Radiation damage analysis in SiC microstructure by transmission electron microscopy

  • Microstructures of monolithic high purity SiC and SiC with sintering additives after neutron irradiation to a fluence of 2.0-2.5 × 1024 n/m2 (E > 0.1 MeV) at 333-363 K and after post-irradiation annealing up to 1673 K were observed using a transmission electron microscopy. Results showed that no black spot defects or dislocation loops in SiC grains were found after the neutron irradiation for all of the specimens owing to the moderate fluence at low irradiation temperature. Thus, it is confirmed that these specimens were swelled mostly by the formation of point defects. Black spots and small dislocation loops were discovered only after the annealing process in PureBeta-SiC and CVD-SiC, where the swelling almost diminished. Anomalous-shaped YAG grains were found in SiC ceramics containing sintering additives. These grains contained dense black spots defects and might lose crystallinity after the neutron irradiation, while these defects may annihilate by recrystallization during annealing up to 1673 K. Amorphous grain boundary phase was also presented in this ceramic, and a large part of it was crystallized through post-irradiation annealing and could affect their recovery behavior.

  • Antioxidative activity of daidzin and puerarin isolated from Puerariae radix against oxidation of low density lipoprotein(LDL) was investigated. The concentration of daidzin at 100$\mu\textrm{g}$/$m\ell$ and puerarin at 60$\mu\textrm{g}$/$m\ell$ inhibited Cu$^{2+}$-mediated oxidation of LDL almost completely. The electrophoretic mobility of oxidized LDL by addition of daidzin(100$\mu\textrm{g}$/$m\ell$) and puerarin(60$\mu\textrm{g}$/$m\ell$) was faster than that of native LDL, but slower than that of oxidized LDL. The oxidized LDL induced by J774 or macrophage was inhibited strongly in the presence of 100$\mu\textrm{g}$/$m\ell$ daidzin and 60$\mu\textrm{g}$/$m\ell$ Puerarin. The formation of conjugated dienes in the oxidized LDL was strongly inhibited by 100$\mu\textrm{g}$/$m\ell$ daidzin and 60$\mu\textrm{g}$/$m\ell$ puerarin.n.

Effects of Feeding Induced Molting on the Performance, Egg Quality, and Visceral Organs in Laying Hens (산란계에서 급이환우가 산란계의 생산성과 계란의 품질 및 주요 장기에 미치는 영향)

  • This study was conducted to compare the effect of feeding molting and fasting molting on the performance, egg quality, and visceral organs in laying hens for animal welfare. Eighty one 62-wk-old White Leghorn hens that egg production was over 80% and average weight was $1.6{\pm}0.3\;kg$ were used in this study. Treatments were control (non-molt treatment), feeding molt treatment, and fasting molt treatment. The three treatments were administered to three replicate group of nine hens wherein each group. All treatment groups were fed the basal diet (CP 15%, ME 2,700 kal/kg) for two weeks as the adaptation period. Induced molt diets contains low CP (6.7%) and low energy (2,200 kal/kg). Test periods were 14 days for feeding molting and 10 days for fasting molting. Egg production decreased to be 0% at 10 days of feeding molting treatment, but at 2 or 3 days of fasting molting treatment. Egg production restarted after 19 days ending molt at feeding molting treatment, while after 24 days at fasting molting treatment. On the egg quality was improved at molting treatments (p<0.05) except egg yolk. Egg shell tissue was crowded at molting treatment to compare to control. Liver weights, heart weight, and oviduct weight of laying hens decreased at molting treatments (p<0.05). Finally, feeding molting might could be replaced fasting molting on the welfare and further studies were needed about molting program.

