Polyphenol-rich Sargassum horneri alleviates atopic dermatitis-like skin lesions in NC/Nga mice by suppressing Th2-mediated cytokine IL-13

  • Atopic dermatitis (AD) is one of major skin inflammatory diseases characterized by excessive Th2-mediated immune responses. Recent evidence provides that interlukin-13 (IL-13) plays the role of a key Th2 cytokine that drives the inflammation underlining AD. Due to adverse effects of commercially available synthetic drugs, the need for treatments based on natural products is gaining much attention. Sargassum horneri is an edible brown algae known for beneficial bioactivities including anti-inflammation. We investigated if polyphenol-rich S. horneri extracts (SHE) could suppress AD-like skin lesions in NC/Nga mice and if that involved inhibition of the infiltration of Th2-mediated cytokine IL-13. We observed markedly increased infiltration of IL-13 positive cells in AD-like skin lesions of mice but SHE treatments decreased it. Also, the dermal expression of IL-13 was sufficient to cause inflammatory responses in mice skin resembling human AD. SHE suppressed the dermal infiltration of inflammatory cells where IL-13 plays a crucial role in skin tissues and in the recruitment of inflammatory cells. Furthermore, it was confirmed that SHE reduced T cell, dendritic cell, and macrophage populations in spleen. Moreover, SHE decreased the collagen deposition in skin and ear dermis resulting in reduced fibrosis that occurs in AD due to excessive collagen. Taken together, our results reveal that SHE suppressed the infiltration of inflammatory cells into skin dermis by decreasing the infiltration of IL-13 positive cells. Therefore, SHE could be taken as a useful therapeutic agent to alleviate AD.

제브라피쉬에서의 제주도 천연추출물의 항스트레스 효과 (Anti-stress Effects of Natural Products from Jeju Island in Zebrafish)

  • Objective: In this study, the anti-stress effects of extract of Hydrangeae Dulcis Folium (EHDF) or ethalonic extract of Opuntiaficus-indica (EOF) of natural extracts from Jeju Island were investigated. Methods: We performed measurement of whole-body cortisol level and behavioral experiments including the novel tank test (NTT) or the open field test (OFT) to assess stress responses in zebrafish. To induce physical stress, we used the net handling stress (NHS). Fish were treated with EOF or EHDF for 6 min before they were exposed to stress. And then, we sacrificed fish for collecting body fluid from whole-body or conducted behavioural tests, including novel tank test and open field test, were evaluated to observe anxiety-like behaviours and locomotion. We used the cortisol enzyme-linked immunoassay kit to measure the amount of cortisol in each zebrafish sample. Results: The results indicate that increased anxiety-like behaviours in novel tank test and open field test under stress were prevented by treatment with both EOF and EHDF (P < 0.05). Moreover, compared with the unstressed group, which was not treated with NHS, the whole-body cortisol level was significantly increased by treatment with NHS. Compared with the NHS-treated stressed control group, pre-treatment with each EHDF and EOF for 6 min significantly prevented the NHS-increased whole-body cortisol level (P < 0.05). Conclusions: In conclusion these results suggest that both EOF and EHDF pretreatment may prevent stress responses and that its mechanism of action may be related to its positive effects on cortisol release.

수국 꽃 추출물 유래 항산화 및 항염 활성 성분 (Anti-oxidative and Anti-inflammatory Constituents from the Extracts of Hydrangea macrophylla Flowers)

  • 본 연구에서는 수국 꽃 추출물 및 용매 분획물의 항산화, 항염 활성을 확인하고 유효성분을 분리하여 화학구조를 규명하였다. 항산화 활성을 확인하기 위하여 DPPH 및 ABTS+ 라디칼 소거 활성을 측정하였으며, 항염 활성은 LPS로 유도된 RAW264.7 세포에서 nitric oxide (NO) 생성 억제 효과로 측정하였다. 실험 결과, 에틸아세테이트 분획물에서 우수한 항산화 및 항염 활성을 확인하였으며 크로마토크래피를 실시하여 3개의 유효 성분을 분리하였고 NMR 데이터 분석을 통하여 화학구조를 동정하였다; Hydrangenol (1), prunin (2), astragalin (3). 분리된 화합물 1-3에 대하여 HPLC 분석을 수행한 결과 hydrangenol이 수국 꽃 추출물의 주요 성분으로 확인되었다. 화합물 1-3의 항염 활성을 확인한 결과 세포 독성 없이 NO의 생성을 감소시키는 것으로 나타났다. 이상의 연구 결과를 바탕으로 수국 꽃 추출물은 천연 항산화 및 항염 소재로써 활용이 가능할 것으로 사료된다.

