Determination of Fire Blight Susceptibility on Wild Rosaceae Plants in Korea by Artificial Inoculation (인공접종을 통한 국내 야생 장미과 식물의 화상병 감수성 검정)

  • The fire blight caused by Erwinia amylovora (Ea) is a devastating disease of Rosaceae plants, including commercially important apple and pear trees. Since the first report in Korea in May 2015, it has been spreading to neighboring regions gradually. Host plants can be infected by pollinators like bees, rainfall accompanied by wind, and cultural practices such as pruning. Many studies have revealed that wild Rosaceae plants such as Cotoneaster spp., Crataegus spp., Pyracantha spp., Prunus spp., and Sorbus spp. can be reservoirs of Ea in nature. However, wild Rosaceae plants in Korea have not been examined yet whether they are susceptible to fire blight. Therefore, the susceptibility to fire blight was examined with 25 species in 10 genera of wild Rosaceae plants, which were collected during 2020-2022, by artificial inoculation. Bacterial suspension (108 cfu/ml) of Ea type strain TS3128 was inoculated artificially in flowers, leaves, stems, and fruits of each plant species, and development of disease symptoms were monitored. Moreover, the presence of Ea bacteria from inoculated samples were checked by conventional polymerase chain reaction. Total 14 species of wild Rosaceae plants showed disease symptoms of fire blight, and Ea bacteria were detected inside of inoculated plant parts. These results suggest that wild Rosaceae plants growing nearby commercial apple and pear orchards in Korea can be Ea reservoirs, and thus they should be monitored regularly to minimize the damage by Ea infection and spreading.

Biochemical Composition of Marine Microalgae and Their Potential Antimicrobial Activity

  • This study is to investigate biochemical compositions of two species of marine microalgae, Chlorella ellipsoidea of Chlorophyta and Tetraselmis suecica of Prasinophyta, and to assess their potential antimicrobial activities. Crude protein, lipid and carbohydrate for C. ellipsoidea were $43.15\%$, $12.63\%$ and $13.09\%$, respectively, and those for T. suecica were $44.95\%$, $4.80\%$ and $24.05\%$, respectively. The major amino acids of the two micro algae were aspartic acid, glutamic acid, glycine, alanine, valine, leucine, lysine and proline, and no significant difference between the amino acid compositions of both micro algae was observed. The major sugars for both microalgae were glucose, galactose and mannose, and glucose contents showed the highest level, $58.70\%$ for C. ellipsoidea and$57.86\%$ for T. suecica. The major mineral contents of both micro algae for 100g were Ca (3,114mg in C. ellipoidea and 3,389mg in T. suecica) and followed by Na (2,881mg), K (548mg) and Mg (545mg) for C. ellipsoidea and Na (1,832 mg), Mg (1,510mg) and K (548mg) for T. suecica. In the content of ATP-related compound, hypoxanthine in C. ellipsoidea and IMP in T. suecica were absolutely dominant compounds. The highest content of fatty acid in C. ellipsoidea was 20:4, $27.15\%$ and that in T. suecica was 18:3 (w-6), $18.10\%$. In case of physiologically important polyunsaturated fatty acids like eicosapentaenoic acid (20: 5) and docosahexaenoic acid (22: 6), both microalgae possessed just trace amounts but was rich in arachidonic acid (20: 4). Vitamin content in both microalgae was significantly high in choline and inositol. In antimicrobial activity by water- and fat-soluble fraction of the micro algae, hexane extract in the fat-soluble fraction of C. elliposidea inhibited the growth of Bacillus subtilis by $96\%$ bactericidal activity and tetrachlorocarbon extract of T. suecica indicated relatively excellent antimicrobial activity $(81\%\;bactericidal\;activity)$ against Escherichia coli. Hot water extract among water-soluble fraction of both micro algae almost suppressed the growth of Staphylococcus aureus by $96\%$ bactericidal activity.

  • To clarify the toxic effect of oxidative stress, hydrogen peroxide $(H_{2}O_{2})-induced$ neurotoxicity was examined in cultured newborn mouse spinal motor neurons after spinal motor neurons were grown in the media containing various concentrations of glucose oxidase (GO). And also, the protective effect of Rhizoma gastrodiae extract against GO-induced neurotoxicity was evaluated. Cytotoxicity was expressed as a cell viability by 3-(4,5-dimethylthiazol-2-yl) -2,5-diphenyltetrazolium bromide (MTT) assay. In this study, exposure of motor neurons to GO-induced cell death significantly, in a dose- and time-dependent manners in spinal motor neuron cultures. The decrease in cell viability of motor neurons damaged by GO was proventioned by Rhizoma gastrodiae extract. These results suggest that the neuroprotective effect of Rhizoma gastrodiae extract on GO-induced neurotoxicity may result from a attenuation of $H_{2}O_{2}-induced$ oxidative stress.

