Biocontrol of Tomato Fusarium Wilt by a Novel Genotype of 2,4-Diacetylphloroglucinol-producing Pseudomonas sp. NJ134

  • The rhizobacterium NJ134, showing strong $in$ $vitro$ antifungal activity against $Fusarium$ $oxysporum$, was isolated from field grown tomato plants and identified as $Pseudomonas$ sp. based on 16S ribosomal DNA sequence and biochemical analyses. The antifungal compound purified by gas chromatography-mass spectrometry, infrared, and nuclear magnetic resonance analyses from NJ134 cultures was polyketide 2,4-diacetylphloroglucinol (DAPG). Analysis of the sequence of part of one of the genes associated with DAPG synthesis, $phlD$, indicated that the DAPG producer NJ134 was a novel genotype or variant of existing genotype termed O that have been categorized based on isolates from Europe and North America. A greenhouse study indicated that about $10^8$ CFU/g of soil NJ134 culture application was required for effective biocontrol of Fusarium wilt in tomato. These results suggest that a new variant genotype of a DAPG-producing strain of $Pseudomonas$ has the potential to control Fusarium wilt under the low disease pressure conditions.

  • To establish a cultivation method of Petalonia fascia, seeds and seedlings cultures and growth tests were performed at the Daeri aquafarm in Haeui, Shinan, Jeollanamdo, Korea. Gametes were easily released from the mature plurilocular sporangia. They developed to crustose discoidal stolons and grew to filamentous and discoidal stolons. The indoor seeding was performed by using the 100-150 ${\mu}m$long fragments of stolons on Porphyra nets and the erect thalli developed from the cuttings when the seawater temperatures were 10-15$^{\circ}C$. In the experimental cultivation in the sea, 1-2mm long plantlets were found after 15 days of cultivation; after two months thalli grew to their maximal size of 215-355 mm long blades; after three months the length of thalli began to decrease due to distal disintegration and the plant color changed to yellow and epiphytic diatoms were attached on the thalli, which deteriorated the quality of products. The cultivation of P. fascia by the regeneration of filamentous-discoidal complexes was carried out successfully for the first time in Korea.

  • Jeollanamdo aims to cultivate Hanok Village as a lodging resource for tourism industry in rural areas. For the purpose, it invited eight models of the Hanok Village publicly and selected our model as the best. This study is to introduce the concepts and characteristics of the village, centering on the model developed by our research team and awarded by the local government. We develop and suggest eleven types of improved-type Hanok model that is to be distributed to each city and county. We compose the space which reflects needs of residents for modern living. And we focus on preservation of decorative beauty to make it a resource of tourism by creating beautiful view of Hanok and reduction of financial burden to activate its distribution. The Hanok developed in this study has a significant meaning as improved Hanok that can be suggested as a new model of rural area housing project.

Induced Systemic Drought and Salt Tolerance by Pseudomonas chlororaphis O6 Root Colonization is Mediated by ABA-independent Stomatal Closure

  • Root colonization by the rhizobacterium Pseudomonas chlororaphis O6 in Arabidopsis thaliana Col-0 plants resulted in induced tolerance to drought and salinity caused by halide salt-generated ionic stress but not by osmotic stress caused by sorbitol. Stomatal apertures decreased following root colonization by P. chlororaphis O6 in both wild-type and ABA-insensitive Arabidopsis mutant plants. These results suggest that an ABA-independent stomatal closure mechanism in the guard cells of P. chlororaphis O6-colonized plants could be a key phenotype for induced systemic tolerance to drought and salt stress.

Quality Properties of Rice Cake Containing Bamboo Sprout Powder (죽순분말을 첨가한 절편(떡)의 특성)

  • Bamboo sprout powder was added to rice cake in an effort to improve the quality and functional properties of dough. The contents of crude protein and carbohydrates increased as the amount of bamboo sprout powder increased, whereas moisture contents decreased. According to increasing contents of bamboo sprout powder, total amino acid contents significantly increased. Hunter's L value decreased as contents of bamboo sprout powder increased, whereas sucrose contents increased. Addition of bamboo sprout powder to rice cake increased dietary fiber contents. Sensory scores of rice cake containing 90 g of bamboo sprout powder were the highest of all rice cakes tested. According to the results, adding bamboo sprout powder could improve the quality and sensory characteristics of rice cake.

  • This study examined the ultraviolet-A blocking effect and antioxidant activity of Gracilariopsis chorda extract for use as sunscreen agents in cosmetics. The maximum absorbance at ultra violet-A320-330 and transmissivity was 20-80%. The 1,1-diphenyl-2-picrylhydrazyl (DPPH)radical scavenging activity of Gracilariopsis chorda extract was 68?96% and increased with the sample concentration. This indicates that Gracilariopsis chorda extract is a promising natural compound that could be useful as a sunscreen.

Application of chitosan resin formulae as a sustained-releasing form adjuvant (키토산 resin formulae의 서방효과(sustained-releasing effect) 보조제로서의 활용)

  • Here, we report the suitability of using a resin-type chitosan formulae as a sustained-releasing form adjuvant in comparison with commercially well-known Freund's adjuvants. To induce the immunological responses, N-terminal region of Pasteurella multocida toxin was used as an antigen, which was found to be protective immunogen against P. multocida infection. Mice immunized with chitosan resin formulae showed statistically significant antibody induction (P<0.001) as much as that of Freund’s adjuvants. As a result, the resin-type sustained-releasing form chitosan formulae is thought to be a good candidate for a new type adjuvant.

  • A new s-wave probe, called "MudFork", has been recently developed and is used in accurate measurements of shear wave velocities of soft soils. In previous researches, some instrumental defects of the probe have been reported. Bending deformation of probe is caused by small section modulus of blades, and open-ended bender elements are vulnerable to damage during penetration. Herein, we improved the MudFork to solve these problems. Field tests at Sinan, Jeollanamdo using the improved MudFork were conducted. Results from MPT are compared with the results from CPT.