감국과 양파껍질 추출물을 이용한 홀치기 천연염색 아동 패션한복 디자인 (Design for Children's Han Bok Product with Variegated Natural Dyeing, Using Mixture Extract of Chrysanthemum indicum Linne and Onion Shell)

  • In this study, the dyeing of Chrysanthemum indicum Linne alone and the combined dyeing of Chrysanthemum indicum Linne and onion shell were first performed to compare dyeing properties. A combination extract of Chrysanthemum indicum Linne and onion shell can be expected to improve coloration. A Hanbok design for children used a natural dyeing fashion dyeing method to make the Chrysanthemum pattern. Chrysanthemum indicum Linne is a type of wild chrysanthemum with various medicinal effects for headache and stress relief, skin diseases, insomnia and depression. Widely used onion shells have medicinal properties that help prevent and treat various adult diseases. The colors of silk dyed exclusively by Chrysanthemum indicum Linne, and the colors of silk dyed by Chrysanthemum indicum Linne and onion shell extract are yellow; in addition, silk dyed by Chrysanthemum indicum Linne and onion shell extracts were almost all unchanged. Four hanboks, which are comfortable for children to wear and use, were produced using Chrysanthemum indicum Linne and onion shell extracts. If children wear hanbok fashion products with auspicious patterns, they can provide a chance to simultaneously experience the traditional symbolism of Korean patterns and culture. In addition, it is expected to develop pride in traditional dress culture.

Protective Activity against Ionizing Radiation of Antioxidative Plants Indigenous to Korea

  • We have screened the cytoprotective effect on ${\gamma}-ray$ radiation induced oxidative stress from forty one Korean plant extracts. Carpinus laxiflora (caulis), Quercus salicina (caulis), and Castanopsis cuspidata (caulis) were found to scavenge 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical and intracellular reactive oxygen species (ROS). As a result, extracts of three plants reduced cell death of Chinese hamster lung fibroblast (V79-4) cells induced by $H_2O_2$ treatment. In addition, these extracts protected cell death of V79-4 cells damaged by ${\gamma}-ray$ radiation. In addition, these extracts scavenged ROS generated by radiation. Taken together, the results suggest that Carpinus laxiflora, Quercus salicina, and Castanopsis cuspidata protect V79-4 cells against oxidative damage by radiation through scavenging ROS.

Whitening and Anti-oxidative Activities of Chemical Components Extracted from Branches of Sorbus alnifolia

  • In this study were evaluated the whitening and anti-oxidative activities from the extracts of Sorbus alnifolia branches, and identified the chemical structures of the active ingredients. In the whitening tests using α-MSH stimulated B16F10 melanoma cells, the 70% ethanol extract and n-butanol (n-BuOH) fractions concentration-dependently inhibited cellular melanogenesis and intracellular tyrosinase activities without causing cell toxicity. The total polyphenol content of n-BuOH and ethyl acetate (EtOAc) fractions were measured to be respectively 241.1 ± 1.1 and 222.9 ± 2.4 (mg/g GAE), and the total flavonoid content of EtOAc fraction was 75.3 ± 2.0 (mg/g QE). Upon anti-oxidant studies with DPPH and ABTS+ radicals, potent radical scavenging activities were observed in the EtOAc and n-BuOH fractions. Moreover, in the study of cell protection efficacy using HaCaT keratinocytes damaged by H2O2, the EtOAc and n-BuOH fractions showed a very positive results on prevention of oxidative stress. Phytochemical studies for this extract resulted in the isolation of four compounds; 2-oxopomolic acid (1), euscaphic acid (2), epi-catechin (3), prunasin (4). These results suggested that the extract of S. alnifolia branches containing compounds 1-4 as natural ingredients could be used as whitening and anti-oxidant ingredients in cosmetic formulations.