The Effect of Exercise Training (EXE) on Myocardium Glucose Metabolic Phenotypic Proteins and HSP-60 Protein Expression after Ischemia/Reperfusion Injury in STZ-induced Rats (지구성 운동이 STZ-당뇨 유발 쥐의 허혈/재 관류 손상 후 심근의 당대사 관련 표현형 단백질과 HSP-60 단백질 발현에 미치는 영향)

  • The objective of this study was to identify EXE (1 hr a day at 21 m/min for 5 day/wk, at 0 % grade for 6 wk) on myocardium glucose metabolic phenotypic proteins (AMPK-PGC-1${\alpha}$-GLUT-4) and HSP-60 protein expression after ischemia/reperfusion injury (IRI) in STZ-induced rats. EXE was performed using STZ-induced diabetic rats on a rodent treadmill (28 m/min, 1 hr/day, 5 day/wk for 6 wk). The results of this study suggest that i) serum insulin level was not changed among groups (p>l0.05). ii) the LVDP level increased significantly in the STZ-EXE-IRI group compared to the STZ-IRI group at 60 min (p<0.01), 70 min (p<0.05) and 80 min (p<0.05) after reperfusion, respectively, and iii) AMPK phosphorylation (p<0.01), PGC-1${\alpha}$ protein (p<0.001), GLUT-4 protein (p<0.001) and HSP-60 protein expressions (p<0.05) increased significantly in the STZ-EXE-IRI group compared to the STZ-IRI group. In conclusion, the findings of the present study reveal that EXE may provide therapeutic value to insulin dependent diabetic patients with peripheral insulin resistance and myocardium injury by improving glucose metabolic proteins (AMPK-PGC-1${\alpha}$-GLUT-4) and heat shock protein-60 (HSP-60), along with increasing LVDP levels and decreasing glucose levels. Therefore, EXE protects the STZ-induced diabetic myocardium injury against ischemia/ reperfusion injury.

  • For preparation of high-quality salted mackerel using enzymatic extracts from Ecklonia cava (EEC) to process fresh mackerel caught off Jeju Island, the optimal concentration of EEC was investigated. There were no differences in proximate composition, salinity, Escherichia coli level, pH, volatile basic nitrogen content, histamine level, or peroxide value between salted mackerel samples prepared with different concentrations of EEC. However, the antiradical properties of salted mackerel, assayed by scavenging of DPPH free radicals, hydroxyl radicals, hydrogen peroxide, and alkyl radicals, increased with increasing concentrations of EEC. The optimal concentration of EEC for preparation of high-quality salted mackerel was 2% (w/w). Salted mackerel soaked in 2% (w/w) EEC was superior to commercial salted mackerel in antiradical properties, biogenic amine content, and other relevant chemical properties.

Food Quality Characterizations of Commercial Salted Mackerel (시판 간고등어의 식품학적 품질 특성)

  • This study was conducted to obtain basic data for developing a special salted mackerel. For this purpose, food quality characterization data on 11 kinds of salted commercial mackerels were gathered. Korean Industrial Standards (KSH 6029) stipulate that a salted mackerel should be less than $1.0{\times}10^6\;CFU/g$ in viable cells, negative for Escherichia coli, less than 50 mg% for volatile basic nitrogen (VBN) and less than 3% for salinity. Only one sample (code 10) among the 11 kinds of commercial salted mackerels is believed to posses acceptable limits according to KSH 6029. The others except code 2 and 4 showed less than 50 mg/kg in histamine content, a safe range for allergies. The peroxide values of 4, 5, 7, 10 and 11 in sample code were lower than 22 meq/kg, which were low compared to the other salted mackerels. The major fatty acids of all salted mackerels were 16:0 (13.2-22.1%), 18:1n-9 (11.7-23.1%), and 22:6n-3 (13.5-20.4%). The Hunter color values ranged from 31.1 to 51.0 (average 37.9) for lightness, from 0.6 to 8.1 (average 3.3) for redness, from -2.9 to 9.3 (average 5.8) for yellowness, and from 46.8 to 65.8 (average 59.5) for color difference. From these results, it was concluded that the code 10 is superior than the other salted mackerels. Thus, a new salted mackerel product should be superior or similar to the food quality characteristics of this sample.

Transcriptional Upregulation of Plasminogen Activator Inhibitor-1 in Rat Primary Astrocytes by a Proteasomal Inhibitor MG132

  • Plasminogen activator inhibitor-1 (PAI-1) is a member of serine protease inhibitor family, which regulates the activity of tissue plasminogen activator (tPA). In CNS, tPA/PAI-1 activity is involved in the regulation of a variety of cellular processes such as neuronal development, synaptic plasticity and cell survival. To gain a more insights into the regulatory mechanism modulating tPA/PAI-1 activity in brain, we investigated the effects of proteasome inhibitors on tPA/PAI-1 expression and activity in rat primary astrocytes, the major cell type expressing both tPA and PAI-1. We found that submicromolar concentration of MG132, a cell permeable peptide-aldehyde inhibitor of ubiquitin proteasome pathway selectively upregulates PAI-1 expression. Upregulation of PAI-1 mRNA as well as increased PAI-1 promoter reporter activity suggested that MG132 transcriptionally increased PAI-1 expression. The induction of PAI-1 downregulated tPA activity in rat primary astrocytes. Another proteasome inhibitor lactacystin similarly increased the expression of PAI-1 in rat primary astrocytes. MG132 activated MAPK pathways as well as PI3K/Akt pathways. Inhibitors of these signaling pathways reduced MG132-mediated upregulation of PAI-1 in varying degrees and most prominent effects were observed with SB203580, a p38 MAPK pathway inhibitor. The regulation of tPA/PAI-1 activity by proteasome inhibitor in rat primary astrocytes may underlie the observed CNS effects of MG132 such as neuroprotection.

