Three lots of mejues were prepared with three different strains of Aspergillus oryzae, Bacillus natto, Bacillus subtilis and one was made by conventional method. The four different soy bean pastes were analyzed for compositions of free amino acids, sugars and organic acids during the period of fermentation. Amino nitrogen contents in the samples of A. oryzae were higher than others throughout the aging period. The amounts of each amino acids were varied markedly among the samples after 20-days, while glutamic acid, aspartic acid, lysine and phenylalanine were dominant in all samples after 90-days. Glucose contents were found to be in the range of $0.46{\sim}2.66%$ and other sugars of fructose, sucrose, rhamnose and maltose were less than 0.35%. The levels of total free sugar were relatively higher in the samples prepared with B. natto than others. Citric, lactic, malic, acetic and oxalic acids were identified, and the content of lactic acid was higher in the samples of A. oryzae, whereas citric acid was higher in conventional method.

Successful cryopreservation of bovine immature oocytes can increase availably of oocytes for the in vitro fertilization or nuclear transfer. However, it was not reported successful development to the blastocyst stage following in vitro fertilization of cryopreserved bovine immature oocytes. The objective of this study was to determine the incidence of survival, meiotic maturation, fertilization and in vitro development of cryopreserved bovine immature by ultra rapid cooling methods. The oocytes were adversely affected by brief exposure to EFS40 solution in electron microscope grids and plunged directly into liquid nitrogen. After such ultra-rapid cooled immature oocytes were warmed, 78% of oocytes were matured to the metaphase II stage, 50% of oocytes were fertilized after insemination, and 5% of oocytes were developed to the blastocyst stage. Different sodium concentration of sodium ion in the freezing medium did not affect survival, maturation, fertilization and in vitro development of cryopreserved oocytes. These results suggested that immature bovine oocytes can be cryopreserved by ultra-rapid cooling methods.
This study was carried out to find heavy metal contents in soil and in native plant in the regions near by zinc-mines, located at the Chilgok and Uljin Gun in Kyeongpook area. In the heavy metal contents during the to growth of native plants, Cd was contained in the order perennial > biennial > annual plant, but Cu was annual > perennial > biennial plant, and there was no difference in Zn contents. The native plants contained heavy metal highest were Osmunda japonica in Cd, Persicaria thunbergii H.G in Cu and Equisetum arvense L. in Zn. Cd was contained highest in Pteridaceae, Cu in Equisetaceae and Zn in Polygonaceae. In the heavy metal contents by the part of plant of Equisetum arvense L. and Erigeron canadensis, Cd and Zn were much contained in the order leaf > stem > root, but Cu was in the order root > leaf > stem. The average contents of Cd, Cu, Zn in soil were 1.27ppm. 12.04ppm. 64.28ppm in Chilgok, and 3.30ppm. 72.93ppm. 194.04ppm in Uljin respectively. There were positive correlations between Cd contents of heavy metals(Cd, Cu, Zn) in soil and in native plant, but not significant. It was estimated that Osmunda japonica, Pteridium aquilinum (KUHN) var and Equisetum arvense L. which most absorbed heavy metal have effect of exclusion of heavy metals near by zine-mines region.

Diffusion processes in tidal swinging coastal waters are studied by releasing one hundred liters of Rhodamine B four times. Dye patches were formed from instantaneous point sources. The patches were sampled with a pump on a tracking boat, and samples were analyzed with fluorospectro-photometer. The patterns of patches were reconstructed and their characteristics were analyzed in terms of variance of concentration, area estimation, and decrease rate of peak concentration. In all of the four experiments, the dye patches were mos시 elongated to the direction of current axis. the elongation rate was 0.34 on the average. Apparent diffusivities were 620 to 3,000 cm super(2) /sec during initial period of 90 minutes. The variance increased by exp(0.047t) on the average, and peak concentration deceased by exp(-0.044t) on the average.
Gas chromatogram of standardized samples for 10 different kinds of polyphenol components, which contained universally in common vegetables and fruits, and those of polyphenol extracts from various ginseng products and Acanthopanax, were revealed, respectively. The consequent results are as follows; 1. There were practically no obvious difference in the polyphenol pattern among white ginseng with skin of either Korean, American, Canadian products, or Korean red ginseng. There was, however, no coincidence in $t_R$ as indicated by peaks of polyphenol pattern for these ginseng products with those expressed by the standard samples. 2. A great similarity existed in the polyphenol pattern between white ginseng and red ginseng extracts, but the number of peaks for the ginseng extracts was far less than dried ginseng. 3. The polyphenol patterns of Russian and Korean Acanthopanax showed a similarity. However, the polyphenol pattern as represented by Acanthopanax was considerably different from that of ginseng products, especially chlorogenic acid which was not present in the ginseng products was identified in Acanthopanax.
