돼지감자 부위별 메탄올 추출물의 항산화 및 항당뇨 활성 (Antioxidative and Antidiabetic Activities of Methanol Extracts from Different Parts of Jerusalem Artichoke (Helianthus tuberosus L.))

  • 본 연구에서는 돼지감자 꽃, 잎, 뿌리가 가진 기능성 식품 소재로서의 활용도를 높이고자 다양한 생리활성을 평가하였다. 돼지감자 꽃, 잎, 뿌리의 폴리페놀 함량은 32.80 mg GAE/extract g, 43.46 mg GAE/extract g, 24.53 mg GAE/extract g으로 나타났으며, 플라보노이드 함량은 각각 15.64, 18.67, 11.65 mg GAE/extract g으로 나타났다. 돼지감자 꽃, 잎, 뿌리의 메탄올추출물의 DPPH 라디칼 소거능 활성은 각각 77.78%, 66.07%, 26.93%로 나타났으며, hydroxyl radical 소거능 활성은 각각 55.33%, 38.30%, 34.89%로 나타났다. 돼지감자 꽃, 잎, 뿌리메탄올 추출물의 아질산염 소거능을 측정한 결과, 돼지감자 추출물의 농도가 증가함에 따라 그 활성 역시 증가하는 경향으로 나타났다. 돼지감자 부위별 메탄올 추출물(5 mg/mL)의 ${\alpha}-glucosidase$ 저해활성은 꽃, 잎, 뿌리에서 각각 41.08%, 55.67%, 60.76%로 유의적인 차이를 나타났으며, ${\alpha}-amylase$ 저해활성은 꽃, 잎, 뿌리에서 각각 49.95%, 59.88%, 66.25%로 유의적인 차이를 나타내었다.

  • Two flavone glucosides have been isolated from the leaves of Helianthus tuberosus (jerusalem artichoke). Their structures were identified as kaempferol 3-O-glucoside (1) and quercetin 7-O-glucoside (2) by spectroscopic analysis and confirmed by comparison with reported data. These flavonoids were isolated for the first time from this plant part.

  • 돼지감자 잎의 활용 및 생리활성을 증가시킬 수 있는 적정 추출방법을 알아보고자 환류냉각, 상온교반, 가압가열 및 저온고압 추출법을 이용하여 추출한 돼지감자 잎 추출물의 생리활성을 비교하였다. 추출수율은 가압가열추출, 환류냉각추출, 상온교반추출, 저온고압추출 순으로 높은 수율을 나타내었다. 폴리페놀 함량은 상온교반추출 및 환류냉각추출의 경우 대등한 함량을 나타낸 반면 가압가열추출 및 저온고압추출에서는 유의적으로 높은 함량을 나타내었으며, 플라보노이드 함량은 가압가열추출 및 저온고압추출에서 높은 함량을 나타내었다. Proanthocyanidin 함량에서는 저온고압추출에서 가장 높은 함량이 검출된 반면 가압가열추출에서는 상온교반 및 환류냉각추출에 비해 낮은 함량을 나타내었다. 항산화 활성에서는 모든 추출물이 농도가 증가함에 따라 활성은 비례적으로 증가하였다. 가압가열추출 및 저온고압추출에서 유의적으로 높은 활성을 나타내었으며, ${\alpha}$-glucosidase, xanthine oxidase 및 angiotensin I-converting enzyme 저해활성에서는 저온고압추출물에서 우수한 활성을 나타내었다. 이러한 결과를 종합해 볼 때 가압가열 및 저온고압추출물이 소재 활용가치가 높을 것으로 사료되며 천연 항산화제 및 기능성 증진을 위한 소재로 이용 가능할 것으로 판단된다.

Extraction and Bioassay of Allelochemicals in Jerusalem Artichoke

  • Helianthus tuberosus has been known to inhibit the growth of weeds and other plants sharing its habitat. This study was conducted to identify the allelochemicals of Helianthus tuberosus which were extracted with water and solvents. Aqueous extracts of leaf, stem, root, tuber and tuber peel of Helianthus tuberosus except tuber did not show significant differences in phytotoxicity to alfalfa seedlings. It was considered that Helianthus tuberosus contained fewer or less potential water-soluble substances that were toxic to alfalfa. Methanol extract of leaves of Helianthus tuberosus was sequentially partitioned in increasing polarity with n-hexane, ethylacetate and n-butanol. Each extract had a yield of 148, 12, 15.7 and 9.5g, respectively. Inhibitory effects on germination of alfalfa seeds treated with four fractions were not significantly different. But the significant reductions on hypocotyl length were observed for all the solvent extracts. Among the four fractions, the ethylacetate fraction showed the most significant inhibition effect on bioassay with alfalfa. Further separation of the active ethylacetate fraction by open column chromatography led to the 25 subfractions. In bioassay of each sub-fraction with alfalfa seeds, sub-fraction No. 13 showed the most inhibitory effect on seedling growth. $^1$H NMR and gas chromatography-mass spectrometry analysis revealed that sub-fraction No. 13 was the mixture of straight-chain saturated fatty acids.

감마선 조사를 이용한 시판 분말원료의 위생화 (Sanitization of Commercial Powdered Products Using Gamma Irradiation)

  • Microbiological populations and the sterility of commercial powdered products treated with gamma irradiation at 0~10 kGy were investigated before using them as ingredients for a non-cooked Saengsik product. We evaluated a total of 14 powdered products: 8 powdered cereals, 3 powdered tubers, and 3 powdered leafy vegetables. The total numbers of bacterial populations in non-irradiated powdered cereals, tubers, and leafy vegetables were 2.7~6.9, 5.6~6.0, and $5.3{\sim}6.8\;log\; CFU{\cdot}g^{-1}$, respectively. Moreover, coliform bacteria were not indicated in adlay, millet, germinated brown rice, soybean, and mulberry leaves powder within detection limit ($2.0\;log\; CFU{\cdot}g^{-1}$). The number of Bacillus cereus exceeded $3.0\;log\; CFU{\cdot}g^{-1}$ (the maximum limit for Saengsik products) in all samples, excluding perilla seeds, buckwheat, barley, oat, potato, and Jerusalem artichoke powder. However, a dose of 6 kGy of gamma irradiation reduced the microbiological populations in all samples, and all the powdered products met the microbial requirements for Saengsik products. Futhermore, it was confirmed that all microorganisms in the 9 powdered products, except fermented brown rice, sweet potatoes, and 3 leafy vegetables, were sterilized by 10 kGy of gamma irradiation.