Estimating the Yield of Potato Non-Mulched Using Climatic Elements (기상자료를 이용한 무피복 재배 감자의 수량 예측)

  • We aimed to evaluate the effects of climatic elements on potato yield and create a model with climatic elements for estimating the potato yield, using the results of the regional adjustment tests of potato. We used 86 data of the yield data of a potato variety, Sumi, from 17 regions over 11 years. According to the results, the climatic elements showed significant level of correlation coefficient with marketable yield appeared to be almost every climatic elements except wind velocity, which was daily average air temperature (Tave), daily minimum air temperature (Tmin), daily maximum air temperature(Tmax), daily range of air temperature (Tm-m), precipitation (Prec.), relative humidity (R.H.), sunshine hours (S.H.) and days of rain over 0.1 mm (D.R.) depending on the periods of days after planting or before harvest. The correlations between these climatic elements and marketable yield of potato were stepwised using SAS, statistical program, and we selected a model to predict the yield of marketable potato, which was $y=7.820{\times}Tmax_-1-6.315{\times}Prec_-4+128.214{\times}DR_-8+91.762{\times}DR_-3+643.965$. The correlation coefficient between the yield derived from the model and the real yield of marketable yield was 0.588 (DF 85).

Migrant Multi-Cultural Family Women's Life Quality Related to Oral Health: Survey in Dae-Gu (다문화가족 이주여성의 구강건강관련 삶의 질: 대구지역 조사)

  • This study conducted oral examinations and individual interviews on migrant multi-cultural family women in Daegu and measured their socio-demographic characters, oral health conditions and OHIP-14 in an aim to investigate the relevance between the oral health of migrant multi-cultural family women living in some big cities and their quality of life. Based on data finally collected from 189 women, the t-test, ANOVA and binary logistic regression analysis were conducted and the conclusions are as follows: The average number of decayed teeth was 2.23, loss teeth was 1.48, and treated teeth was 5.58. Women from the Philippines had more number of loss teeth than those from other countries, and women from China relatively had a small number of filled permanent teeth. The quality of life related to oral health was found to be poor in proportion to the number of loss teeth. A comparison of life quality related to oral health depending on loss teeth showed that life quality related to oral health was lowest in the areas of mental discomfort, physical ability decrease, mental ability decrease, social ability decrease and social disadvantage. Life quality related to oral health was found to be low in proportion to the number of permanent teeth with decay experience and poor monthly household income, which shows that the number of permanent teeth with decay experience and monthly income are mostly related to life quality related to oral health. As migrant multi-cultural family women's life quality related to oral health is low in proportion to the number of loss teeth and decayed teeth, it needs to develop a program to improve their oral healthrelated life quality and conduct follow-up research to verify its effect.

Treatment Effect of Green Manure Crops on Content of γ-Oryzanols from Korean Rice Variety, Unkwangbyeo (녹비 작물 처리에 따른 운광벼 중 감마오리자놀 변화)

  • BACKGROUND: ${\gamma}$-Oryzanol, a mixture of ferulic acid esters of triterpene alcohols and sterols, are a nutritionally important group of rice secondary metabolites. The ${\gamma}$-Oryzanol content and composition were found to vary with enviromental factors such as growth temperature, varietal origin, and cultivation method. Therefore, the effect of green manure treatments will be also be an important factor in their content. METHODS AND RESULTS: The ${\gamma}$-Oryzanols extracted using dichloromethane/methanol were analyzed equipped liquid chromatography with a diode array detector. A total of ten components of ${\gamma}$-oryzanol including ${\Delta}7$-stigmastenyl ferulate were isolated of which, cycloartenyl ferulate, 24-methylenecycloartanyl ferulate, campesteryl ferulate and sitosteryl ferulate were identified as the major components in Korean rice variety, Unkwang. The ${\gamma}$-oryzanol content in rice, in the presence of nitrogen fertilization using green manure was similar to conventional nitrogen fertilization, but was higher than the control(no fertilizations). The ${\gamma}$-oryzanol ontent in rice, in the presence of hairy vetch as green manure crop was the highest compared to other crops (opium-poppy, crimson clover, cornflower). As a result of PLS-DA using SIMCA 11.0 ver. as multivariate analysis program on the basis of total data, in all samples, the specific pattern and cluster of ${\gamma}$-oryzanol scores according to green manure crops and conditions were confirmed with possible distinguishing nitrogen effects. CONCLUSION: The nitrogen contained in the green manure crops is considered to play a major role in the formation of ${\gamma}$-oryzanol. Hairy vetch which contains higher nitrogen increased the concentration of ${\gamma}$-oryzanol in rice.