  • In five varietes of Citrus junos sieb grown in different area in Korea, the general components, total acids and inorganic components were calculated and compared with one other. In the orchard soils, the physical and chemical properties were studied. The results are summarized as follow: 1. Generally, the rind was weighter than tile flesh compared with the other fruits and the proportion of the rind was 49%. 2. Total contents of organic acids in rind and flesh were 12.29me and 39.79me respectively. 3. The order of the contents of inorganic components in the flesh and rind was $CaO>K_2O>MgO>P_2O_5>SO_4$ 4. The contents of crude protein and crude fat were increased with those of MgO and $SO_4$ in fruits. 5. The quantity of protein in the soil seed to influence greatly in rind formation not only in citrus fruits but also Citrus junos sieb. 6. In growing Citrus junos sieb, C.L. seemed to he the most adequate soil. 7. The pH range of the soil was from 5.05 to 7.20 generally. 8. The contents of crude protein and crude fat in citrus fruits seemed to be increased with those of organic material and total-nitrogen in the soil.

Effect of Dietary Fatty Acid and Vitamin E Supplementation in Antioxidant Systmes of the Second Generation Rat Brain Sections (식이지방산 조성 및 비타민 E의 보충이 제 2 세대 흰 쥐 뇌조직의 항산화 체계에 미치는 영향)

  • In this study, we examined the effects of dietary fatty acids and vitamin E supplementation on antioxidant systems in the rat brain regions. The Sprague Dawley rats were fed the experimental diets 3-4 wks prior to the conception. Experimental diet consisted of 10% fat(wt/wt) which were safflower oil(SO, poor in $\omega$3 fatty acids), mixed oil(MO, P/M/S ratio=1.03:1.45:1,$\omega$6/$\omega$3 ratio=6.3) and mixed oil supplemented with vitamin E(ME:MO+500mg vitamin E/kg diet). At 3 and 9 weeks of age of the newborn rats, frontal cortex(FC), corpus striatum(CS), hippocampus(H) cerebellum(CB) were dissected out from the whole brain. Activities of glutathione peroxidase(GSH-P(sub)x, superoxide dismutase(SOD) concentrations of malondialdehyde(MDA) were mesaured. Dietary fatty acids were not effective in antioxidative system for rat brain. However, when vitamin E was supplemented to the diet(ME), the activities of GSH-P(suh)x tended to increase in comparison to MO group. Therefore, the activites of GSH-P(suh)x of FC and H at the age of 3 weeks showed significant differences(p<0.05). The activities of Total-SOD tended to decrease in ME group compared to MO group. There were significant differences(p<0.05) in FC and CS at the age of 3 weeks. The activities of Mn-SOD tended to increase and Cu, Zn-SOD tended to decrease when vitamin E was supplemented. The activity levels of antioxidative enzymes at the age of 3 weeks and 9 weeks were similar. This suggested that the activity level of antioxidative enzymes reached to the adult level at the age of 3 weeks which is the end point of lactation period. The concentrations of MDA were not altered by experimental diets. When the activities of antioxidant enzymes were compared, the activities of antioxidant enzymes were the lowest in H and FC. In conclusion, the antioxidative system were not altered by dietary fatty acid at the age of 3 weeks and 9 weeks, but the supplementation of vitamin E altered the antioxidative systems. Therefore, these findings should be considered comprehensively in scope of the balance of various antioxidative systems and their interactions(Korean J Nutrition 34(1):14-22, 2001)

The Influence of Different Fiber and Starch Types on Nutrient Balance and Energy Metabolism in Growing Pigs