This study was attemtped to improve the conventional processing method and to establish the basic data for evaluating the product suitabilities of dried loach. The semidressed raw material were salted in 15% NaCl solution for 15 minutes and dried to contain about 44% of water, and then heated about 10 minutes at $80^{\circ}C$ controlled by microwave (2,450 MHz). The moisture content of monolayer value for the product showed 5.34% and its water activity was 0.28. The optimum relative humidity for the storage was recognized to be from 32% to 44%. The average shelf life around the year of the Nylon-PVC-PE $(40\;{\mu})$ packed product in domestic circulation market was estimated as 207.4 days. In comparison with raw material, the contents of the major amino acids, glutamic acid, alanine and valine in the product were shown to be slightly increased, while the level of lysine, aspartic acid and methionine were slightly decreased. The contents of saturated fatty acids and oleic acid were shown to be slightly increased, while the other fatty acids tended to be slightly decreased.
Four kinds of polyamicacids(PAAs) were prepared by the condensation reaction of four diamines with different linkages, 3,3'-diaminodiphenyl sulfone(3,3'-$DDSO_2$), 4,4'-diaminodiphenyl sulfone(4,4'-$DDSO_2$), 4,4'-methylene dianiline(4,4'-MDA) and 4,4'-oxydianiline(4,4'-ODA), and dianhydride, 3,3', 4,4'-benzophenone tetracarboxylic dianhydride (BTDA) using the solvent, dimethylacetamide(DMAc). These four PAAs were blended with poly[2,2-(m-phenylene)-5,5'-bibenzimidazole](PBI) from the solution blending. Then called as Blend-I, II, III, and IV, respectively. Cast films or precipitated powders of the PBI/PAA blends were cured at a higher temperature than expected Tg to transform into PBI/PIs blends. Miscibility, specific intermolecular interaction for miscibility and their relative strength as a function of polyimide chemical structure with different four diamines in the PBI/PI systems were investigated. Four blends used in this study were all miscible, and the specific intermolecular interactions existing in these blends was thought to be the hydrogen bonding between the N-H of PBI and the C=O of PIs. The hydrogen bonding in the blends were shown to be stronger in the Blend-III and Blend-IV than Blend-I and II. It is speculated that the differences of hydrogen bonding strength of PBI/PI blends are dependent upon chemical structures of PIs, that is, PIs consisting of $SO_2$ group have a weaker hydrogen bonding strength than those of O or $CH_2$ group because the former has a larger spacer than the latter.
To extract insoluble proteins and to improve functional properties of abolished proteins, a protease producing Aspergillus sp. MS-18 was isolated from soil. The enzyme was purified and its enzymological characteristics were investigated. It was found that production of protease reached to the maximum when the wheat brae medium containing, 3% arabinose, 0.5% polypepton, 0.1% $(NH_4)_2SO_4$ and 0.2% magnesium chloride was cultured for 3 days. Protease was purified 16.9 folds after ion exchange chromatography and gel filtration and the specific activity was 340.4 unit/mg. Purified enzyme was confirmed as a single band by the polyacrylamide gel electrophoresis. The molecular weight of protease was estimated to be 30,000. Crystalization form of purified protease was a stick shape with rounding edges. The optimum pH and temperature for the protease activity were 9.0 and $60^{\circ}C$, respectively. The enzyme was stable in pH 7.0-12.0 at $50^{\circ}C$. The activity of purified enzyme was inhibited by $Hg^{2+}$, $Cu^{2+}$, $Zn^{2+}$ and $Pb^{2+}$, whereas it was activited by $Na^+$, $Mg^{2+}$ and $Mn^{2+}$. The activity of the protease was inhibited by the treatment with ethylenediaminetetraacetic acid and phenylmethane sulfonyl fluoride. The result suggests that the purified enzyme is a serine protease with metal ion at active site. Km and Vmax of purified protease were $29.33\;{\mu}mole/L$ and $5.13\;{\mu}g/min$, respectively.
A carotenoprotein from the muscle of Ascidian (Halocynthia roretzi) was purified by ion exchange chromatography and gel filtration chromatography, and analyzed molecular weight, stability of pH and heat, effect of denaturing agents, amino acids and fatty acids composition. The purified carotenoprotein had absorption maxima of 463 nm and 439 nm. The carotenoprotein had an approxmimate molecular weight of 64.4 kDa in polyacrylamide gel electrophoresis. The amino acid compositions of carotenoprotein were mainly Gly (15.39%), Asn (11.31%), Gln (10.62%) and Ser (13.35%). The major fatty acids composition of carotenoprotein were $C_{16:1(n-7)}\;(15.4%)$, $C_{22:1(n-9)}\;(14.5%)$ and $C_{20:1(n-11)}\;(11.4%)$. The monounsaturated fatty acids (45.2%) contained abundant content compared to other saturated (38.1%) and polyunsaturated (11.7%) fatty acids.
