NMDA (n-methyl-d-aspartate) Change Expression Level of Transcription Factors (Egr-1, c-jun, Junb, Fosb) mRNA in the Cerebellum Tissue of Balb/c Mouse (NMDA투여에 의한 transcription factor (Egr-1, C-Jun, JunB, FosB)의 발현 변화 양상)

  • Glutamate is one of the principle transmitters in the CNS. Ionotropic receptors of glutamate, selectively activated by N-methyl-D-aspartate (NMDA), play an important role in the processes of cell development, learning, memory, and etc. On the other hand, many studies discovered that over-activation of glutamate receptors leads to neurodegeneration and are known to be implicated in major areas of brain pathology. Any sustained effect of a transient NMDA receptor activation is likely to involve signaling to the nucleus and to trigger coordinated changes in gene expression. Classically, a set of immediate-early genes are induced first; some of genes are by themselves transcription factors that control expression of other target genes. This study provides understanding of changes of inducible transcription factors mRNA levels with RT-PCR by inducing over-activation of NMDA receptor with intraperitoneal NMDA injection. The experimental conditions were varied by 1, 5, 25, and 125 g/ of body weight NMDA and measured transcription factors mRNA levels are Egr-1, c-Jun, JunB, and FosB. Based on result obtained, inducible transcription factors mRNA in NMDA injection to mice with 5 g/body weight showed the greatest change. And ITF mRNA showed greatest change 24 hr after injection. The expression level of JunB mRNA was markedly changed. Up to the present days, no study clearly understood how ITF mRNA affected the apoptosis of purkinje cells in the cerebellum. The current study improves the understanding of the mechanism of apoptosis of purkinje cells in the cerebellum.

The Expressive Characteristics of Itami Jun's Interior Space Seen from the Viewpoint of 'Theory of Meeting' ('만남이론' 관점으로 본 이타미 준의 실내공간 표현특성)

  • Korean-Japanese architect Itami Jun that proceeded with primeval architecture based on analogue ideas in the digital era realizing nomad architecture is delivering a totally different meaning in the modern time. Not like the Japanese architectural community equipped with rich technologies showed state-of-the-art architecture after 1980s, Itami Jun' formative consciousness intending to assimilate nature from the original position of things is thought to have started from the relationship with Korean-Japanese painters leading MONO-HA desiring to look at the world as it is and approach the source of existence. The purpose of this study is to analyze the spatial characteristics from the viewpoint of 'Theory of Meeting' which is the essence of the work theory of Lee UFan that made the theoretical foundation of MONO-HA to understand Itami Jun's architectural space in the ideological aspect. Although the characteristics of marginal men appearing in Itami Jun's architectural space as others located on the boundary between Korea and Japan, art and architecture, and functions are valuable research objects, they have not been studied in the aspect of source, so it is needed to research his works and the source of the spatial ideas that led his works. Based on Theory of Meeting claiming that the world becomes the place of experience by the medium of objet which is the structure of meeting, and that human beings can meet the truly opened world by self-awareness through body, the expressive characteristics of Itami Jun's interior space was analyzed. As a result of the analysis, it turned out that Itami Jun's interior space actively expressed Theory of Meeting on the unity of structure and space, and on the trans-boundary by the medium of objet. Conclusively, this study suggests that, in the reality of the modern interior design full of technology-intensive designs, analogue approach moving in the opposite way from technologies could be a better expressive method of design delivering the fundamental human emotions.

Effect of Dietary Capsaicin on Proto-oncogenes Expression in Various in Mice (식이 Capsaicin이 마우스의 주요 장기조직에서의 Proto-oncogenes Expression에 미치는 영향)

  • Capsaicin (8-methyl-N-vanillyl-6-nonenamide: CAP) is a mai or ingredient of hot pepper that has been used as a spicy food additive, preservative, and medicine. In this study, we evaluated the effect of dietary CAP on the selected proto-oncogene(c-jun, c-myc, H-ras, erbB, p53) expressions in various tissues of mice. Male ICR mice were divided into four groups and fed the experimental diets containing CAP at the levels of 0, 5, 20 and 100ppm for four weeks. Steady state RNA levels in various tissues were measured by slot blot hybridization assay. C-jun expression level was enhanced in stomach tissue from mice fed 20ppm CAP and significantly reduced from mice fed 100ppm CAP. The c-jun expression levels were differentially altered in organ-specific manner, Tumor suppressor gene p53 expression level appeared to be slightly increased in the liver from mice fed 20ppm CAP. These results suggested that dietary CAP differentially modulates c-jun and p53 expression in various organs.