  • A repeated $4{\times}4$ Latin square design was conducted with eight ileal cannulated castrates to examine the effect of source of starch and fiber on nutrient balance and energy metabolism. Pigs were fed on one of the four experimental diets: Control diet (C) mainly based on cooked rice; and diets P, S and W with the inclusion of either raw potato starch, sugar beet pulp or wheat bran supplementation, respectively. With the exception of an increased (p<0.05) energy loss from methane production with diet S observed, no significant differences (p>0.05) in the ratio of metabolizable energy (ME)/digestible energy, the utilization of ME for fat deposition and for protein deposition, energy loss as hydrogen and urinary energy were found between diets. The efficiency of utilization of ME for maintenance was lower (p<0.05) with diets P and S than with diet C. The inclusion of fiber sources (sugar beet pulp or wheat bran) or potato starch reduced the maintenance energy requirement. The fecal energy excretion was increased (p<0.05) with either sugar beet pulp or wheat bran supplementation, while it was unaffected (p>0.05) by addition of potato starch. In comparison with diets C and P, a lowered ileal or fecal digestibility of energy with diets S and W was observed (p<0.05). Feeding sugar beet pulp caused increased (p<0.05) daily production of methane and carbon dioxide and consequently increased energy losses from methane and carbon dioxide production, while it did not influence the daily hydrogen production (p>0.05). An increased (p<0.05) proportion of NSP excreted in feces was seen by the supplementation of wheat bran. Higher NSP intake caused an increased daily amount of NSP in the ileum, but the ileal NSP proportion as a percentage of NSP intake was unaffected by diets. Feeding potato starch resulted in increased daily amount of starch measured in the ileum and the proportion of ileal starch as a percentage of starch intake, while no significant influence on fecal starch was found. Higher (p<0.05) daily amount of fecal starch and the proportion of fecal starch as a percentage of starch intake were found with fiber sources supplementation compared with diets C and P. By increasing the dietary NSP content the fecal amount of starch increased (p<0.01).

IN VITRO DEVELOPMENT OF BOVINE ONE-CELL EMBRYOS FERTILIZED IN VITRO IN SERUM- AND FEEDER CELL-FREE CULTURE SYSTEMS

  • The purpose of this study was to evaluate some factors in the bovine embryonic development from one-cell to blastocyst using modified synthetic oviduct fluid medium (mSOFM), after maturation and in vitro fertilization of the oocytes. The embryonic development to the blastocyst stage was assessed at 7-10 days after in vitro fertilization, and the total cells in the blastocysts were counted by staining nuclei with fluorochrome. Some commercial calf sera (CS) and a superovulated cow serum had different effects on the embryonic development to the blastocyst stage (8.6-21.4%), dependent upon their product lots, although the development might not be affected at least by serum progesterone levels. ${\beta}$-Mercaptoethanol (${\beta}$-ME) supplemented into mSOFM was effective to the embryonic development (27.8%), as well as the co-culture system with cumulus cells (19.5%). In a serum- and feeder cell-free culture using mSOFM containing several growth factors and ${\beta}$-ME instead of CS plus co-cultured cumulus cells, bovine serum albumin (BSA, fraction V), but not polyvinyl alcohol (PVA), was highly effective in embryonic development to the blastocyst stage, almost comparable to CS in the serum-contained culture (CS, BSA and PVA; 27.8, 19.5 and 5.7%, respectively). However, fatty acid free BSA rather reduced the number of developed blastocysts, compared with fraction V BSA (7.3 vs 29.4%). In the serum- and feeder cell-free culture, supplement of glucose to the medium (final 2.0 mM) stimulated the cell proliferation of developing embryos 120 hr after in vitro fertilization. These results indicated that a serum-free medium supplemented with ${\beta}$-ME could successfully support the development of bovine one-cell embryos to the blastocyst stage. Moreover, supplement of glucose and fatty acids to the medium might support preferably the development and cell proliferation of embryos.