제주도 자생 노루참나물 (Pimpinella komarovii) 추출물의 항산화 효과 및 멜라닌 억제 효과 (Antioxidant Effects and Melanin Inhibitory Effect of Natural Pimpinella komarovii Extracts in Jeju Island)

  • 본 연구는 제주도의 자생식물인 노루참나물 (Pimpinella komarovii)의 산업적 활용 가능성을 평가하기 위하여 에탄올 추출물 및 순차적 용매분획물을 얻어 항산화 활성 및 항염증 효과를 검색하고자 하였으며, 마우스 유래의 melan-a 세포에 처리하여 멜라닌 생합성 및 tyrosinas 억제활성을 통하여 유용자원으로서 미백 및 기능성 화장료로 활용가능성이 있는지 알아보고자 연구를 시행하였다. 노루참나물 에탄올 추출물은 높은 항산화 활성 (DPPH 소거활성 $IC_{50}$ 값, $231.8{\mu}g/m{\ell}$ ; superoxide 소거 활성 및 xanthine oxidase 억제 활성 $IC_{50}$ 값, 각각 $23.6{\mu}g/m{\ell},\;587.8{\mu}g/m{\ell}$)을 나타내었으며, 순차적 분획물 중에서는 에틸아세테이트 분획물이 가장 높은 항산화 활성을 나타내었다. 그리고 멜라닌 생성 세포인 melan-a 세포에서 노루참나물 에탄올 추출물과 에틸아세테이트 분획물이 tyrosinase와 멜라닌 합성을 농도 의존적으로 억제하였다. 또한 LPS로 자극한 RAW 264.7 세포에서 노루참나물의 헥산 분획물과 에틸아세테이트 분획물이 가장 높은 NO생성 억제 활성을 보였다. 본 연구결과는 노루참나물 추출물이 항산화 및 항염증 활성을 가지는 미백 관련 기능성 소재로서의 활용가치가 있음을 보여준다.

식물 40종 고압용매 추출물의 통합적 항산화 능력 및 항균 활성 (Integral Antioxidative Capacity and Antimicrobial Activity of Pressurized Liquid Extracts from 40 Selected Plant Species)

  • 제주, 전남 고흥, 경북 울릉도에 자생하는 식물자원 40종을 대상으로 고압용매 추출하여 항산화 활성과 항균활성을 측정하여, 식품산업에 응용할 천연소재를 탐색하였다. 총 페놀성 화합물 함량은 짚신나물이 174.4 mg GAE/g로 자생식물 중 가장 높았고, 다음으로 물참나무, 서어나무, 붉나무가 각각 116.9, 113.3, 108.2 mg GAE/g로 높은 함량을 나타내었다. 총고형분 함량에 대한 총 페놀 함량의 비율은 짚신나물이 72.6%로 가장 높았고, 서어나무(47.3%), 물참나무(46.4%), 자금우(40.2%), 바늘꽃(40.1%) 순으로 높은 비율을 나타내었다. 수용성 항산화 능력은 마가목이 5.81 mmol ascorbic acid equivalents/g로 가장 높았고, 다음으로 나도송이풀, 붉나무, 물참나무, 서어나무가 각각 3.96, 3.63, 3.63, 3.34 mmol ascorbic acid equivalents/g를 나타내었다. 지용성 항산화 능력은 서어나무가 8.51 mmol Trolox equivalents/g로 가장 높았고, 다음으로 마가목, 세잎이질풀, 물참나무, 바늘꽃, 섬기린초가 각각 6.57, 5.68, 3.85, 3.83, 3.69 mmol Trolox equivalents/g를 나타내었다. 국내 자생식물 40종 추출물에 대하여 4종의 양식 넙치 질병 세균에 대한 항균활성을 측정한 결과 9종 즉, 바늘꽃, 독활, 짚신나물, 물참나무, 나도송이풀, 세잎이질풀, 서어나무, 섬기린초, 섬바디 만이 항균활성을 나타내었다. 짚신나물은 S. iniae에 대하여 높은 항균활성을 나타내었으며, 독활, 나도송이풀, 섬바디는 S. parauberis과 S. iniae에 대하여, 물참나무, 세잎이질풀, 서어나무는 V. ordalii에 대하여 높은 항균활성을 나타내었다.

