycetes, Dothideomycetes or Ustilaginomycetes. In particular, Lophodermium growing in pine would be an especially important instance of biodiversity for endophytes, suggesting that further study examining its ecological function in the environment is critical.

  • 향나무류를 가해하는 향나무알락명나방(신칭)을 우리나라 미기록종으로 보고하며 성충 및 유충의 외부형태적 특징을 간략히 기술하였다. 금번조사를 통해 향나무, 누운향나무, 옥향나무 등 3종이 기주로 확인되었으며, 유충은 기주의 잎을 엮고 그 안에서 가해하는 것으로 조사되었다.

Diversity of Endophytic Fungi Isolated from Pinus densiflora and Juniperus rigida Distributed in Mt. Baekryeonsan and Mt. Johangsan, Korea

  • We investigated the biodiversity of endophytic fungi in Pinus densiflora and Junipreus rigida from Mt. Baekryeonsan and Mt. Johangsan in Korea. In total, 59 isolates were discovered from 26 host plants and identified using sequences from the rDNA internal transcribed spacer (ITS) region. Twenty taxa of endophytic fungi were identified in this study; 62% of isolates belonged to Leotiomycetes, 34% belonged to Sordariomyetes, and 4% belonged to Agaricomycetes. Results confirmed three previously unreported endophytic fungi in Korea: Phanerochaete concrescens, Creosphaeria sassafras, and Scopuloides rimosa. Community analysis showed that the fungal diversity patterns of the samples from Mt Baekryeonsan and Mt. Johangsan are similar to those from previous studies and varied depending upon host and locality.

Estimation of Strength Loss and Decay Severity of Juniperus procera by Juniper Pocket Rots Fungus, P. demidoffii in Ethiopian Forests

  • A juniper pocket rot fungus, Pyrofomes demidoffii is a basidiomycetous fungus responsible for damage of living Juniperus spp. However, its effect on the residual strength and on the extent of decay of juniper's trunk was not determined in any prior studies. The purpose of this study was to study the features of J. procera infected by P. demdoffii, and to estimate the level of strength loss and decay severity in the trunk at D.B.H height using different five formulas. Infected juniper stands were examined in two Ethiopian forests through Visual Tree Assessment (VTA) followed by a slight destructive drilling of the trunk at D.B.H height. The decayed juniper tree is characterized by partially degraded lignin material at incipient stage of decay to completely degraded lignin material at final stage of decay. In the evaluated formulas, results of ANOVA showed that a significantly higher mean percentage of strength loss and decay severity were recorded in the trees of larger D.B.H categories (p<0.001). The strength loss formulas produced the same to similar patterns of sum of ranks of strength loss or decay severity in the trunk, but the differences varied significantly among D.B.H categories in Kruskal Wallis-test (p<0.001). In conclusion, the employed formulas showed similar to different degree of variability in quantification of strength loss or decay severity in the trunk. The findings of our study could be used as the baseline for further study on juniper's strength loss or decays in the trunk of Juniperus spp. and unequivocally helps to design the corresponding management as result of P. demidoffii.

향나무 추출 색소의 특성 및 염색성 (Characteristics and Dyeability of Juniperus Chinensis Extracts)

  • The purpose of this study was to investigate the characteristics of the pigment and the dyeability of juniperus chinensis needles, berry, bark and heartwood extracts using distilled water, methanol, normal butanol and ethyl acetate as a solvent. Ultraviolet-visible absorption spectrum bands of the extracts were measured at around 280nm to 320nm in all the solvent extracts. The maximum absorption wavelength was able to determine tannin. All the solvent extracts except for distilled water extracts were able to confirm the presence of chlorophyll. Infrared absorption spectra (FT-IR) of all parts of the extracts showed broad absorption bands of OH due to phenolic-OH, benzene CH peak of phenol chemical structure, ether-based stretching vibration peak and the peak of flavonoid compounds that appeared in all the solvent extracts. The yield of juniper needles and heartwood in distilled water and methanol extraction were effective. Extraction of berry yields in distilled water was also effective. The yield of ethanol extraction from the bark showed better efficiency. As a result of using distilled water for the dye solution, the color of the fabrics dyed with all extracts of the needles and Y series berries generally showed light Y progression with a strong red tinge. By using a mixture of 20% ethanol and 80% distilled water for the dye solution, the color of the fabrics dyed with needles and berry extracts showed Y series dominantly. The color of the fabrics dyed with Bark and heartwood extracts were dominantly R series.