Effects of Enzyme Treated Palm Kernel Expeller on Metabolizable Energy, Growth Performance, Villus Height and Digesta Viscosity in Broiler Chickens

  • This study examined whether pre-treating palm kernel expeller (PKE) with exogenous enzyme would degrade its fiber content; thus improving its metabolizable energy (ME), growth performance, villus height and digesta viscosity in broiler chickens fed diets containing PKE. Our results showed that enzyme treatment decreased (p<0.05) hemicellulose and cellulose contents of PKE by 26.26 and 32.62%, respectively; and improved true ME (TME) and its nitrogen corrected value ($TME_n$) by 38% and 33%, respectively, compared to the raw sample. Average daily gain (ADG), feed intake and feed conversion ratio (FCR) of chickens fed on different dietary treatments in the grower period were not significantly different. Although there was no difference in feed intake (p>0.05) among treatment groups in the finisher period, ADG of chickens in the control (PKE-free diet) was higher (p<0.05) than in all treatment groups fed either 20 or 30% PKE, irrespective of with or without enzyme treatment. However, ADG of birds fed with 20% PKE was higher than those fed with 30% PKE. The FCR of chickens in the control was the lowest (2.20) but not significantly different from those fed 20% PKE diets while birds in the 30% PKE diets recorded higher (p>0.05) FCR. The intestinal villus height and crypt depth (duodenum, jejunum and ileum) were not different (p>0.05) among treatments except for duodenal crypt depth. The villus height and crypt depth of birds in enzyme treated PKE diets were higher (p<0.05) than those in the raw PKE groups. Viscosity of the intestinal digesta was not different (p>0.05) among treatments. Results of this study suggest that exogenous enzyme is effective in hydrolyzing the fiber (hemicellulose and cellulose) component and improved the ME values of PKE, however, the above positive effects were not reflected in the growth performance in broiler chickens fed the enzyme treated PKE compared to those received raw PKE. The results suggest that PKE can be included up to 5% in the grower diet and 20% in the finisher diet without any significant negative effect on FCR in broiler chickens.

POTASSIUM REQUIREMENT OF MULE DUCKLINGS

  • Experiments were conducted to determine potassium (K) requirement of mule ducklings. One-day-old ducklings with equal number of both sexes were fed diets containing graded levels of K for three weeks. In experiment 1, corn-soybean meal diet (CP 18.7%, ME 2,890 kcal/kg, K 0.80%) was used. The addition of K (0.00, 0.10, 0.20, 0.30, 0.40%) to the diet by potassium carbonate resulted in a decrease in weight gain and feed efficiency with the group fed on basal diet having the best performance. This means that the existing K in the basal diet is adequate for the growth of ducklings. In experiment 2, low K corn-isolated soy protein diet (K, 0.19%) was used. The supplementation of K (0.00, 0.16, 0.32, 0.48, 0.64%) resulted in an increase in weight gain and feed efficiency with the control group having the lowest performance. The minimum K requirement was found to be 0.49% for both maximum growth and best feed efficiency as determined by using bent-stick model.

BIOCHEMICAL POLYMORPHISM STUDIES IN BREEDS OF WOOL-SHEEP, HAIR-SHEEP AND THEIR HYBRIDS IN MALAYSIA

  • A biochemical genetic study on blood enzyme/protein systems in some breeds/crosses of sheep in Malaysia was carried out using horizontal starch gel electrophoresis. Blood samples were collected from 435 sheep, representing 8 breeds/crosses. These included 5 wool sheep breeds (Thai Longtail, wiltshire, Suffolk, Dorsimal and cMBLx), 1 hair sheep breed (Barbados Blackbelly) and 2 hybrids between wool sheep and hair sheep (Cameroon ${\times}$ Thai Longtail and Bali Bali ${\times}$ Malin). Twenty loci systems were examined. Of these, ten ($HB{\beta}$, ALB, TF, XP, CAT, DIA1, EsA, GPI, ME and NP) exhibited genetic variation whereas the other ten (AAT, CA, DIA2, ${\alpha}GLO$, ${\alpha}GLU$, LDH, MDH, PEP[leu-gly-gly], 6PGD and SOD) were monomorphic. The allelic frequencies which were obtained in 10 polymorphic markers are assessed and compared with the results obtained by previous workers. The estimations of inbreeding coefficient, intrabreed variation and breed relationships have been critically discussed and are used to reveal some important recommendations.