열수 및 효소적 가수분해로 제조된 틸라피아 비늘 젤라틴 가수분해물의 ACE 저해 활성 (Angiotensin I Converting Enzyme Inhibitory Effects of Gelatin Hydrolysates Prepared from Tilapia mossambica Scales by Hot Water and Enzymatic Extraction)

  • Fish scales have potential in functional food preparation due to their antioxidant and antihypertensive properties. We investigated the angiotensin I converting enzyme (ACE) inhibitory activity of Tilapia mossambica scale extracts. Hydrolysates of tilapia scales were prepared by enzymatic extraction using five proteases (${\alpha}$-chymotrypsin, Alcalase, Kojizyme, Protamex and trypsin) after scales were treated with hot water for 3 hr. Scale enzymatic hydrolysates prepared using both hot water and enzyme treatments exhibited elevated hydrolysis (about 25%-55%) compared to only enzyme treatment (about 15%-45%). Enzymatic hydrolysates (1 mg/mL) prepared by both hot water and enzyme treatments also showed significantly increased ACE inhibitory activities from about 20%-75%. The pattern of ACE inhibitory activities was similar to the degree of hydrolysis. Alcalase and ${\alpha}$-chymotrypsin hydrolysates displayed the highest ACE inhibitory activities ($IC_{50}$ = 0.83 mg/mL and 0.68 mg/mL, respectively). In addition, the ACE inhibitory effects of $IC_{50}$-chymotrypsin hydrolysates increased with decreasing molecular weight (5 kDa>, 10 kDa> and 30 kDa>), with the 5 kDa> fraction displaying the highest ACE inhibitory activity (about 89.9% and $IC_{50}$ = 0.1 mg/mL). We suggest that the peptide compounds of enzymatic hydrolysates prepared from tilapia scale enhances ACE inhibitory activity and might be useful as an antihypertensive material.

제주 자생식물 고압용매 추출물의 통합적 항산화 능력 (Integral Antioxidative Capacity of Extracts by Pressurized Organic Solvent from Natural Plants in Jeju)

  • 제주 자생식물 20종을 대상으로 고압용매 추출(추출용매 100% methanol, 추출 온도 $40^{\circ}C$, 추출 압력 13.6 MPa, 추출 시간 10분)하여 총페놀 함량과 통합적 항산화 능력을 측정하고 폴리페놀 성분을 동정하였다. 추출수율은 붉나무, 말오줌때, 사방오리나무, 사람주나무, 팥배나무가 각각 21.8, 21.5, 21.1, 20.7, 20.1%로 가장 높았다. 총페놀 함량은 아그배가 68.3 mg GAE/g로 가장 높았고, 다음으로 사람주나무, 석위, 말오줌때가 각각 57.6, 56.6, 55.1 mg GAE/g을 나타내었다. 수용성 항산화 능력은 이질풀, 사람주나무, 산딸나무, 붉나무가 각각 598, 394, 293, $270\;{\mu}mol$ ascorbic acid equivalent/g로 높았고, 지용성 항산화 능력은 백량금, 새우나무, 이질풀, 붉가시나무가 611, 314, 296, $242\;{\mu}mol$ trolox equivalent/g로 높았다. GC/MS에 의한 폴리페놀성분을 동정한 결과 15개의 주요 피크를 얻었으며, 그 중 2종의 폴리페놀류(gallic acid(체류시간 19.7분)와 quercetin(체류시간 33.5분)), ascorbic acid(체류시간 35.3분) 그리고 다수의 지방산류(체류시간 18.6, 21.0, 21.8, 21.9, 23.6분)를 확인할 수 있었는데, 이 중 gallic acid는 다른 성분보다 peak area가 높은 것으로 나타나 사람주나무의 가장 중요한 폴리페놀 성분으로 추정되었다.