엽록체 DNA matK와 psbA-trnH 염기서열에 기초한 한국산 향나무절(향나무속) 식물의 분자계통학적 연구 (Molecular phylogenetic study of section Sabina (Genus Juniperus) in Korea based on chloroplast DNA matK and psbA-trnH sequences data)

  • 한국산 향나무절 식물에 대한 계통학적 유연관계를 규명하고, 더불어 향나무속 및 향나무절의 계통 및 유연관계를 잘 나타낼 수 있는 분자마커를 찾아내고자 분자계통학적 연구를 수행하였다. 엽록체 DNA matK와 psbA-trnH를 분자마커로 활용하였으며, 두 유전자의 조합분석결과 향나무절이 100%의 BP로 지지되는 분계조를 이루었다. 눈향나무+단천향나무 분계조와 향나무+섬향나무+뚝향나무 분계조는 각각 91%, 100%의 BP로 지지되었다. 따라서 한국산 향나무절은 (1) 눈향나무+단천향나무, (2) 향나무+섬향나무+뚝향나무 두 개의 분계조로 구분하는 것이 가장 적합할 것으로 생각되고, 본 연구에서 이용된 두 개의 분자마커 중 matK가 psbA-trnH 보다 향나무속 및 향나무절의 계통 및 유연관계를 규명하는데 다소 높은 해상력을 나타내었다.

Growth-inhibiting Effects of Juniperus virginiana Leaf-Extracted Components toward Human Intestinal Bacteria

  • The growth responses of materials extracted from Juniperus virginiana leaves against Bifidobacterium bifidum, B. longum, Clostridium perfringens, Escherichia coli, Lactobacillus acidophilus, L. casei, and Streptococcus mutans were examined using impregnated paper disk agar diffusion. The biologically active constituent isolated from the J. virginiana extracts was characterized as ${\alpha}$-cedrene using various spectroscopic analyses including IR, EI-MS, and NMR. The responses varied according to the dose, chemicals, and bacterial strain tested. Methanol extracts of J. virginiana leaves exhibited a strong and moderate inhibitory activity against C. perfringens and E. coli at 5 mg/disk, respectively. However, in tests conducted with B. bifidum, B. longum, L. acidophilus, L. casei, and S. mutans, the methanol extracts showed no or weak inhibitory response. At 2 mg/disk, a-cedrene strongly inhibited the growth of C. perfringens and moderately inhibited the growth of E. coli and S. mutans, without any adverse effects on the growth of four lactic acid-bacteria. Of the commercially available compounds originating from J. virginiana leaves, cedrol and ${\alpha}$-pinene exhibited strong and moderate growth inhibition against C. perfringens, and ${\alpha}$-copaene revealed moderate growth inhibition against E. coli at 1 mg/disk. Furthermore, cedrol exhibited moderate and weak growth inhibition against S. mutans at 2 and 1 mg/disk, respectively. However, little or no activity was observed for camphene, (+)-2-carene, p-cymene, limonene, linalool, and a-phellandrene against B. bifidum, B. longum, C. perfringens, L. acidophilus, L. casei, and S. mutans at 2 mg/disk. The observed inhibitory activity of the J. virginiana leaf-extracted materials against C. perfringens, E. coli, and S. mutans may be an indication of at least one of the pharmacological actions of the J. virginiana leaf.

단양 석회암지역의 식물상에 관한 연구 (A Study on the Flora in Tanyang Limestone Area)

  • Biomass and soil properties of plant communites was investigated in the limestone ares, Tanyang, Ch'ungbuk Province in Korea. Plants was classified in calcicoles and calcifuges within plant communities occurring in the limestone area. Biomass of plant populatins in the limestone area was measured that Stellaria aquatica was the lowest(0.58g/plant), Chrysanthemum boreale was the highest(8.87g/plant) and that Pennisetum alopecuroides, Miscanthus sinensis was 6.67g/plant, 5.76g/plant, respectively. Diameter of breast height among the tree layer communities was investigated that Pinus densiflora was the widest(20cm), Juniperus rigida was the narrowest(6cm) and that Quercus aliena, Populus ${\times}$ tomentiglandulosa was 10cm, 12cm, respectively. The high of density in the tree layer was found out Populus ${\times}$ tomentiglandulosa, Juniperus rigida, Quercus aliena, Pinus densiflora in this order. Dominant species in the study area were Pinus densiflora, Quercus aliena, Juniperus rigida, Quercus dentata in the tree layer, Lespedeza bicolor, Buxus microphylla, Rhus chinensis in the shrub layer, Humulus Japonicus, Erigeron canadensis, Xanthium strumarium, Oenothera odorata Persicaria hypropiper on the riverside, Pennisetum alopecuroides, Vicia amoena, Chrysanthemum boreale on the roadside. Soil properties of the limestone area was investigated around the protrusion of rock in the upper region, around the cave and underlayer accumulated by corrosion in the study area. Lime-chlorosis was observed in the upper region, underlayer and around the cave. It was observed that biota of kum-gul and dungbong-gul was Rhinolophus ferrumequinum korai, Epanerchodus kimi, Antrokoreana gracilipes, Diestrammena japonical. Dominant species were Humulus japonicus, Stellaria aquatica, Hydrocotyle maritima, Phragmites communis, Miscanthus sinensis around the inlet of a kum-gul. It was classified that plants in the study area was 34 order, 53 family, 135 species in all. Calcicoles were 11 order, 16 family, 18 species and calcufuges were 6 order, 8 family, 11 species of them all.
